Glucose-sucrose-potassium tellurite-bacitracin agar, an alternative to mitis salivarius-bacitracin agar for enumeration of Streptococcus mutans.

An agar medium for selective recovery and enumeration of Streptococcus mutans was developed as an alternative to mitis salivarius-bacitracin (MSB) agar. Combinations of dyes, antibiotics, and tellurite were added to a nonselective medium which, because of its sucrose content, allowed easy recognition of S. mutans colonies. Candle jar incubation for 2 days, by comparison with anaerobic incubation, reduced background flora but did not diminish S. mutans recoveries from clinical samples. Quantitative comparisons were made of the simultaneous recoveries of a number of authentic S. mutans serotype representatives and fresh clinical isolates, using various glucose-sucrose-potassium tellurite-bacitracin (GSTB) formulations and mitis salivarius, MSB, and blood agars. Mitis salivarius counts were not detectably different from blood counts, but counts on MSB were distinctly lower. A formulation of the new medium containing 5% glucose 5% sucrose, 0.001% potassium tellurite, 0.3 U of bacitracin per ml (hence GSTB), and 2% agar gave recoveries nearly equal to those on mitis salivarius agar and much greater than those on MSB. The medium yielded readily recognized S. mutans colonies and facilitated detection of intracellular polysaccharide formers upon flooding with I2 reagent. Freshly isolated serotype c, E, and f colonies could often be distinguished from serotype d and g colonies, a distinction made reliable by testing for intracellular polysaccharide. A study of 300 salivary samples revealed GSTB to give significantly higher recoveries than MSB. About 72% of all samples were substantially underestimated for S. mutans with MSB, and 6.7% of samples were falsely negative for S. mutans with MSB. Recovery of background flora on GSTB was as low or lower than on MSB, and both types of agar could be stored for at least 9 weeks without notable change of selectivity. Thus, GSTB agar appears to be simple and reliable to use and requires no anaerobic incubation. Caution is voiced about interpretation of data previously reported which evaluated S. mutans on MSB agar.

[Readjustment of peripheral factors during the unsteady-state phase of dynamic muscular exercise].

The behaviour of the ratio: oxygen extraction (total peripheral resistance (DAV/TPR) in a group of untrained healthy young male students has been bloodless investigated during the unsteady-state phase of the dynamic muscular exercise (2.5 watt/Kg). The results obtained have shown that the kinetics of the ratio, from a resting value of 3.40 ml O2 . min-1 . mmHg-1 to a figure of 17.62 (steady-state) doesn't follow a step function. During the first 30 seconds the percentual increase of the ratio falls to 0.45, becomes 1.5 thirty seconds later and there this exponentially drops down to a value of 1. The observed behaviour agrees with experimental findings of other authors.

[Changes in hemodynamic and metabolic parameters during controlled muscular exertion in a group of international-level professional cyclists].

The behaviour of some cardiovascular and metabolic parameters have been studied in 11 competitive road-cyclists under controlled submaximal work load. The test used was intermittent with load increasing from 100 to 250 watt, independent from age and body weight of the athletes. The results obtained have shown that whereas the estimated VO2 was 4,76 LO2/min, the extrapolated VO2max goes up to 6,08 LO2/min, corresponding to 84,55 m102/Kg X min. Particularly interesting seems to be the way through which the high VO2 levels can be obtained in these subjects: small increment of the mean arterial blood pressure together with large oxygen extraction and high vasodilatation.

Prognostic value of the electroencephalogram in neonatal asphyxia.

In order to determine whether an EEG early in the course of asphyxia neonatorum is of any more value than the neurological examination in predicting outcome we reviewed case histories of 38 infants with asphyxia neonatorum. The EEG background activity was valuable in predicting outcome. Normal and maturationally delayed EEGs were associated with normal outcomes while low voltage, electrocerebral inactivity and burst suppression EEGs were highly correlated with severe neurological sequelae. Epileptiform activity was not as predictive of outcome as background activity. Although initial normal neurological examinations were associated with normal developmental and neurological outcomes, moderately and severely abnormal infants had more variable courses. A single EEG done early in the course of asphyxia neonatorum is a more sensitive predictor of outcome than the neurological examination.

[A new method for determination of ATPase activity in tissue homogenates].

Authors describe a new method to determine Mg2+ and Na+K+ATPase activity on extremely small samples of tissue homogenates. They use specific inhibitors to discern between the two activities and a sensitive colorimetric method to dose Pi released from ATP, Lower limit of sensitivity is about 0,0100 micromoles of Pi/mg proteins/hour.
