[Hypercalciuria and primary hyperparathyroidism in patients with kidney calculi. I. Hypercalciuria].

In 1819 patients with active or non active respectively nephrolithiasis the following parameters were assessed: plasma level of calcium, phosphate, and uric acid and urinary excretion of calcium, phosphate, oxalate, uric acid and creatinine. These parameters were estimated after 5 days of diet containing 400 mg of calcium, 800 mg of phosphate, 100 mg of purines and 40 g of proteins. In 3/4 of all examined patients at least one lithogenic factor was present. More than 40% of patients showed presence of hypercalciuria. Among these patients in 68% renal in 17% absorptive and in 15% undefined hypercalciuria was diagnosed. Patients with active nephrolithiasis showed a similar frequency of hypercalciuria but more profound abnormalities of Ca P metabolism than patients with non active renal stone disease.

[Hypercalciuria and primary hyperparathyroidism in patients with kidney calculi. II. Primary hyperparathyroidism].

Among 1819 patients with renal stone disease 44 cases with primary hyperparathyroidism (p.h.p.) were diagnosed. In all cases the diagnosis of php was confirmed by histomorphological examination. In 34 patients with php solitary adenoma was found, in 5 patients an adenoma with concomitant hyperplasia of the parathyroid glands, in 2 patients hyperplasia and in 3 patients carcinoma of the parathyroid glands. Hypercalcemia was found in 86% of patients, while elevated plasma levels of PTH in 90% of patients with php. Not in all patients PTH secretion was entirely autonomous. No significant correlation was found between plasma levels of PTH and kind of pathology of the parathyroid glands as well as clinical feature of php respectively.

[Injury of the acromial end of the clavicle imitating dislocation of the acromio-clavicular joint].

Two patients have been presented: a 15-years-old boy with rupture of the clavicular-coracoid ligaments as in total acromioclavicular dislocation, in whom dislocation of that joint did not occur but acromial epiphysiolysis of the clavicle (this patient, was successfully treated surgically) and 5-years-old boy with strain of the clavicular-coracoid ligaments and fracture of the acromial end of the clavicle; he was treated conservatively with remaining of angular dislocation of the distal fragment. The hope for rebuilding of the axis was fulfilled in that child. The anatomical and functional result in both patients is good.

[Fibrosing cholangiolitis after administration of methyltestosterone].

A case of intrahepatic cholestasis of great intensity was observed in a patient taking methyltestosterone. In histological examination of liver biopsy specimen evidence was found of fibrosing intralobular cholangiolitis. The histological findings and the clinical course are discussed considering the disease as an atypical liver reaction to methyltestosterone.

Parkinson's syndrome induced in cats by the use of 6-hydroxydopamine. Observations of behavior and motor disorders.

The possibility of inducing Parkinson's syndrome in cats was investigated in three kinds of lesions: by microinjection of 6-hydroxy dopamine (6-OHDA) into the pars compacta of substantia nigra (SNC), bilateral injection into the SNC and globus pallidus (GP) and into the SNC and caput nuclei caudati (NC). In all three kinds of lesions of the dopaminergic system disturbances of behavior involving specially the motor system were obtained, corresponding to the parkinsonism syndrome--in the form of bradykinesia-akinesia, increased muscle tonus of plastic type, vegetative disorders (sialorrhea, pupils) and psychic disorders such as lack of interest in the surroundings and food. The character of the enhanced muscle tonus typical for extrapyramidal disturbances was confirmed by EMG examination. The parkinsonism-like syndrome induced in the cats was transient and receded after several weeks.

Behavioral and neurochemical alterations following 5,6-dihydroxytryptamine administration into the median raphe nucleus in the cat.

