Mechanisms underlying PTEN regulation of vascular endothelial growth factor and angiogenesis.

Inactivation of the tumor suppressor gene PTEN and overexpression of VEGF are two of the most common events observed in high-grade malignant gliomas. The purpose of this study was to determine whether PTEN controls VEGF expression in gliomas under normoxic conditions. Transfer of PTEN to human glioma cells resulted in the transduction of a functional PTEN protein as evidenced by the upregulation of p27 and modification of the phosphorylation status of Akt. Under normoxic conditions, enzyme-linked immunosorbent assay and Northern blot analyses showed downregulation of VEGF in PTEN-treated cells. Moreover, conditioned media from PTEN-treated glioma cells significantly diminished the ability of endothelial cells to grow and migrate. Western blot assays demonstrated that, in a normoxic environment, PTEN downregulates HIF-1 alpha. Finally, promoter activity assays showed that the VEGF promoter region containing the HIF-1alpha binding site is necessary and sufficient for PTEN-mediated downregulation of VEGF. Experiments with PI3-K inhibitors and kinase assays suggested that PI3-K is mediating the effect of PTEN on VEGF, and not the p42/p48 or p38 MAP kinases. These results indicate that restoration of PTEN function in gliomas may induce therapeutic effect by downregulating VEGF. Furthermore, this close functional relationship between PTEN and VEGF suggests that a better understanding of the transduction signal regulated by PTEN might enhance the knowledge of the cause and physiology of vascular and inflammatory diseases.

Molecular epidemiological investigation of Ascaris genotypes in China based on single-strand conformation polymorphism analysis of ribosomal DNA.

Using a single-strand conformation polymorphism-based approach, we investigated nucleotide variation in part of the first internal transcribed spacer (pITS-1) of nuclear ribosomal DNA within and among a large number of Ascaris individuals from humans and pigs from six endemic regions in China, and examined the frequency of the different genotypes of Ascaris in relation to host species and geographical origin. Five different SSCP genotypes (G1-G5) were recorded for human Ascaris (n = 486), of which three (genotypes G1-G3) were detected for pig Ascaris (n = 329). Of the five Ascaris genotypes detected, genotype G1 predominantly infected humans (approximately 63-74%) whereas genotype G3 infected mainly pigs (approximately 79-86%), indicating that each of these genotypes has a particular host affiliation. In contrast, the frequencies of the other three genotypes was substantially lower for each of the two host species. The findings also suggested that the rate of cross infection of Ascaris between humans and pigs is relatively low and that gene flow between the predominant genotypes is limited, consistent with previous proposals for endemic regions in other countries. While the nature and extent of nucleotide variation in the pITS-1 (and the proposal of host affiliated Ascaris populations) may relate to "introgression" or "lineage sorting and retention of ancentral polymorphism", other explanations are possible. Evidence of multiple pITS-1 sequence types in some Ascaris individuals representing particular genotypes (e.g., G2 and G5) may suggest hybridization between human- and pig-affiliated Ascaris. This aspect and the species status of Ascaris (from each host species) warrant future experimental testing, employing the pig/Ascaris model and the present electrophoretic approach.*

Comparative analysis of mitochondrial genome data for Necator americanus from two endemic regions reveals substantial genetic variation.

Necator americanus is a blood-sucking, intestinal nematode of major human health importance in many tropical and subtropical regions of the world. The aim of the present study was to compare the complete mitochondrial genome sequence from one N. americanus individual from Togo with another from China, in order to estimate the magnitude of genetic variability for different mitochondrial genes and non-coding regions. For the 12 protein genes, this comparison revealed sequence differences at both the nucleotide (3-7%) and amino acid (1-7%) levels. The most conserved of these was the nad4L gene, whereas the nad1 gene was least conserved at both the nucleotide and amino acid levels. Nucleotide differences were also detected in 14 of the 22 transfer RNAs (trns) (1-13%), the AT-rich region ( approximately 8%), non-coding regions (8-25%) and in the small (rrnS) and large (rrnL) subunits of mitochondrial ribosomal RNA (rrn) ( approximately 1%). Comparison of the rrnL sequences among multiple individual worms revealed nine unequivocal nucleotide differences between N. americanus from the two countries. Consistent with previous studies, these findings provide evidence for substantial genetic variation within N. americanus, which may have implications for the transmission and control of hookworm disease.

The mitochondrial genome of Strongyloides stercoralis (Nematoda) - idiosyncratic gene order and evolutionary implications.

