Transient replication of bovine mitochondrial DNA and subsequent placental development in mouse embryos.

Mitochondrial DNA (mtDNA) replication during early development is believed to depend on species-specific coordination between the mitochondrial and nuclear genomes. Here, we examined the fate and developmental consequences of bovine mtDNA introduced into mouse embryos using an interspecies mitochondrial transplantation model. Bovine mtDNA exhibited transient amplification during mouse preimplantation development, but declined at the blastocyst stage. Nevertheless, bovine mtDNA persisted in both embryonic and extraembryonic tissues after implantation. Using tetraploid complementation, we further demonstrated that mtB-M embryos developed enlarged placentas with expansion of the spongiotrophoblast layer, accompanied by upregulation of the Sfmbt2-miR-466 m axis. These findings highlight partial engagement of host replication machinery by heterologous mtDNA and species-specific constraints on mtDNA replication in shaping placental development in vivo.

Magnetic resonance voiding cystourethrography for vesicoureteral reflux.

PURPOSE: To assess the feasibility of magnetic resonance voiding cystourethrography (MRVCUG) using MR fluoroscopy for evaluation of vesicoureteral reflux (VUR), and its use as a noninvasive alternative to standard VCUG. MATERIALS AND METHODS: A total of 22 MR studies of 16 patients (five months to 41 years old) with primary VUR diagnosed by standard VCUG were evaluated. Six patients underwent MR studies and standard VCUG pre- and postoperatively. MR fluoroscopy was executed with a non-enhanced heavily T2-weighted single-shot fast spin-echo (FSE) sequence. The MR findings were correlated with those obtained by the gold standard, standard VCUG. RESULTS: Of the 44 kidney-ureter units, 20 were refluxing on MRVCUG and 21 were refluxing on standard VCUG. There were one false-positive and two false-negative units. MRVCUG was 90% sensitive with a specificity of 96% for detecting VURs that were calculated based on kidney-ureter units. Two false-negative units were found in mild cases (grade I and II). For the units of grade III, IV, and V (high-grade reflux), MRVCUG detected all of the refluxing renal collecting systems. CONCLUSION: MRVCUG can demonstrate high-grade reflux without ionizing radiation or catheterization.

Multidetector-row computed tomography coronary angiography: optimization of image reconstruction phase according to the heart rate.

BACKGROUND: To optimize the image reconstruction phase of multidetector-row computed tomography (MDCT) coronary angiography according to the heart rate is crucial. METHODS AND RESULTS: Scan data were reconstructed for 10 different phases in 58 sequential patients who underwent 8-row cardiac MDCT. The obtained images were scored and compared in terms of motion artifacts and visibility of the vessels, and moreover, ECG record-based evaluations were added for clarification of the temporal relationships among these 10 phases. In the cases with lower heart rates (<65 beats/min), the best quality images were obtained when the end of the image reconstruction phase was positioned at the peak of the P wave. In some cases with higher heart rates (>65 beats/min), they were obtained in the late systolic period. CONCLUSION: As the heart rate increased, the optimal image reconstruction phase changed from mid diastole to late systole. However, it is recommended to try to decrease the heart rate of patients before data acquisition.

Highly enantioselective synthesis of atropisomeric anilide derivatives through catalytic asymmetric N-arylation: conformational analysis and application to asymmetric enolate chemistry.

In the presence of (R)-DTBM-SEGPHOS-Pd(OAc)(2) catalyst, N-arylation (aromatic amination) of various o-tert-butylanilides with p-iodonitrobenzene proceeds with high enantioselectivity (88-96% ee) to give atropisomeric N-(p-nitrophenyl)anilides having an N-C chiral axis in good yields. Atropisomeric anilide products highly prefer to exist as the E-rotamer which has trans-disposed o-tert-butylphenyl group and carbonyl oxygen. The application of the present catalytic enantioselective N-arylation to an intramolecular version gives atropisomeric lactam derivatives with high optical purity (92-98% ee). The reaction of the lithium enolate prepared from the atropisomeric anilide and lactam products with various alkyl halides gives alpha-alkylated products with high diastereoselectivity (diastereomer ratio = 13:1 to 46:1).

Refraction contrast 11x-magnified X-ray imaging of large objects by MIRRORCLE-type table-top synchrotron.

