Characterization of lipoxygenase-1 null mutants in barley.

This study describes the discovery and characterization of lipoxygenase-1 (LOX-1) null mutants in barley. Six lines did not exhibit any significant LOX activity in the silenced seed extract. Immunological analysis showed that these lines lacked the authentic LOX-1 protein. Genetic analysis of the F2 population revealed that this trait was governed by a single recessive gene located at the LoxA locus on chromosome 4H. The six LOX-1 null mutants shared similar features and the same unique polymorphism in a structural gene region, implying that these mutants might be derived from the same ancestral origin.

Adsorption to or desorption of beer components from a lipid membrane related to sensory evaluation.

The relationship between the adsorption to or desorption of beer from a lipid membrane and sensory evaluation was studied using a lipid-coated quartz crystal microbalance connected to a flow injection system. The adsorption and duration of adsorption of commercial beers showed a significant correlation with their body and smoothness in a sensory evaluation, respectively. Isohumulones, tartaric acid, NaCl, glutamic acid, and tannic acid were adsorbed onto the lipid membrane. Di- and trihydroxyoctadecenoic acids increased the duration of adsorption of the beer components onto the lipid membrane but not the extent of adsorption. They decreased the astringent duration of beer and the smoothness in the sensory evaluation but did not affect the intensity of bitterness or astringency or the body. It seems that this system, which modifies the electrostatic and hydrophobic interactions of the beer components with the tongue and throat surfaces, can mainly evaluate bitterness and/or astringency which significantly affect the body and smoothness of beer.

The advantage of the osmium conductive metal coating for the detection of the colloidal gold-conjugated antibody by SEM.

For the detection of the colloidal gold-conjugated antibody by scanning electron microscopy, the comparison between the secondary and the back-scattered electron images is necessary. After the various durations of osmium conductive metal coatings, enlargements of the diameters of the antibodies were observed. The thick coating reduced the contrast of the backscattered electron images. By the optimal thickness of the coating, the secondary electron image showed a minimum reduction in the resolution, and the backscattered electron image showed not only the gold particles with high contrast, but also the outline of the basic morphology.

Nucleotide sequence of the uricase gene from Bacillus sp. TB-90.

The nucleotide sequence of the uricase gene from the thermophilic bacterium Bacillus sp. TB-90 was determined. The primary structure of the uricase deduced from the nucleotide sequence comprised 332 amino acids, with a total molecular mass of 37,994 Da. The molecular mass of the subunit of the uricase produced by the transformant of Escherichia coli agreed well with this value. However, the molecular mass of a subunit of the uricase produced by Bacillus sp. TB-90 was found to be 34,000 Da by SDS-PAGE. The difference between these molecular masses was attributed to processing of the C-terminal 13 amino acid residue in Bacillus sp. TB-90. Comparison of the enzymatic properties of both uricases showed that the thermostability of the uricase produced by the transformant was enhanced by about 10 degrees C in comparison to that produced by Bacillus sp. TB-90.

Membrane relationships in murine Meissner corpuscles: cytology of freeze-substituted tissue.

Mechanoreceptive sensory corpuscles (murine Meissner corpuscles) in the toe pad skin of mice, consisting of axon terminals and lamellar cells, were studied following freeze-substitution in order to clarify the plasma membrane relationships between axon terminals and lamellar cells. Tissue preservation of corpuscles was excellent when the corpuscle was located within 10 microns from the contact surface with the precooled metal block. The axolemmata appeared more electron-opaque than did plasma membranes of lamellar cells. The inner leaflet of the unit membranes was thicker than the outer leaflet in the axolemma, and the contour of cell plasma membranes was relatively smooth and straight. Characteristic focal or regional approximations of plasma membranes were noted between the axon and abutting lamellae. Such membrane appositions resembled gap junctions, although no gap junctions were found between the axon and lamellae in chemically fixed materials. Similar gap junction-like close appositions of plasma membranes also were found between neighboring lamellae. These approximations occurred more frequently than typical gap junctions seen in chemically fixed materials. These findings indicate that there may be a relationship of the plasma membranes in the axon terminals and in abutting lamellae as well as between neighboring lamellae that have not been identified as yet in conventional chemically fixed material. Another striking finding was that basal laminae on lamellar cells exhibited the same electron opacity as the surrounding connective tissue matrix and thus the two are indistinguishable from one another. Furthermore, the lamina lucida was not evident, and basal lamina material was directly contiguous with the plasma membrane.(ABSTRACT TRUNCATED AT 250 WORDS)

A re-evaluation of the cytology of cat Pacinian corpuscles II. The extreme tip of the axon.

The present report is the second of two studies re-evaluating the cytological characteristics of Pacinian corpuscles. The extreme tip of the axon of a Pacinian corpuscle has been identified and is quite different from the previously described ultraterminal region. The latter is the site where the inner core lamellae begin to terminate and is characterized by a smooth axolemma. The extreme tip lacks inner core lamellae directly abutting the axolemma and is instead characterized by the presence of many axonal spines projecting into a matrix of basal lamina-like material. The extreme tip of the axon thus resembles the organization of the axolemma facing the clefts of the inner core. The axonal spines at the cleft and extreme tip are proposed as a site of restricted current flow due to the tight apposition of inner core lamellae to the axolemma of X-axis. The hemi-inner cores thus could restrict current flow to the cleft. These anatomical specializations could represent both a source and a sink for K+ ions during mechano-electric transduction and account in part for the exquisite sensitivity of Pacinian corpuscles to complex pressure waves.

An electron microscopical morphometrical study on pinealocytes of pregnant rats.

A morphometrical study was made on the cut-surface of the pinealocytes in electron micrographs of the pineals from pregnant, puerperal and non-pregnant female rats. No significant differences in the cross-sectional area of the pinealocytes and the nuclei or in the nucleo-cytoplasmic ratio were found among the three groups of pregnant, puerperal and non-pregnant rats. The cross-sectional area of the nucleoli was, however, larger in the pinealocytes of the rats in pregnancy and early puerperium. This has been statistically confirmed by the ranking method (the U-test) of Wilcoxon, Mann and Whitney. The enlargement of nucleoli suggests the enhanced function of the pineal in pregnancy and early puerperium.

Scanning electron microscopic studies of ileal epithelial cells in suckling rats.

The ileal epithelial cells containing the tubulo-vacuolar systems and supranuclear vacuoles in suckling rats were investigated with scanning electron microscopy, using specimens treated with osmium-thiocarbohydrazide-osmium staining methods, and critical point drying and cracking. The cracked surface of the apical cytoplasm is seen as irregular and small hollows and pores of the anastomosing and branching tubulo-vacuolar system. The cracked surface of the supranuclear vacuoles shows the ellipsoidal structures. Numerous pores of various size and irregular shape are present on the apical inner surface of the supranuclear vacuole. These pores are clearly the openings from the tubulo-vacuolar system to the supranuclear vacuole. Some small pores are visible on the inner lateral surface of the supranuclear vacuole, especially near the nucleus. They are probably the pathways of the absorbed materials from the supranuclear vacuole into the lateral cytoplasm. Usually, the inclusions of the supranuclear vacuole reveal the globes or coarse and sponge-like networks.

[Balck piedra in chimpanzees from Zaire].

Black piedra was present in 21 of 72 museum pelts of chimpanzees (Pan paniscus and Pan satyrus) from Zaïre. In the absence of culture, the mycological characteristics observed in vivo, although insufficient to allow a definite identification of the Piedraia species involved, are almost identical with those of Piedraia quintanilhae Van Uden, De Barros-Machado & Castelo-Branco, 1963.