The complete mitochondrial genome sequence of the parasitic nematode Strongyloides stercoralis was determined, and its organisation and structure compared with other nematodes for which complete mitochondrial sequence data were available. The mitochondrial genome of S. stercoralis is 13,758 bp in size and contains 36 genes (all transcribed in the clockwise direction) but lacks the atp8 gene. This genome has a high T content (55.9%) and a low C content (8.3%). Corresponding to this T content, there are 16 (poly-T) tracts of >/=12 Ts distributed across the genome. In protein-coding genes, the T bias is greatest (76.4%) at the third codon position compared with the first and second codon positions. Also, the C content is higher at the first (9.3%) and second (13.4%) codon positions than at the third (2%) position. These nucleotide biases have a significant effect on predicted codon usage patterns and, hence, on amino acid compositions of the mitochondrial proteins. Interestingly, six of the 12 protein-coding genes are predicted to employ a unique initiation codon (TTT), which has not yet been reported for any other animal mitochondrial genome. The secondary structures predicted for the 22 transfer RNA (trn) genes and the two ribosomal RNA (rrn) genes are similar to those of other nematodes. In contrast, the gene arrangement in the mitochondrial genome of S. stercoralis is different from all other nematodes studied to date, revealing only a limited number of shared gene boundaries (atp6-nad2 and cox2-rrnL). Evolutionary analyses of mitochondrial nucleotide and amino acid sequence data sets for S. stercoralis and seven other nematodes demonstrate that the mitochondrial genome provides a rich source of phylogenetically informative characters. In conclusion, the S. stercoralis mitochondrial genome, with its unique gene order and characteristics, should provide a resource for comparative mitochondrial genomics and systematics studies of parasitic nematodes.

Population pharmacokinetics of carbamazepine in Chinese epilepsy patients.

AIM: To investigate the pharmacokinetic profile of carbamazepine (CBZ) in Chinese epilepsy patients. MATERIALS AND METHODS: Serum samples through concentrations at steady state (n = 687) were collected prospectively from 585 patients during routine clinical care. Data were analyzed by the non-linear mixed-effect modeling (NONMEM) technique with a one-compartment model of first-order absorption and elimination. RESULTS: The important determinants of clearance (CL) were total body weight (TBW); dose; patient age over 65 years (E); and comedication with phenytoin (PHT), phenobarbital (PB), or valproic acid (VPA) when VPA daily dose was greater than 18 mg/kg. The final pharmacokinetic model for relative CL and apparent distribution volume (V) were: Equation CONCLUSION: A population pharmacokinetic model was proposed to estimate the individual CL for Chinese patients receiving CBZ in terms of patient's dose, TBW, and comedications to establish a priori dosage regimens.

[Induction of lymphangiogenesis by mouse ascites hepatoma cell H22 in vitro].

OBJECTIVE: To construct lymphangiogenesis model of Balb/c mouse in vitro to observe the impact of mouse ascites hepatoma cell H22 on the lymphangiogenesis and lymphatic endothelial proliferation. METHODS: Intraperitoneal benign lymphangioma induced by incomplete Freund's adjuvant in Balb/c mouse was harvested and embedded in fibrin gel to culture in vitro with conditioned medium (CM) of H22 cell. The gels, observed by inverted microscope and transmission electron microscope, were then digested to separate the cells. The expression of Flt-4, c-fos and iNOS in the cell and the content of NO in medium were studied by immunohistochemical and histochemical method. RESULTS: Lymphatic capillary generating from lymphangioma specimen in the gels was observed by inverted microscope and transmission electron microscope. H22 CM was observed to enhance growth of the vessels, induce expressions of Flt-4, c-fos and iNOS in the separated cells and increase content of NO in the medium. CONCLUSION: The model is convenient for in vitro study of lymphangiogenesis. H22 cell can accelerate lymphangiogenesis, which warrants further study for the understanding of the role of lymphangiogenesis in metastasis.

[Using TiNi-SMA distractor for mandibular alveolar ridge augmentation by distraction osteogenesis: a preliminary study].