The high-quality aspects of magnified X-ray images recorded using MIRRORCLE-6X are reported. MIRRORCLE-6X is inherently suitable for hard X-ray imaging owing to its magnified projection X-ray imaging, micrometre-size X-ray source point, wide radiation emission angle, X-ray spectrum ranging from 10 keV to 6 MeV, natural refraction contrast imaging and high flux output. Images produced with 11x geometrical magnification display a sharply enhanced edge effect when generated using a 25 microm rod electron target. Image contrast is enhanced 2x owing to refraction when compared with absorption contrast images. An imitation tumour implanted in a human chest phantom was made clearly visible by using edge enhancement on images. Soft tissue becomes highly visible as a natural consequence of refraction contrast when using hard X-rays for imaging. The authors believe that novel imaging provided by MIRRORCLE makes it a superior instrument for medical diagnosis.

Synthesis, structures and reactions of cyclometallated gold complexes containing (2-diphenylarsino-n-methyl)phenyl (n = 5, 6).

Reaction of (C6H3-2-AsPh2-n-Me)Li (n = 5 or 6) with [AuBr(AsPh3)] at -78 degrees C gives the corresponding cyclometallated gold(I) complexes [Au2[(mu-C6H3-n-Me)AsPh2]2] [n = 5, (1); n = 6, (9)]. 1 undergoes oxidative addition with halogens and with dibenzoyl peroxide to give digold(II) complexes [Au2X2[(mu-C6H3-5-Me)AsPh2]2] [X = Cl (2a), Br (2b), I (2c) and O2CPh (3)] containing a metal-metal bond between the 5d9 metal centres. Reaction of 2a with AgO2CMe or of 3 with C6F5Li gives the corresponding digold(II) complexes in which X = O2CMe (4) and C6F5 (6), respectively. The Au-Au distances increase in the order 4 < 2a < 2b < 2c < 6, following the covalent binding tendency of the axial ligand. Like the analogous phosphine complexes, 2a-2c and 6 in solution rearrange to form C-C coupled digold(I) complexes [Au2X2[mu-2,2-Ph2As(5,5-Me2C6H3C6H3)AsPh2]] [X = Cl (5a), X = Br (5b), X = I (5c) and C6F5 (7)] in which the gold atoms are linearly coordinated by As and X. In contrast, the products of oxidative additions to 9 depend markedly on the halogens. Reaction of 9 with chlorine gives the gold(I)-gold(III) complex, [ClAu[mu-2-Ph2As(C6H3-6-Me)]AuCl[(6-MeC6H3)-2-AsPh2]-kappa2As,C] (10), which contains a four-membered chelate ring, Ph2As(C6H3-6-Me), in the coordination sphere of the gold(III) atom. When 10 is heated, the ring is cleaved, the product being the digold(I) complex [ClAu[mu-2-Ph2As(C6H3-6-Me)]Au[AsPh2(2-Cl-3-Me-C6H3)]] (11). Reaction of 9 with bromine at 50 degrees C gives a monobromo digold(I) complex (12), which is similar to 11 except that the 2-position of the substituted aromatic ring bears hydrogen instead halogen. Reaction of 9 with iodine gives a mixture of a free tertiary arsine, (2-I-3-MeC6H3)AsPh2 (13), a digold diiodo compound (14) analogous to 11, and a gold(I)-gold(III) zwitterionic complex [I2Au(III)[(mu-C6H3-2-AsPh2-6-Me)]2Au(I)] (15) in which the bridging units are arranged head-to-head between the metal atoms. The structures of 2a-2c and 4-15 have been determined by single-crystal X-ray diffraction analysis. The different behaviour of 1 and 9 toward halogens mirrors that of their phosphine analogues; the 6-methyl substituent blocks C-C coupling of the aryl residues in the initially formed oxidative addition product. In the case of 9, the greater lability of the Au-As bond in the initial oxidative addition product may account for the more complex behaviour of this system compared with that of its phosphine analogue.

Tumor necrosis factor-alpha induces apoptosis in cultured porcine luteal cells.

Some studies in mammalian species recently demonstrated that tumor necrosis factor (TNF)-alpha plays an important role for corpus luteum (CL) function by way of apoptosis during the estrous cycle. The objectives of this study were to clarify the induction of apoptosis in cultured porcine luteal cells by TNF-alpha treatment. Luteal cells prepared from porcine ovaries collected from crossbred mature gilts on Days 10-14 of the estrous cycle were isolated and examined as follows: 1) Flow cytometric analysis was carried out to determine apoptosis in cultured luteal cells. 2) Single-cell gel electrophoresis (comet assay) was performed to investigate apoptotic DNA fragmentation in luteal cells. The results of the flow cytometric analysis and comet assay demonstrated coincidentally that TNF-alpha induces DNA fragmentation in luteal cells causing apoptosis. These results revealed that TNF-alpha is an inducing factor of apoptosis in luteal cells.

