Electrophoretic patterns of extracellular deoxyribonuclease (DNase) and their correlation with T-type in group A streptococci.

Molecular heterogeneity of the extracellular deoxyribonuclease (DNase) in group A streptococci was demonstrated in 42 clinical isolates. Although polyacrylamide gel electrophoretic patterns of the extracellular DNase of all the isolates were heterogeneous, they could be divided into five main patterns with respect to the presence or absence of three DNase components including DNase B. By comparing the electrophoretic patterns of DNase in all the isolates with their T-types, we found that the patterns were quite characteristic for their T-types, especially in the prevalent T-types 12 and 1, and that the isolates of T-types 12 and 1 produced DNase B as their major extracellular DNase. Relative DNase B activity in the total extracellular DNase activity of group A, B, and G isolates was determined by the rapid method of neutralization with anti-DNase B antibody. The results showed neutralization of DNase activity in all the isolates of group A streptococci, largely corresponding to their T-types, but not of the isolates of groups B and G. These results indicate that the electrophoretic patterns of the extracellular DNase of group A streptococci are closely correlated with their T-types, suggesting the physicochemical taxonomic value of these properties.

[Antibacterial potency of cefotiam based on the clinical effect, MIC and decrement of organisms in the sputum].

To evaluate the antibacterial potency of cefotiam (CTM) clinical and laboratory studies were carried out and the results were as follows. Clinical evaluation and adverse reaction CTM was given to total of 23 patients, 10 with bronchopneumonia, 10 with bronchitis and one each with cystitis, enteritis and suspected sepsis. Overall efficacy rate was 78.3% (18/23) (excellent 9, good 9, fair 3, poor 2). Only 1 case showed a side effect of slightly elevated GOT and GPT. Antibacterial activities MIC of CTM against isolates from sputum was investigated on those patients mentioned above and was compared with MIC of CEZ and CMZ. CTM showed superior antibacterial activity against almost all strains. Especially on Haemophilus and Klebsiella antibacterial activity of CTM was impressive. Organisms in sputum Four out of 8 causative bacteria disappeared and 1 out of 8 decreased after administration of CTM. Thus CTM is considered to be the useful drug for the treatment of respiratory infection.

The origin of lipid accumulated in liver lysosomes after administration of triton WR-1339.

The origin of the lipids accumulated in liver lysosomes after administration of Triton WR-1339 was investigated. When Triton WR-1339 was injected into rats, serum triglyceride and cholesterol increased markedly. The highest content of triglyceride was observed in the second-day serum, from which very-low-density lipoprotein (VLDL) was isolated. The VLDL was administered to normal rats, then the light mitochondrial fraction of the liver at 24 h was centrifuged in a sucrose density gradient. The activities of lysosomal enzymes, acid phosphatase, N-acetyl-beta-D-glucosaminidase and acid lipase, were all shifted to less dense fractions as compared with those of normal lysosomes. [3H]Triglyceride-labeled VLDL was injected similarly, and at 12 and 24 h after the administration, the light mitochondrial fraction of the liver was fractionated by sucrose gradient centrifugation. Protein content and radioactivity in the immunoprecipitate with anti-VLDL serum at 12 h showed almost the same distribution as acid phosphatase activity. At 24 h, though acid phosphatase activity, immunoprecipitable protein content and radioactivity were all found in less dense fractions than in the case of normal lysosomes, the former two distributions were significantly different from the latter. The anti-VLDL serum reacted in Ouchterlony tests not only with Triton-induced VLDL and normal VLDL but also with the extract from low-density lysosomes. These results suggest that the lipids accumulated in low-density lysosomes following the administration of Triton WR-1339 were probably derived from the elevated serum VLDL induced by the treatment.

A simple procedure for a large-scale preparation of tritium-triglyceride-labeled very low density lipoprotein of the rat.

A simple procedure for the preparation of large quantities of tritium-triglyceride-labeled very low density lipoprotein (VLDL) from rat serum described. Triton WR-1339 (0.10 ml) was intraperitoneally injected into rats weighing 200--300 g, and 100 muCi of tritium-triolein in corn oil (1 ml) was simultaneously administered orally. VLDL was separated by ultracentrifugation from the serum administration of the above compounds for 2 d. At least 10 mg of protein of VLDL was obtained from each animal, and the radioactivity in VLDL was exclusively found in triglyceride. Isolated radioactive VLDL was injected into normal rats, and the decay of the radioactivity in serum was found to be almost identical with that of triglyceride, but different from that of cholesterol.

[Annual changes of sensitivities of Staphylococcus aureus to several antibiotics].

The antibacterial activities of Staph. aureus isolated in 1980 and 1981 at all districts in Japan to ABPC, CEZ, CMZ and GM were tested. Numbers of test strains were 587 strains in 1980 and 700 strains in 1981, respectively. The frequencies of ABPC and CEZ resistant strains of Staph. aureus (MIC: greater than or equal to 25 micrograms/ml) were clearly increased in annual variation. The percentage of CEZ resistant strains in 1981 tested by 10(8) CFU/ml inoculum size was 12.3%, but the same of CMZ was only 0.6%.
