Comparison of antibody titers against borrelial strains isolated in Japan by the microcapsule agglutination test for serological studies of early Lyme disease.

Results of a passive microcapsule agglutination test using 2 Japanese strains, Borrelia garinii and Borrelia afzelii, were comparable to that of the passive microcapsule agglutination test using a Swiss strain Borrelia burgdorferi IRS in detecting antibodies of an early stage of Lyme disease patients in Japan. MCAT with microcapsule sensitized strain IRS seems likely to become one of the important tools for the early diagnosis of Lyme disease in the world. The highest titer of MCAT was detected in the serum samples of patients which were taken around 2 weeks after erythema chronicum migrants. The test is very simple and can be employed easily in diagnostic laboratories.

[Analysis of lymphocyte subsets in human lingual tonsils].

Lymphocytes in the lingual tonsil were analyzed using monoclonal antibodies to cell surface markers. Immunohistologic staining of lingual tonsillar tissue showed localization of the various lymphocyte subsets in the germinal center, mantle zone, and inter-follicular area. Populations of lymphocyte subsets, i.e., T cells, B cells, CD4 positive cells, CD8 positive cells, alpha beta T-cells, gamma delta T-cells, and NK cells were assessed by laser flow cytometry analysis, and we compared the following tissues: peripheral blood, lingual tonsils, palatine tonsils, and pharyngeal tonsils (adenoids).

Effect of recombinant human granulocyte colony-stimulating factor on efficacy of radiation therapy in tumor-bearing rats.

The effect of recombinant human granulocyte colony-stimulating factor (rhG-CSF) on radiation-induced neutropenia and on growth of transplanted tumors treated by irradiation was investigated using tumor-bearing rats as a model for radiation therapy. In a preliminary study using normal rats, neutropenia induced by upper hemi-body irradiation at 3 Gy/day 5 times a week for 3 weeks was prevented by consecutive subcutaneous injections of rhG-CSF at 100 micrograms/kg/day. Rats bearing Walker-256, a mammary tumor, were scheduled to receive upper hemibody irradiation at 3 Gy/day for 15 times in 3 weeks if white blood cell (WBC) counts were maintained above 3,000/microliters. In control tumor-bearing rats not receiving rhG-CSF, irradiation was often withheld because of the decrease in WBC counts below 3,000/microliters. In contrast, a decrease in WBC counts below 3,000/microliters was rarely found in tumor-bearing rats injected daily with rhG-CSF. The average number of radiation treatments in control rats and rats treated with rhG-CSF was about 8 and 14, respectively, out of the scheduled 15 treatments in 3 weeks. Treatment with rhG-CSF made it possible to complete the radiation therapy regimen and thus inhibit the growth of the transplanted tumor more effectively. These results suggest that rhG-CSF may be useful to ensure radiation therapy on schedule in cancer patients.

Relationship between Helicobacter pylori and atrophic gastritis.

OBJECTIVES: To investigate the relationship between Helicobacter pylori and atrophic gastritis. DESIGN AND METHODS: The extent of atrophic gastritis was assessed endoscopically in 97 non-ulcer patients, and their H. pylori status was assessed by enzyme-linked immunosorbent assay. RESULTS: The prevalence of H. pylori infection was greatest in the early stages of atrophic gastritis. Small numbers of H. pylori-negative patients were identified at different stages during the extension of atrophic gastritis. CONCLUSIONS: H. pylori infection precedes the extension of atrophic gastritis, and a minority of patients with atrophic gastritis may never have been infected with H. pylori.

[Bronchogenic cyst of the diaphragm: a case report].

A 30-year-old man was treated surgically for bronchogenic cyst originating from the diaphragm. An abnormal shadow was found on a routine chest roentgenogram. The tumor was located in the left vertebro-phrenic angle. CT scan showed a CT number of 34.2, which MRI examination revealed with low intensity on the T1 weighted image and high intensity on the T2 weighted image. A multilocular cystic tumor filled with viscid fluid, about 3 cm in size, was resected. This was diagnosed as a bronchogenic cyst, as microscopic examination showed bronchial epithelium, one layer of smooth muscle, bronchial glands, and cartilage. This is the 10th case of diaphragm-related bronchogenic cysts reported in Japan. Of these 10, 7 were located at the vertebro-phrenic angle.

