The influence of antigen dose on IgE production in different rat strains.

Strain differences in total serum IgE levels and IgE antibody responses to various amounts of antigen were studied in rats. Five inbred strains (BN, Wistar R/A, DA, LEW, PVG), (Wistar R/A x BN)F1 and (LEW x BN) F1 hybrids were immunized twice at an interval of 28 days by intraperitoneal injection of 10 ng, 100 ng, 1 microgram, 10 microgram, 100 microgram or 1 mg ovalbumin and 1 mg A1(OH)3 in a volume of 0.5 ml. Total serum IgE and IgE antibody levels were followed. Both parameters were measured with a solid phase radioimmunoassay. A significant difference was observed in total IgE levels between the different strains. The IgE antibody response depended both on the strain and on the amount of antigen used for immunization. There was an optimal antigen dose for IgE antibody formation, differing from strain to strain. F1 hybrids had total IgE levels and IgE responses intermediary between those of the two parent strains. A good correlation existed between the total serum IgE level before immunization and the magnitude of the optimum IgE antibody responses in the different strains. These results suggest that there may be a genetic influence on both parameters, the total IgE level and the specific IgE antibody response. The IgE antibody response is also dependent on the dose of antigen.

The effect of age on IgE production in rats.

Inbred BN-rats were immunized at different ages with a standardized amount of ovalbumin and a1(OH)3. The specific IgE antibody response and total serum IgE levels were significantly influenced by age. The IgE antibody response increased to a maximum in 2-month-old rats and decreased progressively at a more advanced age. Total serum IgE levels increased progressively from the age of 1 month to the age of 5 months and decreased again at the age of one year.

Pharmacokinetics of terbutaline after subcutaneous administration.

The pharmacokinetics of terbutaline has been studied after subcutaneous adminsitration of a therapeutic dose of 250 microgram in 14 patients. Short absorption half-lives of about 7 min resulted in a fast uptake of the drug. Serum concentrations of terbutaline were measured up to 10 hr after administration, depending on the individual. An open one-compartment model appeared adequate for the mathematical description of the kinetics in most patients. Elimination constants of 0.27 +/- 0.07 hr-1 were observed. The elimination process was biphasic in 5 patients with a mean elimination constant of the second phase of 0.10 +/- 0.04 hr-1. This resulted in a comparatively high concentration of 0.7 ng/ml 10 hr after administration. Regular use of terbutaline did not influence the pharmacokinetic data in a significant way.

Adult

Mediastinal parathyroid adenoma detected on a routine chest X-ray.

A case is described in which a large tumour was detected in the posterior mediastinum on a routine chest X-ray. Although the parathormone level was very high, no clinical nor radiographic signs of hyperparathyroidism were present. The tumour was removed by lateral thoracotomy and proved to be a parathyroid adenoma. Postoperatively the serum calcium returned to a normal level.

Total serum IgE levels in normals and patients with chronic non-specific lung diseases.

Total serum IgE was measured, using a sensitive sandwich method, in a healthy control population, in healthy persons with a familial history of chronic non-specific lung disease and in patients with chronic non-specific lung disease (CNSLD). In the control population a median total serum IgE of 39 I.U./ml was found. Total serum IgE levels were significantly lower in women that in men. Healthy persons with a familial history of CNSLD had significantly increased total IgE levels compared with the control population. The IgE level was even higher in the group of patients with CNSLD. The presence of specific IgE antibodies and/or an early onset of the symptoms of CNSLD was accompanied by higher total serum IgE levels compared with the IgE values in the other CNSLD patients. But the total IgE levels were increased in all subgroups of CNSLD patients. A significant decrease of the IgE level with age was observed in CNSLD patients.

Allergic granulomatous angiitis.

A 15-year-old boy died after a 3 years' illness; asthmatic bronchitis, recurrent heart failure and eosinophilia were the essential manifestations. The autopsy elicited a diagnosis of allergic granulomatous angiitis, because of angiitis with fibrinoid necrosis and granulomatous lesions in vascular and extravascular regions. The most important differential diagnostic aspects of this disease are discussed, especially the resemblances to Wegener's granulomatosis, hypersensitivity angiitis and polyarteritis nodosa.

The measurement of total serum IgE levels in rats.

A direct solid phase radioimmunoassay for the measurement of total serum IgE in the rat has been developed. The assay is very sensitive with a lower limit of detection of 5 ng/ml. The intra and interassay reproducibility proves to be very acceptable and the specificity is demonstrated by looking at the interferen-e by other immunoglobulins. Preliminary results on total serum IgE levels are given.

Further studies on the effect of oral doxantrazole on allergen-induced bronchospasm.

The protective effect of oral doxantrazole on allergen-induced bronchoconstriction in man has been tested in two different treatment schedules. A single administration of 200 mg or 400 mg doxantrazole did not result in significant protection for a group of 10 patients. Only two patients after 200 mg, and three patients after 400 mg received meaningful protection. A 14-day period of treatment with three times 400 mg doxantrazole per day had no significant different effect compared with placebo treatment on allergen-induced broncho-constriction.

The effect of oral doxantrazole on allergen-induced bronchospasm.

The protective effect of 200 mg doxantrazole given orally on allergen-induced bronchoconstriction has been compared with 20 mg inhaled disodium cromoglycate (DSCG) in 15 patients with bronchial asthma and immediate-type hypersensitivity. Only four out of the 15 patients showed a significant inhibition of their bronchoconstriction after the oral intake of doxantrazole. Nine patients were significantly protected by 20 mg DSCG. All patients responding to doxantrazole also responded well to DSCG.

Delayed hypersensitivity in patients with bronchial carcinoma.

Delayed hypersensitivity skin test reactions have been carried out in patients with bronchial carcinoma. Dinitrochlorobenzene, used as a parameter of newly induced cell-mediated immunity, was clearly suppressed, and this suppression was associated with a poor prognosis. Tuberculin reactivity, used as a parameter of the memory function of the cell-mediated immune system, was not altered, and bears no relation to prognosis.
