Porcine and ovine thyroglobulins were purified from fresh glands by slice extraciton, precipitation with buffered ammonium sulfate and Sepharose 6B gel filtration using 0.1 M sodium phosphate buffer pH 7.2 at all steps. In contrast to thyroglobulin obtained from frozen glands, the proteins purified from unfrozen glands only showed the 19 S and 12 S species by electrophoresis in sodium dodecylsulfate polyacrylamide gels. After full reduction and S-alkylation, no qualitative changes were observed in e gel electrophoresis patterns as compared to the unmodified proteins. Species of apparent molecular weight corresponding to the native 12 S subunit was the major component strongly suggesting a mol. wt. of about 330 000 for the elementary peptide chains of pig and sheep thyroglobulin. This was confirmed by sedimentation equilibrium analyses in 6 M guanidinium. HCl, 0.1 M sodium phosphate pH 7.2 OF FULLY REDUCED AND S-alkylated thyroglobulins which showed a weight average molecular weight of 310 000 daltons.
The C-terminal amidation of calcitonin represents an important technological problem. A method using a serine carboxypeptidase-catalyzed transpeptidation reaction in combination with photochemical cleavage to give the warranted peptide amide is described. The overall yield is higher than 95%.
Structure-activity relationship on cholecystokinin is presented. C-terminal heptapeptide analogues of cholecystokinin exhibiting selective agonist or antagonist activities were synthesized and their biological and pharmacological properties studied. We showed that: 1) Suppression of the C-terminal phenylalanine residue leads to peripheral as well as central cholecystokinin receptor antagonists; 2) Suppression of the C-terminal amide function produces "partial agonists" exhibiting interesting biological and pharmacological activities; 3) Replacement of L-tryptophan residue by D-tryptophan in such "partial agonist analogues" resulted in potent CCK receptor antagonists; 4) The peptide bond between methionine28 and glycine29, as well as the glycine residue are quite significant for the central biological activity; 5) It is possible to obtain highly potent and selective CCK analogues for the central receptor (CCK-B) by cyclization including the C-terminal tetrapeptide. Synthesis and pharmacological studies with these analogues have allowed to precise the significance of some amino acid residues as well as of some peptide bonds. They are significant pharmacological tools for the study of CCK-A (peripheral) and CCK-B (central) receptors, their biological actions and their associated intracellular messengers.
A case of meconium peritonitis associated with materno-fetal infection by human B19 parvovirus is reported. The meconium peritonitis was recognized by ultrasonography at 25 weeks of pregnancy and was confirmed after delivery. Postnatal "opaque index" of the small bowel revealed multiple stenosis. Evolution was favorable and did not require surgery. Diagnosis of B19 parvovirus materno-foetal infection was documented on specific IgM antibodies and viral DNA in the maternal serum and specific IgM antibodies in the infant's serum. Thus, B19 parvovirus appears to be responsible for non lethal fetopathies.
During a period of five years, fifteen thousand platelet counts has been systematically realised at the Hôtel-Dieu Hospital in Marseilles. A thrombocytosis superior to 400 000/mm3 has been observed in 1,2% of cases, in such various etiological cases as mainly cancer (30%), infectious diseases (28%) and post-hemorragic anaemias (11%).
We report our experience with vertically transmitted HIV infection in infants. Fifty-five children were prospectively followed for a period of 3 years. The neonatal diagnosis of HIV infection is not always possible and doubts about contamination may create familial anxiety which requires support by a multidisciplinary structure. Long-term prognosis is difficult to predict and it depends on the severity of the immune deficiency, the early onset of neurologic disease and the frequency of opportunistic infections.

Cerebral arterial occlusion, occurring during monozygotic twin pregnancies, is a well known entity, specially when one twin is stillborn or macerated. Most of the authors consider that the aetiology is either embolization from the dead fetus or intravascular coagulation causing embolization in the surviving co-twin via placental vascular connections. Some data suggest an infectious endarteritis on the basis of angiographic and histological findings. Clinical features may appear several weeks after birth. CT scan and pathological findings depend on the date of the arterial occlusion. Four recent cases are described, two of them concerning monozygotic twin pregnancies. Pathological, CI scan, clinical features and pathogenesis are discussed taking account of the others data in the literature.