[Two cases of tuberculous constrictive pericarditis].

Case 1 was a 79-year-old male suspected of tuberculous constrictive pericarditis. He was admitted to our hospital because of surgical treatment. His heart failure was NYHA IV. Culture of pleural effusion and pericardial effusion was negative. But ADA level in pericardial effusion was found to be increased. So tuberculosis was suspected. Cardiac catheterization date was compatible with constrictive pericarditis. Case 2 was a 73-year-old female. She was admitted because of heart failure (NYHA IV). As RVP wave indicated dip & platou at cardiac catheterization, she was diagnosed as constrictive pericarditis. ADA level in pleural effusion increased. So tuberculosis was suspected as etiology of constrictive pericarditis. In both cases, after pericardiectomy, heart failure improved to NYHA I. Results of pathological examination were tuberculous inflammation.

Effects of electrical stimulation of motor and cutaneous nerves on spinal cord blood flow.

The effects of electrical stimulation of the motor and cutaneous branch of the radial nerve on spinal cord blood flow (SCBF) in the cervical region and the correlation between the changes in SCBF and the amplitudes of spinal cord evoked potentials were investigated in cats anesthetized with alpha-chloralose. When measured by the hydrogen clearance technique, with stimuli of the cutaneous branch, the SCBF significantly (P < 0.01) increased from 37 +/- 8 (mean +/- SD) to 59 +/- 27 ml.100 g-1.min-1 at 10 Hz and from 39 +/- 6 to 54 +/- 9 ml.100 g-1.min-1 at 30 Hz, but stimulation of the motor branch had no effect. When measured by laser Doppler flowmetry, stimulation of the motor branch caused a small, transient, and significant increase in SCBF ending within 2 minutes. Stimulation of the cutaneous branch produced a large, prolonged, and significant increase in SCBF. The increase in SCBF produced by stimulation of the cutaneous nerve was significantly greater than that by stimulation of the motor branch. The SCBF was correlated linearly with the amplitude of the spinal cord evoked potential (r = 0.504; P < 0.01). The increase in SCBF caused by radial nerve stimulation may arise from changes in neuronal metabolism via peripheral nerve activation.

Interaction of cutaneous stromal cells and gamma/delta T cell receptor (TcR)-positive cells. I. V gamma 5-gamma/delta TcR+ T cells migrating from organ-cultured murine skin proliferate by co-culture with cutaneous stromal cells in the presence of interleukin-2.

It has been reported that Thy-1+CD3+CD4-CD8- cells as well as Langerhans cells migrate from organ-cultured murine skin into culture medium. We examined whether these Thy-1+ populations of migrating cells were derived from Thy-1+ dendritic epidermal T cells (Thy-1+ DEC) and found that they were Thy-1+CD3+CD4-CD8-gamma/delta TcR+ (gamma delta+T) cells but did not express V gamma 5TcR, which was used by a vast majority of Thy-1+ DEC. Recently, a unique interaction between stromal cells and lymphohemopoietic progenitors has been reported in bone marrow and thymus. In this study, we established fibroblastoid cutaneous stromal cell (CSC) lines and clones from murine skin and examined the interaction between CSC and gamma delta+T cells. When these gamma delta+T cells were co-cultured with CSC, a marked proliferation of small lymphoid cells was observed only in the presence of interleukin (IL)-2. Neither CSC alone nor IL-2 alone could induce a similar proliferation. Flow cytometry revealed that they were Thy-1+CD3+CD4-CD8-gamma/delta TcR+ but V gamma 5TcR-. Analysis of the major segments of their TcR by polymerase chain reaction demonstrated that V gamma 1, V gamma 2, V gamma 4 and all of the V delta chains from V delta 1 to V delta 7 were used without any predominant pattern. These data indicate the possible presence of gamma/delta+T cells other than V gamma 5TcR+Thy-1+ DEC in the murine skin and the unique capacity of the CSC to support the growth of these migrating gamma/delta+ T cells. The nomenclature of murine T cell receptor gamma chain is according to Reilly et al. (Nature 1986. 321:878). The relationship between the different nomenclature systems is summarized in Takagi et al. (J. Immunol. 1989. 141:2112).

Evaluation of a passive microcapsule agglutination test for the screening of human leptospirosis.

