Single-cell and spatial transcriptomics define a progenitor subpopulation and fibroinflammatory niche at the leading edge of parathyroid carcinoma.

Parathyroid carcinoma (PC) is a rare but clinically aggressive endocrine malignancy with limited treatment options and a poorly defined tumor microenvironment (TME). To elucidate its cellular heterogeneity and spatial architecture, we integrated single-cell and spatial transcriptomic profiling with whole-exome sequencing and multiplex immunohistochemistry on eight parathyroid neoplasm specimens, including PC, parathyroid adenoma, and atypical parathyroid tumor. We identified a distinct progenitor-like endocrine subpopulation (Ca-1) enriched in CDC73-mutant PC, exhibiting stem-like properties, elevated cell cycle activity, and pronounced genomic instability. Spatial mapping revealed that Ca-1 cells preferentially localize at the leading edge, forming a fibroinflammatory niche characterized by the enrichment of inflammatory cancer-associated fibroblasts (iCAFs) and SPP1+ macrophages. Within this niche, the dipeptidyl peptidase 4 (DPP4) is selectively expressed in Ca-1 cells and iCAFs, implicating a potential paracrine axis driving stromal remodeling and immunosuppression. These findings suggest that a spatially organized ecosystem may promote PC progression through TME remodeling and highlight the DPP4-CXCL2 axis as a candidate pathway for future investigation in aggressive parathyroid neoplasms.

Recycling spent zinc manganese dioxide batteries through synthesizing Zn-Mn ferrite magnetic materials.

A novel process to reclaim spent zinc manganese dioxide batteries (SDBs) through synthesizing Zn-Mn ferrite magnetic materials is present. Firstly, the dismantling, watering, magnetism, baking and griddling steps were consecutively carried out to obtain iron battery shells, zinc grains and manganese compounds using the collected SDBs, and then these separated substances were dissolved with 4 mol L(-1) H(2)SO(4) to prepare FeSO(4), ZnSO(4) and MnSO(4) reactant solutions. Secondly, Zn-Mn ferrites with stoichiometric ratio of Mn(0.26)Zn(0.24)FeO(2) were synthesized using chemical co precipitation process with ammonium oxalate precipitator. The XRD results showed that the obtained Zn-Mn ferrites had spinel structure and high purity at the calcining temperatures of 850-1250 degrees C. With the increase of calcining temperature, the finer crystalline structure could be formed, and their intensity of saturation magnetization reached the highest value at 1150 degrees C. The magnetization performances of Zn-Mn ferrites prepared from the SDBs were similar to that of from analysis reagents, suggesting the feasibility to synthesize Zn-Mn ferrites with high properties from SDBs.

Chemical fingerprint and metabolic fingerprint analysis of Danshen injection by HPLC-UV and HPLC-MS methods.

HPLC-UV and HPLC-MS techniques were used in fingerprint analysis of Danshen injection and its raw materials (roots and rhizoma of Salvia miltiorrhiza). HPLC profiles of Danshen injections from a Chinese pharmaceutical factory and their raw materials were established as their characteristic fingerprint and employed to assess their consistency and difference. To develop the representative fingerprint of Danshen injection, 10 batches of samples were analyzed under the same HPLC conditions. The results showed that 10 batches of Danshen injections had very similar HPLC fingerprints. To characterize the major constituents of Danshen injection for quality control, 11 major chromatographic peaks were characterized by their MS spectra and comparison with the reference standards. Through comparison of the HPLC profiles of Danshen injection with its raw material, it was found that they are greatly different, which indicated the changes of major constituents in the course of preparation procedure. In addition, the rat's plasma was analyzed by HPLC-MS technique after intravenous administration of Danshen injection at different time intervals to explore the in vivo metabolism of the major active constituents. Except for protocatechuic aldehyde, the major phenolic acids in Danshen injection appeared in rat's plasma after intravenous administration, but quantity of each phenolic acids was very different from that in Danshen injection. With the administration time prolonged danshensu and salvianolic acid B disappeared quickly, salvianolic D, lithospermic acid and salvianolic A slowly decreased and maintained relatively high concentration after 30 min of intravenous administration. This indicated that polyphenolic acids were significant for biological activity of Danshen injection. It might be concluded that chemical fingerprint combined with metabolic fingerprint is a useful means to control the quality and to clarify the possible mechanism of action of herbal products.

Metabolic studies on the total phenolic acids from the roots of Salvia miltiorrhiza in rats.

Phenolic acids are the main active constituents of Salvia miltiorrhiza Bunge. The metabolism of total phenolic acids from the roots of Salvia miltiorrhiza in rats was investigated. A sample preparation method combining the solid-phase extraction with liquid-liquid extraction was established to separate metabolites from the biological matrix. HPLC-UV and HPLC-MS methods were employed to analyze the metabolites. Five metabolites (M1-M5) were identified by HPLC-MS analysis and comparison with those of the reference standards. The fi ve metabolites were characterized as danshensu (M1), caffeic acid (M2), ferulic acid (M3), isoferulic acid (M4) and methylized ferulic acid (M5), respectively. The possible metabolic pathway of the phenolic acids is proposed.

Predicting change in adolescent adjustment from change in marital problems.

The present prospective, longitudinal study of 451 adolescents and their parents extends earlier research by investigating whether change in marital problems predicts change in adolescent adjustment, after controlling for other marital problems and socioeconomic status. Latent growth curves over a period of 5 years were used, and the results revealed that in general, increases or decreases in marital distress and conflict predicted corresponding increases or decreases in adolescent adjustment problems over time. In addition, the analyses suggested that increases in marital distress are as harmful for adolescent adjustment as increases in marital conflict.
