Impaired neutrophil locomotion during acute bacterial infections.

Neutrophils from patients with acute bacterial infections show a marked decrease in random as well as directional locomotion in vitro. The further analysis of the data suggests that defective directional locomotion is due to impaired locomotion and not to impaired chemotaxis. The defect disappeared almost completely with clinical recovery. There was no direct correlation between impaired directional neutrophil locomotion and the proportion of band forms in the neutrophil population. Plasma obtained from patients with acute bacterial infections showed no 'spontaneous' chemotactic activity and immune complex-induced cytotaxin formation was normal in vitro. Neutrophils from 2 patients with viral pneumonia showed normal locomotion.

Increased lymphocyte load of postcapillary venules in regional lymph nodes following stimulation with antigen -- isologous antibody complexes as compared with antigen alone.

In an attempt to study changing patterns of lymphocyte influx from blood into lymph nodes following regional antigenic stimulation, mice were injected with either fluid tetanus toxoid alone or complexed (at equivalence) with isologous antibody. The mean number of lymphocytes inside the basement membrane of cross-sectioned postcapillary venules (PCV) was evaluated as a function of time after antigen injection. 'Lymphocyte load' of PCV (LL/PCV) began to increase within less than a day after antigenic stimulation. The initial increment of LL/PCV values in both primary and secondary responses was greater when complexed rather than fluid antigen was used. Peak values were reached earlier, i.e., at the end of day 2, in presensitized as compared with unprimed animals (day 4). Maximum values of mean LL/PCV did not reflect the magnitude of subsequent antibody production under all experimental conditions. We conclude that changes in the LL/PCV belong to early lymph node reactions following regional antigenic stimulation. The pattern of this particular response may vary according to the type of immunizing agent used and the state of immunity (sensitization) of the animal as well as other factors still to be elucidated.

Distinct chemokinetic and chemotactic responses in neutrophil granulocytes.

Evidence has been presented to show that chemokinesis and chemotaxis of human neutrophil granulocytes can be controlled by distinct cellular mechanisms and by different chemicals. Materials such as human serum albumin or fibrinogen had chemokinetic but no chemotactic properties. Highly purified preparations of serum-derived peptides containing classical anaphylatoxin had detectable chemotactic activity only. Chemokinetic as well as chemotactic substances were required for the expression of chemotaxis in the form of efficient directional locomotion. The roles of chemokinesis and chemotaxis in the regulation of directional locomotion have been analyzed. Further experiments showed that the chemotactic response conforms to the law of Weber-Fechner. The number of cells which had accumulated in response to a chemotactic simulus was proportional to the logarithm of the concentration of the chemical substance. Readaptation from high to low chemotactic stimulus occurred only to a limited extent.

Early appearance and mitotic activity of multinucleated giant cells in mice after combined injection of talc and prednisolone acetate. A model for studying rapid histiocytic polykarion formation in vivo.

Combined intraperitoneal injection of talc and particulate prednisolone acetate resulted in the formation of significant numbers of multinucleated giant cells within 48 hours. Neither one of these two agents had a comparable effect when injected alone. Evidence is presented showing that the multinucleated giant cells arose by cell fusion. Synchronous nuclear division could be observed in a considerable proportion of newly formed polykarions. Although the late prophases were usually normal, high proportions of abnormal mitotic figures were observed at later stages of mitosis, particularly in anaphase and telophase. The chromosome abnormalities were so severe that it is highly improbable that mitosis led to successful nuclear multiplication within a given syncytium. Similar to prednisolone acetate, prednisone elicited the formation of multinucleated giant cells when injected with talc. No polykarion formation was observed, however, when prednisolone acetate was substituted for other steroid preparations of similar particle size such as cortexon acetate, cortisone, or testosterone isobutyrate.

The chemokinetic effect of serum albumin.

Experiments performed by means of time lapse cinematography or the filter technique show that human serum albumin has marked chemokinetic effects on neutrophil cultured in Gey's solution. The average speed of the cells, as well as the proportion of neutrophils showing locomotion, is increased. Enhanced locomotion correlates with decreased attachment to the substratum as determined by morphological and functional criteria.

