Beta-adrenergic receptor function in affective illness.

In a study of beta-adrenergic receptor sensitivity, the authors determined the response of cyclic AMP synthesis to in vitro addition of norepinephrine (NE) and isoproterenol (IP) in leukocytes of patients with affective illness and schizophrenia and of normal controls. IP-stimulated increase in 3H-cyclic AMP synthesis in depressed patients was significantly lower than in normal subjects and schizophrenic patients. These results suggest that beta-adrenergic receptor sensitivity is reduced in depressive illness.

Intrasubject reproducibility of growth hormone-releasing hormone-stimulated growth hormone in older women, older men, and younger men.

Intrasubject reproducibility of growth hormone (GH) response to growth hormone-releasing hormone (GHRH) was studied in healthy older women (n = 9), older men (n = 8), and younger men (n = 10). Subjects received IV injections of 0.1 ml/kg saline, 1 micrograms/kg GHRH, and 2 micrograms/kg GHRH, three times each, and blood was sampled at 0, 15, 30, 45, 60, and 120 min for GH concentration. There was no significant difference in peak GH response between the 1- and 2-micrograms/kg GHRH dosages. GH responsiveness, group variance of peak GH, and intrasubject variability were greatest for younger men, less for older men, and least for older women at both dosages of GHRH. Because of the large intrasubject variability observed in this study, it appears necessary to test subjects more than once to obtain a valid characterization of GH responsiveness.

Liquid chromatographic determination of reduced haloperidol and haloperidol concentrations in packed red blood cells from humans.

Haloperidol (H) is a neuroleptic drug that has one known biologically active metabolite, reduced haloperidol (RH). A liquid chromatographic method is described for the determination of both compounds in human red blood cell (RBC) samples. The drugs were extracted into hexane at high pH and back-extracted into 0.1M HCl. The acid solution was then analyzed by reversed-phase chromatography under the following conditions: column was ultrasphere ODS; eluant was acetonitrile:0.085M phosphate buffer (30:70), final pH was 3.5, flow rate was 2 mL/min; detection was by light absorption at 246 nm for H and 220 nm for RH. The minimum limits of quantitation for H and RH were 0.25 and 0.1 ng/mL of packed RBC respectively. For six selected patients on 10 or 20 mg per day of oral haloperidol the RBC to plasma concentration ratios for RH and H were 2.20 +/- 0.9 (SD) and 0.81 +/- 0.26, respectively. The data indicate that RH is more concentrated in RBC than in plasma.

Hydergine treatment and psychophysiological measures in primary degenerative dementia.

Changes in smooth pursuit eye movements and the P300 component of the auditory evoked potential were studied in patients with primary degenerative dementia during a double-blind, placebo-controlled study of ergoloid mesylates (Hydergine). After 18 weeks of treatment, P300 latency and amplitude, recorded at three scalp electrode sites, had not changed significantly. Smooth pursuit gain was elevated for the drug group under some stimulus conditions, suggesting a normalization of pursuit eye movement functioning. However, the results of several other measures of the quality of pursuit eye movements failed to corroborate this finding.
