Evaluation of sulfated fungal beta-glucans from the sclerotium of Pleurotus tuber-regium as a potential water-soluble anti-viral agent.

Six water-insoluble fractions of fungal beta-glucans extracted by hot alkali (TM8-1 to TM8-6) from the sclerotia of Pleurotus tuber-regium (PTR) having different molecular weights (M(w)) were sulfated to give their corresponding water-soluble derivatives (S-TM8-1 to S-TM8-6) with the degree of sulfation (DS) ranging from 1.14 to 1.74. The in vitro anti-viral activities of the native beta-glucans (TM8s) and their sulfated derivatives (S-TM8s) were evaluated by the cytopathic effect assay (CPE) and the plaque reduction assay (PRA) against four kinds of viruses, including herpes simplex virus type 1 (HSV-1), herpes simplex virus type 2 (HSV-2), respiratory syncytial virus (RSV), and influenza A virus (Flu A). Although TM8s were inactive in inhibiting the viral replication in cell cultures, the S-TM8 fractions with the defined M(w) range had potent anti-viral activity against HSV-1 and HSV-2 as shown by the CPE assay. The PRA results suggested that S-TM8 fractions seemed to exert their anti-viral effect by binding to the viral particles, preventing the latter from infecting the host cells. It was plausible that the negative charges on the polymer chain of S-TM8 could interact with the positively charged glycoproteins on the surface of HSV, minimizing the interaction between the HSV and the negatively charged host cells. The anti-viral activity of the S-TM8s might also be explained by their more extended chain conformation in solution due to an increase in one of their molecular parameter, persistence length (q), as compared to the native TM8s. The potential use of S-TM8s as a water-soluble anti-HSV agent is discussed.

Determination of rofecoxib in human plasma and breast milk by high-performance liquid chromatographic assay.

A rapid and simple HPLC assay was developed for the determination of rofecoxib in human plasma and breast milk. After solid-phase extraction, rofecoxib was resolved on a C18 column and detected by UV detection at 272 nm. Standard curves were linear over the concentration range 10-2000 microg/L (r2 >0.99). Intra- and inter-day coefficients of variation for both matrices were <10% and the limit of quantification was around 10 microg/L.

[Study of diagnostic criteria of intima-media thickening and effects of drugs on peripheral arteries].

OBJECTIVE: To establish the diagnostic criteria of intima-media thickening (IMT) of the carotid artery, femoral artery and thoracic aorta in normal subjects and evaluate the effects of drugs on IMT in patients with atherosclerosis with high-frequency ultrasound imaging. METHODS: 259 normal subjects and 102 patients with atherosclerosis (AS) were studied. 134 normal subjects were examined by transesophegeal echocardiography. 102 patients with AS were randomly divided into 4 groups:simvastatin group, probucol group, captopril group, and placebo group. The values of IMT, including IMT of the normal segment (IMT(A)) and the maximal IMT of each artery (IMT(B)) of the carotid and femoral arteries, and thoracic aorta, the mean maximal IMT of the 4 arteries (IMT(C)), and the single maximal IMT among the 4 arteries (IMT(D)) were measured. The examinations were repeated in 3 years after treatments. RESULTS: In carotid, and femoral arteries and thoracic aorta, IMT increased with aging. The IMT values of the carotid and femoral arteries and thoracic aorta were 0.63 mm +/- 0.15 mm, 0.68 mm +/- 0.21 mm, 1.02 mm +/- 0.22 mm respectively with the upper limits of 0.93 mm, 1.10 mm, and 1.46 mm respectively. IMT(A) increased 0.023 mm per year. The progression value of IMT(D) in all therapeutic groups were significantly lower than that of the control group (all P < 0.05). CONCLUSION: It is suggested that the IMT > 0.93 mm, > 1.10 mm, and > 1.46 mm is be defined as indicators of IMT in carotid and femoral arteries and thoracic aorta respectively. Drug therapy can decrease the progression of atherosclerosis but has no effects on the IMT in normal segment of arteries. High frequency ultrasound technique is reliable in monitoring the progression or regression of atherosclerosis in peripheral arteries.

