Journey length and high temperatures: effects on rabbit welfare and meat quality.

The transport of domestic animals by road can increase levels of stress and decrease meat quality, especially in unfavourable climates. The purpose of this study was to investigate the effect of journey duration and vertical position on the transport truck on some physiological indicators of stress and on instrumental meat quality parameters in commercial rabbits. In the summer months (June and July, 2003), 78 rabbits were subjected to either long (7 h) (LJ) or short (1 h) (SJ) journeys (3 replicates each, n = 6) between the farm and an abattoir in northern Spain. The position (top, middle, or bottom) occupied by the rabbits on the Multi-Floor cage Rolling Stand (MFRS) of the transport truck was recorded. Blood samples were collected at sticking and meat pH was measured at 24 h post-mortem (pH24). At 48 h post-mortem, samples of the M. longissimus dorsi were used to determine water-holding capacity (WHC) and instrumental tenderness using an INSTRON machine. The levels of corticosterone, glucose, lactate, and creatine kinase were slightly higher in LJ than in SJ samples, but the difference was not statistically significant (p < or = 0.10). Independent of journey length, rabbits in the middle and bottom of the MFRS showed higher levels (p < or = 0.05) of glucose and creatine kinase (middle), and corticosterone (bottom) than the rabbits located at the top floor. The pH 24 and WHC values of the SJ and LJ rabbits did not differ significantly. Journey length had a significant effect (p < or = 0.01) on meat tenderness. The meat from rabbits subjected to LJ had higher values of maximum stress and total toughness than did the SJ rabbits (p < or = 0.05). There were similar differences (p < or = 0.05) between LJ and SJ rabbits in their stress values of compression at 20% and 40% (measures of the tenderness of raw meat). In hot weather, the position on the transport truck appeared to have a greater effect on rabbit welfare than the duration of the journey.

Oestrus synchronisation of rabbit does at early post-partum by doe-litter separation or ECG injection: Reproductive parameters and endocrine profiles.

Inseminating rabbit does at early post-partum, in combination with early weaning, can increase prolificacy (total kits born and still born per parturition) and decrease parturition intervals. Oestrus synchronisation increases fertility and prolificacy, while decreasing the number of inseminations required for gestation. However, little is known about the effectiveness of different oestrus synchronisation methods at early post-partum. In this study, does (n = 138) were artificially inseminated nine times (over a period of 1 year, kits weaned at 25 days), on day 4 post-partum after separation from the litter (for 48 or 24 h) or 48 h after 25 UI eCG injection. Plasma levels of prolactin and estradiol were also evaluated in a subsample of 12 multiparous lactating does per treatment, on days 2, 3 and 4 post-partum. The three treatments increased overall fertility of multiparous females compared to controls (which were not synchronised), but there were no differences among treatments in total kits born or stillborn. Does treated with eCG had a higher culling rate. The interval between parturitions and the number of inseminations required for gestation tended to decrease with increasing number of inseminations. In lactating does, there was an interaction between treatment and insemination order. Fertility decreased with increasing inseminations in eCG does but tended to increase above control values in the separated does until the fourth insemination. Control lactating does had significantly less kits per parturition compared to treatments, but eCG lactating does had more stillborn kits. Oestradiol levels increased on day 4 post-partum in all synchronised lactating does (and immediately before artificial insemination in 48 h doe-litter separation), so ovarian activity could be stimulated at early post-partum using all treatments. However, the increase could not be explained by prolactin levels, since there were no effects of suckling absence on plasma prolactin in separated does. In conclusion, separating does from the litter before insemination can be just as effective as eCG treatment, especially during for the first four inseminations.

Effect of transport stress on physiological responses of male bovines.

Forty-eight slaughter bulls were transported by road in groups of eight for approximately 30 min, 3 h and 6 h in two replicates. Animal welfare during the transport process was assessed. Loadings and unloadings were evaluated with a scoring method. Heart rates were monitored at the farm before loading and during all stages of transport. Blood samples were taken from all animals a week before transport and at sticking and analysed in terms of haematological values: hematocrit, haemoglobin, red and white blood cells (RBC and WBC), differential WBC counts and neutrophil:lymphocyte ratio. Glucose, creatine kinase, lactate and cortisol were also determined. To evaluate differences in meat quality, pH and water-holding capacity (WHC) were measured 24 h after slaughter. The loading and unloading scores were very low (low stress) but were associated with changes in heart rate, especially loading. Animals recovered their resting heart rate during the journey in medium and long transports. On the other hand, animals transported around 30 min maintained an elevated heart rate during the whole journey. All animals showed a stress response with significantly higher (p < 0.05) levels of erythrocyte series, N:L ratio, glucose and lactate. Animals transported for 3 and 6 hours had significantly (P<0.05) higher levels of cortisol than controls or 30 min transports, without differences between control and the shortest journey. Different transport times did not influence meat quality. Under good conditions, the transport had a slight effect on welfare, meat quality or physiological parameters related with stress.

