OBJECTIVE: To investigate the association between polymorphism of CYP17 gene and serum hormone concentrations in aged men. METHODS: Eighty-three healthy men at the average age of 66.7 were divided into a < 66.7 group (n = 36) and a > 66.7 group (n = 47), and the polymorphism of CYP17 gene in the 5' promoter region was investigated by PCR using DNA from the men's peripheral blood lymphocytes. A new recognition site was created for the restriction enzyme MspA1 I by transition (T --> C) in the risk allele (A2). Three genotypes A1/A1, A1/A2, A2/A2 were established, serum sex-hormone levels measured, and mean hormone concentration evaluated in each genotype and age group. RESULTS: No evidence was found that the testosterone (T) level, estrogen (E2) level and T/E2 ratio were associated with the genotype of CYP17 gene. There was no significant difference in T and E2 levels between the two groups, but there was a significant increase in the T/E2 ratio (P < 0.05). CONCLUSION: A2 allele does not increase sex hormone levels in aged men, but the T/E, ratio was higher in the > 66.7 group than in the < 66.7 group. This may be closely associated with the mechanism of benign prostate hyperplasia and prostate cancer in aged men.
This paper describes the use of piezo-driven micropipette for intracytoplasmic sperm injection of mice eggs. The head of fresh spermatozoa from KM (Kunming) fertile mice was individually injected into mature oocytes of hybrid mice B6D2F1. Approximately eighty three percent of sperm-injected oocytes survived, and 84.0% of them fertilized normally (extrusion of the second polar body and formation of male and female pronuclei). The eggs fertilized by sperm injection could develop in vitro to 2-cell (98% vs 94.7%), 4-cell (89.5% vs 92.1%) stages, no significantly (P > 0.05) different from embryos fertilized in vivo but there were significantly (P < 0.01) few morulae (63.8% vs 84.2%) and blastocysts (25.7% vs 68.4%) developed in vitro after further culture in vitro in the group of ICSI. When 120 embryos at the pronuclear stage were transferred to seven pseudopregnant KM female, 23.3% of the embryos (0 - 50%, depending on the host) reached the full term. Except for three that were cannibalized soon after birth, all of the young (25 pups) developed into normal and fertile adult. Here we report the first birth of mouse offspring following ICSI in China. These studies may increase understanding of the fertilization process and of how ICSI works.
OBJECTIVE: To establish a colloid gold-immunochromatography assay (GICA) for detecting Plasmodium falciparum. METHODS: The monoclonal antibodies (mAbs) against lactate dehydrogenase of P. falciparum (LDHpf) were screened for preparation of GICA strips. With microscopic examination of the blood smears and PCR test as control, GICA was evaluated for its sensitivity, specificity and stability in the diagnosis of malaria in the outpatient clinics in China. RESULTS: Four hybridoma cell lines against LDHpf were prepared. Enzyme-linked immunosorbent assay demonstrated that the 4 mAbs reacted only with P. falciparum, but not with protein of normal human red cell, P. vivax, Toxoplasma gondii, or Schistosoma japonicam. All the 4 mAbs recognized a 33 kD protein designated as LDHpf as shown by Western blot analysis. Compared with the microscopic examination of blood smears and PCR test, GICA had the sensitivities of 88.37% and 86.67% and the specificities of 95.65% and 97.78%, respectively. Concordance between microscopic examination and GICA for P. falciparum infection was 91.55%. CONCLUSION: GICA established in this study is simple, rapid, sensitive and specific for detecting P. falciparum, and is potentially useful in developing reagent kits for clinical use.