[A correlation study of serum inflammatory factors and intravascular ultrasound features of atherosclerotic plaques in patients with angina pectoris].

OBJECTIVE: To elucidate the relationship between serum inflammatory factors and intravascular ultrasound (IVUS) findings of atherosclerotic plaques in patients with stable and unstable angina. METHODS: Thirteen patients with stable angina (SA) in group A and nineteen patients with unstable angina (UA) in group B underwent study. Concentrations of hsCRP, sVCAM-1 and sICAM-1 were measured by means of Enzyme-Linked-Immunosorbent Assay (ELISA) and IVUS was used to analysis the coronary lesions. Their results were analyzed by correlate analysis. RESULTS: Concentration of hsCRP, sVCAM-1 and sICAM-1 were significantly higher in group B (4.7 mg/L +/- 2.6 mg/L, 789 micro g/L +/- 65 micro g/L and 365 micro g/L +/- 63 micro g/L) than in group A (2.4 mg/L +/- 1.8 mg/L, 544 micro g/L +/- 70 micro g/L and 264 micro g/L +/- 53 micro g/L, P < 0.01, respectively). IVUS found that 69.2% (18/26) patients in group B had soft lipid plaques, while patients in group A mainly had fibrous and mixed plaques, only 13.3% (2/15) had soft plaques. There were more eccentric plaques and EEMA in group B than in group A (P < 0.05, respectively), and PA and lumen area stenosis ratio (LAS) in group B were larger than those of group A (P < 0.01, respectively). Positive remodeling pattern was observed in 65.4% (17/26) lesions in group B while 66.6% (10/15) lesions in group A showed negative remodeling. sICAM-1 correlated well with RI (r = 0.475, P < 0.05) and C-RP with EEMA (r = 0.448, P < 0.05). CONCLUSION: High levels of hsCRP, sVCAM-1 and sICAM-1 are sensitive predictors of unstable angina. The features of unstable atherosclerotic plaques are eccentric, soft plaques, with large plaque areas. The vessel at the lesion shows positive remodeling. Inflammatory reaction correlated well with vascular enlargement and remodeling.

[Relationship of mitogen-activated protein kinases activation with transdifferentiation of renal tubular epithelial cells in patients with IgA nephropathy].

OBJECTIVE: To investigate the expression patterns of MAPK family (including ERK, JNK and p38) and to analyze the effects of MAPKs on renal tubular epithelial cells (RTEC) transdifferentiation in vivo. METHODS: Renal biopsy specimen were collected from 36 patients with IgA nephropathy (IgAN), which were divided into mild proliferation group (MP), focal proliferation group (FP) and proliferation-sclerosis group (PS) according to the glomerular lesions. Renal tubulointerstitial lesion score was assessed. Positive tubules of PCNA, FN, alpha-SMA, MAPK subtypes expression and phosphorylation in renal tubules were detected by immunohistochemistry staining combined with semi-quantitative method. A correlation analysis was made. RESULTS: In the MP, FP and PS groups of IgAN, the score of tubulointerstitial lesion, the positive stained tubule of PCNA, alpha-SMA and FN expression were more abundant with the severity of glomerular injury. The expression of alpha-SMA in tubule was correlated closely to FN expression and the score of interstitial fibrosis. Distribution of all subtypes of MAPK in renal tissue was no different between IgAN and normal subjects. With the severity of glomerular disease, positive tubules expressed P-ERK or P-JNK increased gradually, especially in PS group, in which expression of P-ERK was 3 fold and P-JNK was 1.6 fold compared to control (P < 0.05). Expression of JNK protein in tubule was enhanced in PS group. Expression of P-p38 had no change in different groups. P-ERK positive tubules correlated to FN expression. CONCLUSION: ERK and JNK are strongly expressed and activated in renal tubules in patients with IgAN. It seems that JNK activation is a critical signal to regulate RTEC transdifferentiation, but ERK activation may play an indirect role on this process.

