Effect of cyclosporin A on B cell maturation and differentiation.

The activation and differentiation of resting B cells into Ig secreting cells are regulated by T cells, macrophages and their secreted factors. The present study evaluated the effect of cyclosporin A (CsA) on this process. Peripheral blood lymphomonocytes (PBMC) drawn from healthy donors were stimulated with protein A (PA) or with lipopolysaccharides plus pokeweed (LPS+PWM) in either the presence or the absence of CsA. Phenotypic B cell changes and immunoglobulin production was then analyzed. The data revealed that CsA decreased the expression of B cell surface receptors of the activation phase, and enhanced the resting phase receptors. Different effects of CsA were found on B cell differentiation, depending on its induction by PA or LPS+PWM. In the first system, CsA decreased the expression of differentiation phase receptors and the secretion of free Ig. In cultures stimulated with LPS+PWM, CsA increased the differentiated phase receptors and Ig secretion. Thus, CsA seemed to act as a blocking agent of the activation phase and as a modulator of the differentiation phase and of IgG secretion, depending upon the antigen used for stimulation.

Membrane interactions involved in the induction of interferon-alpha by Mycoplasma pneumoniae.

Interferon (IFN) induction by Mycoplasma pneumoniae (MP) in human peripheral blood mononuclear cells (PBMC) has been studied. We show that IFN yields depend on the concentration of both lymphocytes and MP. The effective IFN inducer appears to be the mycoplasma membrane, and IFN production occurs without significant lymphocyte proliferation. Data obtained by both positive and negative selection experiments suggest that the PBMC subpopulation induced by MP is B lymphocyte, as opposed to the main subpopulation induced by NDV that is monocyte. Evidence is also provided suggesting that the membrane interaction between MP and the B cells is mediated by MHC Class II antigens.

In vitro interferon production: induction in human leukocytes after exposure to cytomegalovirus antigens.

In vitro lymphocytes from healthy donors with crude (HCMV), heat inactivated (HCMVi) and infected human fetal fibroblasts (HCMVFF) were stimulated. In all the experiments peak titers of alfa Interferon (IFN) were present 24 h after exposure to antigens. On the other hand, at least after seven days, only lymphocytes of seropositive donors stimulated by HCMVFF and HCMV showed gamma IFN production. Our HCMVFF gave much higher IFN yields than free virus. Our data suggest that HCMVFF and HCMV are recognised by lymphocytes of HCMV specific memory and HCMVFF give additional and more reproducible information on HCMV specific cellular immunity.

Heredity and infectious diseases: a twin study.

A concordance study of 6 infectious diseases of childhood has been carried out in a sample of 656 twin pairs classified by sex and zygosity. A new approach is proposed to estimate the respective influence of heredity and of common environment. The estimates thus obtained range from 86% hereditary component in the case of measles to 100% environmental component in the case of scarlet fever.
