Transcription in methanogens. Evidence for specific in vitro transcription of the purified DNA-dependent RNA polymerase of Methanococcus thermolithotrophicus.

The purification of the DNA-dependent RNA polymerase of Methanococcus thermolithotrophicus is described. As the first step of purification the endogenous template was removed from the enzyme by hydrophobic interaction chromatography. The purified enzyme consists of seven components with different molecular masses. Transcription studies on T7 DNA and the recombinant plasmid pMV15, containing rRNA genes of Methanococcus vannielii, revealed that only the methanogen DNA is transcribed specifically, indicating a principal structural difference between archaebacterial and eubacterial promoters. This could be shown both by analysis of ternary transcription complexes and Southern hybridization. The site of initiation was found within a restriction fragment harbouring the first 390 nucleotides of the sequence coding for mature 16S rRNA and 1100 base pairs of upstream sequences. The specific initiation on this fragment strongly suggests that the enzyme can start in vitro transcription from the promoter(s) of rRNA synthesis.

Three new restriction endonucleases MaeI, MaeII and MaeIII from Methanococcus aeolicus.

Three type II restriction endonucleases, MaeI, MaeII and MaeIII, with novel site specificities have been isolated and purified from the archaebacterium Methanococcus aeolicus PL-15/H. The recognition sequences of these enzymes are (formula: see text) with the sites of cleavage as indicated by the arrows. The sequences were confirmed by restriction and computer analyses on sequenced DNA's of plasmid pBR322, bacteriophages lambda and phi X174 and virus SV40.

Structural homology between different archaebacterial DNA-dependent RNA polymerases analyzed by immunological comparison of their components.

The archaebacterial DNA-dependent RNA polymerases have a complex structure containing eight or more components. Immunochemical analysis shows an extensive homology between the components of the enzymes of nine different species. Two enzyme subtypes can be distinguished: that of the thermoacidophilic and/or sulfur-metabolizing archaebacteria with the composition BACDEFGHIJ and that of the methanogenic plus halophilic archaebacteria with the composition ABB'C(D).... Components B and B' of the latter subtype probably evolved by the division of the large component B of the BACD... type enzyme. The existence of the two subtypes corroborates the division of the archaebacteria into two phylogenetic main branches.

Evidence for a plasmid in a methanogenic bacterium.

Among 15 strains of methanogens, one plasmid, pMP1, was identified in the new coccoid isolate PL-12/M. It could not be detected in the cleared lysate, but it was detected in the viscous pellet. The plasmid had a molecular weight of ca. 4.6 x 10(6). A restriction enzyme cleavage map of the cloned plasmid was derived.