KCNE2 protein is expressed in ventricles of different species, and changes in its expression contribute to electrical remodeling in diseased hearts.

BACKGROUND: Mutations in KCNE2 have been linked to long-QT syndrome (LQT6), yet KCNE2 protein expression in the ventricle and its functional role in native channels are not clear. METHODS AND RESULTS: We detected KCNE2 protein in human, dog, and rat ventricles in Western blot experiments. Immunocytochemistry confirmed KCNE2 protein expression in ventricular myocytes. To explore the functional role of KCNE2, we studied how its expression was altered in 2 models of cardiac pathology and whether these alterations could help explain observed changes in the function of native channels, for which KCNE2 is a putative auxiliary (beta) subunit. In canine ventricle injured by coronary microembolizations, the rapid delayed rectifier current (I(Kr)) density was increased. Although the protein level of ERG (I(Kr) pore-forming, alpha, subunit) was not altered, the KCNE2 protein level was markedly reduced. These data are consistent with the effect of heterologously expressed KCNE2 on ERG and suggest that in canine ventricle, KCNE2 may associate with ERG and suppress its current amplitude. In aging rat ventricle, the pacemaker current (I(f)) density was increased. There was a significant increase in the KCNE2 protein level, whereas changes in the alpha-subunit (HCN2) were not significant. These data are consistent with the effect of heterologously expressed KCNE2 on HCN2 and suggest that in aging rat ventricle, KCNE2 may associate with HCN2 and enhance its current amplitude. CONCLUSIONS: KCNE2 protein is expressed in ventricles, and it can play diverse roles in ventricular electrical activity under (patho)physiological conditions.

Stability of benzylpenicillin during continuous home intravenous therapy.

OBJECTIVES: The aim of this study was to investigate the temperature profile of home intravenous (iv) antibiotic reservoirs and the stability of 16 megaunits of benzylpenicillin sodium in 120 mL of sodium chloride 0.9% at constant and variable temperatures. METHODS: A Tinytag computerized thermometer recorded temperatures every minute in the home iv antibiotic reservoir pouches of nine patients over a 24 h period. Similar bags containing benzylpenicillin sodium (16 megaunits) were maintained either at a constant 36 degrees C, 26 degrees C or 21-22 degrees C or were worn in a pouch by five healthy volunteers for a 24 h period. Other bags were stored at 3-5 degrees C for 10 days. The bags were sampled at timed intervals and benzylpenicillin concentrations assayed by HPLC. RESULTS: Median temperatures recorded in the infusion bags worn by the nine patients were in the range 16.7-34.1 degrees C. For infusion bags maintained at 36 degrees C, 26 degrees C and 21-22 degrees C, the concentrations of benzylpenicillin dropped below 90% of the initial concentration at a mean time of 5 h 18 min, 12 h 54 min and 13 h 20 min, respectively, whereas for bags worn by the healthy volunteers the mean time for 10% loss of benzylpenicillin was 9 h 20 min. In contrast, at 3-5 degrees C, concentrations of benzylpenicillin only dropped below 90% of the initial concentration at 8 days. CONCLUSIONS: Significant temperature-dependent degradation of benzylpenicillin occurs during continuous home iv antibiotic programme infusions, which could result in loss of efficacy.

Radiosensitization of vinorelbine and gemcitabine in NCI-H460 non-small-cell lung cancer cells.