Pluronic F127: application in arterial embolization.

PURPOSE: Pluronic is a substance that is widely used in medical and pharmaceutical fields. In particular, 20% Pluronic F127 solution is a unique substance that is liquid at less than 15 degrees C and gelatinous at 25 to 60 degrees C. In this study, the authors took advantage of the gelation property of Pluronic F127 at human body temperature to simulate embolization and dissolution of the embolism in the renal artery and the superior mesenteric artery (SMA) using a rabbit model. MATERIALS AND METHODS: Four female Japanese rabbits (weight, 2.5-3 kg each) were used. The renal artery was fitted with a 4-F cobra-type catheter and embolized with a 20% Pluronic F127 solution at a temperature of 20 degrees C. The embolic effect was evaluated by angiography immediately after the initial injection and every 15 minutes for 2.5 hours after embolization. After 24 hours, pathologic changes of the renal parenchyma were also evaluated. The embolic effect for SMA and ischemic changes of the intestine were evaluated in the same manner. RESULTS: Angiographic findings showed that Pluronic F127 caused embolization immediately after injection and dissolved in the renal artery and the SMA after 90 to 120 minutes. The pathologic findings showed no ischemic change in the renal parenchyma. Necrosis was not found in the intestine, but focal hemorrhagic changes were extensively present when the gel had dissolved. This suggested that Pluronic F127 dissolved before severe tissue damage could occur. CONCLUSION: Pluronic F127 can potentially be used as a temporary embolic material.

Vasoconstrictor administration during cardiopulmonary bypass affects acid-base balance in infants and children.

BACKGROUND: In experimental reports, blood flow redistribution occurred during cardiopulmonary bypass (CPB) and perfusion pressure was restored by vasoconstrictor administration without improving splanchnic perfusion. The influence of vasoconstrictor administration during CPB was clinically examined. MATERIALS AND METHODS: Twenty-two consecutive pediatric CPB cases of ventricular septal defect without blood transfusion were divided into two groups, depending upon whether a vasoconstrictor was administered during CPB or not (n = 7 vs. 15). Bypass flow and systemic perfusion pressure during CPB were maintained at 2.5 L/m(2)/min and not lower than 30 mm Hg by vasoconstrictor administration, respectively. RESULTS: Although preoperative state and CPB conditions were comparable between the two groups, more sodium bicarbonate was administered (P < 0.05); duration from the operation to extubation was longer (P < 0.05); and bowel movement occurred later in the vasoconstrictor-administered group than in the control group. CONCLUSIONS: Vasoconstrictor administration during CPB may deteriorate the acid-base balance and the postoperative state in infants and children.

[Plasma platinum concentration and anti-tumor effects after intra-arterial infusion of lipiodol-CDDP suspension evaluation with VX 2 rabbit liver cancer model and 7.0 Tesla MRI system].

To evaluate the usefulness of combination chemotherapy with cisplatin powder, which was newly designed for intra-arterial infusion (IA CALL) and lipiodol, for the VX 2 liver cancer model of the rabbit, sequential change of the plasma platinum concentration within the first 24 hours, as well as the tissue platinum concentration at 24 hours was measured after intra-arterial infusion of IA CALL. The infused materials were either IA CALL alone (C) or the combination of lipiodol+IA CALL (CL). In addition,the reduction rate of the VX 2 tumor was calculated among four therapeutic groups (C, CL, lipiodol alone (L), and saline alone (S)) after one week of intra-arterial infusion on the basis of 7.0 Tesla MR images. Total plasma platinum concentrations within the first 24 hours were kept low in group CL. No increase in the tissue platinum concentration in group CL was observed. On the other hand, the tumor reduction rate tended to be higher in group CL (group CL>group L=group C>group S). These results suggested that the intra-arterial infusion of IA CALL with lipiodol is more effective than that of IA CALL alone.

Biferrocene-M(mnt)2 charge-transfer complexes (M = Ni, Co; mnt = maleonitriledithiolate). structure, valence states, and magnetic properties.

