Sublocalisation of the X breakpoint in the translocation (X; 18)(p11.2; q11.2) primary change in synovial sarcomas.

A specific translocation between chromosomes X and 18 was identified in synovial sarcomas. From a girl with synovial sarcoma, we isolated two clones with t(X; 18)(p11.2; q11.2) and which had lost the normal X chromosome. Southern blot analysis of DNA from the tumor, the patient and her parents demonstrated that the normal X chromosome, lost in the tumor, was the paternal one. A somatic hybrid cell line was established by fusing tumor cells (after passages on athymic mice) to an HPRT deficient hamster cell line. By cytogenetic, in situ hybridization and molecular analysis, it was found to contain the derivative (X) chromosome in the absence of the der (18) chromosome. To determine the position of the breakpoint on the X chromosome, Southern blots of DNA from this hybrid were hybridized to [32P]-labelled X chromosome probes. DXS146 and DXS255 were retained in the hybrid cell line whereas GAPDP1, the ARAF1 and TIMP proto-oncogenes were not present, indicating that the breakpoint lies proximal to GAPD1, ARAF1 and TIMP and distal to DXS255 and DXS146. Results obtained from other authors are compared. Further studies will be necessary to determine the extent of variation of the breakpoint in different tumors.

Heterogeneous distribution of nuclear triiodothyronine receptors in liver and preadipose cells as evidenced by their reactivity to antibodies against different protein products of erb A oncogenes.

Polyclonal antibodies raised in rabbits against bacterially produced peptides in the C-terminal region of v-erb A or human c-erb A alpha oncogenes recognize the nuclear triiodothyronine (T3) receptors in the T3-sensitive Ob 17 mouse preadipocyte cell line and not in mouse or rat liver. The results confirm the existence of different T3 receptors in different tissues. The results also suggest a heterogeneous receptor distribution within the preadipose cell line, with a predominance of c-erb A alpha-type species. Antibodies raised against domain 149 227, but not against domain 245-325, impair T3 binding, suggesting a role for this domain in ligand binding.

[Variability of blood pressure measured using an oscillometric method in children 4 to 48 months of age].

The usual method of measuring arterial blood pressure cannot be used in children less than 4 years of age because of lack of cooperation. We investigated variations of blood pressure measured during 20 minutes by an oscillometric method (Dinamap) in 100 children aged 4 to 48 months and tried to come up with some recommendations for measuring and interpreting blood pressure. Serial blood pressure recordings showed poor reproducibility due to movements which are impossible to avoid and which account for wide variations. Because of this variability, a single measurement is inadequate and must be repeated. The average of repeated recordings does not seem to be a reliable parameter. We suggest that the lower measured value (minima) as the only parameter allowing us to track the progressive increase in blood pressure with age. The data probably explain the large discrepancies between published studies taking into account the use of different methods.

[The role of cellular immunity in gingival inflammation through a study of the cells of Langerhans].

The purpose of this semiquantitative study was to evaluate the number of Langerhans cells in correlation with gingival inflammation and the use of two antisera: anti-T6 and anti-HLA/DR reacting with two different membrane antigens. Results indicate that HLA/DR is a better functional marker, and that the number of Langerhans cells is higher in the moderate inflammation of the gingiva (G.I. 1 to 2).

[Experimental study of a metronidazole gel inserted in periodontal pockets].

The aim of this study was to evaluate the effects of a gel containing 4% metronidazole and collagen in periodontal pockets deeper than 4 mm. The treatment was utilized in 10 subjects with pocketing in the premolar-molar regions. The experimental design was as follows: one quadrant was treated with scaling and root planning (S), one with the metronidazole gel (M), one with scaling, root planning and metronidazole gel (S & M), and the last quadrant remained untreated. Pocket depths, gingival index (GI), sulcular bleeding index (SBI) and plaque indices were recorded at 7, 14 and 30 days after the experiment began. Significant improvement of the parameters studied was noted in the S & M quadrants.

Quantification of T6+ and HLA/DR+ Langerhans cells in normal and inflamed human gingiva.

Langerhans cells are dendritic non epithelial cells found in the gingiva. The latest research in immunohistochemistry suggests that they may play an important role in the cellular immune response. The purpose of this quantitative study was to evaluate the number of Langerhans cells in association with gingival inflammation and the use of monoclonal antibodies reacting with two different antigens: T6 and HLA-DR, present on the cell membrane. Results indicate that Langerhans cells are mostly T6+; but HLA-DR appears as a good functional marker. Langerhans cells may play an important role in the cellular immune response to bacterial antigens in moderate inflammation of the gingiva.

