Long-term lowering of tumour interstitial fluid pressure reduces Ki-67 expression.

High tumour interstitial fluid pressure (TIFP) is a characteristic of most solid tumours. Recent data give first evidence that mechanical stretch induced by TIFP triggers proliferation in solid tumours. In the present study we compared two protocols of TIFP reduction on the expression of the tumour proliferation marker Ki-67: (a) short-term lowering of TIFP by a singular puncture and (b) long-term lowering of TIFP by catheterization. Utilizing two experimental tumours (A431, A549) it was found that the TIFP broke down rapidly after a singular puncture but recovered within 6h. In case of permanent catheterization no TIFP recovery was observed. After 24h tumours were excised and stained against the proliferation marker Ki-67. While a singular puncture had no effect catheterized tumours showed a significant decrease in Ki-67 expression. Our data suggest that long-term lowering of TIFP is required to reduce tumour proliferation.

Cluster increments for macropolyhedral boranes.

Cluster increments derived for individual cluster fragments reproduce the DFT computed relative stabilities of macropolyhedral boranes usually within +/-6 kcal mol(-1). A simple summation procedure helps to select the best partner for a given cluster fragment in order to construct the thermodynamically most stable macropolyhedral borane. Cluster increments are considerably smaller for nido-cluster fragments with an even number of vertexes than for odd nido-cluster fragments pointing towards high thermodynamic stability of macropolyhedral boranes with even numbered nido-units.

FTY720 preferentially depletes naive T cells from peripheral and lymphoid organs.

The sphingosine-1-phosphate receptor agonist FTY720 induces lymphopenia by inhibiting lymphocyte egress from thymus and lymph nodes. The immediate effect of the drug on T cells in blood and lymphoid tissues is well documented, however effects on peripheral T cell sub-populations have not been studied. We therefore analyzed the changes in T cell subset compositions in liver, lung, kidney, spleen, lymph nodes and blood induced by FTY720-treatment using 9-parameter flow cytometry. In untreated mice, naive T cells were present in all peripheral organs. Naive T cells were depleted from peripheral organs within 3 days by FTY720, and with slower kinetics from lymphoid organs. Antigen-experienced T cell subsets were less affected by FTY720-treatment and substantial numbers were retained in the periphery. The proportion of CD8(+)CD44(+)CD43(+) Gr-1(+) effector memory cells increased after FTY720-treatment, while that of CD8(+)CD44(+)CD62L(+) central memory cells was unchanged. Our data demonstrate that naive T cells pass peripheral tissues as part of their default recirculation pathway. FTY720 treatment primarily affects the recirculation of naive and central memory cells, both of which re-circulate through lymph nodes on a regular basis, but does not influence effector memory cells. This suggests that treatment with FTY720 may not interfere with immune functions mediated locally by tissue-resident peripheral effector/memory T cells.

Which nido:nido-macropolyhedral boranes are most stable?

A comparison of the relative stabilities computed at RB3LYP/6-311+G(d,p)//RB3LYP/6-31G(d)+ZPE of the neutral nido-single clusters and two vertex-sharing macropolyhedral nido:nido-clusters shows single-cluster nido-boranes with up to 11 vertexes to be energetically more favorable than isomeric macropolyhedral boranes. Extra hydrogen atoms at the open face have a significant influence on the relative stabilities of the single cluster nido-boranes vs nido:nido-macropolyhedral boranes. For anionic species, a clear-cut turning point for macropolyhedral preference is shifted to no less than 17 vertexes. Thermodynamically most stable neutral and anionic nido:nido-macropolyhedral boranes usually consist of a nido-10-vertex and a nido-11-vertex unit, respectively. The relative stabilities of isomeric neutral macropolyhedra reflect the patterns exhibited by the sum of thermodynamic stabilities of the individual clusters.

Identification of organ-specific T cell populations by analysis of multiparameter flow cytometry data using DNA-chip analysis software.

