Cells in G2/M phase increased in human nasopharyngeal carcinoma cell line by EBV-LMP1 through activation of NF-kappaB and AP-1.

Although previous studies showed that the principal oncoprotein encoded by Epstein-Barr virus, latent membrane protein 1(LMP1), could induce the nasopharyngeal carcinoma cells in G2/M phase increased, little is known about the target molecules and mechanisms. The present study demonstrated that LMP1 could induce the accumulation of p53 protein and upregulate its transactivity in a dose dependent manner, which resulted in the decrease of the kinase activity of cdc2/cyclin B complex and inducing arrest at G2/M phase through the activation of NF-kappaB and AP-1 signaling pathways, and the effect of NF-kappaB was more obvious than that of AP-1. This study provided some significant evidence for further elucidating the molecular mechanisms that LMP1 had effects on the surveillance mechanism of cell cycle and promoting the survival of transformed cells and tumorigenesis.

Effects of weaning and rearing environment on intestinal gene expression of IGF-I, IGFBP (1-6), and the IGF receptor and on specific binding of IGF-I to mucosal membranes in the pig.

The objective of this study was to investigate changes in gene expression of intestinal IGF-I, IGFBPs, and IGF-I receptor in pigs in response to weaning and different rearing environment. Pigs were weaned early at 12 days of age and either remained on-site in a separate facility (CON) or were moved to a segregated site with reduced infection pressure (segregated early weaning; SEW). Small intestinal samples were collected from a total of 15 pigs killed at 11 (pre-weaning), 15 (3 days post-weaning), and 34 days of age. Intestinal IGF-I mRNA levels were higher (P < 0.01) in SEW than in CON pigs at 3 days post-weaning, but not at 34 days of age. Weaning reduced (P < 0.05) both IGF-IR mRNA levels and specific binding of IGF-1 in the jejunum in both groups at day 34, but only in SEW pigs (P < 0.05) at day 3 post-weaning. Weaning resulted in a major reduction (P < 0.05) in intestinal IGFBP-2 mRNA, with no difference between SEW and CON. Intestinal IGFBP-3 mRNA levels were unaffected by weaning or post-weaning environment. Weaning did not affect intestinal IGFBP-4 mRNA levels, except for an increase (P < 0.05) in CON pigs compared to pre-weaning, and to SEW pigs at 3 days post-weaning. The abundance of IGFBP-5 mRNA in the gut was highly variable with no apparent treatment effect. Intestinal IGFBP-6 mRNA levels were reduced (P < 0.05) after weaning, with lower (P < 0.05) levels in SEW pigs than in CON pigs at 34 days of age. This study documents the changes in IGF-1, IGF-IR, and IGFBP mRNA abundance, and in IGF-1 binding during post-weaning adaptation of the intestine in early-weaned pigs. In addition, the relative differences observed in intestinal expression of IGF-1, IGF-IR, and in IGF-1 binding between the post-weaning environments are consistent with previous observations in a companion study indicating that segregated early weaning enhances post-weaning intestinal maturation in pigs.

Epstein-Barr virus induces human nasopharyngeal epithelial cells to escape from the replicative senescence.

OBJECTIVE: To observe the biological changes of primary human nasopharyngeal epithelial cells in the early stage of immortalization. METHODS: The morphological changes of nasopharyngeal epithelial cells were observed by phase contrast microscopy, and the activity profile of senescence-associated beta-galactosidase (SA-beta-Gal) was detected by SA-beta-Gal staining. The expression of p16(INK4a) protein was tested by immunochemical assay, and the life span in vitro of nasopharyngeal epithelial cells was calculated as population doublings. In addition, the expression of Epstein-Barr (EB) virus latent membrane protein 1 (LMP1) was also detected by immunofluorescence staining. RESULTS: Morphologically, cells treated with EB virus and 12-o-tetradecanoyl-phorbol-13-acetate (TPA) formed multi-layer foci, and their cellular life span in vitro was extended (about 155 days of culture). A low percentage of cells (about 4.8%) expressed SA-beta-Gal activity at late primary culture, and did not always express p16(INK4a) protein in the progression of culture. CONCLUSIONS: Nasopharyngeal epithelial cells treated with EB virus in cooperation with TPA can pass through the stage of senescence and enter the early stage of immortalization. Some changes of phenotype occur in these cells. Our results provide data for further studying the mechanism of immortalization and the establishment of a human nasopharyngeal epithelial cell line.

[EB virus encoded latent membrane protein 1 modulates the phosphorylation of epidermal growth factor receptor in nasopharyngeal carcinoma cell line].