Relation between stimulus intensity and neutrophil chemotactic response.

The effect of chemotactic peptides which lack chemokinetic activity has been investigated. The neutrophil response is proportional to the logarithm of the stimulus intensity, or alternatively a power function with an exponent of 0.3. Equal responses are obtained for equal ratios between the peptide concentration in the lower compartment and the threshold concentration. The significance of Weber-Fechner's law in leucocyte chemotaxis is discussed.

Humoral antibodies against spermatopoietic cells associated with mediastinal germinoma.

A case of mediastinal germinoma in a 21-yr-old male is reported. The neoplasm exhibited a morphology similar to that of a testicular seminoma and was heavily infiltrated by lymphocytes and plasma cells. Germinal centres were also present in lymph follicles. Humoral antibodies (IgG), specific for spermatopoietic tissue, were present in the patient's serum. Antibody titres declined after surgical removal of the germinoma over a period of 2 yr, and antibody is no longer detectable at the present time. The implications of this observation are discussed with regard to defence mechanisms of the tumour host, and to the histogenesis of these rare neoplasms.

Significance of Hageman factor (coagulation factor XII) for neutrophil locomotion and chemotaxis.

Neutrophils separated from Hageman factor-deficient blood exhibit normal random and directional locomotion. No defect in cytotaxin formation could be demonstrated in Hageman factor-deficient plasma, nor did kallikrein inhibitor interfere with the chemotactic activity of normal immune complex-activated human plasma. The results suggest that the chemotactic activity which is detectable in unfractionated human plasma is not derived from Hageman factor-dependent pathways.

Histopathological and immunohistological changes in the rabbit lung after experimental exposure to a purified enzyme of Micropolyspora faeni.

Rabbits were exposed intratracheally to enzyme 1, a highly immunogenic esterase isolated from Micropolyspora faeni. A single exposure to active enzyme 1 induced no histologically or immunohistochemically detectable changes in the lungs of experimental animals, while signs of focal interstitial and perivascular inflammatory reactions were evident following a course of three exposures to the enzyme. Interstitial pneumonia with characteristic generalized vasculitis and perivasculitis was produced following seven or nine inoculations. An enzymatically inactive preparation of enzyme 1, even by repeated administration, proved ineffective in eliciting pneumonia. Intracellular antigen within macrophages/histiocytes was demonstrated in the lungs of all experimental animals, including those which had been exposed to inhibited enzyme. Repeated exposure to the enzymatically active preparation resulted in the deposition of immunoglobulin and complement in association with vascular endothelia and in the walls of small- and medium-sized blood vessels; both immunoglobulin and complement could also be demonstrated within macrophages/histiocytes. On the basis of these findings it is concluded that (1) Farmer's lung-like interstitial pneumonia may be produced in rabbits by exposure to a purified, enzymatically active derivative of M. faeni, (2) an important pathogenic principle of the disease may consist in the rapid vascular deposition of immune complexes (type III reaction), and (3) damage by direct enzyme action may prove to contribute significantly in eliciting tissue damage by (an) ancillary mechanism(s) not yet understood.

[Functional lymph node diagnosis].

This short review focuses on recent developments in the field of functional morphology of peripheral lymphoid tissues, in particular of lymph nodes. Emphasis is given to cellular and histological characteristics of lymph nodes in the course of cell-mediated and humoral immune responses. Against the background of regional and age-related changes, selected examples of a functional interpretation of structural changes are presented.

[Epithelial barriers of the intestine. Significance and function in defense against infection].

The epithelial surface of the intestinal tract provides for an important barrier between the organism and its environment. However, it permits the passage into the tissues of limited amounts of macromolecules and/or particles, especially bacteria. These functions are age- and species-dependent, and they are closely related to nonspecific and specific immune reactions. Continuous and intimate contact between antigenic material from the gut lumen and immunocompetent cells takes place in gut-associated lymphoid tissues.

Mast cells in the skin of normal, hairless and athymic mice.

The skin of congenitally athymic nu/nu mice is rich in mast cells which stain metachromatically, contain histamine and 5-hydroxytryptamine, and participate in the PCA reaction. Mast cells of athymic mice have thus the attributes of normal mast cells.
