Derivatives of 2-mercaptobenzenesulphonamide. VIII. Synthesis and some pharmacological properties of 1,3-dialkyl-2-(4-chloro-2-carboxyalkylthio-5-methylbenzenesulphonyl) guanidines.

Synthesis of 1,3-dialkyl-2-(4-chloro-2-carboxyalkylthio-5-methylbenzenesulphonyl)guanidines (IX-XVII) and their easily soluble in water sodium (Xa-XIVa, XVIIa) or 2,6-dimethylpiperidine salts (XIb, XVIIb) have been described. The results of preliminary pharmacological examinations such as acute toxicity and influence on circulatory system have been presented.

Synthesis and pharmacological investigations of derivatives 6-azabenzodiazepines.

Acylated derivatives 4-6 of 2-aminomethylene-4-phenyl-1H-tetrahydro-6-azabenzo-1,5-diazepine 3, and carbonyl derivatives 7-10 of 6-azabenzo-1,5-diazepines were obtained. They did not display any particular pharmacological activity in screening tests: 2 exerted a weak hypotensive action and 8 possessed negligible anticonvulsant properties.

Catecholamines concentration in brain and heart tissue of rats and its blood pressure during chronic treatment with a new tricyclic antidepressant 10-(3-dimethylaminopropyl)-7-chlorobenzo-(b), (1,8)-naphthyridone-5 monohydrochloride (IF-C-45).

DA concentration in brain tissue decreased statisticaly after 3, 6 and 9 weeks, NA concentration also statisticaly decreased but after 3 weeks only. The NA concentration in heart tissue decreased statisticaly significant in all investigated animals. There were no statisticaly significant changes in blood pressure.

Influence of plasma substitutes on the concentration of tissue catecholamines in rats.

The concentration of catecholamines was determined in the brain, heart and adrenals in normovolaemic and hypovolaemic rats after reinfusion of the lost blood and after infusion of equivalent volumes of plasma substitutes (dextran and modified gelatins). After infusion of these preparations in normovolaemic rats noradrenaline concentration increased significantly in the myocardium. It was found also that restoration of normal blood volume by infusion of plasma substitutes prevented changes in catecholamine concentrations induced by hypovolaemia in the brain but not in the adrenals.

Assessment of haemodynamic changes and acid-base equilibrium during hypovolaemia and after infusion of plasma substitutes in dogs.

The following haemodynamic values were determined in anaesthetized mongrel dogs: heart rate, systolic blood pressure in the ascending aorta, left ventricular pressure at the peak dp/dt, left ventricular end-diastolic pressure, time interval from Q in ECG to the onset of the systolic wave of dp/dt, time interval from Q in ECG to peak dp/dt, maximum rate of left ventricular pressure rise, femoral arterial flow, and certain indices of left ventricular contractility. It was concluded from the results of these experiments that infusion of a modified gelatin solution Fluigel prevented haemodynamic and metabolic changes produced by experimental hypovolaemia more effectively than infusion of Plasmagel.

Tissue catecholamine and arterial blood pressure changes in non-anaesthetised rats following administration of a new derivative of furanocarboxylic acid with pressor effect.

The effect of a new derivative of furanocarboxylic acid MePAF-4 on the arterial blood pressure, heart rate and the concentration of catecholamines in the brain, heart and adrenals was assessed in non-anaesthetised rats. MePAF-4 was given orally in a single dose of 60 mg/kg (1/30 of LD50)administered during 9 days (at the same daily dose), of injected intravenously in a dose of 10 mg/kg. After intravenous injection MePAF-4 had a significant pressor effect associated with bradycardia but without changes in the tissue concentration of catecholamines. After a single oral dose a slight, transient (60 min) rise of the arterial blood pressure was observed, while the heart rate showed no statistically significant changes. Repeated oral administration of MePAF-4 raised the brain dopamine level and adrenal adrenaline level with simultaneous increase in the weight of these organs. No detectable effects of the long term MePAF-4 administration on the arterial blood pressure and heart rate were observed.

Relationship between hemodynamic changes and endogenous catecholamine release caused by a new antiarrhythmic agent, AB-2, in cats and dogs.

An antiarrhythmic agent, AB-2, given iv produced in cats and dogs a biphasic action on blood pressure: rapid and short-lasting hypertension followed by depression of blood pressure. The first phase was accompanied by a sharp rise of serum catecholamines (6-fold increase in adrenaline, and 4.5-fold increase in noradrenaline level) and of indices of left ventricular contractility. These effects were more pronounced in cats than in dogs. In cats, but not dogs, the biphasic effect was observed also after po administration. The elevated endogenous catecholamine release was not accompanied by an increase in plasma 11-hydroxycorticosteroids levels.

Hypotensive effect of clonidine and regional noradrenaline concentration in the brain of normotensive and hypertensive rats after acute and long-term treatment.

The following experiments were performed: a) the effects of increasing doses (0.025-0.2 mg/kg p.o.) of clonidine on the blood pressure and heart rate in SHR and normotensive rats (acute treatment), b) the influence of an effective dose (0.05 mg/kg) of clonidine on the same parameters during 3-week administration, c) the effect of clonidine (0.05 mg/kg) on tissue noradrenaline (NA) concentration in the medulla, striatum and hypothalamus after acute and long-term treatment of SHR and normotensive rats. The experiments were carried out on unanaesthetized animals. Blood pressure and heart rate were measured by on indirect method. Tissue NA was assayed fluorimetrically. Clonidine in a single dose caused a significant fall of blood pressure and heart rate. These effects were much more pronounced in SHR. The effective dose (0.05 mg/kg) in acute experiments was ineffective in long-term (3 weeks) treated rats. This dose of clonidine had no detectable effect on regional NA concentration in rat brain with the exception of the hypothalamus where the concentration of NA was markedly increased (SHR in acute experiments).

Tissue catecholamine concentrations in the brain, heart and adrenals in experimental hypovolaemia in rats.

The experiments were carried out on Wistar rats with graded hypovolaemia ranging from 10% to 30% of blood volume loss. Determinations were done 10, 60 and 120 minutes after haemorrhage. The concentrations of catecholamines in the tissues were determined fluorimetrically. It was found that even moderate hypovolaemia evoked changes in catecholamine content of the brain, heart and adrenals.

Biosynthesis of catecholamines, uptake of 3H-noradrenaline, and reactivity of cardiovascular system of the rat after chronic and acute treatment with a new antidepressant agent, IPF C-45.

The effect of acute and chronic treatment of rats with IPF C-45 on the following biochemical and pharmacological parameters was tested: catecholamine concentration in brain, heart and adrenals, 3H-noradrenaline uptake by cardiac muscle, the action on catecholamine synthesis following treatment with alpha-methyl-p-tyrosine, reactivity of cardiovascular system to catecholamines and isoproterenol-induced tachycardia. Given chronically, IPF C-45 does not affect noradrenaline uptake by the cardiac muscle. Both chronic and acute administration of the drug decelerates catecholamine biosynthesis by inhibition of tyrosine hydroxylase activity. It seems that the action of IPF C-45, a benzonaphthyridone derivative, affects catecholamine metabolism in a manner distinctly different from that of tricyclic antidepressant drugs.