PURPOSE: Results from recent clinical studies of gemcitabine and vinorelbine have encouraged the use of this combination concurrently with radiotherapy in the treatment of non-small-cell lung cancer, although preclinical data are limited. The present study aimed to quantify the in vitro interaction and radiosensitizing effect of gemcitabine and vinorelbine individually and in combination. METHODS AND MATERIALS: Cytotoxicity was measured by exposing NCI-H460 cells to gemcitabine and/or vinorelbine simultaneously or sequentially, followed by irradiation at 0-10 Gy. Clonogenic cell survival assays were performed. Flow cytometry was used to measure the effects of both drug and radiation on cell cycle distribution. Apoptosis was assessed by morphologic criteria, by sub-G1 changes using flow cytometry assay, and by Annexin-V binding assay. RESULTS: Both drugs showed single-agent activity against NCI-H460 cells and targeted different phases of the cell cycle. When both drugs were used in combination, they showed schedule-dependent interaction. An antagonistic effect was observed with simultaneous exposure to the two drugs. The optimum combination schedule was sequential exposure to vinorelbine followed by gemcitabine 24 h later. Both drugs showed radiosensitization effects. The radiosensitization effect of gemcitabine was evident when radiation was given immediately after 4-h incubation. However, the radiosensitization effect of vinorelbine was time dependent and observed with radiation given at 24 h postincubation. Apoptosis induced by gemcitabine increased gradually, reaching 20% at 72 h posttreatment. In contrast, apoptotic cell death was an early feature in vinorelbine-treated cells, reaching approximately 40% at 24 h. CONCLUSIONS: The individual cytotoxic effects of gemcitabine and vinorelbine on NCI-H460 cells are phase specific, and the combined effect of gemcitabine and vinorelbine is sequence dependent. The radiosensitizing effects of both drugs seem to be related to enhanced apoptosis.

Chemical components and molecular mass of six polysaccharides isolated from the sclerotium of Poria cocos.

Six polysaccharides were extracted sequentially from the fresh sclerotium of Poria cocos cultivated in China using 0.9% NaCl (PCS1), hot water (PCS2), 0.5M NaOH (PCS3-I and PCS3-II), and 88% formic acid (PCS4-I and PCS4-II). Their chemical and physical characteristics were determined using infrared spectroscopy (IR), gas chromatography (GC), GC-MS methylation analysis, 13C NMR spectroscopy, elementary analysis (EA), protein analysis, size exclusion chromatography combined with laser light scattering (SEC-LLS), light scattering (LS), and viscometry. The results indicated that the polysaccharides PCS1, PCS2, and PCS3-I were heteropolysaccharides containing D-glucose, D-galactose, D-mannose, D-fucose, and D-xylose; the predominant monosaccharide was D-glucose except for PCS1 where it was D-galactose. PCS3-II, the main component of the sclerotium of P. cocos, was a linear (1-->3)-beta-D-glucan of high purity. PCS4-I consisted of (1-->3)-beta-D-glucan with some beta-(1-->6) linked branches. PCS4-II was mainly composed of (1-->3)-beta-D-glucan containing some glucose branches. The M(w) values of the six polysaccharides PCS1, PCS2, PCS3-I, PCS4-I in 0.2M NaCl aqueous solution, PCS3-II, and PCS4-II in dimethyl sulfoxide (Me(2)SO) were determined to be 11.6 x 10(4), 20.8 x 10(4), 17.1 x 10(4), 9.1 x 10(4), 12.3 x 10(4), and 21.1 x 10(4), respectively. The six polysaccharides in aqueous solution or Me(2)SO exist as flexible chains.

[Induction of atherosclerotic plaque instability in rabbits after transfection of human wild-type p53 gene].