The 5,6-dihydroxytryptamine administration into the median raphe nucleus in the cat resulted in an increase in the carbachol-induced emotional-defensive behavior and a decrease of 5-hydroxyindoleacetic acid and dopamine level in the hypothalamus, midbrain and amygdala. Lesions of median raphe nucleus resulted in similar, but more subtle alterations of post-carbachol emotional-defensive response similarly to dorsal raphe nucleus lesions, and they had also different biochemical effects in the "emotional regions" of the brain (hypothalamus, midbrain, amygdala). The absence of differentiated participation of both dorsal raphe nucleus and median raphe nucleus in the regulation of different aspect of the cat's behavior, and the role of both nuclei in the modulation of post-carbachol response is discussed.

5,6-Dihydroxytryptamine

Further evidence against using sorbitans in the brain mechanisms investigation.

In both experimental groups an intraperitoneal injection of a single dose of 450 mg/kg p-Chlorophenylalanine (p-CPA) containing 1.0% arabic gum and 450 mg/kg p-CPA containing 0.1% Tween 80 resulted in a similar drop of the serotonin (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) level in the anterior (HA) and posterior (HP) hypothalamus, midbrain (GC) and amygdala (AM). Despite the fact that just the same decrease in the 5-HT turnover occurred in both groups, only in the group treated with p-CPA containing arabic gum a rise in postcarbachol emotional-defensive response was observed. The level of dopamine (DA) did not change in both groups. The level of noradrenaline (NA) in the p-CPA with arabic gum treated group was not subject to any change, whereas in the p-CPA with Tween 80 treated group its considerable decrease was noticed, though no essential statistical differences appeared: in HA by 26%, in HP by 38%, in GC by 23% and in AM by 62% as compared to control group (NaCl treated) and in HA by 35%, in HP by 41%, in GC by 33% and in AM by 50% as compared to the p-CPA with arabic gum treated group. In all groups of animals there occurred neither spontaneous aggressive behavior nor changes in spontaneous motility. The results obtained make it clear that Tween 80 exerts its effect both on biochemical and behavioral processes. Thus, it should be acknowledged that using sorbitans in the investigation of brain mechanisms is inadvisable.

Collagen and elastin in the liver of rats intoxicated with mercuric chloride.

Intoxication of rats with mercuric chloride (0.5 mg Hg/kg of body weight, daily for 10 weeks) increased the hepatic contents of soluble and insoluble collagen and elastin. The increase was associated with elevated serum aminotransferase and alkaline phosphatase activities, and decreased total protein level in serum. Inflammatory changes were found in the liver. An increase in the fibrous protein content suggests that inflammatory reaction to mercuric chloride can result in hepatic fibrosis.

Is it advisable to use sorbitans in the investigation of the brain mechanisms?

Injection of 0.1% saline solution of Tween 80 i.p. (3 X 3 cm3/kg/12 h) in the cat decreased the carbachol-induced growling response 36 h after the last injection. The content of 5-hydroxyindoleacetic acid (5-HIAA) increased in the hypothalamus, midbrain and amygdala, which indicates the increase in serotonin (5-HT) turnover and the 5-HT system activity. The results indicate that Tween 80 causes per se both behavioral and neurochemical changes. Therefore, it is inadvisable to add sorbitans to the solutions used in the investigation of the brain mechanisms.

[Giant benign osteoblastoma of the first metacarpal bone. Late results of its removal together with the whole metacarpal bone].

A case that is a recurrence of the localization of the first described case: within the first metacarpal bone, has been presented in detail. The tumor was of enormous size, eliminating grip function of the hand. It was removed surgically together with the whole first metacarpal bone, which original borders could not be determined. In the place of the removed tumor, freeze-dried cortical-spongy bone was inserted. The follow-up at 5 years after operation revealed full healing of the graft, without any symptoms of tumor metastasis and excellent hand function.

Serotonin depletion with P-chlorophenylalanine in the cat: effects on carbachol-induced defensive behavior and regional brain amine content.

