[Carnosine as a stimulator of cytotoxic and phagocytic function of peritoneal macrophages].

Biochemical changes in peritoneal macrophages and their relatedness to the cytostatic and phagocytotic function in C3HA mice injected with a single intraperitoneal dose of 0.45 mM carnosine and 4-methyluracil or stimulated with peptone have been studied. During the first 24 hours after injection both carnosine and 4-methyluracil increase the activity of adenosine deaminase and purine nucleoside phosphorylase, the key enzymes of purine catabolism which is the main source of O2-. radicals in macrophages. In carnosine-stimulated macrophages the activity of membrane 5'-AMP nucleotidase decreases on days 1-3 after injection which points to alleviation of adenosine-induced inhibition as well as to macrophage activation. Carnosine increases the cytostatic and phagocytotic activities of macrophage coupled to O2-. production. The mechanism of the stimulating effect of carnosine on macrophages seems to consist in the dipeptide interaction with specific receptors localized on the plasma membrane of macrophagal cells.

[Effect of carnosine and 4-methyluracil on the development of experimental hepatitis in rats].

A comparative study of the hepatoprotective effect of carnosine and 4-methyluracil under CCl4-induced acute toxic hepatitis has been carried out. The extent of liver injury and its regeneration were established from morphological data as well as from changes in the activities of alanine aminotransferase (ALT) and histidase and the bilirubin content in blood serum. Hyperlipoperoxidation in the liver and serum was assessed by the amount of TBA-active products. It was found that by day 10 of experimental hepatitis ALT and histidase levels in blood sera of untreated animals exceeded the normal values 1.3- and 3.9-fold, whereas those in the carnosine-treated group approximated the values characteristic of intact animals. The activity of serum ALT in animals treated with vitamin B12 or 4-methyluracil exceeded normal values 1.5 and 1.6 times, whereas that of histidase was 2.5 and 2.7 times as high. Carnosine and 4-methyluracil inhibited (in approximately the same degree) the formation of TBA-active products in the liver. According to morphological dta, cessation of CCl4 injections was accompanied by rapid regeneration of liver tissues in all animal groups. Carnosine enhanced regenerative processes in parenchymatous and connective tissues in a far greater degree in comparison with other drugs. The mitotic index in the carnosine-treated group exceeded more than twofold the corresponding parameters in untreated animals. Possible mechanisms of carnosine action on liver repair are discussed.

[Effects of 4-methyluracil and carnosine on healing of skin wounds in rats].

In experiments in vivo and in vitro the authors studied antioxidative properties of 4-methyluracil and carnosine, their capacity to inhibit sex and accelerate healing of skin wounds. 4-methyluracil and carnosine discover almost the same capacity to decrease in the tissues of the wound and the blood serum in the formation of various intermediate products of free radical oxidation. Data are given on the study of the dynamics of wound healing after a 5-day treatment with equimolar quantities.

[The delayed effects of systemic circulatory arrest on the free-radical processes in the brain structures of rats with different types of behavior in the emotional resonance test].

Male Wistar rats with different types of behavior in "emotional resonance" test ("active" and "passive") were studied one week after the global ischemia induced by cardiac arrest. Recovery of some physiological functions as well as free-radical-mediated processes and NO-synthase activity were studied in cerebral structures and blood serum. The "open-field" behavior normalized more rapidly in the "active" rats than in the "passive" ones, though the time course of the neurologic deficit compensation did not differ in these groups. A decrease in superoxide scavenging activity and in the content of 2-thiobarbituric acid-reactive material was revealed in the cerebral structures of both "active" and "passive" rats. Increased levels of free-radical generation in the hippocampus of the "passive" rats and in the cerebellum of the "active" rats were found. Higher NO-synthase activity was demonstrated in the cerebellum of the "passive" rats. Taken together, these data suggest that there are specific patterns of free-radical-mediated processes in the brain of rats with different types of behavior in "emotional resonance" test.

[The indices of free-radical oxidation in the umbilical cord blood and placenta of inhabitants of the Altai Territory].