OBJECTIVE: To summarize operational effect with surgical treatment of aortic regurgitation caused by Behcet's disease and discuss relevant surgical techniques for treatment of these conditions. METHODS: Eight patients with aortic regurgitation secondary to Behcet's disease and received surgery between April 1997 and August 2003 were retrospetively analyzed. Among them, two patients had their aortic valves replaced in other hospital before admitted to our hospital where one undertook aortic valve replacement (AVR), and the other undertook aortic root replacement (ARR). In six patients who were initially treated in our hospital, the surgical procedures for aortic regurgitation included AVR in three patients and ARR operation in other three patients in whom Bentall-type operation was conducted in two patients and Cabrol-type operation in one. RESULTS: One patient died during hospital stay. The follow-up periods ranged from 3 months to 36 months. In five patients with prosthetic valve detachment or suture detachment, redo homograft replacement was required in one patient and redo AVR in 3, one patient had redo AVR twice, and the remaining one patient had no surgery at present. Three patients primarily operated by ARR operation have no complications. CONCLUSIONS: The rate of prosthetic valve detachment is high in patients with Behcet's disease. ARR should be a first-line therapy for operation promised these patients.
Apolipoprotein A-I-Milano(AIM), a natural variant, not only inhibits the initiation and progression of atherosclerosis, but also makes the preexisting atherosclerotic lesions regress. AIM gene, at which N-terminal codens were optimized, was subcloned into the expression vector of pET22b. Recombiant plasmids were transformed into E. coli strain BL21 (DE3) and induced with IPTG. The expressed apoliprotein A-I-Milano was soluble in E. coli and was about 38% of total cell lysate. Purified by Butyl Sepharose 4F. F hydrophobic chromatography and Q Sepharose H.P. anion exchange chromatography, followed by ultrafiltration with Vivaspin 20 (30 000MW), AIM monomer was obtained in a purity of more than 95%. Activity assay of binding of AIM monomer to lipid indicates that association of AIM monomer with DMPC is slower than normal apoA-I but DMPC number associated by AIM monomer is more than by apoA-I. This results will be important for studying structure, function of AIM, specially clinical application.
This study was aimed to explore the feasibility of transplanting human cord blood stem cells (HSC) into pre-immune fetal and neonatal pigs, and to investigate the self-renewal of HSC in the recipient pigs. The fetus and neonate were manipulated in sterile separated room and human donor cells were injected into fetus via fetus muscle or umbilical vein (dissectted womb) or into neonate via umbilical vein before cutting it. Human CD45(+) cells s were detected by labeling with human anti-CD45 antibody and analyzed by fluorescence activated cell sorting (FACS). The results showed that tested pigs developed as well as control and a definite proportion of human cells existed in peripheral blood of chimeric pig on day 60 after transplantation. In conclusion, the fetus and neonate pigs can tolerate a definite proportion of human antigens, and to establish the human/pig model of hematopoietic chimerism is possible.
OBJECTIVE: To assess the indication and efficacy of surgical treatment for patients with hepatic metastases from gastric cancer. METHODS: Clinical data of 43 patients with hepatic metastases from gastric cancer undergoing surgery from September 1995 to May 2002 were analyzed retrospectively. RESULTS: No relationships were found between the number of hepatic metastases and patient's gender, age, tumor invasion depth, lymphatic node metastases and differentiation P> 0.05. All of 43 patients underwent surgery. Four cases undergoing hepatic resection for metachronous hepatic metastases had a higher survival rate than those who had curative resections for synchronous hepatic metastases (median survival 35 months vs. 10 months) (P=0.0233). 39 patients had synchronous hepatic metastases, of whom 32 patients received gastric resection only and 7 patients received both gastric and hepatic resections, there was significant difference of median survival between synchronous group and metachronous group(median survival 6.0 vs. median survival 10.0 months)(P=0.2799). There was significant difference of the postoperative survival rate among H (1) (7.5 months), H (2) (6 months) and H (3) (4 months) in the patients with palliative gastric resections (P=0.0007). CONCLUSION: Hepatic resection for metachronous hepatic metastases from gastric cancer has a better prognosis. Resections of gastric and hepatic lesions at the same time may not benefit the patients with liver metastases from gastric cancer. H(3) is not feasible for palliative gastric resections.