Scoring system for evaluating the stress to cattle of commercial loading and unloading.

The loading and unloading of cattle for road transport is stressful but the stress is difficult to evaluate in terms of the welfare of the cattle. Over one year, 40 normal commercial journeys in northern Spain were analysed in terms of time limits and behavioural events in order to design an objective method for assessing the stresses imposed on the animals, and a scoring method was developed for assessing the welfare of the animals during loading and unloading. Several definitions of time intervals were assessed to calculate a time score per animal, and easily observable behavioural events were scored and combined with the time score to obtain a total loading/unloading score. More than half of the loadings and unloadings involved turns, slips and vocalisations. Mounts and bouts of fighting were infrequent and balks and falls were significantly more frequent during loading than unloading. The plasma concentrations of cortisol, glucose and lactate, the activity of creatine kinase and the pH of the meat 24 hours after the animals were slaughtered were also measured. The results indicated that loading was more stressful than unloading and that higher scores implied significantly higher levels of stress.

Effect of commercial transport in Spain on cattle welfare and meat quality.

The purpose of this work was to determine the effect of transport of cattle from the farm to the abattoir on the animal's welfare and post mortem meat quality (instrumental and sensorial). Thirty-nine commercial journeys were followed in Spain from July 2000 to July 2001 at varying distances from a local abattoir in the city of Zaragoza. Blood samples were taken at the farm and at sticking and meat pH24 was measured at 24 hours post-mortem. Samples from the M. longissimus dorsi thoracis were taken from 48 slaughter bulls and analysed in terms of instrumental and sensorial qualities. Cortisol, glucose, CPK, lactate and N:L ratio increased during transport compared to control values. Cortisol was higher after 1-2 h transport compared to journeys that were less than 1 h or more than 2 h long. In general the parameters indicated slightly more stressful conditions under winter conditions than in the summer. Meat tenderness measured by a Warner-Bratzler device demonstrated that there was no significant effect of either transport time or season. There was no significant change in pH24 or meat colour at 24 h with transport time. Transport time had a significant effect (p < 0.05) on overall liking and animals transported in summer had slightly more tender meat and higher overall liking.

Prospective evaluation of a model of prediction of invasive bacterial infection risk among children with cancer, fever, and neutropenia.

A risk prediction model for invasive bacterial infection (IBI) was prospectively evaluated among children presenting with cancer, fever, and neutropenia. The model incorporated assessment of 5 previously identified risk factors: serum level of C-reactive protein (CRP) >/=90 mg/L, hypotension, identification of relapse of leukemia as the cancer type, platelet count of </=50,000 platelets/mm(3), and recent receipt of chemotherapy [16]. Children were uniformly evaluated at enrollment and were classified as having high or low risk for IBI according to a model that considers the number and type of variables present. Of the 263 febrile episodes evaluated during a 17-month period, 140 (53%) were in IBI-positive children. The sensitivity, specificity, and positive and negative predictive values of the model were 92%, 76%, 82%, and 90%, respectively. Identification of these 5 risk factors during the first 24 h of hospitalization was helpful in discriminating between children with a high or low risk for IBI.

Critical points in the transport of cattle to slaughter in Spain that may compromise the animals' welfare.

The welfare of cattle depends greatly on the attitudes and training of stockpersons and on the availability of appropriate facilities. Much has been learned about stress during transport, but less attention has been paid to identifying and correcting critical points, partly because they vary widely both nationally and internationally. A survey of cattle transport in Spain was made in an effort to determine which parts of the process most compromised the animals' welfare. Data were collected on the methods and facilities for loading and unloading, transport times, types of vehicle and slaughterhouse practices. Loading facilities were adequate and loading times generally short but some farms continued to use an electric goad and weather-proofing was generally poor. The average journey time within Spain was three-and-a-half hours, but many trips were made abroad (especially to Italy), few drivers received specific training courses and the types and quality of vehicles varied widely. The average unloading time was very short but the animals were not always inspected for injuries or dirtiness. Lairage times were normally more than eight hours but few slaughterhouses had air-conditioning equipment to prevent excessive heat or dehydration. Almost all stockpersons avoided either regrouping animals or housing or transporting animals at high densities.