OBJECTIVE: To elucidate the regulation of the phosphorylation of epidermal growth factor receptor (EGFR) by the EB virus encoded latent membrane protein 1 (LMP1) in nasopharyngeal carcinoma cell line. METHODS: The levels of EGFR expression and phosphorylation in pTet-on LMP1 HNE2 cell, a nasopharyngeal carcinoma (NPC) cell line, in the dynamic expression of LMP1 induced by different concentrations of doxycycline (Dox) were observed. The EGFR dominant negative mutant and LMP1 antisense expression plasmid were transiently transfected into pTet-on LMP1 HNE2 cells by lipofectamine, and the changes in EGFR phosphorylation were observed by immunocoprecitation and Western blot. The changes in EGFR phosphorylation were observed after EGF treatment. RESULTS: In pTet-on LMP1 HNE2 cells, Dox-induced LMP1 upregulated EGFR expression and phosphorylation in a dose-dependent manner. After EGFR dominant negative mutant was transfected into pTet-on LMP1 HNE2 cells, the increase of EGFR phosphorylation was inhibited completely. When LMP1 antisense expression plasmid was transfected into pTet-on LMP1 HNE2 cells, the levels of EGFR phosphorylation were also inhibited significantly. Meanwhile, after EGF had been added into pTet-on LMP1 HNE2 cells, increase of EGFR phosphorylation was induced, but it was completely blocked by EGFR dominant negative mutant and the introduction of LMP1 antisense. CONCLUSION: EB virus encoded LMP1 not only induces the dose-dependent expression of EGFR, but also the dose-dependent phosphorylation of EGFR. The phosporylation of EGFR may play a vital role in the development of nasopharyngeal carcinoma.

[The prevalence of permanent caries in adolescent in Sichuan province].

OBJECTIVE: The aim of this survey was to obtain some information of the prevalence of dental caries in the youth aged 12, 15 and 18 years of Sichuan province. METHODS: Based on WHO and national oral health survey standards, dental caries prevalence and treatment needs were investigated randomly in selected young individuals of Sichuan province in 1995. RESULTS: The prevalent rates of caries of 12-, 15-, and 18-year-old adolescents were 28.47% and 36.40% and 39.54% respectively. The mean DMFT of 12-, 15- and 18-year-old were 0.52, 0.81, and 0.99. There are more pit and fissure caries than smooth caries in the young of 12-year-old than those of 15- and 18-year-old. CONCLUSION: Despite improvement can be seen in adolescents' oral health in Sichuan, there remains a proportion of active untreated decays. Some simple prevention programs are needed to stabilize the caries.

[Study of infrared spectra of polyaluminum ferric chloride].

Mid-IR spectra of polyaluminum ferric chloride (PAFC) with different Al/Fe molar ratio were studied by Fourier transform infrared spectroscopy. Some vibration bands were assigned. In the range of Al/Fe molar ratio < 3:7 and > 6:4, the delta bending vibration frequencies at 850-880 cm-1 of [formula: see text] and 930-970 cm-1 of [formula: see text] in PAFC vs Al/Fe molar ratio are linearly relalional. It shows that Fe(III) and Al(III) were taken place each other in [formula: see text] and [formula: see text], and these were the evidences for the presence of [formula: see text]. When Al/Fe molar ratio was close, [formula: see text] and [formula: see text] Al bulk bending vibration bands at 680 cm-1 and 625 cm-1 respectively became an overlapped and broad band at 640 cm-1. Correspondingly, the [formula: see text] and [formula: see text] delta bending vibration bands were weak markedly or even disappear. And these were the evidences for the presence of co-aggragation of Fe(III) and Al(III) hydroxyl polymers (Aluminous ferrihydrite). Intensity and frequency change of H-OH delta vibrations bands at 1,610-1,630 cm-1 vs Al/Fe molar ratio show: the amounts of coordinating water in Al-Fe hydroxyl co-aggragation compounds of PAFC are the highest and no longer change, as well as, the force of banding of coordinating water with center atoms is increasing when Al/Fe molar ratio > 6:4. These suggested that there are species structural Al-Fe hydroxyl co-aggragatin compounds and this is one of the causes that stability of PAFC solution can be keeped for a long time.

Solving geometric constraints with genetic simulated annealing algorithm.

This paper applies genetic simulated annealing algorithm (SAGA) to solving geometric constraint problems. This method makes full use of the advantages of SAGA and can handle under-/over- constraint problems naturally. It has advantages (due to its not being sensitive to the initial values) over the Newton-Raphson method, and its yielding of multiple solutions, is an advantage over other optimal methods for multi-solution constraint system. Our experiments have proved the robustness and efficiency of this method.
