Gene expression profile in immunologically injured liver cell of mice.

To study the gene expression profiles between immunologically injured liver cell and normal liver cell of mice and to screen on a large scale the differentially expressed genes associated with the formation of liver injury, the experimental mice were randomly divided into the normal group for controlling and the immunologically liver-injured group induced by BCG and LPS. The liver mRNA of the two groups were extracted respectively and reversely-transcribed to cDNA with the incorporation of different fluorescence (Cy3, Cy5) labeled dUTP as the hybridization probes. The mixed probes were hybridized to the cDNA microarray chips. The fluorescent signal results were acquired by scanner ScanArray 4000 and analyzed with software GenePix Pro 3.0. Among the 14112 target genes, 293 genes were found to be significantly differentially expressed, in which 188 genes were up-regulated and 105 genes were down-regulated. Based on the analysis of biological functions of those differentially expressed genes, it was indicated that the occurrence and development of mouse liver damage induced by BCG and LPS were highly correlated with the processes of immune reactions, cell synthesis, metabolism, apoptosis and transportation in liver cell, which might be quite important for elucidating the regulatory network of gene expression associated with the liver damage, also important for finally discovering the pathogenic mechanisms of immunological liver damage.

[The value of dynamic color duplex scanning in the diagnosis of vascular erectile dysfunction].

OBJECTIVE: To evaluate deep penile arterial flow after an intracavernosal injection with prostaglandin E1 in patients with erectile dysfunction (ED). METHODS: 527 patients with ED were evaluated using Color Doppler Ultrasonography. Diagnostic tests were undertaken after an intracavernosal injection with 20 microg prostaglandin El. The peak systolic velocity (PSV), end diastolic velocity (EDV) and resistive index (RI) were analysed. RESULTS: There were 112 patients (26.99%) with nonvasculogenic ED, 207 patients (49.88%) with arteriogenic ED, 144 patients (34.70%)with venogenic ED, 64 patients (15.42%)with mixed ED. CONCLUSION: The Color Doppler Ultrasonography technique allows accurate location and evaluation of deep penile arteries. Vascular pathology may be differentiated after an intracavernosal injection with a vasomotor agent. Recognising the pathological pattern assists in choosing the best method of treatment.

Differential activation of LFA-1 and Mac-1 ligand binding domains.

Nine integrin alpha subunits contain an 'inserted' or I-domain, known to involve in ligand binding. Mutation of an invariant isoleucine residue in the I-domains of alphaL and alphaM has previously been reported to activate LFA-1 and Mac-1, respectively. In this article, we report notable differences in the regulation of adhesion of these two integrins. We find that mutation of the isoleucine residue in the proposed "socket for isoleucine" in full-length alphaL does not lead to an active LFA-1, although mutation of the equivalent residue in alphaM does convey constitutive activity to Mac-1. In addition, we observe the isolated I-domain of alphaL to be constitutively active. This challenges reports that state the alphaL I-domain exists in an inactive, closed conformation, and requires the presence of activating agents for ligand binding. These results shed further light on the many questions surrounding regulation of integrin activation.

[Comparing studies on the differences of CCl4 liver injury and immunity liver injury mice models].

Comparison between CCl4 liver injury model and immunity liver injury model was studied through physiological, biochemical and gene expression profile methods, and the pathological mechanism of them were discussed. There are significant changes about the physiological, biochemical parameters including ALT,AST between two model groups and normal control group. Among the 14112 target genes, 379 and 293 differentially expressed genes were screened out from the both two model groups respectively in gene expression profile experiments, including 105 common, differentially expressed genes. The results indicate that the two models of CCl4 and immunity have some similarities. However, the differences between them are also obvious. It is useful for the further investigation on the liver-injury mechanism and pathological process of the two different liver-injury models.

[Changes of gene expression profiles in CCl4 injured liver of mice].

AIM: To study the gene expression profiles between the CCl4 injured liver and normal liver in mice, and to screen the differentially expressed genes that relate to liver injury by CCl4 on a large scale using cDNA microarrays. METHODS: Male Kunming strain mice were divided into two groups: one was control group and another was CCl4 injured liver group that was given 0.1% CCI4 oil solution ip at dose of 10 mL x kg(-1) every three days, totally for ten times. Then mRNA in livers of the two groups of mice was extracted, separately, and reversely transcribed to cDNA with the incorporation of different fluorescent-labeled dUTP as the hybridization probes. The mixed probes were hybridized to the cDNA microarrays. The fluorescent signal values were acquired by scanner and analyzed with statistical software. RESULTS: Among the 14 100 target genes, 379 genes were differentially expressed, in which 163 genes were up-regulated and the other 216 genes were down-regulated. They are closely related to a range of biological functions. CONCLUSION: Using the cDNA microarray and experimental animal modeling technique, the differentially expressed genes of CCl4 injured liver in mice on a large scale could be studied. It is useful for further investigation of the injury mechanism of CCl4.

The integrin alpha L beta 2 hybrid domain serves as a link for the propagation of activation signal from its stalk regions to the I-like domain.

