Synthesis of dihydronaphthalenes via aryne Diels-Alder reactions: scope and diastereoselectivity.

Novel aryne Diels-Alder reactions with functionalized acyclic dienes are reported. These give useful cis-substituted dihydronaphthalene building blocks in good yield which are not easily accessible via other means, as demonstrated in the synthesis of sertraline. The first asymmetric aryne cycloaddition with an acyclic diene is also reported, giving excellent diastereoselectivities with Oppolzer's sultam as a chiral auxiliary.

Intermolecular Heck-type coupling of aryl iodides and allylic acetates.

[reaction: see text] A palladium-catalyzed arylation of allylic acetates followed by beta-acetoxy elimination was shown to produce Heck-type coupling products. Optimal reaction conditions employed ligand-free palladium on carbon in the presence of tetrabutylammonium chloride, a trialkylamine base, and water.

Palladium-catalyzed alkylation-hydride reduction sequence: synthesis of meta-substituted arenes.

A new three-component, palladium-catalyzed domino reaction which gives access to meta-substituted arenes using aryl iodides and primary alkyl halides is reported. Various functional groups are tolerated on both the aryl iodide and alkyl halide. In addition, isotopic labeling studies provide insight into the mechanism of this Catellani-type reaction. Pd-catalyzed tandem C-N/C-C coupling of gem-dihalovinyl systems: a modular synthesis of 2-substituted indoles.

2-substituted indoles were synthesized via a Pd-catalyzed tandem C-N/Suzuki-Miyaura coupling from readily prepared ortho-gem-dihalovinylanilines. Optimal conditions used a Pd(OAc)(2)/S-Phos catalyst in the presence of K(3)PO(4).H(2)O and an organoboron reagent, which included boronic acids, esters, alkyl 9-BBN derivatives, and trialkylboranes. Yields of the desired indoles were good to excellent using low catalyst loadings (typically 1 mol %). [reaction: see text]

Preparation of homoallylic homopropargylic alcohols from 2-vinyloxiranes.

Beta,gamma-unsaturated aldehydes generated in situ by treatment of 2-vinyloxiranes with a catalytic amount of Sc(OTf)(3) or BF.OEt(2) are effectively trapped by B-allenyl-9-BBN to afford homoallylic homopropargylic alcohols in high yield. An enantioselective version has been demonstrated, and a convenient synthesis of 9-allenyl-9-BBN is described. [reaction: see text]

Total synthesis of (-)-(alpha)-kainic acid via a diastereoselective methylenecyclopropane ring expansion.

[reaction: see text] A concise and enantioselective synthesis of (-)-(alpha)-kainic acid in 13 steps with an overall yield of 15% is reported. The pyrrolidine kainoid precursor with the required C2/C3 trans stereochemistry was prepared with excellent diastereoselectivity (>20:1) via a MgI(2)-mediated ring expansion of a tertiary methylenecyclopropyl amide. A selective hydroboration was then employed to set the remaining stereochemistry at the C4 position en route to (-)-(alpha)-kainic acid.

Palladium-catalyzed intramolecular coupling between aryl iodides and allyl moieties via thermal and microwave-assisted conditions.

Palladium-catalyzed intramolecular cross-coupling reactions between aryl iodides and allyl moieties were successfully demonstrated in the presence of palladium catalyst, tri-o-tolylphosphine, a tertiary amine, and water. Several kinds of trans-2,4-disubstituted 1,2,3,4-tetrahydroquinolines were synthesized in 73-88% yields with excellent diastereoselectivities. This method was further applied to a large variety of substrates to form five-, six-, and seven-membered carbo- and heterocycles in good yields, regardless of the ring-containing atom, via microwave-assisted conditions.

A highly diastereoselective MgI2-mediated ring expansion of methylenecyclopropanes.

[reaction: see text] A highly diastereoselective MgI(2)-mediated ring expansion of methylenecyclopropane amides to functionalized pyrrolidines has been developed using chiral aromatic sulfinimines. The 2,3,4-trisubstituted pyrrolidines were isolated in generally good to excellent yields and in excellent diastereoselectivities for aromatic and heterocyclic sulfinimines.

