Development of interaction measures based on adaptive non-linear time series analysis of biomedical signals.

An important feature of interaction between two signal components is the direction of the interaction. Recently, different methods have been developed and applied for detecting the direction of interactions. Besides frequency-dependent methods, Granger causality is a well-known frequency-independent approach. One popular linear approach is based on autoregressive modeling of the underlying process and evaluates prediction errors under different past assumptions. In the present study, this linear concept is extended to self-exciting threshold autoregressive models, which cover a wider class of processes. An approach for the definition of a state-dependent Granger causality is given and applied to simulated data.

Methods and milliliter scale devices for high-throughput bioprocess design.

Based on electromagnetic simulations as well as on computational fluid dynamics simulations gas-inducing impellers and their magnetic inductive drive were optimized for stirred-tank reactors on a 10 ml-scale arranged in a bioreaction block with 48 bioreactors. High impeller speeds of up to 4,000 rpm were achieved at very small electrical power inputs (63 W with 48 bioreactors). The maxima of local energy dissipation in the reaction medium were estimated to be up to 50 W L(-1) at 2,800 rpm. Total power input and local energy dissipation are thus well comparable to standard stirred-tank bioreactors. A prototype fluorescence reader for 8 bioreactors with immobilized fluorometric sensor spots was applied for online measurement of dissolved oxygen concentration making use of the phase detection method. A self-optimizing scheduling software was developed for parallel control of 48 bioreactors with a liquid-handling system for automation of titration and sampling. It was shown on the examples of simple parallel batch cultivations of Escherichia coli with different media compositions that high cell densities of up to 16.5 g L(-1) dry cell mass can be achieved without pH-control within 5 h with a high parallel reproducibility (standard deviation<3.5%, n=48) due to the high oxygen transfer capability of the gas-inducing stirred-tank bioreactors.

The use of time-variant EEG Granger causality for inspecting directed interdependencies of neural assemblies.

Understanding of brain functioning requires the investigation of activated cortical networks, in particular the detection of interactions between different cortical sites. Commonly, coherence and correlation are used to describe interrelations between EEG signals. However, on this basis, no statements on causality or the direction of their interrelations are possible. Causality between two signals may be expressed in terms of upgrading the predictability of one signal by the knowledge of the immediate past of the other signal. The best-established approach in this context is the so-called Granger causality. The classical estimation of Granger causality requires the stationarity of the signals. In this way, transient pathways of information transfer stay hidden. The study presents an adaptive estimation of Granger causality. Simulations demonstrate the usefulness of the time-variant Granger causality for detecting dynamic causal relations within time intervals of less than 100 ms. The time-variant Granger causality is applied to EEG data from the Stroop task. It was shown that conflict situations generate dense webs of interactions directed from posterior to anterior cortical sites. The web of directed interactions occurs mainly 400 ms after the stimulus onset and lasts up to the end of the task.

Time-variant investigation of quadratic phase couplings caused by amplitude modulation in electroencephalic burst-suppression patterns.

OBJECTIVE: Several studies suggest that parameters derived from bispectral analysis of the EEG can be used to characterize specific cortical activation states represented by non linear interaction between brain electrical oscillations. For example, it was shown that so-called bispectral indices can be used to assess the depth of sedation. The methods applied so far are based on interval-related procedures of bispectral analysis. Consequently, the use of the resulting parameters for on-line monitoring or investigations of signal properties during transition periods, e.g., when entering burst-suppression periods, is restricted. The objective of this paper is to provide the methodological basis for a time-continuous (on-line) investigation of quadratic phase coupling induced by amplitude modulation. METHODS: To accomplish this aim an algorithm is presented which enables the analysis of the temporal development in the degree of amplitude modulation (DAM), e.g., during the transition to burst-suppression periods in patients with severe neurological diseases. RESULTS: It was found that these periods are associated with increasing DAM compared with the baseline conditions.
