Establishment of induced pluripotent stem cell line TRNDi045-A-38 carrying homozygous DOK7-related Congenital Myasthenia patient-mutation knock-in variant from parental KOLF2.1J.

DOK7-related Congenital Myasthenic Syndrome (CMS) is a rare genetic neuromuscular junction disorder. This is one of the most common of the recessive forms of CMS, often presenting with more static proximal weakness (hence also referred to as limb girdle CMS). Whole-genome sequencing of affected patients implicates frameshift duplication mutations in DOK7 as drivers of impaired neuromuscular-junction signaling. In this study, we generated a human induced pluripotent stem cell (hiPSC) line TRNDi045-A-38 from the KOLF2.1J reference line, engineered to carry homozygous DOK7 c.1124_1127dupTGCC mutation knock-in using CRISPR/Cas9. This iPSC line could be used for in vitro disease modeling to study disease pathophysiology and for therapeutic development.

Enhanced Prediction of Peripheral Artery Disease Using Plasma Proteomics Among Individuals Without Diabetes.

BACKGROUND: Although peripheral artery disease (PAD) is an important diabetes complication, a substantial proportion of cases occur among individuals without diabetes. This study aimed to assess the predictive value of plasma proteomics in the long-term risk of PAD among individuals initially free of diabetes. METHODS: Included were 46 508 participants (6046 with prediabetes) without diabetes or major cardiovascular disease at recruitment of the UK Biobank. Using multivariable Cox regression models, a total of 2923 unique plasma proteins were assessed for the associations with incident PAD. Significant proteins were subsequently processed by a trained light gradient boosting machine classifier to determine important proteins. Using receiver operating characteristic analyses, the performance of these important proteins in predicting incident PAD were evaluated, in the whole sample and by glycemic status (normoglycemia and prediabetes). RESULTS: During a median follow-up of 12.7 years, 461 participants developed PAD. There were 107 proteins associated with incident PAD, with 103 positive associations. The LGBM approach identified 9 proteins (eg, WFDC2 [WAP 4-disulfide core domain protein 2], MMP12 [macrophage metalloelastase], and GDF15 [growth differentiation factor 15]) as the top-ranked proteins based on their importance ordering. Whereas glycated hemoglobin showed very modest predictive accuracy, a panel incorporating these top proteins showed good performance in the prediction of PAD risk (area under the curve 0.820), and it significantly enhanced the prediction beyond traditional risk factors (raising area under the curve from 0.803 to 0.837, DeLong test P=5.21×10-3). These observations were consistent for participants with normoglycemia or prediabetes. CONCLUSIONS: Plasma protein biomarkers enhance the prediction of long-term risk for PAD among individuals without diabetes, regardless of glycemic status.

Epstein-Barr Virus Sequence Variations Among the Understudied Nasopharyngeal Carcinoma Patients of Diverse Ancestries in Southeast Asia.

Epstein-Barr virus (EBV) is associated with cancers, including lymphomas and nasopharyngeal carcinoma (NPC). To date, risk variants for NPC were mainly identified from Chinese populations, which dominated the world's total number of cases. Although Southeast Asia (SEA) countries have among the world's top yet intriguingly diverse NPC age-standardized incidence rates across subpopulations, data on EBV from SEA remains scarce. In this study, we examined 83 NPC patients of different ancestries for the presence of risk haplotypes associated with the Southern Chinese NPC and generated and analyzed 67 EBV sequences (from tissue, patient-derived xenografts and lymphoblastoid cell lines of 60 NPC patients) together with 838 published EBV genomes. Our study revealed that NPC patients of non-Chinese ancestry had fewer risk variants and haplotypes that are associated with Southern Chinese NPC and clustered distinctly from lymphomas, Southern Chinese NPC, and non-cancer controls. The distribution of non-synonymous variants was similar among NPC patients of Chinese ancestry, irrespective of geographical location. Meanwhile, non-synonymous variants in genes related to packaging, latency, and structural proteins such as BPLF1, LF3, and LMP1 varied across different ancestries. Our findings suggest possibilities of EBV adaptation to host genetics for NPC pathogenesis and warrant further research for the understudied NPC subpopulations.

Histone modifications and Sp1 promote GPR160 expression in bone cancer pain within rodent models.

Bone cancer pain (BCP) affects ~70% of patients in advanced stages, primarily due to bone metastasis, presenting a substantial therapeutic challenge. Here, we profile orphan G protein-coupled receptors in the dorsal root ganglia (DRG) following tumor infiltration, and observe a notable increase in GPR160 expression. Elevated Gpr160 mRNA and protein levels persist from postoperative day 6 for over 18 days in the affected DRG, predominantly in small-diameter C-fiber type neurons specific to the tibia. Targeted interventions, including DRG microinjection of siRNA or AAV delivery, mitigate mechanical allodynia, cold, and heat hyperalgesia induced by the tumor. Tumor infiltration increases DRG neuron excitability in wild-type mice, but not in Gpr160 gene knockout mice. Tumor infiltration results in reduced H3K27me3 and increased H3K27ac modifications, enhanced binding of the transcription activator Sp1 to the Gpr160 gene promoter region, and induction of GPR160 expression. Modulating histone-modifying enzymes effectively alleviated pain behavior. Our study delineates a novel mechanism wherein elevated Sp1 levels facilitate Gpr160 gene transcription in nociceptive DRG neurons during BCP in rodents.

