Cardiovascular effects of dibutyryl cyclic AMP and milrinone under right heart bypass in anesthetized dogs.

To compare the cardiovascular effects of dibutyryl cyclic AMP (DBcAMP) and milrinone, we used the right heart bypass method in intact anesthetized dogs. The preload of the left ventricle was maintained constant throughout the experiment. Therefore, the effects of DBcAMP and milrinone on cardiac contractility and afterload of the left ventricle were investigated respectively without preload influence. DBcAMP and milrinone exhibited marked positive chronotropic-, positive inotropic-, vasodilative- and diuretic-activity. DBcAMP expressed these effects gradually and continuously, whereas milrinone expressed its pharmacological actions rapidly, but for a short duration of time. Both the afterload-reducing and the cardiac contractile force-enhancing activities of DBcAMP were longer lasting than those of milrinone. Mean arterial pressure was not altered by DBcAMP infusion, whereas it was increased significantly (p < 0.05) by milrinone infusion. The results of this study suggested that DBcAMP appeared to be indicated when there is a need to increase cardiac output by reducing afterload and increasing cardiac contractile force gradually and continuously. On the other hand, milrinone would seem to be indicated when the goal is to raise blood pressure rapidly and to increase cardiac output simultaneously.

Visualization of single molecules of RNA polymerase sliding along DNA.

Transcription requires that RNA polymerase binds to promoters buried in nonspecific sites on DNA. The search for promoters may be facilitated if the polymerase slides along the molecule of DNA. Single molecules of Escherichia coli RNA polymerase were visualized, and their movements on immobilized bacteriophage lambda and T7 DNAs were examined. Deviating from drifts by bulk flow, about 40 percent of the enzyme molecules moved along the extended DNA. The results provide direct evidence for sliding as a mechanism for relocation of the enzyme on DNA.

Higher-order dielectrophoretic effects: levitation at a field null.

Experiments with certain new micro-electrode structures used to achieve passive dielectrophoretic levitation of small particles and biological cells reveal a pronounced size-dependent effect not anticipated by the conventional dipole-based model. The conventional theory fails to predict this size effect because it neglects higher-order moments such as the quadrupole, hexapole, and octupole. These higher-order moments are in fact responsible for the levitation force achieved by azimuthally periodic electrode structures because, in such geometries, the electric field is zero along the axis so that the induced dipole moment must be zero. For example, the planar quadrupole levitates particles passively along the central axis through the interaction of its field with the induced quadrupolar moment of the particle. The size effect reported with this structure is readily explained in terms of this quadrupolar component of the ponderomotive force exerted on the particle.

Surgery of hepatic cysts in a cat.

A 2-year-old female Japanese domestic cat weighing 3.6 kg was presented due to marked abdominal distention and was diagnosed as hepatic cyst. The bile acid concentration of the hepatic cyst was as low as the serum bile acid concentration. The membrane of the cysts was excised from the edge of the compressed hepatic tissue and no other surgical manipulation was performed. This surgical procedure is simple and quick compared to hepatic lobectomy, however, it is necessary to evaluate the fluid of the cyst before performing this procedure.

Arrhythmogenicity of dopamine and its effects on hemodynamics in dogs under halothane anesthesia.

The arrhythmogenicity of dopamine, its effects on cardiac function, hemodynamics, and diuresis under halothane anesthesia were evaluated in dogs. The induction time of arrhythemias and the effect of arrhythmias on cardiac function, hemodynamics, and diuresis were determined after infusion of dopamine for 30-min period at increasing doses of 3, 5, 7, 10, and 15 micrograms/kg/min. The results were as follows. 1. Arrhythmia induction percentage was 28.6% at 5 micrograms/kg/mn, 42.9% at 7 micrograms/kg/min, 25% at 10 micrograms/kg/min, and 41.7% at 15 micrograms/kg/min. The induction time of arrhythemias (sec) was 459 at 5 micrograms/kg/min, 332 at 7 micrograms/kg/min, 152 at 10 micrograms/kg/min, and 279 at 15 micrograms/kg/min. No arrhythmias were present at 3 micrograms/kg/min. 2. Heart rate and myocardial oxygen consumption was increased in the arrhythmia-induced group compared to the non-arrhythmia-induced group. 3. Myocardial contractility, mean aortic pressure, mean pulmonary arterial pressure, and diuresis increased dose-dependently in the non-arrhythmia-induced group; however, these measures were increased in the arrhythmia-induced group without regard to dose.

