[An investigation of the relationship between Lepr gene Gln223Arg polymorphism and obstructive sleep apnea hypopnea syndrome].

OBJECTIVE: To investigate the relationship between leptin receptor (Lepr) gene Gln223Arg polymorphism and obstructive sleep apnea hypopnea syndrome (OSAHS). METHODS: The genotypes of Gln223Arg polymorphism in Lepr gene were determined by polymerase chain reaction-restriction fragment length polymorphisms (PCR-RFLP) assay in 181 unrelated subjects of North region "Han" population (including 78 non-OSAHS subjects and 103 OSAHS subjects). RESULTS: The OSAHS patients with GG genotype had significantly larger neck circumference (NC) than did OSAHS patients with (GA + AA) genotype (P = 0.031); The study revealed no difference in genotype of Gln223Arg polymorphism between OSAHS group and non-OSAHS group (P = 0.676); There was also no significant difference in body mass index (BMI), waist hip rate (WHR), blood pressure (Bp), leptin (Lep), true insulin (TI), fast blood glucose (FBG), triglyceral (TG), cholesterol (Chol) and polysomnography (PSG) between GG and (GA + AA) genotype of OSAHS patients (P > 0.05). CONCLUSIONS: Lepr gene Gln223Arg polymorphism may be involved in the regulation of the neck regional fat distribution in OSAHS patients. It is unlikely that Lepr gene Gln223Arg polymorphism may contribute to the pathogenesis of OSAHS.

Arsenic trioxide-induced apoptosis of human malignant lymphoma cell lines and its mechanisms.

OBJECTIVE: To investigate the responses of human Burkitt lymphoma cells to arsenic trioxide (As2O3) and the possible mechanisms. METHODS: Epstein-Barr virus (EBV)-positive human B-lymphoma Raji cell line and EBV-negative human B-lymphoma BJAB cell line were used as in vitro models to assess the cell apoptosis by morphology and DNA agarose gel electrophoresis. Protein expression was analyzed using Western blotting. RESULTS: After 24-hour treatment with the 2, 5 and 10 micromol/L As2O3, the concentrations of As2O3 achievable in vivo, cell apoptosis was induced in human Burkitt lymphoma BJAB cells at the rates of 47.6%+/-4.8% (Mean+/-SD, n=3), 66.4%+/-5.1%, 87.0%+/-7.3% and at 35.5%+/-3.8%, 51.5%+/-6.2%, 62.2%+/-7.9% respectively in Raji cells, corresponding to the concentration of As2O3. EBV-infected Raji cell line was less sensitive to As2O3 than EBV-negative BJAB cell line (P<0.05). As2O3-induced apoptosis was accompanied by down-regulation of Bcl-xL protein expression and activation of apoptosis protein caspase-3, as identified by Western blotting. CONCLUSION: As2O3 exerts apoptosis-inducing effects on human Burkitt lymphoma cells through down-regulation of Bcl-xL protein expression and activation of apoptosis protein caspase-3, and may serve as a candidate therapeutic agent against malignant lymphoma for both systemic and local therapies.

[Vitamin D receptor gene Bsm I polymorphism and the susceptibility to prostate cancer in northern Chinese Han population].

OBJECTIVES: To investigate Bsm I single nucleotide polymorphism (SNP) of vitamin D receptor gene (VDRG) in low-risk Chinese Han population and its relationship to the susceptibility to prostate cancer (PCa), and to discuss the possible reason for the racial difference of PCa. METHODS: One hundred and three patients with PCa and 106 normal controls, mainly from Northern Chinese Han population, were enrolled in this study. Their blood samples were obtained, all of which were genotyped for Bsm I SNP by denaturing high performance liquid chromatography(DHPLC) methods using case-control study. RESULTS: The distribution of genotype and allele had no significant difference between PCa patients and normal controls (P > 0.05). The frequencies for the bb, Bb and BB genotypes in PCa patients and normal controls were 92.23%/94.34%, 7.77%/5.66%, and 0/0, respectively. The frequencies for B and b allele were 3.88%, 96.12% and 2.91%, 97.09%, respectively. CONCLUSIONS: The results indicate no significant relationship between the VDRG polymorphisms and PCa in Northern Chinese Han population. The distribution of VDRG Bsm I SNP varies in different ethnic populations, which may be one reason for the racial difference of PCa.