Morphology-tunable and photoresponsive properties in a self-assembled two-component gel system.

Photoresponsive C3-symmetrical trisurea self-assembling building blocks containing three azobenzene groups (LC10 and LC4) at the rim were designed and synthesized. By introducing a trisamide gelator (G18), which can self-aggregate through hydrogen bonds of acylamino moieties to form a fibrous network, the mixture of LC10 (or LC4) and G18 forms an organogel with coral-like supramolecular structure from 1,4-dioxane. The cooperation of hydrogen bonding and the hydrophobic diversity between these components are the main contributions to the specific superstructure. The two-component gel exhibits reversible photoisomerization from trans to cis transition without breakage of the gel state.

Self-organized microporous structures based on surfactant-encapsulated polyoxometalate complexes.

Self-organized microporous structures based on a series of surfactant-encapsulated polyoxometalate complexes (SECs) have been prepared by using ordered condensed droplets as a template. Among these structures, ordered honeycomb structures were obtained and characterized in detail by taking (DODA)(12)H[Eu(SiW(11)O(39))2] (SEC-1) as an example. Optical microscope, atomic force microscopic, and scanning electron microscopic measurements confirmed the formation of three-dimensional microporous structure, in which the top surface shows a highly ordered honeycomb structure. As compared to common solvent-casting films, the corresponding honeycomb films are more hydrophobic and possess more ordered lamellar structures. Both the wettability and the size of SECs exert significant influence on the formation of microporous structures. The proper hydrophobicity of SECs was proposed to be an essential factor for the formation of honeycomb films, and large-sized SECs are favorable for the fabrication of highly ordered honeycomb structures. The conditions for the formation of different surface morphologies have been discussed in terms of the contact angle of SECs at the interface between water and chloroform, and a contact angle slightly greater than 90 degrees is found to be a prerequisite for the formation of honeycomb structures. The results reported in this paper not only help to further comprehend the mechanism of the formation of honeycomb structures, but also provide some guidance for the design of ordered microporous films based on organic/inorganic hybrid materials, exemplified by the organic/nanoparticle complexes.

Self-assembling structures and thin-film microscopic morphologies of amphiphilic rod-coil block oligomers.

This paper examines the influences of solvent evaporation and atmosphere humidity on self-assembling structures and thin-film microscopic morphologies of amphiphilic rod-coil block oligomers (EOnOPV) containing conjugated oligo(phenylene vinylene) dimer (OPV) coupled to poly(ethylene oxide) (PEO; n, the average number of ethylene oxides, is 16, 12, 7, and 3, respectively) on hydrophilic substrates. Atomic force microscopy (AFM), UV-vis absorption, and small angle X-ray diffraction are employed to investigate the thin-film morphology and structure. Solvent evaporation and atmosphere humidity are found to exert a strong influence on thin-film morphology and structure. Under the condition of quick evaporation and dry atmosphere, all EOnOPV oligomers form the monolayer islands. Increasing the solute volume, both EO16OPV and EO12OPV oligomers can form the polar lamellas with a head-to-tail packing arrangement. Under the condition of slow evaporation and humid atmosphere, EO16OPV and EO12OPV may self-assembly into curvy nanoribbons with well-defined width and curvature radii on mica, while EO7OPV and EO3OPV with the shorter PEO coils do not form. A symmetric bilayer structure for the ribbons is proposed. Plausible reasons for the variation in thin-film morphology are discussed, based on the results obtained from investigation of PEO coil length, solvent evaporation, and atmosphere humidity effects.

The miscibility of poly(D,L-lactide-co-glycolide) with amphiphilic molecules and the interaction of their mixtures with DNA at air/water interface.

The miscibility of poly(D,L-lactide-co-glycolide) (PLG) with three amphiphilic molecules and the interaction of the PLG/surfactant mixtures with DNA at air/water interface are investigated by pi-A isotherms, Brewster angle microscopy (BAM) and atomic force microscopy (AFM) techniques. The pi-A isotherms of the PLG mixtures with cationic C(12)AzoC(6)PyBr, and C(12)AzoC(6)N(CH(3))(3)Br, are quite different from the pi-A isotherm of pure PLG on water subphase. In contrast to the case, the pi-A isotherm of PLG mixed with nonionic C(12)AzoC(6)OPy is almost identical to the pure PLG except some increasing of molecular area. Similar phenomena are observed on DNA subphase. The in situ BAM and ex situ AFM observations demonstrate that the dispersion of PLG at air/water interface becomes good when it mixes with the two cationic surfactants, whereas quite poor due to the phase separation when it mixes with the nonionic amphiphilic molecule. Based on these results we conclude that the cationic surfactants can affect the conformation change of PLG at air/water interface and figure a well miscibility with polymer whereas the nonionic amphiphilic molecule presents poor miscibility. In addition, the even mixing of the PLG and the cationic surfactants is favorable for the adsorption to DNA more effectively.