Prospective, multicenter evaluation of risk factors associated with invasive bacterial infection in children with cancer, neutropenia, and fever.

PURPOSE: To identify clinical and laboratory parameters present at the time of a first evaluation that could help predict which children with cancer, fever, and neutropenia were at high risk or low risk for an invasive bacterial infection. PATIENTS AND METHODS: Over a 17-month period, all children with cancer, fever, and neutropenia admitted to five hospitals in Santiago, Chile, were enrolled onto a prospective protocol. Associations between admission parameters and risk for invasive bacterial infection were assessed by univariate and logistic regression analyses. RESULTS: A total of 447 febrile neutropenic episodes occurred in 257 children. Five parameters were statistically independent risk factors for an invasive bacterial infection. Ranked by order of significance, they were as follows: C-reactive protein levels of 90 mg/L or higher (relative risk [RR], 4.2; 95% confidence interval [CI], 3.6 to 4.8); presence of hypotension (RR, 2.7; 95% CI, 2.3 to 3.2); relapse of leukemia as cancer type (RR, 1.8, 95% CI, 1.7 to 2.3); platelet count less than or equal to 50,000/mm(3) (RR, 1.7; 95% CI, 1.4 to 2.2); and recent (< or = 7 days) chemotherapy (RR, 1.3; 95% CI, 1.1 to 1.6). Other previously postulated risk factors (magnitude of fever, monocyte count) were not independent risk factors in this study population. CONCLUSION: In a large population of children, common clinical and laboratory admission parameters were identified that can help predict the risk for an invasive bacterial infection. These results encourage the possibility of a more selective management strategy for these children.

[Causative agents of bloodstream infections in children with neoplasm, in 5 hospitals of Santiago (1994-1998)].

BACKGROUND: Pediatric patients in treatment for cancer can have fatal bacterial infections. Thus, in the presence of fever or other signs infection, antimicrobials have to be prescribed empirically. AIM: To know the causative agents of bacteremia in children with cancer, their changes with time and between different hospitals and their patterns of susceptibility. MATERIAL AND METHODS: We reviewed the blood cultores of children with cancer in five hospitals of Santiago, from 1994 at 1998. RESULTS: During the study period, 707 agents were isolated. The most frequently isolated species or genus were coagulase negative Staphylococcus (43%), Staphylococcus aureus (16%), Escherichia coli (9%), Klebsiella spp. (8%), Pseudomonas spp. (5%) and Candida spp. (4%). Coagulase negative Staphylococcus was 55% resistant to meticilin and S. aureus was 44% resistant. Enterobacteriaceae had 15% resistance to gentamicin and amikacin, 2% to imipenem, 26% to ceftriaxone, 21% to cefotaxime and 20% to ceftazidime. Among non fermenting agents resistance was 6% for imipenem, 9% for amikacin 10% for ciprofloxacin, 19% for ceftazidime and 22% for cefoperazone. The resistance of Streptococcus spp. (non pneumoniae) to penicillin reached 50% and that of Enterococcus spp. was of 33%. CONCLUSIONS: Treatment for pediatric patients with cancer must be modified and new guidelines including more active medications for patients at risk for bacteremia, should be devised.

[Optimization of formulations for dietetic pastry products].

Optimized formulations of dietetic pastry products such as cake and sponge cake premixes were formulated using the surface response methodology. % Emulsifier agent and baking time were the selected independent variables for cake, as well as % emulsifier agent % chlorinated flour the variables selected for sponge cake. Three different level of each variable summing up thirteen experimental formulae of each product were assessed to optimize the variables that could have some influence in the sensory characteristics of these dietetic products. The total sensory quality was determined for both dietetic products using the composite scoring test and a panel of 18 trained judges. Looking at the contour graphic and considering economic aspects the best combination of variables for cake formulation was 2% emulsifier agent and 48 minutes for baking time, With respect to sponge cake, the best combination was 6% emulsifier agent and 48% chlorinated flour. Shelf life studies showed that both dietetic formulations remained stable during storage conditions of 75 days at 30 degrees C. During this period, significant differences in sensory characteristics were not found (p < 0.05). Data of peroxide values were kept under the critical value reported for detection of organoleptic rancidity. Reported values of hedonic test showed that these dietetics pastry products had good acceptability, and open up marketing opportunities for new products with potential health benefits to consumers.

[Sensory characterization of pate from waste salmon (Salmo salar L.) products utilizing response surface methodology].