Serotonin (5-HT) depletion with p-chlorophenylalanine (p-CPA) (3 X 150 mg/kg/12 h i.p.) in the cat resulted in an increase in the carbachol-induced growing response 36 h after the last injection. Besides important depletion of 5-HT in the anterior and posterior hypothalamus, midbrain and amygdala, and 5-hydroxyindoleacetic acid (5-HIAA) in the anterior and posterior hypothalamus and midbrain, dopamine (DA) level significantly increased in the amygdala and posterior hypothalamus with tendency to increase in the anterior hypothalamus and midbrain. No changes in noradrenaline (NA) level were observed. The results indicate that the carbachol-induced defensive behavior significantly increased as a consequence of reduction in the 5-HT system activity. The changes in DA content indicate that there are interactions between the 5-HT/DA systems.

[Isolated fractures of the pisiform bone].

The literature has been discussed and two patients followed up for this injury have been presented. The patients were treated conservatively. Good results were obtained.

[Use of an immunoenzyme method with monoclonal antibodies in cytologic studies for the optimal diagnosis of non-Hodgkin's lymphoma in children].

From January 1987 to April 1988 six children we studied cytologically with the use of alkaline phosphatase-anti-alkaline phosphatase (APAAP) method and monoclonal antibodies (MoP) in order to establish the diagnosis of Non-Hodgkin Lymphoma. (NHL). Cytologic studies concerned pleural fluid (3 patients) imprints of the lymph node (3 patients), bone marrow smears (2 patients) and cerebrospinal fluid (1 patient). We performed simultaneously routine cytologic and histopathologic studies of the lymph nodes. Antigen T6 (thymocytes) was present in blasts of all patients, which permitted us to classify the blasts as common stage II group according to Reinherz. In all cases we found at least two positive antigen detected by MoP pan T (CD2, CD3, CD5, CD7) in one case--no expression of antigen T3 (CD3) and in two cases no antigen detected by an antibody CD2. Antigen Ia was found in one patient, and weak expression of antigen CALLA (CD10) in one patients. In three patients we showed a simultaneous expression of antigens T4, T8, whereas in two patients they were not observed. One child possessed mature phenotype T4, T6. By using APAAP method with MoP in cytologic studies it was possible to diagnose T-lymphoblastic lymphoma in six children before the results of histopathologic examination of the lymph nodes.

Alkaline Phosphatase

Covalent hybrids of ovomucoid third domains made from one synthetic and one natural peptide chain.

We have obtained two semisynthetic covalent hybrids (Wieczorek, M. & Laskowski, M., Jr., (1983) Biochemistry 22, 2630-2636) of turkey ovomucoid third domain by coupling the natural 19-56 peptide fragment with crude, synthetic peptides 1-18 and 6-18, respectively. We have reformed all of the disulfide bridges and then we have enzymatically synthesized the 18-19 peptide bond. The enzyme-inhibitor association constants for interaction with five different serine proteinases were the same for the semisynthetic proteins 1-56 and 6-56 and for natural proteins 1-56 and 4-56. Further, the semisynthetic 1-56 and natural 1-56 proteins were indistinguishable in analytical ion exchange and reverse-phase chromatography. This work shows that 1) making the covalent hybrids from synthetic and natural material is a facile and efficient method for preparing variants for highly quantitative sequence to reactivity studies, 2) the first five NH2-terminal residues of avian ovomucoid third domains have no effect on inhibitory activity, and 3) it is sufficient and convenient to prepare 6-56 proteins rather than 1-56 for inhibitory activity studies.

Effect of isoprenaline on tissue elastin content and serum elastolytic activity in normal and hydrazinophthalazine-treated rats.

Cardiomyopathy was produced by isoprenaline injections to healthy and hydrazinophthalazine-treated rats. The elastin content in the heart muscle and the aortic wall as well as the level of serum elastolytic activity were determined. An increase of elastin content in both tissues was found, and its progress was concomitant with the development of isoprenaline injury. It was also found that the elastolytic activity increased on the second day after the last isoprenaline injection, and on the seventh day of the experiment it began to decrease. Changes in hydrazinophthalazine-treated rats after dosing them with isoprenaline had a similar tendency but they were significantly lower than those found in healthy animals receiving isoprenaline only.