OBJECTIVE: To study the possibility of mandibular alveolar ridge augmentation by distraction osteogenesis using TiNi-SMA (shape memory alloy) distractor. METHODS: All of the mandibular premolars and the first molar were extracted from ten adult mongrel canines. One month later, box-shaped osteotomies of the alveolar bone were performed out and TiNi-SMA distractors were buried in experimental group. The bone height was measured before and 1, 5, 13 weeks after the operation. The radiographs were taken on the day of, and 1, 5, 13 weeks after operation. The animals were killed at one or three months after distraction and examined histologically. RESULTS: The bone segments were lifted up immediately after the insertion of distractors and the height achieved 7.5 - 11.5 mm one week after the operation. One month after distraction, the radiographs showed there was regeneration of bone in distraction gap. The new bone and the normal bone could not be differentiated clearly three months after distraction. No bone resorption was found during this period. In histological sections, there were collagen bundles in early distraction, then these bundles were calcified and became traeculaes. CONCLUSION: It suggests that the canine mandibular posterior alveolar ridge can be augmented by distraction osteogenesis using TiNi-SMA distractor, and the new bone is formed by intramembranous ossification.

[Hypermethylation of hMLH1 and microsatellite instability in ovarian mucinous tumors].

OBJECTIVES: To investigate the role of hMLH1 promoter hypermethylation and microsatellite instability (MSI) in the development of ovarian mucinous tumors. METHODS: One hundred and seven of paraffin-embedded specimens of ovarian mucinous tumors (malignant 49, borderline 35, and benign 23) were collected from Women's Hospital, School of Medicine, Zhejiang University from 1995 to 2001. The assessment of MSI was based on the use of a panel of six microsatellite markers (BAT-25, BAT-26, BAT-40, D5S346, D17S250, and D2S123) by polymerase chain reaction (PCR). Hypermethylation of hMLH1 promoter region was detected using restriction cut analysis. RESULTS: 4.3% (1/23), 14.3% (5/35), and 36.7% (18/49) of benign tumors, borderline tumors, and malignant tumors respectively displayed hypermethylation of the hMLH1 promoter. The hMLH1 promoter hypermethylation rate of malignant group was significantly higher than that of borderline and benign group (P = 0.023, 0.004), but no significant difference between the borderline group and the benign group (P = 0.438); 4.3% (1/23), 8.6% (3/35), and 16.3% (8.49) of benign tumors, borderline tumors, and malignant tumors showed MSI positive phenotype. But there were no significant differences each other in the MSI positive phenotype rate; 75% (9/12) MSI positive phenotype ovarian mucinous tumors were hypermethylated at hMLH1 promoter, while the MSI-phenotype tumors were unmethylated in 84.2% (80.95) of cases. There was significant correlation between MSI positive phenotype and hMLH1 promoter hypermethylation (P = 0.000). CONCLUSIONS: In ovarian mucinous tumors, malignant, borderline, and benign tumors exist hMLH1 promoter hypermethylation. Hypermethylation of hMLH1 promoter results MSI in ovarian mucinous tumors. Methylation of hMLH1 promoter and MSI may be involved in the carcinogenesis of ovarian mucinous cancer.

[Purification of recombinant Schistosoma japonicum signaling protein 14-3-3 and antibody preparation].

OBJECTIVE: To prepare and characterize the polyclonal and monoclonal antibodies(McAbs) against Schistosoma japonicum(Sj) signaling protein 14-3-3. METHODS: Recombinant Sj14-3-3 protein was expressed and the gel slices were lyophilized. Rabbits were immunized with the ground gel powder to prepare polyclonal antibodies against Sj14-3-3. BALB/c mice were immunized with recombinant Sj14-3-3 purified by electroelution. The anti-rSj14-3-3 monoclonal antibody was obtained by using hybridoma technique. Characterization of the antibodies was performed by enzyme linked immunosorbent assay (ELISA) and Western blotting analysis. RESULTS: Purified recombinant Sj14-3-3 protein was obtained. The titers of immune sera from the animals were 1:8-1:64. The monoclonal antibody(McAb) against 14-3-3 antigen was obtained and its subclass was found to be IgG1. The McAb reacted strongly and specifically with the expressed Sj14-3-3 protein. CONCLUSION: The anti-14-3-3 sera were obtained and the hybridoma cell lines which secreted stable monoclonal antibodies against Sj14-3-3 were established, which provides a new approach to study the role of Sj14-3-3 in the signal transduction of S. japonicum.

[Particle enhanced immunoturbidimetic assay of high sensitive C-reactive protein].