Microcapsules, absorbed with two mixed antigens--each composed of 3 sonicated leptospira serovars--were developed in the past as a microcapsule agglutination test (MCA-LS) for the screening of clinical leptospirosis. For this study, fifty serum samples, taken at an earlier and at a later stage of illness from 25 Italian in-patients with clinical symptoms of leptospirosis, were tested with both the MCA-LS one-dilution test and the microscopic agglutination (MA) test, the confirmatory test for leptospirosis, with 18 leptospira strains circulating in Italy. Compared with MA, MCA-LS showed a sensitivity of 91.7%, and a specificity of 92.3%, including leptospirosis not sustained by the diagnostic strains used in the MCA-LS kit.

Exacerbating factors of radiation-induced myeloid leukemogenesis.

The spontaneous incidence of myeloid leukemia in female mice was slightly higher than in male mice, whereas the radiation-induced incidence was significantly lower than in male mice. We also examined whether the incidence of myeloid leukemia was related to inflammatory response. Mice had a piece of cellulose acetate membrane inserted into the peritoneal cavity to cause inflammation. This did not affect the incidence of myeloid leukemia in unirradiated mice at all, but in 2.84 Gy irradiated mice the incidence (35.9% in male, 26.0% in female mice) increased significantly compared with irradiated-only mice (23.9% and 12.0%, respectively). From these results, the physiological fluctuation of humoral factors by means of inflammatory response is considered to increase the development of radiation-induced myeloid leukemia.

Age-related changes in the microvessels of the human stomach: an ultrastructural study.

The ultrastructure of microvessels in the gastric mucosa of patients with atrophic gastritis was investigated. In the gastric mucosa of atrophic gastritis, the endothelial cells of microvessels were irregularly swollen and subendothelial spaces were enlarged. The basement membrane underlying the endothelium was thickened. Degeneration of smooth-muscle cells was also evident in the media. These changes appear to correspond to sclerosis or aging of the microvessels. Such sclerotic changes in microvessels may initiate deterioration of the microenvironment in the gastric mucosa and may consequently affect metabolic activity of the gastric mucosal cells. Those sequences might induce degeneration or abnormal differentiation of mucosal cells. Such vascular changes may advance slowly and increase in intensity with advancing age. The final result would be the histologic changes associated with atrophic gastritis.

Histologic differentiation and motility of rat stomach.

An ultrastructural and immunohistochemical study was done to investigate the normal developmental process of rat gastric wall. Neuronal processes were observed on the external side of immature muscle cells on gestational day 15, and Auerbach's plexus was found on gestational day 18, after which neuronal elements extended just under the subepithelial area. Myofilaments appeared in undifferentiated mesenchyme cells on gestational day 15. On day 17, stomach was responsive to Ach stimulation. On the day after the initiation of gastric motility, gastric glands started to form on the mucosal surface. At the same developmental stage, some parts of the mucosal epithelium and the submucosal connective tissues were stained with antiserum against tenascin. Endocrine and parietal cells appeared in the mucosal epithelium on gestational day 19. Dense secretory granules on the apical surface of mucosal epithelium were increased between day 20 and 21. The cytoplasmic organelles gradually developed with advancing fetal age. We suggest that tenascin may play a role in gastric gland formation and that there are some relationships between gastric gland formation and gastric motility. We believe that the development of subepithelial tissues may be closely associated with epithelial growth.

Impaired sensory conduction in a mixed nerve induced by high frequency stimulation of the muscle fibres. An experimental study in cats.

To see if symptoms such as sensory disturbance or numbness worsen during exercise as is thought to occur in entrapment neuropathy, we studied the interaction between sensory and muscle nerve fibres in a mixed nerve in cats. Stimulation electrodes were placed on both the deep (muscle) and superficial (sensory) branch of the radial nerve at the elbow. High frequency stimulation at 100 Hz was applied to one branch for five minutes and the nerve action potential of the other branch was recorded at the axilla where the radial nerve is mixed. The stimulation decreased the amplitude of the nerve action potential and prolonged its latency. Recovery from these changes was gradual, and changes were the same whichever branch was stimulated. When a cat's leg was made ischaemic, the changes were more pronounced. The changes in the nerve action potential may have arisen from ischaemia in the nerve, possibly caused by high frequency stimulation.