OBJECTIVE: To prepare and characterize the monoclonal antibodies (mAbs) against erythrocyte-binding antigen 175 of Plasmodium falciparum (EBA-175). METHODS: BALB/ c mice were immunized with purified recombinant EBA-175 and mAbs against EBA-175 were prepared by means of hybridoma technique. Enzyme-linked immunosorbent assay (ELISA) and Western blotting were employed for characterization of the mAbs. RESULTS: Six McAbs against EBA-175 antigen were obtained, 5 of which were identified as IgG1 and one as IgG2a. The titer of these aAbs was 1:12,800 to 1:25,600 in the ascites and 1:256 to 1:512 in supernatant, and ELISA demonstrated specific binding of the 4 mAbs (1F3, 2H5, 4A1 and 4H9)with Plasmodium falciparum. Three of these mAbs recognized the protein of EBA-175 as shown by Western blotting. CONCLUSION: Six hybridoma cell lines secreting the mAbs against EBA175 with high specificity are successfully established.
OBJECTIVE: To investigate effects of supplementation of folate on the expression of monocyte chemoattractant protein-1 (MCP-1) in aortic endothelium and release from peripheral blood mononuclear cells (PBMC) in rats with hyperhomocysteinemia induced by ingestion of excess methionine. METHODS: Thirty male SD rats were randomly divided into 3 groups (n = 10 for each group): control group (Control), high homocysteinemia group (Hhcy), and folate supplementation group (FA). They were fed with nomal diet, normal diet enriched by 1.7% methionine, and normal diet plus 1.7% methionine and 0.006% folate, respectively, for 45 days. The levels of total plasma homocysteine (Thcy) were measured by high performance liquid chromatography and the concentrations of chemokine MCP-1 released from PBMC stimulated by oxidized low density lipoprotein were detected by enzyme immunoassays. The expression of MCP-1 on aortas of rats was detected by immunohistochemistry and Western blot. RESULTS: A high methionine diet for 45 days induced hyperhomocysteinemia. Folate supplementation to high-methionine diet significantly decreased plasma Thcy levels (P < 0.01). The expression of MCP-1 in aortic endothelium and the levels of MCP-1 released from PBMC stimulated by oxidized low density lipoprotein were significantly higher in rats of Hhcy group than in rats of control group (P < 0.05, P < 0.01). During supplementation of folate, normalization of Thcy levels was accompanied by a marked reduction of MCP-1 expression in aortic endothelium and by a significant decrease of MCP-1 released from PBMC stimulated by oxidized low density lipoprotein (P < 0.05, P < 0.01). CONCLUSION: Folate supplementation can prevent an elevation of homocysteine levels in the blood and decrease the expression MCP-1 in aortic endothelium and release of MCP-1 from PBMC in rats with hyperhomocysteinemia.

OBJECTIVE: To find a new way of treating otitis media with effusion by studying the curative effect of pulmonary surfactant on secretory otitis media in guinea pigs. METHOD: Nonviable heat-killed pneumococci (HKP) solution was inoculated into the middle ear cavity in guinea pig via a transeardrum approach, set up models of otitis media with effusion in guinea pigs, after being injected with pulmonary surfactant by transeardrum approach, observed changes of serous effusion in tympanum and response threshold. RESULT: Five days after inoculation of HKB, serous effusion was present in the middle ear cavity of guinea pigs with disappearance of light cone. ABR response threshold raised from (14.00 +/- 3.08) dB to (45.00 +/- 5.67) dB. After seven days following injection of pulmonary surfactant by transeardrum approach, serous effusion of tympanum reduced or disappeared. ABR response threshold decreased from (45.00 +/- 5.67) dB to (23.53 +/- 6.32) dB, there was significantly difference between them (P < 0.001). In guinea pigs suffering from otitis media with effusion were not treated by PS, the serous effusion of tympanum did'nt change. Eardrum was more muddy and ABR response threshold raised from (45.00 +/- 5.67) dB before treatment (65.50 +/- 6.85) dB after being treated by PS, respectively, with statistically significant difference between them (P < 0.01). CONCLUSION: Pulmonary surfactant plays an important role in treating otitis media with effusion in guinea pigs.
