[The use of flow cytofluorometry for determining antisperm antibodies by sperm immobilization and sperm toxicity methods].

During antisperm antibody detection by conventional methods of immobilization and cytotoxicity investigators used flow cytometry to detect dead and/or immovable sperm cells. It was shown that after incubation of sperm cells with low antiserum dilution immovable spermatozoa are still living. Evidently, cytotoxicity test in this case brings about false-negative results. Therefore, flow cytometry will be a success for ASAB detection through immobilization test.

[Isolated symplasts and stromal syncytial buds of the normal human placenta at 6 to 14 weeks of pregnancy].

Formation of isolated symplasts (IS) ("deported trophoblast", according to English nomenclature) and stromal syncytial buds (SSB) has been studied in 10 normal patients at pregnancy of 6-14 weeks by means of the indirect immunoperoxidase method. Monoclonal antibodies against beta-subunit of the human chorionic gonadotropin (beta-HCG), serial paraffin slices, stained with hematoxylin-eosin, semithin slices, stained with toluidin blue-pyronine and scanning electron microscopy have been used. A great amount of syncytiotrophoblastic processes and IS, analogous by their structure to syncytiotrophoblast are revealed. Presence of beta-HCG in the syncytiotrophoblast and its absence in the cytotrophoblast are proved. Concentration of beta-HCG in the IS is higher than in the syncytiotrophoblast of the villi. Maximal concentration of beta-HCG in the SSB is described for the first time. The functional role of the IS and SSB is discussed in the system mother-placenta-fetus.

[Obtaining of monoclonal antibodies to human growth hormone and their characteristics].

The aim of present study was to obtain the hybridomas producing monoclonal antibodies against human growth hormone (Mabs hGH), to investigate the properties of the obtained Mabs and the possibility of their application in immunoanalytical systems. Two hybridomas secreting Mabs against hGH and belonging to the IgGI subclass have been obtained. The cross-reactivity of the Mabs with structurally similar to hGH hormones (hGHbio, hPL, hPRL, bGH, bPRL, pPRL) using indirect IFA has been studied. It has been shown that Mabs hGHI and Mabs hGH2 are directed to common specific antigenic determinant i.e. they have the same epitope specificity and don't react with other structurally related hormones, i.-e. this determinant is unique for hGH. The obtained Mabs hGH2 would find application for determination hGH by immunochemical methods in fractions while the hormone isolation from pituitaries and hGH obtained recombinant DNA methodology. The development of immunosorbents on the basis Mabs hGH2 seems to be perspective. Application of this immunosorbent may give possibility to optimize hormone isolation process.

[Monoclonal antibodies against the beta subunit of human chorionic gonadotropin].

The aim of investigation was creation of hybridomas, which produce monoclonal antibodies to the beta-subunit of human chorionic gonadotropin (beta-hCG), characterization of monoclonal antibodies, which necessary for hCG immunoassay in biological fluids, as an immunological methods of detection of early stage of pregnancy and choriocarcinoma. 4 hybridomas, producing monoclonal antibodies to-hCG of IgG1 isotype, were created. On the base of monoclonal antibodies, which produced by D2 hybridoma cell line, test-systems for RIA of hCG in blood serum and urine were elaborated. These test-systems can be used in medical practice for diagnosis of early stages of pregnancy and choriocarcinoma.

[Regional graft vs host reaction to H-Y antigen (immunological mechanisms)].

Structural changes in the regional popliteal lymph nodes have been studied in C57Bl/6 male mice at the peak of the reaction "graft-versus-host" to H-Y antigen. Morphological and morphometrical investigations have been carried out in three groups of males (10 animals in each group). The first group includes intact animals (the first control group). To the males of the second group (the second control group) lymphoid cells are inoculated from intact C57Bl/6 females. To the males of the third group (experimental group) lymphoid cells are inoculated from H-Y antigen immunized C57Bl/6 females (anti-H-Y effector lymphocytes). The popliteal lymph nodes of the male mice from the third group twice increase in their size comparing to those in the control (the first and the second groups). Miotic activity increases in them 4.5 times, amount of cellular blast forms in medullary cords--4 times and 10 times--in the light zone of the cortical substance. Ratio of macrophages and eosinophils in structural components of the lymph nodes studied changes; this is, evidently, connected with massive destructive progresses, that take place in the lymph nodes of the animals from the third group. The results of the morphological investigations are in agreement with the hypothesis suggested, explaining the mechanism of development of the regional reaction "graft-versus-host" to H-Y antigen, basing on idiotype-antiidiotype interaction (the idiotypic network in the immune system).

[Ultrastructure of the ovaries of guinea pigs immunized with preparations of purified and unpurified antigens of the zona pellucida of xenogenic ova].

The state of the guinea pig ovaries has been studied electron microscopically in 8-15 and 30 days after an active immunization course with purified and non-purified antigens of the ovum zona pellucida has been completed. Both antigens produce early total morphofunctional rearrangements in the ovaries: increasing process of follicular atresia and disturbance of folliculogenesis, accompanied with hyperplasia of the interstitial tissue. The non-purified antigen produces more pronounced changes in the cavitary follicles: disturbances of cellular communications, barrier function of the follicular epithelium becomes weak, its respiratory, plastic and secretory function changes with subsequent distension of the follicle and with an enhanced development of connective tissue in its membranes. Injection of the purified antigen results in progressive atresia of the cavitary follicles and in hyperplasia of the interstitial tissue. Reduction of the corpus luteum is not observed.