OBJECTIVE: To evaluate the particle enhanced immunoturbidimetic assay of high sensitive C-reactive protein by the automated chemistry analyzer (Hitachi 7170A). METHODS: The detection limit of quantification, measurement range, precision, interference and reference interval were assessed. RESULTS: The measurement range of the method was from 0.125 to 12.5 mg/L. The detection limitation was 0.115 mg/L. The internal and external accuracy were 0.89% and 0.90% respectively. The reference range of the assay was from 0.20 mg/L to 2.60 mg/L. CONCLUSION: The particle enhanced immunoturbidimetic assay is sensitive and stable, and can be used in clinical diagnosis.

[Reconstruction of segmental mandibular defect of canine using titanium-nickel distractor].

OBJECTIVE: To establish a continuous and automatic distraction osteogenesis technique in reconstruction of segmental mandibular defect by using embedded titanium-nickel alloy distractor with characteristics of shape-memory and super-elasticity. METHODS: Adult hybrid canines were used as the animal model. Segmental defects of 1-3 cm in the body of mandible were created by surgical osteotomy. Bi-focal distraction osteogenesis was applied using embedded titanium-nickel distractor designed by the authors. The canines were sacrificed 3 months after the operation and the mandibles were harvested to examine the results of bone regeneration. RESULTS: The histocompatibility of titanium-nickel distractors was good. Distraction osteogenesis was completed automatically and the defects were elementarily restored. Radiological and histological examination showed well bone regeneration in distraction area. CONCLUSIONS: Distraction osteogenesis using embedded self-loading titanium-nickel distractor could be a hopeful and useful technique. It might help to solve the problems of functional mandibular reconstruction in the near future.

Distributions of dimethylsulfide in the Bohai sea and Yellow Sea of China.

Dimethylsulfide (DMS) measurements in the surface seawater of China eastern coastline were conducted during March 9-10, 1993 in Bohai Sea along the cruise from Dalian to Tianjin and during September 24-25, 1994 in Yellow Sea along the cruise from Shanghai to Qingdao. On the cruise in Bohai Sea DMS concentrations varied from 0.11 to 2.63 nmol/L with an average of 1.31 nmol/L, while DMS flux was estimated to be 0.85 micromol/(m2 x d) with the range of 0.04-3.12 micromol/(m2 x d). On the cruise in Yellow Sea DMS concentrations varied from 0.95 to 7.48 nmol/L with an average of 2.89 nmol/L, and DMS flux was estimated to be 7.94 micromol/(m2 x d) with the range of 0.11-18.88 micromol/(m2 x d). Variations in DMS concentrations along the latitude in Yellow Sea were observed larger than those along the longitude in Bohai Sea. DMS concentrations and fluxes had a similar spatial trend both in Bohai Sea and Yellow Sea with the correlation coefficients of 0.75 and 0.64, respectively.

Immunoblot analysis of Schistosoma japonicum egg antigens with sera from patients with acute and chronic schistosomiasis japonica.

Humoral immune responses of IgG, IgM, IgA, IgE and IgG subclass antibodies to Schistosoma japonicum egg antigens were determined by immunoblotting with serum samples from individuals in China with acute (n=24) or chronic (n=35) schistosomiasis. In general, IgM, IgA, and IgE in sera from acute patients exhibited strong binding to antigens but binding was much weaker in chronic cases. Reaction of IgG4 of chronic cases was stronger than that of IgG4 of acute cases. The recognition profile of each antibody isotype in sera was analyzed for 11 major antigen molecules (antigens with apparent molecular weights of 82, 76, 61, 57, 53, 46, 40, 32, 27, 10 and less than 6.5 kDa). Except for the 10 kDa molecule, they were well-recognized by IgA and IgE in sera of acute cases. In other combinations of antibody class and clinical phase, recognition patterns against these molecules differed among individuals. Notably, the 10 kDa molecule was specifically recognized by total IgG and IgG4 in sera from most of the chronic patients, but in sera from only one acute case. This result suggests that the 10 kDa molecule is one of the major target antigens of IgG4 and may be useful as a marker antigen to characterize the clinical phases of S. japonicum infection.

Crystal structure of a UBP-family deubiquitinating enzyme in isolation and in complex with ubiquitin aldehyde.

The ubiquitin-specific processing protease (UBP) family of deubiquitinating enzymes plays an essential role in numerous cellular processes. HAUSP, a representative UBP, specifically deubiquitinates and hence stabilizes the tumor suppressor protein p53. Here, we report the crystal structures of the 40 kDa catalytic core domain of HAUSP in isolation and in complex with ubiquitin aldehyde. These studies reveal that the UBP deubiquitinating enzymes exhibit a conserved three-domain architecture, comprising Fingers, Palm, and Thumb. The leaving ubiquitin moiety is specifically coordinated by the Fingers, with its C terminus placed in the active site between the Palm and the Thumb. Binding by ubiquitin aldehyde induces a drastic conformational change in the active site that realigns the catalytic triad residues for catalysis.