Charge-transfer salts of branched-alkyl biferrocenes, (1',1' ''-R2-1,1' '-biferrocene)[Ni(mnt)2] (1a, R = isopropyl; 2a, R = dineopentyl) and (1',1' ''-R2-1,1' '-biferrocene)2[Co(mnt)2]2 (1b, R = isopropyl; 2b, R = dineopentyl), were prepared. Their valence states were investigated using X-ray crystallography and Mössbauer spectroscopy. Complexes 1a and 1b show segregated-stack crystal structures that contain columns of acceptors, whereas structures of 2a and 2b, which contain bulky donors, are rather discrete. All of the complexes contain mixed-valent biferrocenium monocations. A two-step valence transition was found in complex 1a. The crystal contains two crystallographically independent cations: one undergoes valence localization below room temperature; the other undergoes valence localization below ca. 130 K. The former transition is derived from asymmetry of the crystal environment around the cation, whereas the latter one is caused by symmetry lowering coupled with a spin-Peierls transition (T(C) = 133.2 K) associated with the dimerization of the acceptors. This compound was found to exhibit a dielectric response based on valence tautomerization. Other complexes (1b, 2a, and 2b) show a valence-trapped state. In all complexes, charge localization was found to occur through local electrostatic interactions between the donor's cationic moiety and the acceptor's electronegative moieties.

Effect of packing on orientation and cis-trans isomerization of azobenzene chromophore in Langmuir-Blodgett film.

Photoactive film material of long-chain azobenzene derivative, p-(omega-trimethylammoniodecyloxy)-p'-octyloxyazobenzene bromide (TAOAB), was fabricated into a Langmuir-Blodgett (LB) film by a polyion-complex technique using poly(sodium 4-styrenesulfonate) as a polyanion. To investigate the effect of the packing state of the azobenzene chromophore on its orientation and cis-trans isomerization, TAOAB was mixed with methyl stearate in the LB film matrix at various mole fractions (X(TAOAB)), and structural characterizations were subsequently carried out by means of Fourier transform infrared and ultraviolet-visible spectroscopies, X-ray diffraction analysis, and atomic force microscopy. The results obtained show that as the degree of packing increases, both the azobenzene chromophores and the hydrocarbon chains orient more perpendicularly to the surface of the LB film. In addition, reversible cis-trans photoisomerization of TAOAB took place upon alternate irradiation with UV and visible light even in a mixed LB film with the chromophores in a dense lateral packing state. In the process of thermal cis-to-trans isomerization, we found that the reaction rate is strongly affected by the packing state of TAOAB at 20 degrees C, reflecting the differences in steric hindrance among LB films of various X(TAOAB). In addition, higher activation energy was obtained for thermal cis-to-trans isomerization when the free volume around the chromophores became smaller.

Efficient synthesis of optically active atropisomeric anilides through catalytic asymmetric N-arylation reaction.

In the presence of (R)-DTBM-SEGPHOS-Pd(OAc)2 catalyst, N-arylation of ortho-tert-butyl-NH-anilides with 4-nitroiodobenzene proceeds with high enantioselectivity (89-95% ee) to give optically active atropisomeric anilides possessing N-C chiral axis. Furthermore, the intramolecular version of the present catalytic asymmetric N-arylation gave atropisomeric lactams with high optical purity (94-96% ee).

Establishment of a specific radioimmunoassay for bovine interferon tau.

A radioimmunoassay (RIA) was developed for quantification of bovine interferon (bIFN) tau, conceptus secretory protein, which allows for the maintenance of the corpus luteum during early pregnancy. A cDNA coding bIFN tau was derived from cultured trophoblast cells (TBs). Recombinant (r) bIFN tau was produced in a baculovirus expression system with two different viruses. The RIA was a double-antibody competitive binding assay that used anti-bIFN tau antiserum (raised in rabbits) as the primary antibody, a radioiodinated derivative of bIFNtau as the radioactive tracer, and goat anti-rabbit IgG as the secondary antibody. The antibody did not cross-react with rbIFN alpha, recombinant human IFN beta or recombinant ovine IFN tau. The correct recovery of amounts of rbIFN tau indicated good accuracy. Serially concentrated TB conditioned media, paralleled the standard curve for bIFN tau. The intra-assay and inter-assay coefficients of variation at bIFN tau levels of 7.8 and 15.6 ng/mL were 7.1 and 8.1%, and 11.0 and 8.5%, respectively. bIFN tau was directly detected in uterine flushings obtained from cows at Day 16 of pregnancy. In summary, this assay was suitable for the measurement of bIFN tau.

MR imaging of the normal appendix and acute appendicitis.