Atrial fibrillation in the cow, pig, dog, and cat.

Atrial fibrillation (AF) is the most frequently observed arrhythmia in milking cows. About 50% of AF-affected cows have a history of ketosis, suggesting a relationship between AF and ketosis. Clinical signs of AF include a sudden decrease in milk production and an unstable appetite. Oral administration of quinidine is effective in the treatment of AF. AF in the pig has been observed only in one breeding animal. The clinical signs included anorexia and abnormal heart sounds. The AF was successfully terminated by the administration of 10 mg/kg quinidine every 8 h. AF in dogs and cats appears as a sign of cardiac failure, although AF may also be observed in large dogs without any underlying cardiac disease. Direct current defibrillation is effective in the treatment of these animals.

Nucleotide sequence at the 3' end of Japanese encephalitis virus genomic RNA.

Japanese encephalitis (JE) virus genomic RNAs were purified from virions. Two hundred nucleotides at the 3' end of JE virus genomic RNA were directly sequenced by using reverse transcriptase. The nucleotide sequence at the 3' end of the viral RNA was conserved among four kinds of JE virus strains. The sequence has no AU-rich region that is present at the 3' termini of alphavirus RNAs. We also compared the nucleotide sequences at the 3' ends of RNAs from three different flaviviruses and found several common sequence elements. A secondary structure at the 3' end of JE virus genomic RNA was proposed that may be common among flavivirus genomic RNAs. Such structures and other common stretches of nucleotide sequences may be related to the biological properties of flaviviruses.

Induction of brain tumors by a newly isolated JC virus (Tokyo-1 strain).

A newly isolated virus from a patient with progressive multifocal leukoencephalopathy (PML) (Tokyo-1 strain) was found serologically identical to JC virus (Mad-1 strain) and showed high neurooncogenicity in hamsters. Twenty-one animals inoculated intracerebrally with the virus developed brain tumors during a period that averaged 5 months. The tumors were cerebellar medulloblastoma (n = 20); plexus tumor (n = 2) occurred in 1 animal as a single tumor and in another in combination with a medulloblastoma. Thalamic gliomatosis was also present in 6 animals with medulloblastoma. Five mock-infected animals did not develop tumors. Medulloblastoma cells were shown to contain papovavirus T-antigen. In 20 animals examined the medulloblastoma showed a close resemblance to the human medulloblastoma in its histologic, immunocytochemical, and ultrastructural features. Examination of the incipient tumors indicated that the hamster medulloblastoma originated in cells in the neonatal external granular layer. Following infection the cells apparently migrated into the internal granular layer, carrying integrated virus genes and expressing phenotypical transformation. These findings confirm previous reports on the oncogenicity of virus isolates from PML (ZuRhein and Varakis, 1979), but are novel in that with this new isolate tumors could be induced with comparatively low levels of virus inocula.

Changes in glucose transport activities in mammary adenocarcinoma of dogs.

The activities of D-glucose transport (D-GT) and cytosolic enzymes were significantly higher in mammary adenocarcinoma of dogs than in mammary gland from normal dogs. The activities of D-GT in adenocarcinoma were over three-and-a-half times higher than in the controls. The K(m) value of the D-GT activity for glucose in both the adenocarcinoma and normal mammary gland was approximately 0.9 mM. The activities of the key glycolytic enzymes, hexokinase and pyruvate kinase, in the adenocarcinoma were also more than three-and-a-half times higher than in the controls. The increased activities of D-GT are considered to be accompanied by an acceleration of glucose utilisation in the adenocarcinoma of dogs.
