From Genetically Altered Mice to Integrative Physiology.

Transgenic and gene-targeted mice permit the study of the function(s) of the single gene(s) in a whole organism, thereby relating molecular biology and integrative physiology. To demonstrate the potential of transgenic models, we present in this review some physiologically relevant information obtained from genetically engineered mice.

Permanently increased conductance of the murine uterine arcade after the first pregnancy.

Mechanical forces due to increased blood flow during the first pregnancy might induce a permanently higher conductance of the uterine arcade. Number of endothelial and smooth muscle cells, cross-sectional area and wall thickness of the uterine artery were measured in nulliparous mice (n = 11) and until the 93rd day after parturition in primiparous mice (n = 44). Inner diameter and wall area were calculated. Three months after the first pregnancy, uterine artery wall thickness was not altered compared to nulliparous mice. In contrast, inner diameter increased 1.6-fold, wall area 1.5-fold and the numbers of endothelial and smooth muscle cells increased 1.5 times. These changes were completely stable during the whole observation period. The increased blood flow during the first pregnancy might be a mandatory mechanical stimulus for uterine arcade maturation. This physiological maturation process could result in abortion explaining the higher prevalence of unexplained pregnancy losses in primiparous women.

Simultaneous exposure of rats to dioxin and carbon monoxide reduces the xenobiotic but not the hypoxic response.

Aryl hydrocarbon receptor (AhR) and hypoxia-inducible factor-1alpha (HIF-1alpha) are conditionally regulated transcription factor subunits that form heterodimeric complexes with their common partner, AhR nuclear translocator (ARNT/HIF-1beta). Whereas the environmentally toxic compound 2,3,7,8-tetra-chlorodibenzo-p-dioxin (TCDD) initiates the trans-activation activity of AhR:ARNT/HIF-1beta, hypoxic exposure stabilizes HIF-1alpha and functionally activates the HIF-1alpha:ARNT/HIF-1beta complex. To analyze a possible crosstalk between these two pathways in vivo, rats were given dioxin orally and/or were exposed to carbon monoxide (CO), causing functional anemia. We found that exposure to CO inhibited the xenobiotic response while dioxin application had no significant negative impact on hypoxia-mediated gene transcription.

Ultrasound guided, pre-radiation oxygen measurements using polarographic oxygen needle electrodes in spontaneous canine soft tissue sarcomas.

BACKGROUND: Poor tumour oxygenation is associated with radiation resistance. The recording of pre-treatment oxygenation status has been shown to be of prognostic relevance. MATERIAL AND METHODS: Eleven dogs with spontaneously arising soft tissue sarcomas were included in this study. Oxygen partial pressure measurements (pO2) were performed with the Eppendorf method. RESULTS: The mean of median pO2 was 9.6 mmHg (range: 0.1-30 mmHg). Four of the nine dogs included in the statistical analysis showed a median pO2 < or = 2.5 mmHg. The natural logarithm of the hypoxic subvolume correlated with the hypoxic fraction < or = 2.5 mmHg (p = 0.0712) and < or 5 mmHg (p = 0.0988). Mean corpuscular hemoglobin (MCH) and mean corpuscular volume (MCV) correlated significantly to several oxygen parameters. CONCLUSION: Hypoxia exists in spontaneous canine soft tissue sarcomas and the dog can be used as a reliable model for repeated oxygenation measurements. Ultrasonography assures reliability of needle placement.