OBJECTIVE: Bilateral pheochromocytomas are rare diseases. The purpose of this retrospective study was to elucidate the clinical characteristics of patients with bilateral pheochromocytomas. METHODS: We analyze the clinical data of 25 patients with bilateral pheochromocytomas who were treated at Peking Union Medical College Hospital between 1952 and 2004. RESULTS: The average age at diagnosis was (32 +/- 14) years. 19 cases (76%) were familiar type, and among the 19 cases, 13 cases were multiple endocrine neoplasia (MEN), 5 cases von Hippel-Lindau (vHL) disease and 1 case of isolated familial pheochromocytomas. In the 25 patients, bilateral pheochromocytomas were discovered at the same time in 88%, and multiple tumors existed in at least one side of the adrenal gland in 56%. 50% of cases recurred after resection of pheochromocytomas. CONCLUSIONS: Hereditary syndromes should be screened when pheochromocytoma is bilateral, and the patients' family members also should be screened for hereditary syndromes. During operation for bilateral adrenal pheochromocytoma, multiple tumors in one side should be considered. Long-term follow-up is necessary because recurrence may develop many years after operation.

OBJECTIVE: The purpose of the study is to establish a new process which unites the seropharmacology and cytobiological experiments to reflect the exact effects of herb-food extract (HFE) on, lung cancer, and provides some scientific evidence for the actions of anti-cancer by foods. METHODS: The optimal conditions of the seropharmacology method were explored by an orthogonal experimental design. The effects of HFE on lung cancer cell YTLMC-90 were proceeded by means of three assays as follows: Methyl thiazolyl tetrazolium (MTT) assay, cell double time analysis and colony forming assay. RESULTS: The method was successfully built up and the best combination was, as to the HFE, giving to the rats 2 times a day (2%), totally lasting 3 days, and adding 10% of the serum as in the culture medium. The next study illustrated that all the HFEs, including 0.5 g/ml, 1.0 g/ml with 2.0 g/ml ones, had inhibitive capabilities on the growth and proliferation of lung cancer cells, specially those of giving 2.0 g/ml in 3 days and 0.5 g/ml in 10 days. And there was no significant difference between the last two. CONCLUSION: Seropharmacology is a useful method to the study of the effects of mixtures combined with cytobiological experiments. The inhibitive ability of HFE on lung cancer cells suggests that it has effective value in cancer prevention.
OBJECTIVE: To investigate the molecular mechanism ot lingual epithelial cell (LEC) apoptosis and its related genes expression in different tongue furs. METHODS: The LEC apoptosis and its related genes expression including bax, fas, TGF-beta 3 mRNA and protein product in tongue fur was determined using terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick end labelling technique (TUNEL), in situ hybridization, immunohistochemical technique and image analysis. RESULTS: LEC apoptosis could always be seen in 4 types commonly encountered tongue fur. The tendency of changing in thickness of tongue fur was opposite to that of apoptotic index. Compared with normal thin fur, bax and fas genes were over-expressed in exfoliative fur with increased apoptosis, while in thick fur, bax and TGF-beta 3 genes were low-expressed and accompanied with decreased apoptosis. The level of apoptosis promoting genes, bax, fas, TGF-beta 3 gene expression in LEC showed a tendency parallel to that of LEC apoptosis. CONCLUSION: Change of apoptosis related genes expression, bax, fas and TGF-beta 3 may effect the LEC apoptosis and be the important factor for changing of the thickness of tongue fur.
CD134, a member of the tumor necrosis factor receptor superfamily, plays a crucial role in T cell survival. In this study, peripheral blood mononuclear cells (PBMCs) stimulated with phytohemagglutinin (PHA, 50 microg/ml) were treated by CD134 mAb (1 microg/ml, 5 microg/ml, 10 microg/ml) for 6 h, 12 h, 24 h and 48 h. The level of perforin mRNA was measured by reverse transcription-polymerase chain reaction (RT-PCR) technique. Our data showed that the expression of perforin mRNA in PBMCs was down-regulated by CD134 mAb in a dose-dependent manner in range of 1 microg/ml to 5 microg/ml and dropped down to its minimum on 24 h (p<0.05). The level of perforin mRNA reached a plateau when the concentration of CD134 mAb exceeded 5 microg/ml. In conclusion, CD134 mAb can inhibit perforin expression, which may enhance the ability of T cells for survival.
