Rats' preferences for corn versus wood-based bedding and nesting materials.

Corn by-products can be used as bedding and nesting products. Corn-cob bedding resists ammonia build-up and corn-husk nesting material resists dampness. It is not clear whether these advantages are at the expense of animal comfort. Corn cob was compared to aspen chip bedding, and corn husk to paper strip nesting material. Data from 20 rats with differential early bedding experience suggested that they prefer aspen chip, but are also biased towards the bedding they were raised on. Data from 10 rats with no prior nesting material experience suggested that paper strip was preferred over cornhusk. Thus, corn-cob products are not recommended except in situations where air quality and/or flooding are significant problems.

Vasogenic oedema and brain infarction in an experimental penumbra model.

The aim of this study was to modify the photochemical stroke model of Watson et al. [23] so as to make possible microscopical investigation of the so-called penumbra, a tissue zone at risk that surrounds an infarction. The idea was to minimize photochemical challenge to endothelial membranes in such a way that thrombotic vascular obstruction is avoided but destabilization of the blood-brain barrier is still obtained. Morphological examination of the challenged area revealed open blood vessels, overt blood-brain barrier leakage over the entire area, severely swollen glial cells and structurally intact neurons. The lesion expanded over time due to progressive extravasation, formation of perivascular edema and consequent development of secondary ischemia through mechanical compression and microvascular congestion. In contrast to a photothrombotic infarct, in which the ischemic insult is more severe and blood vessels are completely congested by aggregated platelets, with this approach blood flow is partially preserved. In this way, an ischemic penumbra is created that mimics pathologic conditions secondary to stroke and trauma. The model may be useful in studying effects of drugs on pathologic phenomena that are characteristic of a penumbra, e.g. vasogenic and cellular edema, inflammation and infarction.

Psychopathological correlates of self-reported behavioural inhibition in normal children.

The present study examined the relationship between self-reported behavioural inhibition and psychopathological symptoms in a sample of 152 children aged 12-14 years. Children were provided with a definition of behavioural inhibition and then asked to classify themselves as low, middle or high on behavioural inhibition. Furthermore, children completed questionnaires of worry, depression and anxiety symptoms. Results showed that children who endorsed the high behavioural inhibition category had elevated levels of anxiety, worry and depression compared to children who endorsed the low or middle behavioural inhibition categories. Moreover, children high on behavioural inhibition more frequently reported anxiety disorders symptoms in the subclinical range. These findings fit well with those of previous studies on behavioural inhibition.

Domains of the TCR beta-chain required for early thymocyte development.

The T cell receptor beta (TCR beta) chain controls the developmental transition from CD4-CD8- to CD4+8+thymocytes. We show that the extracellular constant region and the transmembrane region, but not the variable domain or cytoplasmic tail of the TCR beta chain are required for this differentiation step. TCR beta mutant chains lacking the cytoplasmic tail can be found at the cell surface both in functional TCR/CD3 complexes and in a GPI-anchored monomeric form indicating that the cytoplasmic tail of the TCR beta chain functions as an ER retention signal. The concordance between cell surface expression of the mutant chains as TCR/CD3 complexes and their capacity to mediate thymocyte differentiation supports the CD3 mediated feedback model in which preTCR/CD3 complexes control the developmental transition from CD4-CD8- to CD4+CD8+thymocytes.

[Endosonography of the esophagus and the stomach].

Endosonography of the oesophagus and stomach is important for staging of carcinomas, for judging the degree of thickening of the wall, and for assessment of submucosal and intramural lesions and of space-occupying processes in extra-oesophageal and extragastric organs.

Chlorophyll a fluorescence lifetime distributions in open and closed photosystem II reaction center preparations.