BACKGROUND: The analysis of cells from multiple experimental groups by multiparameter flow cytometry leads to the generation of complex data sets, for which adequate analysis tools are not commonly available. We report here that software designed for transcriptomics applications can be used in multiparameter flow cytometry. METHODS: Lymphocytes isolated from nine different mouse organs were stained and subjected to 10-parameter flow cytometry. The resulting data set contained 594 different T cell subsets per organ per mouse and was organized into a so-called flow cytometry array (FCA). RESULTS: Computation of a hierarchical tree revealed that lymph nodes and spleen were populated by similar T cell subsets, while T cells from peripheral organs displayed a diverse subset composition. Furthermore, organ-specific T cell subsets were identified. CONCLUSIONS: This new FCA concept in flow cytomics proved to be a valuable tool for the fast and unbiased analysis of complex multiparameter flow cytometry data sets. It can be used for assessing disease progression and therapeutic intervention, and for the association of disease-related biomarkers on the protein level.

Precursor frequency can compensate for lower TCR expression in T cell competition during priming in vivo.

The factors controlling clonal dominance of cytotoxic T lymphocyte (CTL) responses are currently not well understood. To study the functional impact of the strength of the interaction of a T cell with an antigen-presenting cell in this context, we established a new mouse model comprised of two T cell receptor (TCR)-transgenic strains expressing the identical TCR in differing amounts, hence providing two CTL clones with different avidities but identical specificity and affinity. Utilizing this new model, we show that upon antigen challenge higher-avidity CTL expand at the expense of moderate-avidity CTL in vivo if present in equal numbers. Beyond this, moderate-avidity T cells can also contribute to a CTL response when present in excess. These results suggest that in addition to a proposed affinity/avidity threshold, the precursor frequency is important in defining clonal dominance. A new model in which TCR density and precursor frequency define the outcome of a CTL response is discussed.

Lowering of tumor interstitial fluid pressure reduces tumor cell proliferation in a xenograft tumor model.

High tumor interstitial fluid pressure (TIFP) is a characteristic of most solid tumors. TIFP may hamper adequate uptake of macromolecular therapeutics in tumor tissue. In addition, TIFP generates mechanical forces affecting the tumor cortex, which might influence the growth parameters of tumor cells. This seems likely as, in other tissues (namely, blood vessels or the skin), mechanical stretch is known to trigger proliferation. Therefore, we hypothesize that TIFP-induced stretch modulates proliferation-associated parameters. Solid epithelial tumors (A431 and A549) were grown in Naval Medical Research Institute nude mice, generating a TIFP of about 10 mm Hg (A431) or 5 mm Hg (A549). Tumor drainage of the central cystic area led to a rapid decline of TIFP, together with visible relaxation of the tumor cortex. It was found by sodium dodecyl sulfate polyacrylamide gel electrophoresis and Western blot analysis that TIFP lowering yields a decreased phosphorylation of proliferation-associated p44/42 mitogen-activated protein kinase and tumor relaxation. In confirmation, immunohistochemical staining showed a decrease of tumor-associated proliferation marker Ki-67 after TIFP lowering. These data suggest that the mechanical stretch induced by TIFP is a positive modulator of tumor proliferation.

The relative stabilities of 11-vertex nido- and 12-vertex closo-heteroboranes and -borates: facile estimation by structural or connection increments.

The relative thermodynamic stabilities of ortho-, meta- and para-isomers of 12-vertex closo-heteroboranes and -borates with different p-block heteroatoms were determined using density functional theory. More electronegative (smaller) heteroatoms tend to occupy non-adjacent, whereas less electronegative (larger) heteroatoms tend to occupy adjacent vertices in the thermodynamically most stable closo-diheterododecaborane isomers. The computed relative stabilities agree perfectly with experimental observations. The energy differences of para- and meta- relative to ortho-isomers of 12-vertex closo-heteroboranes generally depend on the extent of electron localization by a given heteroatom and show highly periodic trends, i.e. increase along the period and decrease down the group. The energy penalties for the HetHet structural feature (two heteroatoms adjacent to each other) for the 12-vertex closo-cluster are apparently significantly different from those for the 11-vertex nido-cluster. Reformulating two 11-vertex nido-structural features, i.e. Het(5k)(2) and HetHet, in terms of connection increments along with the additional structural feature HetHet(m) give the relative stabilities of various isomeric 11-vertex nido- as well as 12-vertex closo-heteroboranes and -borates, using one unique set of increments.

