Oxidative aggression in atherosclerosis associated to degenerative psychoorganic disturbances.

We had in view the effect of the oxidative aggression as determinant factor in atherogenic process associated with degenerative psychoorganic disturbances. It was studied a group of old people distributed in two subgroups: a) 51 old people with atherosclerosis (AS) and b) 57 old people with atherosclerosis associated with degenerative psychoorganic disturbances determined by chronic ethylism. The results were compared to those of a 40 healthy adults group. Concomitantly, it were evaluated the supervened modifications in the antioxidant defense systems of the organism, by determining both of some nonenzymatic defense system components (reduced glutathione, ceruloplasmine and transferrin) and of some enzymatic defense system components (superoxide dismutase, catalase and glutathione peroxidase). The obtained results emphasize an important aggression exerted by the lipid peroxides against the old people organism, aggression that is amplified by the chronic ethylism with appearance of the degenerative psychoorganic disturbances. Generally, the antioxidant defense capacity of the old people organism is depressed, especially in the group with atherosclerosis associated with degenerative psychoorganic disturbances engendered by excessive and chronic alcohol intake.

[Bilateral retinal venous thrombosis after 8 years in a young patient with congenital coagulopathy].

The issue discusses the case of a young male patient (35 years old), with bilateral retinal vein thrombosis. The onset of the ocular disease was eight years ago, in the left eye, with decreased visual acuity and complicated with neovascular glaucoma; with an adequate treatment, anatomical and functional evolution was good. A new retinal vein thrombosis, this time in the right eye, make us to concentrate our efforts on the etiology of the disease. Thus, we found a blood hyperviscosity syndrome, caused by a congenital deficiency of protein "S" and resistance at the activated protein "C", which may produce recurrent venous thrombosis.

Molecular biology and clinical manifestation of hereditary factor VII deficiency.

Inherited factor VII (FVII) deficiency is a rare autosomal recessive disorder. Mutations and polymorphisms of the FVII gene were characterized in more than 40 unrelated patients with FVII deficiency. Among the 29 different mutations, the most frequent were Ala294 Val, Ala294Val;404delC, IVS7+7, and Val281 Phe. Four novel mutations (IVS2+1G>C, Arg247 Cys, Glu265 Lys, Asp343 His) were detected. The relationships between genotypes of mutations and polymorphisms of the FVII gene, FVII deficiency, and clinical phenotype were investigated. Homozygosity of the Phe4 Leu, IVS4+1G>A, Cys135 Arg, Ala244 Val, and Ala294 Val;404delC and the double heterozygosity of Tyr68 Cys / IVS3-1G>A, Val252 Met / IVS2+5G>T, Val281 Phe / Cys135 Arg, Ala294 Val / Val281 Phe, Ala294 Val;404delC / Val281Phe, Ala294 Val;404delC / Arg152 stop, Ala294Val;404delC / Gln(-35) stop, Ala294 Val / Val252 Met, Ala294 Val / Gly156 Asp, and Thr359 Met / Asp242 His were related to clinical symptoms. Double heterozygotes for Arg247 Cys / IVS2+1G>C, Ala206 Thr / Pro303 Arg, Leu(-20) Pro / Val252 Met as well as IVS7+7 /Ala294 Val, IVS7+7 /Ala206 Thr, and IVS7+7 / Met298 Ile were asymptomatic. The clinical symptomatology is rather poor in correlation with the FVII activity. Concerning the clinical phanotype, a correlation seems to exist between specific mutations and clinical symptoms.

Progress-curve equations for reversible enzyme-catalysed reactions inhibited by tight-binding inhibitors.

The rate equation for a tight-binding inhibitor of an enzyme-catalysed first-order reversible reaction was used to derive two integrated equations. One of them covers the situations in which competitive, uncompetitive or non-competitive inhibition occurs and the other refers to the special non-competitive case where the two inhibition constants are equal. For these equations, graphical and non-linear regression methods are proposed for distinguishing between types of inhibition and for calculating inhibition constants from progress-curve data. The application of the non-linear regression to the analysis of stimulated progress curves in the presence of a tight-binding inhibitor is also presented. The results obtained are valid for any type of 'dead-end'-complex-forming inhibitor and can be used to characterize an unknown inhibitor on the basis of progress curves.

[Ultrastructural modifications in the hypophyseal-adrenocortical system in rabbits during experimentally induced parodontosis (author's transl)].

During parodontosis induced experimentally by irritation of dental pulp with carbolic acid in rabbits, the participation in the pathological process of adenohypophyseal ACTH-producing cells and of adrenocortical fasciculata cells was studied. Ultrastructural alterations suggesting stimulation of ACTH and glycocorticoid synthesis were observed. The data obtained prove that via afferent pathways to the central nervous system, irritation of the dental pulp induces morphofunctional alterations in the hypothalamo-adenohypophyseal neurosecretory system, by means of functional perturbation of the cerebral cortex leads to hypothalamus (CRF) leads to adenohypophysis (ACTH) leads to adrenal cortex (glucocorticoids)axis.

[The india ink phagocytosis by monocytes and granulocytes during acute and chronic consumption coagulopathy].

During generalized intravascular coagulation (GIC) the polimerisation products of fibrinogen are phagocytized by the reticulo-endothelial system of the liver and spleen. Thus an inhibition of the clearance of RES could be detected in a progressed stage of the shock and GIC; this can also be represented by a decrease of India ink storage. In the present work the authors investigate the behavior of monocytes and granulocytes in the peripheral blood in GIC. Approximately 80% of cells will usually phagocytize and concentrate India ink granules, if heparinized blood mixed with India ink is incubated for two hours at 37 degrees C. A decrease of the India ink phagocytosis was detected in 6 patients and in 10 dogs during the shock. This observation corresponds to the behavior of hepatolienal RES during this condition.