Free-radical processes were studied in the umbilical blood and placenta of women from the regions of the Altai Territory, which were affected to different extents by nuclear tests on the Semipalatinsk grounds in 1949-1965. The data was obtained, which suggest changes of free-radical processes, from studied materials from women in labor in the regions most affected by the consequences of tests. The activity of erythrocytic superoxide dismutase was decreased, thus suggesting the formation of structural-functional defects of the erythrocytes. The data corresponds to the results obtained earlier when studying free-radical processes in the venous blood samples from female residents of the Altai Territory.

[Administration of aggregated beta-amyloid peptide (25-35) causes change in long-term potentiation in the hippocampus].

A month after intracerebroventricular injection of aggregated beta-amyloid fragment 25-35 (7.5 nmol/ventricle), a significant change in in vivo long-term potentiation in hippocampus was demonstrated. The time course of the long-term potentiation was compared with that in sham-operated animals, a powerful and stable increase in the evoked potential amplitude was observed. This phenomenon can be related with the oxidative stress that was revealed in this model in our previous studies, and, as a consequence, with deterioration of ion homeostasis.

[Effects of morphine withdrawal on the indices of free radical homeostasis and nitric oxide system in rat liver and thymus].

Indices of oxidative stress, nitric oxide (NO) metabolism as well as the activity of caspase-3, an important enzyme of apoptotic cell death, were measured during the morphine withdrawal syndrome in liver and thymus of rats. Male Wistar rats were administered with morphine hydrochloride (i.p., at increasing doses from 10 to 100 mg/kg, twice a day, for 6 days). Thirty-six hours after the last administration the withdrawal syndrome was monitored using the specific autonomic and locomotor indices. During this period, weights of body and thymus significantly decreased. Oxidative stress in liver was accompanied by an increase in aspartate aminotransferase and gamma-glutamyl transferase in blood serum. No signs of oxidative stress could be demonstrated in thymus. The activity of the Ca(2+)-dependent isoform of nitric oxide synthase (NOS) in liver increased, while, the activity of the Ca(2+)-independent NOS diminished, the total activity of NOS in liver and thymus remained unchanged. The concentration of nitrates/nitrites in blood was decreased, in thymus increased, and in liver unchanged. Caspase-3 activity changed neither in liver, nor in thymus. The results are discussed from the perspective of possible antioxidant and antiapoptotic role of NO during morphine withdrawal syndrome.

[Nitric oxide synthase activity and nitrates/nitrites level in brain regions during the spontaneous morphine withdrawal in rats].

Nitric oxide synthase (NOS) activity and nitrate/nitrites (NO(x)-) concentrations were measured in brain regions of rats during the spontaneous morphine withdrawal. Male Wistar rats were injected intraperotoneally with morphine hydrochloride at increasing doses (10-100 mg/kg) during 6 days twice a day. Thirty six hours after the last injection the severity of the spontaneous morphine withdrawal syndrome was determined by specific autonomic and locomotor indices. Both NOS activity and NO(x)- levels increased in the midbrain and the hippocampus, decreased in the striatum and the hypothalamus, and did not change in the cerebral cortex and the brain stem. NO(x)- concentrations in the cerebellum did not change, while NOS activity decreased. Both NOS activities and NO(x)- concentrations in the cerebral cortex, striatum, midbrain, and cerebellum correlated with withdrawal syndrome severity on the whole, and with the specific signs of abstinence.

[Effects of sodium succinate on functional, biochemical and morphological indicators of CNS repair in rats after 10-minute circulation arrest].

Therapeutic effect of sodium succinate on various functional, biochemical, and morphological parameters of CNS repair was studied in experiments on rats exposed to 10-min circulation arrest. The first series of experiments was devoted to studies of the effects of the drug, injected intraperitoneally directly after recovery of effective cardiac activity and during the subsequent 5 days in doses 20, 100, and 200 mg/kg, on the survival and recovery of the external neurologic status. The dose of 20 mg/kg proved to be the most effective. The second series of experiments was devoted to therapeutic effect of sodium succinate in the same dose injected from day 3 to day 7 after revival on the orientation and investigation behaviour in an "open field" test, on changes in radical formation in the blood serum and the brain, and on the cholesterol/lipid ratio in the brain, as well as on the morphologic changes in the cerebral hemispheres and the cerebellum. In contrast to untreated animals, the treated ones had a less intensive reaction in the "open field" test in response to acute stressor exposure, their cerebral and blood serum levels of free radical processes were reduced, the destruction of neuronal membranous elements was less intensive, as were dystrophic changes in the cerebral cortex and the cerebellum. The data permit a conclusion about antistressor and protective effect of sodium succinate in the postresuscitation period at the functional, biochemical, and morphological levels.