Periodic trends and easy estimation of relative stabilities in 11-vertex nido-p-block-heteroboranes and -borates.

Density functional theory computations were carried out for 11-vertex nido-p-block-hetero(carba)boranes and -borates containing silicon, germanium, tin, arsenic, antimony, sulfur, selenium and tellurium heteroatoms. A set of quantitative values called "estimated energy penalties" was derived by comparing the energies of two reference structures that differ with respect to one structural feature only. These energy penalties behave additively, i.e., they allow us to reproduce the DFT-computed relative stabilities of 11-vertex nido-heteroboranes in general with good accuracy and to predict the thermodynamic stabilities of unknown structures easily. Energy penalties for neighboring heteroatoms (HetHet and HetHet') decrease down the group and increase along the period (indirectly proportional to covalent radii). Energy penalties for a five- rather than four-coordinate heteroatom, [Het(5k)(1) and Het(5k)(2)], generally, increase down group 14 but decrease down group 16, while there are mixed trends for group 15 heteroatoms. The sum of HetHet' energy penalties results in different but easily predictable open-face heteroatom positions in the thermodynamically most stable mixed heterocarbaboranes and -borates with more than two heteroatoms.

The mechanism of Mo-/Cu-dependent CO dehydrogenase.

Density functional theory computations at the B3LYP/SDDp//B3LYP/Lanl2DZ level were performed on model complexes derived from [(Me(2)C(2)S(2))Mo(O)(2)-S-CuSMe](2-) or its oxo protonated form to gain insight into the reaction steps involved in substrate oxidation of a Mo-/Cu-dependent CO dehydrogenase. Only the bisoxo but not the hydroxo oxo complex was found to oxidize CO exothermically. A thiocarbamate complex structurally characterized as the reaction product of the enzyme with the inhibitor n-butylisonitrile corresponds to a thermodynamic well on the potential energy surface. For the formation of the analogous thiocarbonate complex from CO oxidation, however, we do not find a significant thermodynamic driving force. In the protein matrix of the enzyme this species should be further destabilized, as it requires the metal centers to move apart considerably from each other.

Structural increments for 11-vertex nido-phospha- and aza(carba)boranes and -borates; dependence of energy penalties on the extent of Electron Localization.

Relevant structural features and corresponding energy penalties were determined that allow to easily estimate the relative stabilities of 11-vertex nido-phospha- and aza-substituted boranes, borates, carbaboranes, and carbaborates. For this purpose, density functional theory computations at the B3LYP/6-311+G(d,p)//B3LYP/6-31G(d)+ZPE level were carried out to determine the relative energies of 95 phospha- and 46 aza(carba)boranes and -borates. Energy penalties assigned to disfavoring structural features show additive behavior and excellent precision with respect to the computed results, as in the case of 6- and 11-vertex nido-carboranes and -borates. An unsubstituted phosphorus atom was found to possess energy penalties quite similar to those of the three-electron-donating H-C group. A bare nitrogen atom has energy penalties much larger than those of a bare phosphorus atom. Four-electron-donating RP and RN moieties, however, have even more adverse energy penalties. The disfavoring effects of heteroatoms in a borane cluster are determined by the amount of electron localization, that is, primarily by the number of skeletal electrons that formally originate from the heterogroup and secondarily by the electronegativity. Heteroatom energy penalties are independent of the type of the other heteroatoms present in the same cluster. Some novel phospha(carba)borane geometries with bare and exo-substituted phosphorus atoms in the same cluster have favorable thermodynamic stabilities competitive with those of known isomers.

Investigation of high power effects on Ti/Al and Ta-Si-N/Cu/Ta-Si-N electrodes for SAW devices.

Damage behavior of two different metallization systems (Ti/Al bilayer and Ta-Si-N/Cu/Ta-Si-N multilayer) as finger electrodes in surface acoustic waves (SAW) devices was investigated. A special test structure was developed for this reason. The samples were loaded with traveling SAWs varying input power and loading time. Simultaneously during these experiments, the electric behavior of the SAW structure was measured and damage development by voids and hillock formation was observed using optical microscopy, too. The damaged structures were investigated by means of different microscopy techniques. Results show that the Cu-based metallization system has a significantly higher acoustomigration resistance and power durability in comparison with the Al thin film system.