Treatment of six cases of left renal nutcracker phenomenon: surgery and endografting.

OBJECTIVE: A retrospective study was undertaken to examine therapeutic strategies used in 6 patients with left renal venal nutcracker phenomenon. METHODS: Three patients underwent surgical transposition of the superior mesenteric artery. Three patients underwent endograft stent implantations with intervention. RESULTS: All 6 cases were completely relieved of left renal vein compression after the operations, leading to improved clinical symptoms and normal urine tests. CONCLUSIONS: Transposition of the superior mesenteric artery is a feasible method, but leads to complications and great trauma. Treating left renal venal nutcracker phenomenon with endografting techniques is minimally invasive in certain cases, and seems to have a prospective future.

[Refolding and purification of Plasmodium falciparum glutamate dehydrogenase fusion protein].

OBJECTIVE: To establish the method for renaturation and purification of the fusion protein of Plasmodium falfiparum (FCC1/HN ) glutamate dehydrogenase (GDH) with glutathione S-transferase (GST). METHODS: The recombinant plasmid GDH/pGEX-4T-1, encoding the full-length GDH gene, was transformed into E.coli BL21 (DE3) to achieve IPTG-induced high expression of GDH/GST in the form of inclusion bodies identified by SDS-PAGE. After denaturation with 8 mol/L urea, the inclusion bodies were subjected to 3 different renaturation methods, namely Sephacryl S-200 chromatography, dialysis and dilution, for refolding of the fusion protein. The refolded GDH/GST was then purified by different chromatographic approaches. RESULTS: SDS-PAGE analysis showed that the expression GDH/GST fusion protein mounted up to approximately 25% of the total bacterial protein. The dilution was better than the other two methods for the refolding of the fusion protein, with the optimized renaturation condition necessitating the presence of 20 mmol/L Tris-HCl and 1 mmol/L EDTA at pH8.5 with GSSG/GSH ratio of 1 10, which resulted in a recovery rate exceeding 90%. Two-step ion exchange chromotography was optimal for purification of the fusion protein. CONCLUSION: The high-purity and biologically active GDH/GST can be acquired by dilution renaturation followed by two-step ion exchange chromatography.

[Analysis of LRP15 Gene Expression Pattern and Its Expression in Leukemia Cells].

To explore the possible function of LRP15 gene in carcinogenesis and its significance in the classification and prognosis of leukemia, the expression pattern of LRP15 in normal tissues, tumor tissues and cell lines was detected with SAGE and gene expression database provided by NCBI and NCI respectively. RT-PCR was used to detect the expression of LRP15 in leukemia patients. The results showed that LRP15 was expressed in different tissues and tumor cell lines, the positive rate of LRP15 in immature blood cells was higher than that of mature blood cells and the positive rate of M(1), M(2) and M(3) was higher than that of other AML subtypes (P < 0.01), the expression of LRP15 in refractory leukemia was higher than that of de novo leukemia. The results suggest that LRP15 may play an important role in carcinogenesis, AML classification and acute leukemia prognosis.

[Diagnosis and therapy of the nutcracker phenomenon: long-term follow-up].

