[The clinical administration of the opioid antagonist naloxone in dogs].

The short introduction gives a review on the complex of exogen and endogen opioids and their receptors as well as on their pharmacodynamics, pharmacokinetics and toxicity of naloxone. The clinical efficacy of naloxone as an opioid antagonist is described. Applications of naloxone for the dog are specified: antagonisation of etorphine, morphine, levomethadone and fentanyl, antagonisation of exogen and endogen opioids in puppies and treatment of lactomania in the bitch. The mean effective dose to antagonize morphines is 0.003 mg/kg bodyweight. If persisting analgesia is indicated the dose of naloxone in titrating steps in 0.001 mg/kg bw. To antagonize postpartal hypoxia in puppies 0.02 mg per animal naloxone have to be injected. For treatment of lactomania a dose of 0.01 mg/kg bodyweight twice a day for a couple of days is recommended. The clinical effectivity of naloxone is proved doubtlessly. Compatibility and safety are very high.

Prenatal diagnosis of genetically determined early manifestation of autosomal dominant polycystic kidney disease?

A case of an unusually early manifestation of autosomal dominant polycystic kidney disease (ADPKD) is reported that was prenatally diagnosed by ultrasound. The ultrasonographic picture showed greatly enlarged kidneys and increased echogenicity that was indistinguishable from cases of autosomal recessive polycystic kidney disease or Meckel syndrome without further information. Because of two further cases of early manifestation of ADPKD within the family reported (brother and cousin), as well as several other "familial" cases reported in the literature, we postulate that genetic factors are involved (modifying alleles). When reported observations of "familial" cases of early manifestations of ADPKD are made, genetic counseling should be considered.

Localization and characteristics of ferritin in human bone marrow.

It has been found possible to test ferritin concentrations in the reticulum and hemopoietic cells from human bone marrow by an immunoradiometric assay. Ferritin concentration in healthy test persons amounts to 0.92 +/- 0.38 ng/microgram protein in the reticulum and 0.084 +/- 0.031 ng/microgram protein in hemopoietic cells. In healthy test persons about 90% of the ferritin is localized in the reticulum and about 10% in the hemopoietic cells. The ferritin concentration in the reticulum is decreased in patients having iron deficiency anemia. In these patients only 50-70% of the bone marrow ferritin is localized in the reticulum. Bone marrow ferritin was characterized by a combination of anion-exchange chromatography using Sephadex A-50 and an immunoradiometric assay. Marrow ferritin from healthy persons is eluted at chloride concentrations between 200 and 300 mM. The anion-exchange chromatographic properties of ferritin in the reticulum and hemopoietic cells are identical.

Comparison of brain structure volumes in Insectivora and Primates. I. Neocortex.

Based on volume measurements the total neocortex increases enormously from the lower ('basal') Insectivora, through prosimians, monkeys and apes up to man. In man it is about 132 times larger than in the average basal Insectivora of (theoretical) equal body weight and 232 times larger than in the insectivoran species which has the least developed neocortex. Within the neocortex the white matter increases more markedly than the grey matter. For man the white matter reaches a value 298 times that of the basal Insectivora and the grey matter reaches a value 198 times greater. Within the grey matter the cell dense layers (laminae 2-6) increase distinctly more than the molecular layer (lamina 1). Thus in man the volume of layers 2-6 is 272 times greater than that in basal Insectivora and the molecular layer is 68 times greater. When related to the total grey matter the percentage of the molecular layer clearly decreases from about 32% in the lower Insectivora to 12% in higher primates.

Ferritin in bone marrow and serum in iron deficiency and iron overload.

Nonheme iron and ferritin in the bone marrow and serum ferritin was investigated in patients with iron deficiency anaemia or iron overload. As controls served patients without any disturbance of the iron metabolism. There is a precise correlation between the nonheme iron and ferritin in the bone marrow of patients with and without disturbance of iron metabolism. A correlation was also found between the ferritin in the bone marrow and the serum. Nonheme iron and ferritin in the bone marrow and serum ferritin was decreased in patients with iron deficiency anaemia. Conversely, the same parameters were increased in patients with iron overload.

Deep hyperthermia with radiofrequencies in patients with liver metastases from colorectal cancer.

Patients at advanced stage of colorectal cancer with liver metastases have been treated with deep hyperthermia alone or in combination with chemotherapy (5-FU + FA + MMC). Hyperthermia was achieved by arrangements of capacitive electrodes with a radiofrequency field of 13.56 MHz (RF-DHT). This prospective open single-arm clinical study with 80 patients suffering from liver metastases from colorectal cancer gives some first hints, that deep RF-hyperthermia alone may have a substantial beneficial effect on overall survival time of patients with liver metastases from colorectal cancer. Long lasting no-change, partial and even some complete remissions could be observed. The overall median survival time from progression of metastases or relapse was 24.5 months and survival rates at 1, 2 or 3 years from first diagnosis of metastases or progression were twice as high as expected from patients treated with chemotherapy. The combination of hyperthermia with delayed chemotherapy did not change overall survival time. These encouraging results deserve to be confirmed in randomized clinical studies.

A total organic carbon analysis method for validating cleaning between products in biopharmaceutical manufacturing.

The validation of cleaning procedures for biopharmaceutical products produced by recombinant DNA (rDNA) technology presents a diverse analytical challenge. This is because of the need for quantitation of a broad range of potential residual cellular components, including proteins, carbohydrates, and nucleic acids, as well as trace levels of detergents at various manufacturing stages. The validation of a Total Organic Carbon (TOC) analysis method for use in cleaning validation studies is presented. The method has a limit of detection of approximately 0.1 ppm, with a limit of quantitation of 0.5 ppm. TOC analysis has an accuracy of 50 to 70% or better in the 0.5- to 10-ppm range and demonstrates an overall variability of approximately 5%. The method is broadly applicable to a variety of impurities and contaminants that are likely to be encountered following the manufacture of rDNA products.