PURPOSE: To describe the MR appearance of the normal appendix and the MR imaging characteristics of acute appendicitis with correlation to pathological severity. MATERIALS AND METHODS: A total of 20 volunteers participated in this study to demonstrate normal appendices by MR imaging. A total of 37 consecutive patients with clinically diagnosed acute appendicitis were also scanned. T1-weighted (T1WI) spin-echo images, T2-weighted (T2WI) fast spin-echo, and fat-suppressed spectral presaturation inversion recovery T2-weighted (T2SPIR) fast spin-echo images were obtained. The MR criteria for considering acute appendicitis were as follows: 1) thickening of the appendiceal wall with high intensity on T2WI or T2SPIR; 2) dilated lumen filled with high intensity material on T2WI or T2SPIR; and 3) increased intensity of periappendiceal tissue on T2WI or T2SPIR. RESULTS: The visibility of a normal appendix on MR imaging was 90% (18/20). It appeared as a cord-like structure of medium intensity without fluid collection in the lumen. A total of 30 cases with clinically diagnosed acute appendicitis had positive MR findings and all except one were pathologically proven. The one had cecal diverticulitis. These cases demonstrated filled lumen, with a hypointense wall on T1WI and slightly hyperintense on T2WI or T2SPIR. MR findings correlated well with pathological severity, especially a thicker wall, periappendiceal high intensity, and ascites were useful for suspecting severe appendicitis. CONCLUSION: Correct diagnosis of acute appendicitis was obtained with MRI, and correlated well with its pathological severity. MRI is a powerful alternative for diagnosing acute appendicitis especially for the patients in whom the radiation is major concern.

The first trial of phase contrast imaging for digital full-field mammography using a practical molybdenum x-ray tube.

RATIONALE AND OBJECTIVES: The image quality of a newly developed full-field digital phase contrast mammography (PCM) system and of a conventional screen-film (SF) mammography system were compared via images of a phantom and receiver operating characteristic (ROC) analysis of clinical images. METHODS: Magnified (1.75X) PCM images were scanned (sampling rate, 43.75 microm) and then reduced to original-sized, 25-micron pixel images printed on photothermographic film. Along with corresponding SF images, the phantom images were evaluated subjectively, and the clinical images of 38 patients were subjected to ROC analysis of mass and microcalcification. RESULTS: In the image quality of a phantom, the PCM exceeded the SF. In both mass and microcalcification, the ROC analysis Az values of the PCM clinical images surpassed those of the SF images. CONCLUSION: The PCM provides better images than the SF. Clinical trials suggest superior detection of both mass and microcalcification by full-field digital PCM over conventional SF mammography.

In vitro attachment of bovine hatched blastocysts on fibronectin is mediated by integrin in a RGD dependent manner.

We investigated the effect of extracellular matrix protein on in vitro attachment and outgrowth of bovine hatched blastocysts. In vitro produced bovine hatched blastocysts were cultured on a fibronectin- or laminin-coated Petri dishes. Hatched blastocysts adhered and outgrew on the fibronectin-coated dish whereas no attachment was observed on the laminin-coated dish. The attachment and outgrowth on fibronectin were significantly inhibited in the presence of synthetic peptides containing the Arg-Gly-Asp (RGD) sequence, which interacts with the fibronectin receptor (integrin alpha5beta1), but were not inhibited by the control peptides containing the Arg-Gly-Glu (RGE) sequence. Addition of anti-fibronectin receptor (integrin alpha5beta1) antibody to the culture medium also inhibited the attachment and outgrowth on fibronectin-coated Petri dishes. Subsequently we examined mRNA expression and protein expression of alpha5 and beta1 integrin subunit in the hatched blastocyst by reverse transcription-polymerase chain reaction (RT-PCR) and immunostaining, respectively. Expression of both mRNA and protein were detected in blastocysts. These results indicate that trophectoderm cells of bovine hatched blastocysts have already acquired the ability to adhere and outgrow on fibronectin in vitro by an integrin- mediated manner.

Molecular cloning of canine nm23 cDNAs and their expression in normal and tumor tissues.

Two canine nm-23 cDNAs, designated as nm23-C1 and -C2, were isolated and characterized. Both have a putative open reading frame consisting of 459-bp encoding 152 amino acids and are highly similar to human, mouse and rat homologues. To understand the potential role of nm23-C1 and -C2 in the development of mammary gland tumors (MGT), we analyzed the mRNA expression in 14 MGT samples by RT-PCR. The samples were divided into categories according to their histopathology (benign/malignant) and metastasis. No significant difference in the mRNA expression levels of either nm23-C1 or -C2 were observed between the benign and malignant groups or the metastatic and non-metastatic groups. These results suggest that nm23-C1 and -C2 are not related to the establishment of malignancy and metastatic lesions in canine MGT cases.