Length-controlled rodlike self-assemblies in binary mixed langmuir-blodgett monolayers on mica.

In this paper, atomic force microscopy (AFM) has been used to investigate the morphology of monolayers of the amphiphilic rod-coil diblock molecule (EO7OPV) containing oligo(phenylene vinylene) dimer (OPV) and poly(ethylene oxide) (PEO) as well as the morphology of mixed monolayers of EO7OPV and palmitic acid (PA) deposited onto mica by the Langmuir-Blodgett technique. At surface pressures higher than 3 mN/m, EO7OPV forms regular-shaped aggregates with a monomolecular layer structure, where the hydrophilic PEO blocks are adsorbed onto the mica substrate and the hydrophobic OPV blocks form an ordered crystalline OPV layer on the top of the PEO layer through the strong pi-pi stacking interaction. In the mixed LB monolayers of EO7OPV and PA, the phase separation occurs. At a certain mixed ratio, EO7OPV molecules form rodlike domains with regular shape and uniform size at surface pressures higher than 3 mN/m. With the increase of the molar fraction of PA, the rodlike domains consisting of EO7OPV are elongated. The length of the rodlike domains can be tuned easily in a large range by altering the molar ratio of EO7OPV and PA. In addition, the rodlike domains are oriented to specific directions, corresponding to the directions of the potassium ion array on the mica surface having 6-fold symmetry. We demonstrate the possible formation mechanism and the elongation origin of rodlike domains in mixed LB monolayers and propose the two-step formation process of oriented rodlike domains deposited onto the mica substrate.

Surfactant encapsulated DNA: structure characterization and interaction with dye molecules in organic media.

The recognition of electrostatically-bound DNA-didodecyldimethylammonium (DNA-DDDA) complex by three dye molecules, acridine orange (AO), ethidium bromide (EB) and 5,10,15,20-tetrakis(4-N-methylpyridyl) porphyrin tetra(p-toluenesulfonate) (TMPyP) in organic media was investigated through 1H NMR, UV-vis, and circular dichroism (CD) spectroscopies. When the organic solvent in which DNA-DDDA complex dissolves is changed from ethanol to chloroform, the adsorbed AO undergoes a reversible transformation from a monomer to a highly aggregated state at the interface between DNA and DDDA. EB also adsorbs at the interface between DNA and DDDA when EB interacts with the DNA-DDDA complex in organic media, but its existing state is independent of the used solvents. The third dye, TMPyP cation can intercalate into the G-C region while its anionic p-tosylate counterion remains unbound when it mixes with DNA complex in organic media. The complexes of DDDA with previously recognized DNA by the three dye molecules (DNA-dye), respectively, are also investigated. AO seems having changed its location from the grooves of DNA to the interface between DNA and DDDA after DNA-AO complex was electrostatically encapsulated with DDDA. The aggregation behavior of AO also shows a dependence on the polarity of the organic solvent. EB molecules are believed to intercalate into the base pairs of DNA in aqueous solution. The intercalation mode is still maintained after the encapsulation for DNA-EB in organic solvents, which is different from the situation between DNA-DDDA complex and EB. But in both cases, the existing states of EB are independent of the polarity of the organic solvents. Finally, TMPyP in the complex of DNA-TMPyP and DDDA is also judged to intercalate into the G-C region of DNA while its anionic p-tosylate counterion remains separated from DNA complex, which is similar to its interaction with DNA-DDDA complex in organic media. These data also strongly suggest that the intercalation state of TMPyP is more stable than its adsorption state in grooves when it is recognized with DNA. The present results are significant for the designs of both laser dye and conductive materials.

[Mechanism of isinglass in prevention and treatment of chronic atrophic gastritis in rats].

OBJECTIVE: To investigate the mechanism of isinglass in the prevention and treatment of chronic atrophic gastritis (CAG) in rats. METHOD: Animal models of SD rats with CAG were made in accordance with the previous experience of combined administration of 60% ethanol, 20 mmol x L(-1) sodium deoxycholate and 0.1% ammonia water. In prevention groups, sucralfate and isinglass were used as preventive therapy while CAG rat model was being made. In the reverse groups, sucralfate and isinglass were used to treat rats after establishment of CAG rat model. Finally all the rats were executed. Then the length of the proliferation zone of the gastric mucosa and serum epidermal growth factors (EGF) and growth hormones (GH)level were studied. RESULT: In isinglass prevention groups and high dose isinglass reverse group, the length of the proliferation zone of the gastric mucosa was very close to that in normal control group (P > 0.05), much better than model control group (P < 0.01). In low dose isinglass reverse group, it was lower than that in normal control group (P < 0.01), but much better than model control group (P < 0.01). In both prevention and reverse groups, serum EGF level was higher than that in normal (P < 0.01) and model control group (P < 0.05). Serum GH level was the same in every group (P > 0.05). CONCLUSION: The mechanism of isinglass in the prevention and treatment of CAG rats lies in revitalizing and proliferating gastric mucosal cells by stimulating endogenous EGF secretion.