Structural increment system for 11-vertex nido-boranes and carboranes.

An increment system forming a set of quantitative rules that govern the relative stabilities of 11-vertex nido-boranes and carboranes is presented. Density functional theory computations at the B3LYP/6-311+G//B3LYP/6-31G level with ZPE corrections were carried out for 61 different boron hydride and carborane structures from [B(11)H(14)](-) to C(4)B(7)H(11) to determine their relative stabilities. Disfavored structural features that destabilize a cluster structure relative to a hypothetical ideal situation were identified and weighted by so-called energy penalties. The latter show additive behavior and allow us to reproduce (within 5 kcal mol(-)(1)) the DFT computed relative energies. Energy penalties for four structural features, i.e., adjacent carbon atoms, CC, a hydrogen atom bridging between a carbon and a boron atom, CH-B, an endo-terminal hydrogen atom at an open face carbon atom, CH(2) and an endo-H between two carbon atoms, C(BH(2))C for the 11-vertex nido-cluster are quite similar to those reported for the 6-vertex nido-cluster, thus showing a behavior independent of the cluster size. Hydrogen structural features, however, vary strongly with the cluster size. Two unknown 11-vertex nido-carboranes were identified which are thermodynamically more stable than known positional isomers.

On, off and intermediate coordination of a bridgehead triarylamine donor in tripodal complexes: towards the tuning of coordinative bond distance.

A series of first row transition metal complexes of the tripodal ligand 2,2',2"-nitrilotribenzoic acid H3L has been prepared and characterised by X-ray crystallography: Mononuclear [M(L)]- species [Cu(H2O)4]3[Cu(L)(H2O)]6.25H2O (2), [Co(H2O)6][Co(L)(H2O)].8H2O (4), [Zn(H2O)6][Zn(L)(H2O)].8H2O (5) and a neutral [M(L)] complex [Fe(III)2(L)(H2O)3].5H2O (8) are formed as well as dimeric [M(L)]2 2- species (HNEt3)2[Cu(L)]2.2CH3CN (1), (HNEt3)3[Ni(L)]2(ClO4).H2O (3), (HNEt3)2[Fe(II)(L)]2.2CH3CN (6) and (HNEt3)2[Fe(III)2(L)2(mu-O)](7). The complexes display a unique variation in the M-N distance (2.09 A for Cu(II) to 3.29 A for Fe(III)) to the bridgehead triphenylamine donor and are classified into compounds with "On","Off" and "Intermediate" N-coordination. The trigonal-bipyramidal coordination polyhedron changes towards tetrahedral in the intermediate and octahedral in the Off-state. The M-N distance of individual complexes is reversibly tuned by external chemical input such as changes of metal ion oxidation state (Fe(II)/Fe(III)) or variation of the axial coligand as a consequence of solvent or pH variation. Possible reasons for the exceptional tolerance of the M-N bond to distance variations are discussed under consideration of gas phase DFT calculations of [Zn(L)]-.

Mechanical pressure-induced phosphorylation of p38 mitogen-activated protein kinase in epithelial cells via Src and protein kinase C.

Mechanical stimulation is known to modulate cell physiology in a variety of different tissues. Particularly, epithelial cells are permanently exposed to mechanical stimulation generated by externally applied forces. The present in vitro study demonstrated mechanical pressure as a trigger-factor of the p38 mitogen-activated protein kinase (MAPK) pathway in epithelial cells. Mechanical pressure applied by teflon weights (1.02g/cm(2)) led to a rapid phosphorylation of p38 peaking between 5 and 10min. Furthermore, phosphorylation of the small heat shock protein 27 (HSP27) was shown in response to mechanical pressure. Suppression of p38 function by using specific inhibitors blocked the pressure-mediated phosphorylation of HSP27. In order to identify upstream regulators of p38, a contribution of Src and protein kinase C (PKC) in pressure-signaling was investigated. We could demonstrate that inhibition of Src or PKC suppressed the pressure-induced phosphorylation of p38. These findings suggest mechanical pressure as a new type of effector stimulus for the p38 pathway with implications to (patho-) physiological conditions.
