Serum procollagen peptides and collagen type VI for the assessment of activity and degree of hepatic fibrosis in schistosomiasis and alcoholic liver disease.

Schistosomiasis, in contrast to alcoholic liver disease, leads to presinusoidal hepatic fibrosis, which determines the prognosis of the disease. Because conventional liver function tests and liver biopsy specimens provide little information about the dynamics of the fibrotic process, we measured the serum concentrations of procollagen type III N-propeptide and procollagen type I C-propeptide, believed to mainly reflect collagen synthesis, and procollagen type IV C-propeptide and collagen type VI, two presumptive markers of collagen degradation. Determinations were performed in 15 healthy control subjects, 69 patients with various stages of infection with Schistosoma mansoni/Schistosoma haematobium (28 with an early active infection and no organ involvement, 27 with hepatosplenic involvement and 14 with complications of portal hypertension) and 16 patients with alcoholic cirrhosis. In addition, liver biopsy specimens were obtained from 30 schistosomal patients (18 with hepatosplenic involvement and 12 with complications of portal hypertension for histopathological grading and collagen histochemistry. Procollagen type III N-propeptide was significantly elevated in the three patient groups with schistosomiasis when compared with controls (p less than 0.01). Also, patients with higher histological grading showed significantly higher procollagen type III N-propeptide values (p less than 0.05). In alcoholic patients, procollagen type III N-propeptide was even higher and increased parallel to the severity of the disease, determined by using a combined clinical and laboratory index. Procollagen type I C-propeptide was only elevated in early infection (p less than 0.05) and steadily decreased with disease progression.(ABSTRACT TRUNCATED AT 250 WORDS)

Polyelectrolyte effects on 9-aminoacridine-DNA binding.

The 9-aminoacridine-DNA binding curve is analyzed in two ways: with polyelectrolyte effects neglected and with polyelectrolyte effects included. It is found that the analysis which includes polyelectrolyte effects is consistent with the violation of neighbor exclusion displayed by diacridine complexes as observed by Atwell et al. and by Zimmerman and coworkers. However the analysis which neglects polyelectrolyte effects is inconsistent with the diacridine results. This comparison supports the necessity of including polyelectrolyte effects in the analysis of drug-DNA binding curves.

Antiparietal cell antibodies in gastritis associated with chronic liver diseases.

Thirty patients with gastritis associated with chronic liver disease and ten normal controls were the subject of this study. Pathologically about 18 were diagnosed as chronic atrophic gastritis, 10 as chronic superficial gastritis, 1 as gastric atrophy and 1 as hypertrophic gastritis. Parietal cell antibodies (PCA) were detected (IFA) in 2 cases with no significant relation.

Influence of hormonal imprinting on hormone level. Permanent receptor damaging effect of pituitary hormones administered to newly-hatched cockerels.

The overlapping effect of TSH and FSH on the gonad and on the thyroid gland can be demonstrated in cockerels even at the age of five weeks. These hormones influence the secretion of testosterone in a similar way and to a similar extent, while on the thyroxine level the influence of the specific hormone is more pronounced. Neonatal FSH and TSH treatment considerably decreased the basal testosterone level measured at the age of five weeks. Neonatal FSH treatment increased the basal T4 level while TSH treatment decreased it. The effect of TSH treatment administered at the age of five weeks in increasing the testosterone level was weakened after neonatal pretreatment with any iodine hormone. The effect of TSH treatment could only be inhibited by neonatal FSH pretreatment. Neonatal pretreatment with any of the trophormones caused a diminution of the T4 level augmenting of FSH and TSH administered at the age of five weeks.

Investigation of gonadotropin-thyrotropin overlapping and hormonal imprinting in the rat testis.

In the neonatal period both gonadotropin and thyrotropin increase the weight of the testis, influence considerably the diameter of the contorted tubules increase the occurrence of Sertoli cells and decrease the number of spermatogonia. These phenomena can still be observed at the age of seven days but they disappear by the age of six weeks; at that time the hormones decrease the weight of the testis. One single TSH treatment administered in the neonatal period considerably increased the weight of the testis and the diameter of the channels when investigated at the age of six weeks. Gonadotropin had none of these effects. The phenomenon of imprinting could be proved as in the animals pretreated with TSH, gonadotropin given at the age of six weeks decreased the weight of the testis and both hormones decreased the diameter of the seminiferous cords.

Structure--activity relationships within a series of 2,4-diaminoalkoxybenzene compounds.

Six 2,4-diaminoalkoxybenzenes were examined for their ability to induce mutation in Salmonella typhimurium. Each compound was tested at 8 concentrations in 5 strains. The mutagenicity was influenced by the size of the alkoxy group substituted at the C1 position of 2,4-diaminobenzene. When S9 induced by Aroclor 1254 was present, 2,4-diaminoanisole (the methoxy derivative) exhibited the highest mutagenic activity. The compounds 2,4-diaminoethoxybenzene, 2,4-diaminoisopropoxybenzene and and 2,4-diamino-n-propoxybenzene were also mutagenic, but were distinctly less active than 2,4-diaminoanisole. With the compounds 2,4-diaminophenoxyethanol and 2,4-diamino-n-butoxybenzene, the increases in numbers of revertant colonies above control levels were slight or absent. The mutagenicity of 2,4-diaminoalkoxybenzenes was detected in strains TA98, TA1538 and in some cases TA1537. None of the compounds was active in strain TA1535. The relative response of the various strains suggests that 2,4-diaminoalkoxybenzenes induce frameshift mutations but not base-pair substitutions. None of the compounds was active without metabolic activation. In addition to conducting the standard plate test, we tested the urine of rats exposed to 2,4-diaminophenoxyethanol and 2,4-diaminoanisole for mutagenicity in Salmonella typhimurium. The rats were exposed by topical application, oral administration or intraperitoneal injection. The results were positive for 2,4-diaminoanisole and negative for 2,4-diaminophenoxyethanol.
