Pharmacological studies on a new 1-benzoyloxy-alkylaminocycloalkane derivative (YAU-17) with special reference to its mucosa anesthetic activity.

A new compound, dl-cis-1-benzoyloxy-2-dimethylamino-1,2,3,4-tetrahydronaphthalane (YAU-17) was found to produce sciatic nerve block, corneal and intradermal anesthesia in guinea-pigs, which markedly exceeded the effect of procaine and lidocaine. YAU-17, when given orally to mice, was much less toxic than any other drugs tested, though YAU-17 showed a considerably high acute toxicity by i.v. route. Oral administration of YAU-17 was effective in antagonizing the emetic response of dogs to intragastric phenylalanine isopropyl ester, but produced no distinct inhibition against vomiting elicited by oral copper sulfate. Furthermore, it possessed a spasmolytic activity which was comparable to that of papaverine, and showed an effect of suppressing the mucosal intrinsic reflex in the dog intestine. These pharmacological properties of YAU-17 were qualitatively similar to those of oxetacaine which was used as one of the reference compounds.

Hydrogenated benzo[f]quinolines and benz[e]indoles as analgetics. 1.

A series of octahydrobenzo[f]quinolines(IIIa) and hexahydro-1H-benz[e]indoles(IIIb), rigid structures related to 3-phenylpiperidine and pyrrolidine analgetics, has been synthesized. Structure-activity relationships were investigated by varying the structural parameters including a change in the stereochemistry of the ring junction. Several of the resulting compounds had analgetic activity on the order of meperidine.

Fine structure of taste buds in the rat.

The taste buds of rat circumvallate papillae contain three distinct types of cells. The type I (dark) cell is characterized by the presence of dense round granules, which are precursor to the dense substance of the taste pore. The granules are discharged into the pore by exocytosis. The type II (light) cell is filled with numerous vesicles and smooth-surfaced endoplasmic reticulum. The type III cell contains in its basal cytoplasm characteristic dark-cored vesicles and masses of clear vesicles, and makes synapse-like contacts with nerve fibers. The fine structure of foliate buds corresponds to that of circumvallate papillae, while fungiform buds differ in their apical regions. In the latter the pore is filled with vesicles alone, and the type I cell contains rod-shaped granules of moderate density. When polysaccharides were examined by means of the periodic acid-silver methenamine and the periodic acid-thiocarbohydrazide methods, slightly positive reactions were found on the dense granules of the type I cell and the dense substance in the pore, whereas the membranes of the pore vesicles, apical cytoplasmic processes and cytoplasmic vesicles in type II cells showed intense reactions.

Comparative study of methods for the extraction of eleven organophosphorus pesticide residues in rice.

Several extraction methods are compared for the simultaneous analysis of organophosphorus pesticides in unpolished rice. Four stationary phases were used for the subsequent gas-liquid chromatographic (GLC) determination of the selected pesticides. Using 3 different GLC columns, 11 pesticides were completely separated and identified. The efficiency of the cleanup and the sensitivity of the analytical method were evaluated by using powdered unpolished rice samples fortified with the pesticides and also wheat and dried bean samples. Average recoveries ranged from 74.7% for disulfoton to 97.4% for malathion in unpolished rice and from 68.1% for disulfoton to 108.3% for malathion in other crops. The method described is applicable to the analysis of selected organophosphorus pesticide residues in unpolished rice, wheat, buckwheat, and dried beans.

A new colimetric assay for monoamine oxidase in serum and its clinical application.

A new colimetric assay for serum monoamine oxidase and clinical findings based on this method are described. This rapid, precise procedure employs 1-[(4-aminomethylphenyl)azol]-2-naphthol as a substrate. The enzymatically generated 1-[(4-formyl phenyl)azo]-2-naphthol is extracted into cyclohexane to form a colored solution whose absorbance is determined at 500 nm. The method proved to be extremely simple, and the results obtained correlated well with those given by the standard method of McEwen.