Integrin activation involves global conformational changes as demonstrated by various functional and structural analyses. The integrin beta hybrid domain is proposed to be involved in the propagation of this activation signal. Our previous study showed that the integrin beta(2)-specific monoclonal antibody 7E4 abrogates monoclonal antibody KIM185-activated but not Mg(2+)/EGTA-activated leukocyte function-associated antigen-1 (LFA-1; alpha(L)beta(2))-mediated adhesion to ICAM-1. Here we investigated the allosteric inhibitory property of 7E4. By using human/mouse chimeras and substitution mutations, the epitope of 7E4 was mapped to Val(407), located in the mid-region of the beta(2) hybrid domain. Two sets of constitutively active LFA-1 variants were used to examine the effect of 7E4 on LFA-1/ICAM-1 binding. 7E4 attenuated the binding of variants that have modifications to regions membrane proximal with respect to the beta(2) hybrid domain. In contrast, the inhibitory effect was minimal on variants with alterations in the alpha(L) I- and beta(2) I-like domains preceding the hybrid domain. Furthermore, 7E4 abrogated LFA-1/ICAM-1 adhesion of phorbol 12-myristate 13-acetate-treated MOLT-4 cells. Our data demonstrate that interaction between the hybrid and I-like domain is critical for the regulation of LFA-1-mediated adhesion.

Mu rhythm-based cursor control: an offline analysis.

OBJECTIVE: To classify the EEG data recorded in mu rhythm-based cursor control experiments with 4 possible choices. METHODS: The algorithm included preprocessing, feature extraction, and classification. Two spatial filters, common average reference and common spatial subspace decomposition, were used in preprocessing to improve the signal-to-noise ratio, and then two features were extracted based on the power spectrum and the time course of the mu rhythm respectively. A Fisher ratio was defined to select channels in feature extraction. A 2-dimensional linear classifier was trained for final classification. RESULTS: Two types of classifiers were trained for the training dataset. The uniform classifier gave a classification accuracy of 76.4%, and the classifier trained by leave-one-out method gave a classification accuracy of 74.4%, both higher than the online accuracy 69.5%. The uniform classifier was applied to the test dataset and the classification accuracy was 65.9%, lower than the online accuracy 73.2%. CONCLUSIONS: Spatial filtering can give a notable improvement in classification accuracy. The time course of the mu rhythm, as well as the power of the mu rhythm, shows difference between the 4 targets, and can contribute to the classification. SIGNIFICANCE: The spatial filtering, feature extraction and channel selection methods in the algorithm will provide some practical suggestions for further study on the mu rhythm-based brain-computer interface.

Mechanism of the alternating copolymerization of epoxides and CO2 using beta-diiminate zinc catalysts: evidence for a bimetallic epoxide enchainment.

A series of zinc beta-diiminate (BDI) complexes and their solid-state structures, solution dynamics, and copolymerization behavior with CO(2) and cyclohexene oxide (CHO) are reported. Stoichiometric reactions of the copolymerization initiation steps show that zinc alkoxide and bis(trimethylsilyl)amido complexes insert CO(2), whereas zinc acetates react with CHO. [(BDI-2)ZnOMe](2) [(BDI-2) = 2-((2,6-diethylphenyl)amido)-4-((2,6-diethylphenyl)imino)-2-pentene] and (BDI-1)ZnO(i)Pr [(BDI-1) = 2-((2,6-diisopropylphenyl)amido)-4-((2,6-diisopropylphenyl)imino)-2-pentene] react with CO(2) to form [(BDI-2)Zn(mu-OMe)(mu,eta(2)-O(2)COMe)Zn(BDI-2)] and [(BDI-1)Zn(mu,eta(2)-O(2)CO(i)Pr)](2), respectively. (BDI-2)ZnN(SiMe(3))(2) inserts CO(2) and eliminates trimethylsilyl isocyanate to give [(BDI-2)Zn(mu-OSiMe(3))](2). [(BDI-7)Zn(mu-OAc)](2) [(BDI-7) = 3-cyano-2-((2,6-diethylphenyl)amido)-4-((2,6-diethylphenyl)imino)-2-pentene] reacts with 1.0 equiv of CHO to yield [(BDI-7)Zn(mu,eta(2)-OAc)(mu,eta(1)-OCyOAc)Zn(BDI-7)]. Under typical polymerization conditions, rate studies on the copolymerization exhibit no dependence in [CO(2)], a first-order dependence in [CHO], and orders in [Zn](tot) ranging from 1.0 to 1.8 for [(BDI)ZnOAc] complexes. The copolymerizations of CHO (1.98 M in toluene) and 300 psi CO(2) at 50 degrees C using [(BDI-1)ZnOAc] and [(BDI-2)ZnOAc] show orders in [Zn](tot) of 1.73 +/- 0.06 and 1.02 +/- 0.03, respectively. We propose that two zinc complexes are involved in the transition state of the epoxide ring-opening event.

A BCI-based environmental controller for the motion-disabled.