Echo-histological "Acino-Ductal Analysis". Preliminary results.

Echo-histological "Acino-Ductal Analysis" allows a display of lesions based on anatomo-physio-pathology, their micro-sampling for histological evaluation by interventional echography, and their direct visual observation by intra-ductal echo-guided endoscopy. This combination complements other means of investigation and shows an unequaled power of diagnosis by using the two most efficient means of diagnosis for malignancies: detection of cellular abnormalities and observance of evolution over time.

A new concept in breast investigation: echo-histological acino-ductal analysis or analytic echography.

"Echo-histological acino-ductal analysis of the breast" or "analytic echography" allows a display of lesions based on anatomo-physio-pathology, their micro-sampling by interventional echography for histological evaluation and their direct visual observation by intra-ductal echo-guided endoscopy. This method efficiently complements the other means of investigation by evaluating mammographic densities and palpable masses, and shows maximum diagnostic efficiency by improving and using the 2 most efficient means of diagnosis for malignancies, namely: the immediate detection of cellular abnormalities by more precise sampling; or the display of tumor evolution in a shorter time (3 months), with repeated sampling.

Haematological and hormonal responses to dynamic exercise in patients with chronic airway obstruction.

The influence of exercise on hormonal and total white blood cells (WBC), lymphocytes (L). Granulocytes (GR), and platelet (P) count responses was studied in: twenty-five patients with chronic airway obstruction (CAO, 47 +/- 1.8 years, mean +/- SEM) and thirteen normal subjects (N, 36 +/- 2.6 years). They performed a submaximal (40 W) and a maximal exercise (VO2max). Arterial blood samples were taken at rest, 40 W, and VO2max. [H+], PaCO2, PaO2 haematocrit (Hct), [Hb], P, total platelet volume (TPV), WBC, GR, L, and total red blood cells (RBC) were measured. At rest, WBC, GR, P and TPV were higher in CAO patients, whilst PaO2 and cortisol were lower. At 40 W, when compared to values obtained at rest, WBC, GR, L, P and TPV were increased in both groups; WBC, GR, P and TPV were higher in CAO patients. VO2max of CAO patients represented 54% of that of controls. At VO2max, Hct, [Hb] and RBC were approximately 10% higher than at rest in both groups, whilst changes were more significant in normals for WBC (CAO = 55%, N = 76%), lymphocytes (CAO = 83%, N = 105%), GR, (CAO = 37%; N = 51%), platelets (CAO = 23%, N = 29%), TPV (CAO = 25.4%, N = 35%), [H+] (CAO = 43%, N = 38%) and ACTH (CAO = 82%, N = 139%). PaO2 and cortisol did not differ between groups. PaCO2 and platelets however, were higher in the CAO group.(ABSTRACT TRUNCATED AT 250 WORDS)

Effects of high-fat diet, angiotensinogen (agt) gene inactivation, and targeted expression to adipose tissue on lipid metabolism and renal gene expression.

To address the role of angiotensinogen (agt) in lipid metabolism and its potential endocrine effects in vivo, we studied the effects of high-fat diet (HFD) on adult, 28-week-old agt knockout (KO) mice compared to wild type (WT) mice. Recent studies (Massiera et al., 2001) have demonstrated that reexpression of agt in adipose tissue of KO mice normalized adiposity, blood pressure, and kidney abnormalities. We therefore used microarray analysis to investigate changes in gene expression profile in kidneys of KO vs. Tg-KO mice, where agt expression is restricted to adipose tissue. Body weight, adiposity and insulin levels were significantly decreased (p < 0.05) in KO mice on a chow diet (CD) compared to WT mice, while circulating leptin levels were similar. On a high-fat diet, KO mice exhibited significantly lower bodyweight (p < 0.05), adiposity (p < 0.05), leptin, and insulin levels (p < 0.05) compared to WT mice. In agreement with previously reported changes in kidney histology, agt KO mice displayed altered expressions of genes involved in blood pressure regulation and renal function, but these levels were corrected by reexpression of agt in adipose tissue. Collectively, these findings further document important endocrine roles of adipocyte agt, in part via regulation of lipid metabolism and kidney homeostasis.