The goal of the present study was the development of a salmon pate formulation with excellent nutritional and sensory properties, using by product components from de salmon industry. The optimized formulation was obtained using the response surface methodology. A 2n factorial and a composite central rotatable design was applied in the experience. Salt and xanthan hydrocolloid gum were the selected independent variables and sensory quality the response variable. Statistical analysis was utilized to estimate the fitted model. The optimum combination of selected experimental variables were 1.5% salt and 1.35% xanthan gum. Data from the chemical characterization of salmon pate showed an important protein content (20%) greater than liver pate (11.5%) and trout pate (14.7%). With regard to fat content, the concentration of this component in salmon pate was half the concentration found in liver pate. The shelf life of the optimized formula determined at 3 degrees C and 18 degrees C were 18 days and 8 days respectively.

Copulatory behaviour and paternity in the American kestrel: the adaptive significance of frequent copulations.

The adaptive significance of repeated withinpair copulations is not well understood. We analysed the copulatory behaviour of 16 pairs of solitary-nesting American kestrels, Falco sparverius, in southern Quebec (Canada), and the achieved reproductive success (paternity) of 21 kestrel families determined by DNA fingerprinting, in terms of four hypotheses. (1) The paternity assurance hypothesis, which suggests that males copulate frequently to avoid being cuckolded, was rejected because there were few extrapair copulation attempts (<1% of all copulations observed), withinpair copulations were not timed during the fertile period and mate attendance did not increase as the fertile period approached. (2) The immediate material benefits hypothesis, which suggests that females trade copulations for food, was refuted because copulations most often occurred without food transfers, especially outside the fertile period. (3) The female mate guarding of males hypothesis, which suggests that females distract their mates from other mating opportunities by copulating frequently, was rejected because extrapair copulation attempts were infrequent, male and female solicitation frequencies were similar and females did not differ in the timing or frequency of solicitations. (4) The mate assessment hypothesis, which suggests that assessment of mate quality is mediated via copulation, most closely predicted the behaviour observed since withinpair copulations were frequent outside the fertile period and at pair formation, males and females solicited similar numbers of copulations and pairs did not differ significantly in solicitation or copulation frequency. In line with this hypothesis we found that only one brood was extrapair, probably the result of mate replacement.Copyright 1998 The Association for the Study of Animal Behaviour

Multimodal therapy for children and adolescents with Ewing sarcoma: results of the First National Chilean Trial (1986-1991).

Thirty-seven patients with Ewing sarcoma were treated in the First National Chilean Trial for Ewing's Sarcoma (1986-1991), which comprised the St. Jude Ewing's 78 Study. All patients received cyclophosphamide, doxorubicin, vincristine, and Dactinomycin for a total treatment period of about 10 months, and all prescribed therapy was administered. Local therapy consisted of irradiation (RT) to the primary tumor, complete surgical resection, or a combination of both surgery and RT. Twenty-nine of these patients had localized tumors, 24% had pelvic primary tumors, 21 were males, and 20 were greater than 10 years of age at diagnosis. Twenty-one patients had tumors that were greater than 8 cm in largest diameter. Fourteen of the 29 patients with localized disease remain disease free at 23 to 91 months from diagnosis. Fourteen patients have died of-tumor-related complications and 1 of a secondary malignancy. Relapse was local only in 4, metastatic in 9, and local plus metastatic in 1. Only 1 of the 8 patients with metastatic disease at presentation remains disease free. Toxicity consisted primarily of myelosuppression and mucositis. We conclude that this form of relative intense multimodal therapy for children/adolescents with localized Ewing sarcoma is curative in about half of affected children as in the original St. Jude study, and that it can be safely given in a developing country, provided that careful attention to supportive care and treatment planning is given. Although these results represent improvement in outcome for our patients, more effective therapy is needed for children with Ewing sarcoma, especially those with metastatic disease at presentation.

Discontinuation of antimicrobial therapy for febrile, neutropenic children with cancer: a prospective study.

During a 2-year period, all children with cancer, neutropenia, and fever who were admitted to Hospital de Niños Luis Calvo Mackenna (Santiago, Chile) were enrolled in a study of the safety of stopping antibiotic therapy on day 3 of treatment. Children who met predefined criteria for nonbacterial fever were randomized on day 3 to stop (group A) or continue (group B) antibiotic therapy. A total of 220 children with cancer had 238 episodes of fever and neutropenia; 68 children with 75 episodes met entry criteria for nonbacterial fever (group A, 36; group B, 39). Both groups were comparable in terms of age, gender, oncological disease, chemotherapy status, and initial neutrophil count. Resolution of symptoms occurred in 34 of 36 episodes in group A and 36 of 39 episodes in group B (P > .05). No deaths occurred, and bacterial superinfections were uncommon. For children with cancer as well as episodes of fever and neutropenia without an identifiable bacterial etiology at admission, stopping antibiotic therapy on day 3 was safe and not associated with a higher risk of bacterial superinfections.

