[Use of a caudal epidural sympathetic nerve block and a combination injection in chronic arterial circulatory disorders of the lower extremities].

In 30 patients with organic disturbances of the arterial blood supply of the lower extremities the caudal epidural combination injection (one per cent procaine and radecol comp.) in comparison to the caudal epidural sympathetic blocking one per cent procaine) was performed. By means of experiments on corpses of adults the spreading of the anaesthetic drug (in form of a model imitated by one per cent methylene blue) into the thoracic epidural space could be proved. The method was objectified according to clinical and plethysmographic points of view. A particular indication is present in the ischaemic ulcer depending on arteriosclerosis. During the plethysmographic investigations clear differences appeared in favour of the caudal epidural combination injection.

Specific chromosome markers involved with chronic T lymphocyte tumors.

In a series of 12 patients with chronic T cell tumors, 9 showed abnormalities of the long arm of chromosome #14. The most common aberration was an inversion with breakpoints in q11 and q32, which was seen in 7 patients. One patient had a translocation between chromosomes #9 and #14, with breaks in 9q34 and 14q11, and in another patient, the two long arms of chromosome #14 were fused together in q32, forming a dicentric chromosome. The results confirm earlier suggestions that rearrangements of band 14q11 are involved with T cell tumors and may be of importance for the development of the disease. Chromosome #8 aberrations were seen in seven patients, six of whom had an extra 8q in common. Abnormalities of chromosome #7 occurred in five patients, but the breaks were localized in different bands. The significance of the karyotypic changes in our present and previous series of patients is discussed, together with data from other laboratories.

Direct evidence for the non-random localization of mammalian chromosomes in the interphase nucleus.

Indirect immunofluorescence staining with human anti-centromere autoantibodies from a patient (LU 851) suffering from the CREST form of scleroderma was used to analyse chromosome topology in interphase nuclei of rat-kangaroo (PTO) and Indian muntjac (IM) cells. In some cells, centromeres were arranged in pairs suggesting association of homologous chromosomes. Clustering of centromeres at one pole of the nucleus (Rabl configuration) and other patterns suggesting higher order organization were also observed. In one fifth of the IM cells it was possible to identify the intranuclear location of each single chromosome on the basis of the morphology of the immunostained centromeres. In 30% of the IM cells in which centromeres could be identified, homologous chromosomes occupied adjacent territories within the interphase chromatin.

Study of DNA repair on a xeroderma pigmentosum patient and his heterozygotic parents.

Unscheduled DNA synthesis, photoreplication repair capacity, and photoreactivating enzyme levels were examined in cells of individuals of a family with one case of XP and otherwise clinically normal parents. The patient's parents were first cousins. The activity of three paths of DNA repair was depressed in the XP cells. The clinically normal parents showed normal levels of unscheduled DNA synthesis as well as postreplication repair, however their photoreactivating enzyme level was as low as 30% of normal levels.

Effect of immunization on the RNA polymerase activity of guinea-pig macrophage nuclei.

Macrophages from peritoneal exudate contain three types of DNA dependent RNA polymerase. The activity of RNA polymerases in macrophages derived from normal animals is very low. Guinea-pigs were immunized by sheep red blood cells. The immunization enhanced the activity of the RNA polymerase of macrophages derived from peritoneal exudate. The RNA polymerase activity was tested after the solubilization and chromatographic resolution of the three types of polymerases with exogenous template. The results obtained indicated that the immunization enhances the levels of polymerase I and III 10 fold while the level of polymerase II increased 5 fold.