Rhodium-catalyzed tandem cyclization: formation of 1H-indenes and 1-alkylideneindans from arylboronate esters in aqueous media.

Arylboronate esters bearing a pendant Michael-type acceptor olefin or acetylene linkage undergo transmetalation with a rhodium-based catalytic complex to generate a functionalized organorhodium intermediate which can cyclize onto nonterminal acetylenes in good to excellent yields. The catalytic system involves the use of electron-rich, sterically bulky ligands as tri-tert-butylphosphonium tetrafluoroborate stabilizing the organorhodium intermediates and reduces the incidence of protodeboronation in aqueous media.

Chemoselective cross metathesis of bishomoallylic alcohols: rapid access to fragment a of the cryptophycins.

[reaction: see text] The racemic or enantioselective allylation of in situ formed beta,gamma-unsaturated aldehydes provides efficient access to bishomoallylic alcohols from readily available 2-vinyloxiranes. These products, when subjected to modified Grubbs cross metathesis conditions, afforded terminally homologated products in moderate to good yields with high E selectivity and without degradation of the enantiomeric excess. The compounds obtained through this two-step sequence yield fragments of an important and pharmacologically active family of cryptophycins.

Rhodium-catalyzed asymmetric ring opening reactions of oxabicyclic alkenes: catalyst and substrate studies leading to a mechanistic working model.

Catalyst and substrate studies have been performed on the rhodium-catalyzed asymmetric ring opening reaction. A working model is advanced that involves oxidative insertion with retention to form an organorhodium intermediate that then undergoes nucleophilic attack with inversion. Kinetic and competition experiments have uncovered evidence for a proton transfer step in the catalytic cycle that may activate both the allylrhodium intermediate and the nucleophile. We have also conducted experiments designed to understand which properties of the PPF-P(t)Bu(2) ligand contribute to the high reactivities and enantioselectivities.

Rhodium-catalyzed asymmetric ring opening of oxabicyclic alkenes with sulfur nucleophiles.

The synthesis of 2-sulfanyl-1,2-dihydro-naphthalen-1-ols is described. This methodology is based on rhodium catalysis and enables various thiols to undergo an asymmetric SN2' ring opening of oxabenzonorbornadiene. Under the reaction conditions ([Rh(COD)Cl](2) (2.5 mol %), (S)-(R)-PPF-P(t)Bu(2) (6 mol %), AgOTf (7 mol %), NH(4)I (1.7 equiv), galvinoxyl (5 mol %), THF, 85 degrees C), aryl- and alkyl-sulfide adducts are obtained in good to excellent yield and in high enantiomeric excess (>90% ee).

Scope of palladium-catalyzed alkylative ring opening.

We have explored the scope of the palladium-catalyzed nucleophilic ring opening methodology. New highly selective and highly active catalysts have been found for the ring opening of oxabenzonorbornadienes. Employing these catalysts, the addition of various alkyl nucleophiles to oxabenzonorbornadiene has been achieved. In addition, reaction of diethylzinc with [3.2.1] oxabicyclic alkenes has been accomplished to yield ring-opened products as well as functionalized alkene addition products.

Rhodium-catalyzed asymmetric ring opening reactions of oxabicyclic alkenes: application of halide effects in the development of a general process.

We have demonstrated halide effects in the rhodium-catalyzed asymmetric ring opening reaction of oxabicyclic alkenes. By employing halide and protic additives, the catalyst poisoning effect of aliphatic amines is reversed allowing the amount nucleophile to react in high yield and ee. Second, by simply changing the halide ligand on the rhodium catalyst from chloride to iodide, the reactivity and enantioselectivity of reactions employing an aromatic amine, malonate or carboxylate nucleophile are dramatically improved. Third, through the application of halide effects and more forcing reaction conditions, less reactive oxabicycle [2.2.1] substrates react to generate synthetically useful enantioenriched cyclohexenol products. Application of these new conditions to the more reactive oxabenzonorbornadiene permits the reaction to be run with very low catalyst loadings (0.01 mol %).