Kinetics of reduction of ferrylmyoglobin by (-)-epigallocatechin gallate and green tea extract.

The hypervalent heme pigment ferrylmyoglobin, a potential prooxidant in muscle tissue and meat, is efficiently reduced by epigallocatechin gallate (EGCG) from green tea and by green tea polyphenol extract (GTP) in neutral or moderately acidic aqueous solution (0.16 M NaCl) to yield metmyoglobin in two parallel processes. The second-order rate constant for direct reduction at pH 7.4 and 25 degrees C was found to have the value 1170 +/- 83 M(-1).s(-1) and activation parameters DeltaH(#) = 70.6 +/- 7.2 kJ.mol(-1) and DeltaS(#) = 50.7 +/- 24.1 J.mol(-1).K(-1) for EGCG and the value 2300 +/- 77 M(-1).s(-1) and parameters DeltaH(#) = 60.6 +/- 2.6 kJ.mol(-1) and DeltaS(#) = 23 +/- 9 J.mol(-1).K(-1) for GTP (based on EGCG concentration). For decreasing pH, the rate increased moderately due to a parallel reduction of protonated ferrylmyoglobin. At physiological pH, EGCG is more efficient in deactivating ferrylmyoglobin than other plant phenols investigated, and the relatively high enthalpy and positive entropy of activation suggest an outer-sphere electron transfer mechanism. The interaction between EGCG and other tea catechins in GTP could be responsible for the even stronger ability for GTP to deactivate ferrylmyoglobin.

[Concentrations of aerosol and related gases in Beijing].

The measurement was made in 3 periods from May to September of 2000 in Beijing. The measurement species included SO4(2-), NO3-, NO2-, Cl- of aerosol and related gases. The hourly variation of the concentration was presented. It showed that NO3-, NO2-, Cl- of aerosol and HCl, HNO2, HNO3 of gas varied with regularity. The variation of gas SO2 and aerosol SO4(2-) depended on the emission of SO2. The peak time of SO4(2-) was about 2 hours later than SO2.

Detection of hepatitis B virus by piezoelectric biosensor.

A highly sensitive piezoelectric HBV DNA biosensor has been developed based on the sensitive mass-transducing function of the quartz crystal microbalance and the speciality of nucleic acid hybridization reaction. HBV nucleic acid probe was immobilized onto the gold electrodes of a 9 MHz AT-cut piezoelectric quartz crystal with the polyethyleneimine adhesion, glutaraldehyde cross-linking (PEI-Glu) method or the physical adsorption method. The coated crystal with the PEI-Glu method to immobilized HBV nucleic acid probe showed the better results than the physical adsorption method with respect to sensitivity reproducibility and stability. The frequency shifts of hybridization have better linear relationship with the amount of HBV DNA, when the amount was in range 0.02-0.14 microg/ml. The crystal could be regenerated nearly five times without perceptible decrease of sensitivity.

Single-strand conformation polymorphism-based analysis of mitochondrial cytochrome c oxidase subunit 1 reveals significant substructuring in hookworm populations.

Sequence heterogeneity in a portion of the mitochondrial cytochrome c oxidase subunit 1 gene (pcox1) was measured for the hookworms, Ancylostoma caninum from Australia, A. duodenale from China, and Necator americanus from China and Togo using single-strand conformation polymorphism (SSCP) analysis combined with DNA sequencing. The pcox1 sequences were characterised for individual nematodes displaying genetic variation within each of the three species, and those were compared with pcox1 sequences of four other species of hookworm. While intraspecific variation in the pcox1 sequence ranged from 0.5 to 8.6% for A. caninum, 0.3 to 3.3% forA. duodenale, and 0.3 to 4.3% for N. americanus, interspecific differences varied from 4.8 to 12.9%. Sequence data also provided information on nucleotide compositions and substitution patterns. Genetically distinct groups were detected within A. caninum and A. duodenale, indicating significant population substructuring within these species. Also, N. americanus individuals from China all differed from those from Togo at four nucleotide positions, supporting a previous proposal (based on ribosomal DNA sequence data) that N. americanus may represent a species complex. The findings indicated the value of pcox1 sequence data and the mutation scanning approach for studying the genetic structures of hookworm populations, which should have important epidemiological relevance.