[The neurochemical characteristics of rats differing by their behavior in the emotional resonance test. Free-radical processes and lipids of the cerebral cortex in older rats].

Free radical processes and lipid content in brain membranes were investigated in old male Wistar rats with differences in individual behaviour in "emotional resonance" test. Two groups of rats were selected, i.e. 1--which stayed for more than 2/3 of the total test time in the dark part of the experimental chamber and 2--which spent the total test time in the light part of the chamber. Free radical processes and lipid content in the cortex of the left and right brain hemispheres did not differ in control animals of groups 1 and 2. Acute immobilization stress induced changes of the studied parameters depending on behavioural type. Rats of group 2 were more reactive than those of group 1.

[The binding of [3H]aldosterone by brain corticosteroid receptors in rats with different individual-typological behavioral characteristics].

[3H]Aldosterone (AS) binding to cytoplasmic corticosteroid receptors (CR) of the hippocampus and brain (without cerebellum and hippocampus) was studied in rats of two behavioral types under normal conditions and within two weeks after electric-pain stress. No asymmetry was shown in the distribution of CRs that bound [3H]AS in the rats of both groups. A relationship was established between the hormone binding by CRs of the hippocampus, rather than the rest of the brain, and type of animal behavior. The action of the stress factor led to decreased binding of [3H]AS to CRs of both hippocampus and brain. Thus, a relationship between the state of CRs of the hippocampus and individual behavior of animals was first established. The results obtained agree with the published data on relationship between the emotional reactivity of the animals and state of CRs in the limbic structures of the brain.

[The effect of succinic acid on postresuscitation pathology of the CNS and of the body as a whole].

The effect of oral succinic acid was studied in rats exposed to 10-min heart arrest followed by resuscitation. The drug was administered for 5 days in a dose of 30 mg/kg starting from day 3 up to day 7 after resuscitation. Succinic acid was found to normalize the orientation and exploration behavior of rats in the "open field" test, decreased the intensity of response to stress (electric shock), and normalized the radical formation in the brain tissue and blood serum, thus reducing the morphological changes in the brain. In addition, succinic acid prevented the development of risk factors of atherogenesis, namely, increase of the levels of blood cholesterol, triglycerides, and low and very low density lipoproteins. Further studies are needed to validate the addition of succinic acid to the armory of drugs preventing the development of postresuscitation encephalopathies in remote (3 months) periods.

The effect of systemic circulatory arrest on the process of free-radical oxidation and on NO-synthase activity in the brain structures of rats of different behavioral types in an emotional resonance test: the acute effects.

Free radical-mediated processes and NO-synthase activity were studied in cerebral structures and blood serum of male Wistar rats with different types of behavior in emotional resonance test one hour after global ischemia induced by cardiac arrest. Oxidative stress accompanied by loss in NO-synthase activity was revealed in cerebral cortex after the ischemia. The oxidative stress was also evident in cerebellum and to a lesser extent in hippocampus. The majority of behavior-related biochemical differences were induced by cardiac arrest. These differences could be global or related to specific brain structures. Sometimes they became apparent in cerebral lateralization of biochemical indices.

[Parameters of free radical oxidation in blood plasma of women from the Altaĭ region].

We studied the indices of free radical-mediated processes in blood plasma of women from the Altai district affected by the nuclear weapon tests at the Semipalatinsk ground in 1949-1962 and 1965. The data obtained suggest modifications of free radical-mediated processes in inhabitants of the regions most affected by the tests. A significant decrease in the activity of erythrocytic superoxide dismutase indicates structural and functional disturbances of the erythrocytes.
