Proliferation of multiple sclerosis cerebrospinal fluid lymphocytes after stimulation with measles virus antigens.

A method for measuring the proliferative response of cerebrospinal fluid (CSF) lymphocytes upon stimulation and mitogens and viral antigens has been developed. The number of lymphoid cells in DNA synthesis after in vitro stimulation is counted and the results expressed as a percentage of the total number of surviving lymphocytes. Preliminary results on four MS patients indicated that their CSF lymphocytes responded considerably more strongly to purified measles virus antigen than did corresponding peripheral blood lymphocytes (PBL). The responses to crude herpes virus antigen were at least as strong in PBL as in CSF lymphocyte cultures from the same patients. The CSF lymphocytes responded slightly stronger to phytohemagglutinin than PBL did.

Münchausen syndrome: a review and two case reports.

In the light of the pertinent literature, 2 illustrative cases of the Münchausen syndrome are presented. In both patients there had been dynamically significant losses in the oedipal age leading to an intense feeling of being rejected. Both patients also exhibited problems of dependence, perfectionistic attitudes toward the self, and fear of social failure. In addition, characteristics typical of alexithymia could be noted. As a dynamical interpretation it is proposed that in these patients basic helplessness and inadequacy of expressing feelings have been channelled into multiform psychosomatic symptoms. The latter in turn have caused continuous seeking for acceptable somatic etiology and treatment of the symptoms. Thus, gradually, the clinical picture of the Münchausen syndrome has developed.

A rapid and simple gas-liquid chromatographic determination of valproic acid (alpha-propyl-valeric acid) in serum.

A simple and rapid gas-liquid chromatographic method for the determination of valproic acid in serum is described. Valproic acid is extracted from acidified serum into chloroform and analyzed by gas-liquid chromatography using an FFAP column. The sensitivity of the method is 5 mumol/l and the within-run and day-to-day precisions (CV) are 4.5% or better. Analytical recovery is estimated to exceed 98% and no interference from other drugs or constituents of serum has been observed. The method was used to study the half-life of valproic acid in five healthy persons receiving a single oral dose of the drug, and the technique is well adapted to the routine monitoring of epileptic patients. In routine analysis, the determination of 50 serum samples may be performed by one technician during a normal working day.

Measles and rubella virus antibodies in patients with multiple sclerosis. A longitudinal study of serum and CSF specimens by radioimmunoassay.

A longitudinal study on rubella, measles, and respiratory syncytial virus antibodies in serial serum and CSF specimens from 20 multiple sclerosis (MS) patients was performed, using solid-phase radioimmunoassay. Albumin and immunoglobulin G (IgG) levels were also measured to check the integrity of the blood-brain barrier and the intrathecal IgG production. All the patients had local IgG production in their CNA. A local antibody production against one or more of the viruses studied was evident in 15 patients. Fluctuations in the intrathecal viral antibody synthesis were evident in eight patients. No correlation was found between these changes and the clinical course of the disease. The results suggest that the intrathecal antibody synthesis in MS is only partially against any given virus, and in most patients the bulk of the oligoclonal CSF antibodies is against antigens other than those studied here.

HLA antigens and antibody responses to measles and rubella viruses in multiple sclerosis.

Twenty-four HLA-antigens coded by loci A and B were determined serologically for 60 multiple sclerosis patients from Northern Finland. Forty-five of the patients were also typed by mixed leucocyte culture reactivity for HLA-Dw2. The only statistically significant difference in antigen frequencies in these patients compared with those in controls was the decrease of HLA-A2 among patients. Measles and rubella antibody titres in patients with and without HLA-Dw2 and HLA-B7 antigens were compared using a sensitive radioimmunoassay method. There was no difference between age- and sex-matched patient groups.

Arachnoid cyst of the intraorbital portion of the optic nerve with unilateral disc oedema and transient shallowing of the anterior chamber. A case report.

An unusual occurrence of chronic monocular disc oedema, visual loss and shallowing of the anterior chamber in a patient with an arachnoid cyst involving a portion of the intraorbital optic nerve was reported. Decompression of the optic nerve sheath through a Krönlein approach was followed by prompt deepening of the anterior chamber and a gradual, delayed relief of the disc oedema. It is concluded that orbitotomy and decompression of the optic nerve sheath should be done before atrophic changes of the optic nerve and visual loss begin to develop.

Characterisation of volatile organic compounds in stemwood using solid-phase microextraction.

Solid-phase microextraction (SPME), hydrodistillation and dynamic headspace combined with GC and GC-MS were applied and compared for the analysis of volatile organic compounds (VOCs) from coniferous wood. The SPME conditions (type of fibre, size of wood sample, temperature and exposure time) were optimised, and more than 100 VOCs and semi-volatile compounds extracted and identified from the sapwood and heartwood of Norway spruce (Picea abies). The total number of mono- and sesquiterpenes eluted and identified was similar for the SPME and hydrodistillation methods, but more semi-volatile compounds were released by hydrodistillation. By applying dynamic headspace at room temperature, it was possible to analyse only the most volatile compounds. The qualitative composition of VOCs was similar in spruce sapwood and heartwood, although Z-beta-ocimene occurred only in sapwood while fenchol was present only in heartwood. SPME sampling coupled with GC, applied here to the analysis of VOCs released from stemwood of firs for the first time, is a convenient, sensitive, fast, solvent-free and simple method for the determination of wood volatiles. The technique requires much smaller sample amounts compared with hydrodistillation, and the total amount of VOCs extracted and identified is higher than that obtained by hydrodistillation or dynamic headspace. The relative ratios of the main mono- and sesquiterpenes and -terpenoids were similar using the SPME-GC and hydrodistillation methods.
