[Basal lipid energy utilization in obese women and men].

In 16 abdominal type obese males, in 16 gluteal-femoral type obese females and in 10 nonobese males and 9 nonobese females, as controls, the influence of body fat distribution on basal lipid oxidation was evaluated by indirect calorimetry. Basal lipid oxidation was significantly higher in the obese males (1.21 +/- 0.12 g/kg-FFM.die) than in the nonobese males (0.47 +/- 0.13 g/kg-FFM.die; p less than 0.001), but not significantly different between obese and nonobese females (0.98 +/- 0.16 and 0.89 +/- 0.13 g/kg-FFM.die; p ns). A significant correlation between basal lipid oxidation and percentage of body fat was found only in the male subjects (r = 0.51; p less than 0.01). These results suggest that basal lipid oxidation differs in the two sexes according to adipose tissue anatomic location.

The inhibitory effect of dithiothreitol on the assembly of the filamentous phage fd.

Assembly of the filamentous phage fd is preceded by the formation of a complex between the viral single-stranded (ss) DNA and the virally coded gene 5 protein (gene 5 protein-ssDNA complex). The presence of 5 mM dithiothreitol in the growth medium prevents phage production; however, phage infection, phage DNA replication and phage genome expression are still observed. In contrast, the gene 5 protein-ssDNA complex is not formed in the presence of dithiothreitol in vivo, although the complex is not affected by the disulfide reducing agent in vitro. Furthermore, host lipid composition is altered by growth in the presence of dithiothreitol. The zwitterionic lipid, phosphatidylethanolamine, increases while the cationic phospholipid content, cardiolipin and phosphatidylglycerol, decreases. This suggests a role for lipids or membranous structures in the process of gene 5 protein-ssDNA complex formation.

The effects of muscular exercise on glucose, free fatty acids, alanine and lactate in type I diabetic subjects in relation to metabolic control.

Metabolic effects of muscular exercise were studied in eleven subjects with type I diabetes mellitus during poor metabolic control, and again during good metabolic control, and in ten healthy control subjects. All the subjects were submitted to a submaximal gradual triangular test on an electrically braked bicycle ergometer; glucose, FFA, alanine and lactate were measured at rest, and after exercise. In poorly controlled patients, glucose and FFA were unchanged after exercise, whereas blood alanine and lactate increased by a percentage similar to that of the controls, and well-controlled diabetic patients. Baseline alanine concentrations were lower and lactate concentrations higher than in the controls and well-controlled patients. After adequate metabolic control was achieved, in the well-controlled diabetic patients a normalization of pre-exercise alanine and lactate levels and a decrease in blood glucose and FFA after exercise was observed.

The dermoepidermal junction in psoriatic skin as revealed by scanning electron microscopy.

Our previous ultrastructural and immunohistochemical studies, in vivo and in vitro, have shown important modifications of the basement membrane of psoriatic skin, which could play a key role in the alterations of keratinocyte adhesion, migration, proliferation and differentiation. In order to complete the morphological examination of all the structures in the dermoepidermal junction of psoriatic skin, we carried out a scanning electron microscopic study using biopsies taken from eight psoriatic patients. The biopsies were fixed in a mixture of 0.2% paraformaldehyde and 0.25% glutaraldehyde in 0.1 M cacodylate buffer at pH 7.4. The specimens were then frozen in liquid nitrogen and fractured following the natural cleavage planes and observed under a Jeol JSM-6301F field emission scanning electron microscope operating at 1.8-2.0 kV. The basal keratinocytes observed showed pore-like depressions on the lateral plasmalemma and villous-like projections in very dilated intercellular spaces. Moreover the basal cell plasma membrane was seen to rest on the papillary dermis without interposition of the lamina densa. The detachment of some keratinocytes enabled the examination of the lamina densa, which appeared slightly granular with numerous focal interruptions through which it was possible to observe the underlying collagen fibres. These findings, together with previously reported findings, support the hypothesis that in psoriasis molecular and structural alterations of the dermoepidermal junction are present, that could fundamentally alter the regulation of the cytomorphological processes and the normal functions of the basement membrane.

Phototrichogram using videomicroscopy: a useful technique in the evaluation of scalp hair.

The purpose of our work is to contribute to the definitive standardization of the phototrichogram in view of the new possibilities offered by information technology. We placed under investigation a 1 cm2 scalp area on the vertex of 28 healthy volunteers (20 women and 8 men) between the ages of 18 and 30 (average age was 23.32 +/- 3.48 years). We performed the examination using a computerised videomicroscope equipped with lenses x 20 and x 50. The results obtained can be summarized as follows: total hair density was 260 +/- 30/cm2 in men and 300 +/- 20/cm2 in women; telogen percentage was higher in men (16.5 +/- 1.5% vs 11 +/- 1% of women); thin hairs (< 0.040 mm) were 9.4 +/- 1.8% in men and 9 +/- 2% in women; growth rate was 0.35 +/- 0.03 mm/day in men and 0.38 +/- 0.03mm/day in women. Thanks to its integration with digital imaging, the phototrichogram has undoubtedly become an examination of primary importance in trichology because of its simplicity and sensitivity even if it cannot yet be considered the ideal technique to replace all the others.

[Combined fetal alcohol and hydantoin syndromes].

Ethanol and hydantoin are both teratogenic drugs of common usage and are associated with specific syndromes. This is a report on two children born to different heavy drinking, mentally abnormal epileptic women, that were also under treatment with 300 mg/day of hydantoin (mother of case 1 on a regular basis and that of case 2 sporadically) during pregnancy. Both infants displayed the syndrome due to both of these drugs. Particularly relevant were failure to thrive, severe mental retardation, microcephaly, blepharophimosis, hypertelorism, and long philtrum. Additional findings in case 1 included nail hypoplasia and in case 2 palatine fissure. Case 1 died unexpectedly at age three months. Case 2 is still alive and he is severely mentally retarded at age four years.