OBJECTIVE: To develop an animal model of unstable atherosclerotic plaques. METHODS: Sixty-four New Zealand white rabbits were randomly divided into two groups: group A (n = 54, undergoing balloon-induced abdominal aortic wall injury and then fed on a diet of 1% cholesterol) and group B (n = 10, fed on a diet of 1% cholesterol only). At the end of the eighth week, the rabbits in group A were randomly divided into two subgroups: group A1 (n = 27) and group A2 (n = 27). Recombinant adenovirus carrying human wild-type p53 gene and beta galactosidase (LacZ) genes were injected through a catheter into the aortic segments rich in plaques in group A1 and A2 respectively. Two weeks later, 10 rabbits each in group A1 and A2 were killed to observe the occurrence rate of spontaneous plaque rupture, and the remaining rabbits in group A1, A2 and B all underwent pharmacological triggering by Chinese Russell's viper venom injected subperitoneally and histamine injected intravenously two times with an interval of 24 hours. Twenty-four hours after the second pharmacological triggering the remaining rabbits were all killed and their abdominal aortae were taken out to undergo macropathologic observation, staining, and immunohistochemistry to examine the accumulation of macrophages and the expression of p53 protein, in the plaques with and without rupture respectively. At the beginning of the experiment and before being killed the serum total cholesterol (TC), triglyceride (TG), low-density lipoprotein choloesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) were examined. RESULTS: Before the rabbits were killed the blood-lipid levels were significantly increased in comparison with the baseline levels (all P < 0.01), however, without significant differences among the three groups. The rate of cells positive in p53 transfection was 32.4% +/- 10.2% in the group A1, significantly higher than those in the group A2 and group B (15.8% +/- 3.6% and 16.2% +/- 6.7% respectively, both P < 0.001). The fibrous cap of the plaque was significantly thinner in the group A1 than in the other 2 groups (both P < 0.05). The apoptosis rate was 2.5% +/- 0.8% in the group A1, significantly higher than those in the group A2 and group B (1.0% +/- 0.3% and 0.9% +/- 0.4% respective, both P < 0.01). The accumulation of macrophages within the plaques was significantly remarkable and the number of vessel smooth muscle cells was much smaller in the group A1. Plaque rupture and thrombosis occurred in 12 rabbits, numbering 20 lesions, after pharmacological triggering in the group A1, and occurred in only 5 rabbits, numbering 7 lesions, in the group A2. None of the rabbits in group B showed any lesions after the pharmacological triggering. CONCLUSION: With transfection of human wild-type p53 genes and pharmacological triggering, plaque rupture and thrombosis may occur in most atherosclerotic lesions in rabbits, which offered a reliable model for the further study of unstable plaques.

Calcium signal-dependent plasticity of neuronal excitability developed postnatally.

Neuronal plasticity and its development were investigated at pyramidal neurons in the cortical slices of rats. The threshold and probability of firing spikes were measured by using whole-cell recording to assess neuronal excitability. Postsynaptic high frequency activity (HFA) at the pyramidal neurons, evoked by 20 trains (250-ms interval) of five depolarization-pulses (1 ms) at 100 Hz, persistently lowered the threshold and increased the probability of firing spikes. After long-term enhancement of neuronal excitability by HFA was stable, another HFA induced further enhancement. Infusing 1 mM 1,2-bis(2-aminophenoxy)-ethane-N, N,N',N'-tetraacetic acid or 100 microM CaMKII(281-301) into the recording neurons prevented HFA-induced long-term enhancement of neuronal excitability. The infusion of 40 microM calcineurin autoinhibitory peptide enhanced neuronal excitability, which occluded HFA effect. HFA-induced long-term enhancement of intrinsic excitability expressed at most pyramidal neurons after postnatal day (PND) 14, but not at those before PND 9. Our results show a new type of neuronal plasticity induced by physiological activity at cortical neurons, which requires calcium-dependent protein phosphorylation and develops during postnatal period. An upregulation of intrinsic excitability at cortical neurons facilitates their activity and broadens signal codes; consequently, their computational ability is upgraded.

The mechanism of action of platinum(IV) complexes in ovarian cancer cell lines.

