Single Nucleotide Polymorphisms in RUNX2 and BMP2 contributes to different vertical facial profile.

The vertical facial profile is a crucial factor for facial harmony with significant implications for both aesthetic satisfaction and orthodontic treatment planning. However, the role of single nucleotide polymorphisms (SNPs) in the development of vertical facial proportions is still poorly understood. This study aimed to investigate the potential impact of some SNPs in genes associated with craniofacial bone development on the establishment of different vertical facial profiles. Vertical facial profiles were assessed by two senior orthodontists through pre-treatment digital lateral cephalograms. The vertical facial profile type was determined by recommended measurement according to the American Board of Orthodontics. Healthy orthodontic patients were divided into the following groups: "Normodivergent" (control group), "Hyperdivergent" and "Hypodivergent". Patients with a history of orthodontic or facial surgical intervention were excluded. Genomic DNA extracted from saliva samples was used for the genotyping of 7 SNPs in RUNX2, BMP2, BMP4 and SMAD6 genes using real-time polymerase chain reactions (PCR). The genotype distribution between groups was evaluated by uni- and multivariate analysis adjusted by age (alpha = 5%). A total of 272 patients were included, 158 (58.1%) were "Normodivergent", 68 (25.0%) were "Hyperdivergent", and 46 (16.9%) were "Hypodivergent". The SNPs rs1200425 (RUNX2) and rs1005464 (BMP2) were associated with a hyperdivergent vertical profile in uni- and multivariate analysis (p-value < 0.05). Synergistic effect was observed when evaluating both SNPs rs1200425- rs1005464 simultaneously (Prevalence Ratio = 4.0; 95% Confidence Interval = 1.2-13.4; p-value = 0.022). In conclusion, this study supports a link between genetic factors and the establishment of vertical facial profiles. SNPs in RUNX2 and BMP2 genes were identified as potential contributors to hyperdivergent facial profiles.

Initial changes in pulpal microvasculature during orthodontic tooth movement: a stereological study.

Any alteration in blood flow or vascular pressure caused by a trauma may damage the pulp tissue. The aim of this study was to evaluate the vascular changes during the initial period of tooth movement. These alterations were assessed in coronal molar pulp tissue of 20 male Wistar rats, 90 days of age, submitted to mesial inclination movement by a closed coil spring, placed from the right maxillary first molar to the maxillary incisors. The animals were divided into three experimental groups of 6, 24, and 72 hours of 0.4 N force application, with five animals in each group, and a control group of five animals without tooth movement. The volume density of blood vessels (V(v)) of the coronal pulp tissue in the experimental groups was calculated by stereology and compared with the control group. The results demonstrated a significant increase in V(v) at 6 hours of 10.2 per cent compared with 7.2 per cent for the control group (P 0.05). These results demonstrate the high capacity of adaptation of the pulp tissue to an aggression, provided the biological limits of tolerance of the pulp are respected.

Anterior open bite--cephalometric evaluation of the dental pattern.

The purpose of this study was to compare the dental pattern of patients with anterior open bite malocclusion to that of individuals with normal overbite by utilization of lateral cephalograms, panoramic radiographs and study casts. The findings showed that there was no significant difference in the inclination of the occlusal plane (SN.PlO) and position of the maxillary and mandibular incisors (1-NA, 1-NB) between both groups of individuals; but the angles of inclination of the maxillary and mandibular incisors (1.1, 1.NA and 1.NB) differed statistically between patients with anterior open bite of the individuals that presented normal overbite, which suggests that the anterior open bite may be of dental origin.

Modified Thurow appliance: a clinical alternative for correcting skeletal open bite.

Open bite malocclusion is frequently discussed in orthodontics; diagnosis, treatment, and retention can be difficult because this malocclusion has numerous correlated etiological factors. The earlier this malocclusion is corrected, the better the prognosis will be, especially when the problem is skeletal. This article presents a patient with skeletal open bite who was treated in the mixed dentition with an orthodontic appliance that included an acrylic occlusal splint and an expansion screw, based on the original Thurow appliance, to guide the vertical force against the posterior teeth and the alveolar process.

Bonding brackets to porcelain--in vitro study.

The aim of this research was to verify, in vitro, the effect of various porcelain surface treatment on the shear strength of orthodontic brackets bonded to porcelain and the mode of fracture after debonding. Eighty-eight samples of metallic supported feldspathic porcelain were randomly divided into four groups according to their surface preparation as follows: the porcelain was maintained intact (GI), roughened with a diamond bur (GII), etched with 10% hydrofluoric acid (GIII), or sandblasted with aluminum oxide (GIV). The specimens were treated with silane (Scotchprime) and brackets were bonded with Concise. Each sample was subjected to a shear load at a crosshead speed of 1 mm/min and a recording was made at the point of failure. Bond strengths, adequate to withstand the application of orthodontic forces, were achieved in all groups. The Kruskal-Wallis statistical test showed no significant differences in bond strength between the groups (p > 0.05). However, many more porcelain fractures occurred on deglazed porcelain. This study indicates that with the appropriate material selection, the silane/composite procedure alone may be adequate for bonding.