We have measured the decay of chlorophyll a fluorescence at 4 degrees C under anaerobic conditions in stabilized photosystem II reaction center complex isolated from spinach, using multifrequency (2-400 MHz) cross-correlation phase fluorometry. Examination of our data shows that although the fluorescence decay of open reaction centers (i.e., when both the electron donor P-680 and the electron acceptor pheophytin are capable of engaging in charge separation) can be analyzed as a multiexponential decay, another representation of the data is obtained when the decay is analyzed using a continuous distribution of lifetimes. Our results on the open reaction center differ from the two lifetime components of 25 ps and 35 ns published by Mimuro et al. (Biochim. Biophys. Acta 933 (1988) 478-486) for the D1-D2-cytochrome b-559 complex, obtained for F682 at 4 degrees C by a time-resolved photon-counting spectrofluorometer. When the reaction centers are closed by pretreatment with sodium dithionite and methyl viologen followed by exposure to laser excitation, conditions known to result in accumulation of reduced pheophytin, a dramatic decrease in the contribution of the slow lifetime component(s) is observed. These results suggest that the slow distribution lifetime component(s) in the 5-20 ns range originate(s) in the back reaction of the charge separated state. On the other hand, the fast lifetime component(s) in the picosecond range may be only partially related to the charge separation, since no dramatic change is observed upon closure of the reaction center. Perhaps, this component is related, in part, to the excitation energy migration among the various chromophores in the reaction center preparations.

DPH lifetime distributions in vesicles containing phospholipid hydroperoxides.

Multifrequency phase fluorometry was used to determine the lifetime distributions of 1,6 diphenyl-1,3,5-hexatriene in 1-palmitoyl-2 linoleoyl phosphatidylcholine small unilamellar vesicles containing 2% incorporation of phospholipid hydroperoxides. A biexponential decay was observed in both vesicle preparations over a temperature range of 5 to 35 degrees C. Vesicles containing phospholipid hydroperoxides showed an overall longer lifetime as well as a greater distribution in width. These findings suggest that phospholipid hydroperoxides create structural heterogeneity in membrane structure.

The elimination of spermidine from plasma in rats.

Infused spermidine in rats with bypassed livers was eliminated very slowly, whereas endogenous spermidine in saline-infused controls with bypassed livers increased. In the same animals endogenous putrescine plasma levels did not differ significantly between spermidine- or saline-infused animals, whereas endogenous cadaverine levels increased during spermidine infusion and decreased afterwards. It is concluded that the liver is the most important site for plasma spermidine elimination.

Angle resolved fluorescence depolarisation of 1,6-diphenyl-1,3,5 hexatriene (DPH) in unsaturated digalactosyl diacylglycerol (DGDG) lipid bilayers.

The orientational ordering and dynamics of DPH molecules embedded in macroscopically ordered bilayers of the unsaturated galactolipid digalactosyl diacylglycerol (DGDG) were studied by angle-resolved fluorescence depolarisation at room temperature. The depolarisation ratios exhibited a weaker angular dependence than those observed for DPH incorporated in multi-bilayers of saturated lecithins above their phase transition. The results reflect the differences in the modes of reorientational motion of the DPH molecules in these systems while a similar orientational order is found. It is suggested that DPH molecules in bilayers of DGDG cannot be considered to possess an effective cylindrical symmetry in contrast to DPH in bilayers of saturated lecithins.

Angle resolved depolarised resonance Raman studies of beta-carotene in lipid/water mixtures.

The orientational ordering of beta-carotene incorporated in lamellar model membranes above their main phase transitions has been studied by angle resolved depolarised Resonance Raman scattering. The ordering strongly depends on the chemical composition of the lipid bilayers: in di-oleoyl lecithin (DOPC) the carotenoid molecules lie parallel to the bilayer plane, while in di-galactosyl diglyceride ( DGDG ) a bimodal distribution was found with molecules lying both parallel and perpendicular to the membrane phase.

Study of the orientational ordering of carotenoids in lipid bilayers by resonance-Raman spectroscopy.

The orientational ordering of beta-carotene and crocetin embedded in lamellar model membranes has been investigated by angle-resolved resonance Raman scattering at a temperature well above the phase transition of the lipid chains. It is shown that the ordering of the carotenoids is dependent on the chemical composition of the lipid bilayers. The orientational distribution functions found clearly show that beta-carotene is oriented parallel to the bilayer plane (dioleoyl lecithin) or perpendicular to it (soybean lecithin). For dimyristoyl lecithin at 40 degrees C, egg-lecithin, and digalactosyl diacylglycerol two maxima were found in the orientational distribution: one parallel and one perpendicular to the bilayer surface. Crocetin embedded in soybean lecithin bilayers yields a similar bimodal distribution function. Because of rapid photodegradation no results could be obtained for spirilloxanthin.