With the development of brain-computer interface (BCI) technology, researchers are now attempting to put current BCI techniques into practical application. This paper presents an environmental controller using a BCI technique based on steady-state visual evoked potential. The system is composed of a stimulator, a digital signal processor, and a trainable infrared remote-controller. The attractive features of this system include noninvasive signal recording, little training requirement, and a high information transfer rate. Our test results have shown that this system can distinguish at least 48 targets and provide a transfer rate up to 68 b/min. The system has been applied to the control of an electric apparatus successfully.

Ectopic intracavernous sinus adrenocorticotropic hormone-secreting microadenoma: could this be a common cause of failed transsphenoidal surgery in Cushing disease? Case report.

Despite diagnostic advances, it remains difficult to identify intrasellar and ectopic parasellar adrenocorticotropic hormone (ACTH)-secreting microadenomas. The authors present the case of a 61-year-old woman with Cushing disease in whom a significant central-to-peripheral and lateralized right-sided ACTH gradient was demonstrated on inferior petrosal sinus sampling; no discernible abnormality was seen on magnetic resonance imaging. She underwent transnasal transsphenoidal surgery. No tumor was found on sellar exploration and a total hypophysectomy was performed, yet her hypercortisolemia persisted. The patient died of cardiac events 17 days postsurgery. Autopsy revealed an isolated, right-sided, intracavernous ACTH-secreting adenoma with no intrasellar communication. This case represents the first failed transsphenoidal surgery for Cushing disease in which there is postmortem confirmation of a suspected intracavernous sinus lesion. It supports the hypothesis that Cushing disease associated with nondiagnostic imaging studies, a strong ACTH gradient on venography, and negative findings on sellar exploration may be caused by an ectopic intracavernous ACTH-secreting adenoma. There are no premortem means of confirming the presence of such lesions, but these tumors could underlie similar cases of failed surgery. Radiation therapy targeting the sella turcica and both cavernous sinuses, possibly supplemented with medical treatment, is suggested for similar patients in whom transsphenoidal hypophysectomy has failed. Adrenalectomy may also be appropriate if a rapid reduction in ACTH is necessary.

Sulfo-SADP (sulfosuccinimidyl[4-azidophenyldithio]propionate) an active site directed reagent inhibiting the NADPH dependent O2- generation of leukocyte cytochrome b(558).

Functional reagents known to bring about the formation of a distinct membrane molecular complex of the subunits of cytochrome b(558) (gp 91(phox) and p22(phox)) were investigated for their influence on the O2- generating capability of liposome incorporated cytochrome b(558) preparations. One, ethyleneglycolbis[sulfo-succinimidylsuccinate], (sulfo-EGS) was found to inhibit O2- generation at concentrations which are known to result in cross-linking the two subunits of cytochrome b(558). Sulfosuccinimidyl [4-azidophenyldithio] propionate, (sulfo-SADP) on the other hand, was found to be a powerful inhibitor of the cytochrome b(558) dependent O2- production at concentrations not able to result in cross linking of the two subunits. Sulfo-SADP inhibits the cytochrome b(558) O2- production 50% at 25 microM, while sulfo-EGS requires 400 microM. For these reagents, the succinimidyl group of sulfo-SADP and sulfo-EGS is the reactive group, which inhibit irreversibly, cytochrome b(558) generation of O2-. Both sulfo-SADP and sulfo-EGS have similar linker arms of 13.9 and 16.1 A, respectively. The difference, accounting for the strong inhibitory profile for sulfo-SADP as compared with sulfo-EGS, resides in the aryl group associated with the sulfo-SADP. The aryl group of sulfo-SADP has been found to be important in directing the specificity of the probe in its inhibition of O2- generation. When the disulfide bond linking the aromatic portion of the probe to the succinimidyl ring is cleaved by DTT (dithiothreitol), the product loses its specificity and has an inhibitory activity with respect to O2- generation comparable to that of sulfo-EGS. The partial protection against the inhibitory influence of sulfo-SADP by NADP(+) indicates that the reagent may interact at the pyridine nucleotide-binding domain of cytochrome b(558). Its low inhibitory titer and its water solubility suggest that sulfo-SADP reacts with a specific amine (the primary reactant for the succinimidyl group) on cytochrome b(558).

Design and implementation of a brain-computer interface with high transfer rates.

This paper presents a brain-computer interface (BCI) that can help users to input phone numbers. The system is based on the steady-state visual evoked potential (SSVEP). Twelve buttons illuminated at different rates were displayed on a computer monitor. The buttons constituted a virtual telephone keypad, representing the ten digits 0-9, BACKSPACE, and ENTER. Users could input phone number by gazing at these buttons. The frequency-coded SSVEP was used to judge which button the user desired. Eight of the thirteen subjects succeeded in ringing the mobile phone using the system. The average transfer rate over all subjects was 27.15 bits/min. The attractive features of the system are noninvasive signal recording, little training required for use, and high information transfer rate. Approaches to improve the performance of the system are discussed.