The reduction potentials, lipophilicities, cellular uptake and cytotoxicity have been examined for two series of platinum(IV) complexes that yield common platinum(II) complexes on reduction: cis-[PtCl(4)(NH(3))(2)], cis,trans,cis-[PtCl(2)(OAc)(2)(NH(3))(2)], cis,trans,cis-[PtCl(2)(OH)(2)(NH(3))(2)], [PtCl(4)(en)], cis,trans-[PtCl(2)(OAc)(2)(en)] and cis,trans-[PtCl(2)(OH)(2)(en)] (en=ethane-1,2-diamine, OAc=acetate). As previously reported, the reduction occurs most readily when the axial ligand is chloride and least readily when it is hydroxide. The en series of complexes are marginally more lipophilic than their ammine analogues. The presence of axial chloride or acetate ligands results in a slighter higher lipophilicity compared with the platinum(II) analogue whereas hydroxide ligands lead to a substantially lower lipophilicity. The cellular uptake is similar for the platinum(II) species and their analogous tetrachloro complexes, but is substantially lower for the acetato and hydroxo complexes, resulting in a correlation with the reduction potential. The activities are also correlated with the reduction potentials with the tetrachloro complexes being the most active of the platinum(IV) series and the hydroxo being the least active. These results are interpreted in terms of reduction, followed by aquation reducing the amount of efflux from the cells resulting in an increase in net uptake.

Rabbit knee model of post-traumatic joint contractures: the long-term natural history of motion loss and myofibroblasts.

Our objective is to describe the natural history of motion loss with time and myofibroblast numbers in a rabbit knee model of post-traumatic joint contractures. Twenty-eight skeletally mature New Zealand White female rabbits had five-mm-squares of cortical bone removed from the medial and lateral femoral condyles of the right knee. A Kirschner wire (K-wire) was used to immobilize the knee joint in maximum flexion. A second operation was performed 8 weeks later to remove the K-wire. The rabbits were divided into four groups depending on the time of remobilization; 0, 8, 16 or 32 weeks. The average flexion contracture of the experimental knees in the 0-week and 8-week remobilization groups (38 degrees and 33 degrees, respectively) were significantly greater when compared with the values of the unoperated contralateral knees (8 degrees). The average flexion contractures of the experimental knees in the 16-week and 32-week remobilization groups were also greater than the unoperated contralateral knees, although they were not statistically significant. The average flexion contractures of the 16-week and 32-week groups were 19 degrees and 18 degrees, respectively, indicating a stabilization of the motion loss. Myofibroblast numbers in the posterior joint capsules were elevated 4-5x in the knees with contractures when compared to the contralateral knees. The initial decrease in severity followed by stabilization of motion loss and the association of motion loss with myofibroblasts mimics the human scenario of permanent post-traumatic joint contractures.

Myofibroblast numbers are elevated in human elbow capsules after trauma.

Elbow contractures, a frequent problem after injury, can be treated by excision of the joint capsule. However, the underlying changes in the joint capsule are poorly understood. Based on skin healing work, we examined the hypotheses that myofibroblast numbers and expression of a myofibroblast marker alpha-smooth muscle actin, are elevated in patients with posttraumatic joint contractures. Anterior capsules were obtained from six patients who had operative release of posttraumatic contractures greater than 5 months after injury and six elbows of organ donors free of contractures. Immunohistochemical studies revealed that myofibroblast numbers and percentage of total cells that were myofibroblasts were significantly elevated in the joint capsules from patients with contractures (326 +/- 61 cells per field, 36% +/- 4% total cells) when compared with similar tissues of the organ donors (69 +/- 41 cells per field, 9% +/- 4% total cells). Western blot analysis showed that protein levels of alpha-smooth muscle actin were significantly elevated in patients with posttraumatic joint contractures. However, analysis with reverse transcription-polymerase chain reaction determined that messenger ribonucleic acid levels for smooth muscle actin normalized to the housekeeping gene glyceraldehyde-3-phosphate dehydrogenase were not significantly different between the two groups. An association between increased numbers of myofibroblasts and posttraumatic joint contractures has been established in the human elbow capsule. Additional work is required to determine whether myofibroblast regulators may be targets for adjuvant therapies of posttraumatic contractures.

