Stereochemistry of protected ornithine side chains of gramicidin S derivatives: X-ray crystal structure of the bis-Boc-tetra-N-methyl derivative of gramicidin S.

An X-ray crystallographic analysis of the bis-Ndelta-Boc-tetra-Nalpha-methyl derivative of gramicidin S, cyclo(-Val-MeOrn(Boc)-Leu-d-MePhe-Pro-)2, was undertaken successfully (R-factor = 0.088). As expected, the main chain adopts an antiparallel pleated beta-sheet conformation, but the pleated sheet is slightly twisted, and the sense of twisting is opposite to that found in the reported crystal structures of the gramicidin S-urea complex and the bis-Ndelta-(trichloroacetyl) and bis-Ndelta-(m-bromobenzoyl) derivatives of gramicidin S. In agreement with the observed resistance toward N-methylation, the urethane NH groups of the protected Orn side chains are hydrogen bonded to the carbonyl groups of the d-Phe residues. However, the side-chain-main-chain hydrogen bonding is in the i --> i - 3 mode, although hydrogen bonding in the i --> i + 2 mode was deduced from a 1H NMR study of protected gramicidin S derivatives and was actually found in the crystal structures of the diacylated gramicidin S.

Pentacyclic laddersiloxane.

A novel method which effects ring extension of the ladder oligosilsesquioxanes was developed. By applying this method, we synthesized the first functionalized tricyclic laddersiloxanes and pentacyclic laddersiloxane. The reaction of (i-PrSi(OH)O)4 with (i-PrPhClSiO)2 in pyridine gave the tetraphenyl tricyclic laddersiloxane. Following dephenylchlorination with AlCl3/HCl, and hydrolysis enabled us to isolate the tetrahydroxyl tricyclic laddersiloxane in good yield. We repeated the similar reaction from this tetrahydroxyl laddersiloxane, and the first pentacyclic laddersiloxane was obtained. The structures of the tetraphenyl and tetrahydroxyl tricyclic laddersiloxanes, and pentacyclic laddersiloxane were determined by X-ray crystallography.

Chiral crystallization of anti-dodecaisopropyltricyclo-[4.2.00.0(2,5)] octasilane.

The aim of this article is to describe the results of our investigation on the chiral crystallization of anti-tricyclic ladder polysilane, anti-dodecaisopropyltricyclo [4.2.0.0(2,5)] octasilane (1). In solid state, the silicon framework of 1 adopts twisted conformation in which the Si4 rings are puckered in the same direction. Crystallization of 1 afforded a large conglomerate crystal in high yield. X-ray crystallography revealed that the crystal consists of either right-handed (P) or left-handed (M) molecules. The ultraviolet and circular dichroism spectra of the chiral crystals of 1 reveal two absorption bands in the 400-250 nm region.