[Validity of routine serum calcium measurements].

BACKGROUND: The frequency of diagnosis of primary hyperparathyroidism increased dramatically with the advent of routine serum calcium measurements. The reliability of colorimetric serum calcium measurements is not well known. AIM: To know the frequency of hyper and hypocalcemia in a population of healthy people and the reliability of serum calcium measurements. SUBJECTS AND METHODS: Serum calcium was measured in 731 subjects participating in a preventive medical examination, using an automated colorimetric method. Serum albumin was also measured. In 31 randomly chosen serum samples, calcium was also measured by atomic absorption spectrometry. Normal serum calcium ranges were established as the mean +/- 1.34 SD of the sample. RESULTS: Mean serum calcium was 9 +/- 0.7 mg/dl, serum albumin was 4.3 +/- 0.5 g/dl and albumin corrected calcium levels were 8.7 +/- 0.7 mg/dl. Excluding lipemic sera, colorimetric calcium measurement had a correlation of 0.6 with atomic absorption spectrometry and a reliability of 17.9%. Using the proposed normal ranges (7.3-10.6 mg/dl), the frequency of hypercalcemia and hypocalcemia was 0.14 and 0.4% respectively. CONCLUSIONS: Most subjects found to have hyper or hypocalcemia in this study were asymptomatic.

[Sensory characterization of lupin pudding using surface response methodology].

Full fat sweet lupin flour (Lupinus albus c.v. Multolupa) with 39.6% protein and 13.0% lipid content, and carrageenan were used to incorporate them to commercial pudding formula with the specific purpose to prepare an optimized pudding for diabetic people. Using response surface methodology, several experimental pudding trails were prepared and sensorially assessed to optimize the variables that may influence the overall sensory quality of pudding meals. Different concentrations of lupin flour and carrageenan were tested simultaneously at three levels each parameter, requiring nine combinations to be analyzed for a panel of eleven trained judges. Appearance, aroma, taste, color, texture were tested to determine total sensory quality using a composite scoring test. It was concluded that appropriate concentration of lupin flour range from 7% to 11%, and carrageenan from 0.4% to 0.5% to be added to the pudding formulation. This result was confirmed by hedonic test of acceptability.

[Chemical, sensory and clinical characterization of lupin marmalades].

Legumes are characterized as showing interesting hypoglicemic properties. Their presence in the diet is very useful in controlling the level of blood glucose in diabetic people. Taking in mind this property different formulations of prune marmalades were developed using 5, -7.5, 10 and 15% of full fat sweet lupin flour (FFLF). These samples were submitted to chemical, physical, sensory and clinical analysis. The protein content increased from 0.6 to 5.2% in direct proportion to lupin flour concentration. Energy content remained relatively constant in all formulations. The incorporation of FFLF did not affect the natural dark red prune color and pH, acidity and soluble solids values remained under the limits of Chilean food regulation laws. The samples were analyzed by a Rank preference test. Reported data showed a significative preference for the prune marmalade with 10% HEL (p < 0.05). A facial Hedonic test was used to compare the degree of acceptability between this formula and a control marmalade with fructose as edulcorant. Statistical analysis using the student test did not show significant difference (p < 0.05) between them. The hypoglycemic effect of the lupin flour was tested in seven adult non insulin dependent diabetic patients using the glucose tolerance test. Postprandial blood glucose concentration data was lower in individuals after a test meal of lupin marmalade compared to fructose marmalade. This result would allow diabetic people to eat foods such as marmalades which are known to contain a large amount of carbohydrates.

Chilean hazelnut butter, a new alternative for consumers.

Four formulations of Chilean hazelnut butter were prepared containing Chilean hazelnut paste and 5%, 10%, 15% and 20% margarine. As the level of margarine was increased to 20%, the protein and crude fiber content decreased markedly, while those of moisture, crude fat and calories increased. After 90 days of storage, neither the samples stored at 5 degrees C nor those stored at 15 degrees C showed any objectionable effects both from the bacteriological and chemical point of view. Sensory analyses, including quality and acceptability studies, were performed on the various blends. Flavor, color and taste were improved by the addition of margarine to the butter formulas. It is concluded, therefore, that Chilean hazelnut butter represents a new and interesting alternative for human nourishment.