Identification of acyloxyacyl hydrolase, a lipopolysaccharide-detoxifying enzyme, in the murine urinary tract.

Acyloxyacyl hydrolase (AOAH) is an unusual but highly conserved lipase, previously described only in myeloid cells, that removes secondary fatty acyl chains from bacterial lipopolysaccharides (LPS) and may also act on various glycero(phospho)lipids. Deacylation by AOAH greatly reduces the ability of LPS to stimulate cells via CD14-MD-2-Toll-like receptor 4. We report here that renal cortical tubule cells produce AOAH and secrete it into urine, where it can deacylate LPS. In vitro studies revealed that proximal tubule cells secrete pro-AOAH, which can be taken up by bladder cells and processed to the heterodimeric, more enzymatically active, mature form of AOAH. AOAH can then be used by the recipient cells to deacylate LPS. The enzyme produced by proximal tubule epithelium may thus be shared with downstream cells. In addition, mature AOAH is found in the urine. We suggest that cortical tubule cells may produce and secrete AOAH to limit inflammatory responses to gram-negative bacteria throughout the urinary tract.

[Evaluation on clinical application and long term outcomes of transcervical resection of endometrium].

OBJECTIVE: To evaluate the short term and long term outcomes of abnormal uterine bleeding (AUB) treated by transcervical resection of endometrium (TCRE) as well as the factors which influence the outcomes. METHODS: From May 1990 to September 2002 there were 1431 cases who suffered from menorrhagia. A total of 1468 times of TCRE were performed. Hysteroscopic diagnosis and endometrium biopsy was performed to rule out malignant diseases in 1203 cases. One stage TCRE was performed in 265 cases. Resistance index (RI) and pulse index (PI) of every level uterine artery were measured by ultrasound and serum levels of were 6 steroid sex hormones were measured by laboratory in 32 cases. The mean duration of follow-up was 68.5 months (3 - 148 months). RESULTS: Three cases of uterine perforation were encountered. Smog like echo in the anterior uterine wall was found in 714 cases (49.9%) by B ultrasonography, among them 21 cases (63.6%) were proved adenomyosis by pathological examination. One hundred and fifty nine cases (10.8%) had been treated by drugs, such as homeostasis, analgesic agent, antibiotics, progesterone and endometrium inhibiting agents, etc. Repeat TCRE was performed in 37 cases. Owing to recurrent bleeding, dysmenorrhea or uterine myoma 87 cases (6.1%) were finally received hysterectomy. CONCLUSIONS: (1) The main cause of recurrence was incomplete destruction of or leak from basal layer of endometrium. Shift wire loop with the sheath together to resect endometrium from the fundus to cervix smoothly can solve the problem of incomplete resection. Upon completion of operations, "blind area" and "blind point" should be examined and resected again if there is some endometrium left behind which could solve leak from basal layer of endometrium. (2) The duration of follow-up and adenomyosis are the main factors, which influence the long-term successful rate. (3) TCRE dose not influence the ovarian function obviously.

[Results after laser epithelial keratectomileusis for myopia].

PURPOSE: To study the results of laser epithelial keratectomileusis (LASEK) for myopia. METHODS: LASEK was performed with the Nidek EC-5000 excimer laser. 39 patients (78 eyes) with a refraction of -1.50 to -12.00 diopters were treated and followed for 3-12 months. Postoperrative pain, uncorrected visual acuity (UCVA), refraction, corneal haze, and surgical preference were examined. RESULTS: During 3-12 months follow-up, 96.2% UCVA were above 0.5, 92.3% eyes were at least 1.0 in UCVA. The last refraction were whthin +1.00 D and -1.00 D in 18 eyes except 4 eyes. The II grade haze was found in four eye. CONCLUSIONS: LASEK was one of methods to treat eyes with myopia safely and effectively, but also studied in operation, experiment, and further effect.