OBJECTIVE: To study the diagnosis of adult nutcracker phenomenon (NCP) and assess the therapeutic value of endovascular stenting (ES) and superior mesentery artery transposition (SMAT) for the treatment of NCP in long-term follow-up. METHODS: Six patients (6 men) aged 16 and 34 years old (mean age, 22.7 +/- 18.0 years) were diagnosed as having NCP using the examination of the doppler ultrasound and/or magnetic resonance of artery/digital radiography (MRA/DSA). Three patients underwent ES and 3 patients received SMAT for the treatment of the NCP patients associated with recurrent gross hematuria and left flank pain. Doppler ultrasound and urine examination were used at pre- and post-operation. RESULTS: In 3 patients who underwent SMAT, the postoperative complications comprised retroperitoneal hematoma necessitating surgical revision (n = 1). Functional disorder of intestine (n = 1) and paralytic ileus (n = 1) that were resolved by conservative management. In 3 patients who experienced ES, 1 patient received surgical revision because the position of stent was not suitable in left renal vein but no other complication took place. During the follow-up of 6 approximately 50 months (mean 24.7 +/- 18.0 months), except that 1 patient's hematuria disappears at rest and reappears after motion while the other 5 patients remain asymptomatic and free of hematuria. The dopplar ultrasound showed the left renal vein diameters of the angel segment between superior mesentery artery and aorta were (0.18 +/- 0.05) cm preoperation and (0.65 +/- 0.17) cm postoperation, P < 0.001; the left renal vein diameters of the portal segment were (0.89 +/- 0.22) cm preoperation and (0.79 +/- 0.20) cm postoperation, P = 0.003; the left renal vein diameter's ratio between portal and angel segment were (4.99 +/- 0.79) preoperation and (1.23 +/- 0.16) postoperation, P < 0.001. CONCLUSIONS: Dopplar ultrasound plays a very important role in the diagnosis of adult NCP. SMAT is an efficient surgical approach to the treatment of the nutcracker phenomenon and is associated with an acceptable risk of complications. But ES is safer and more efficient than SMAT and may represent a valuable approach to lessening the morbidity of surgical procedures. ES is a new therapeutic method for adult NCP.

[Treatment of Wilson's disease with penicillamine and zinc salts: a follow-up study].

OBJECTIVE: Wilson's disease (WD) is an autosomal recessive disorder characterized by excessive accumulation of copper in the liver and later in the brain and other organs. Penicillamine acts as a reductive chelator. Zinc salts induce the synthesis of metallothionein in cells. Thus these two drugs are theoretically synergistic for the treatment of the disease. However, the two drugs may also have some unfavorable interactions. In this study, the effect of the therapy with combined penicillamine and zinc salts was evaluated based on the follow-up observations of 21 patients with Wilson's disease. METHODS: Using the combined therapy of penicillamine [10-30 mg/(kg.d)] and zinc (22.5 mg, 3 times per day), follow-up study by hospitalization or communication with telephone or mail. RESULTS: Before treatment, all the 21 patients were suffered from chronic liver disorder. Among them, 13 patients (62%) showed to be reactive to the treatment for their liver disorder, 5 patients (24%) died, and 3 patients (14%) dropped off our follow-up study. Among the 5 patients who died, 3 died within 40 days after treatment, one had taken penicillamine only 8 mg/(kg.d), and one died after discontinuation of the treatment by the parents. Of the 12 patients having neurological involvement, neurological symptoms disappeared or markedly improved in 11 patients after treatment. One patient dropped off the follow-up study. The patient with renal tubular acidosis responded well to the treatment. Urine routine analysis was followed up in 6 of the 7 patients with hematuria. Hematuria disappeared in one, became less severe in 1, and remained unchanged in 4 patients. Hypersensitivity to penicillamine was found in one patient. WBC and platelet were found decreased further in 3 patients after the medications. CONCLUSIONS: The combined therapy with penicillamine and zinc salts was effective in treatment of patients with Wilson's disease.

[Effects of organophosphates on cell proliferation and 45Ca uptake of neuroblastoma SH-SY5Y cells].

OBJECTIVE: To explore the mechanism of cytotoxic effects of the organophosphates (OPs) with delayed neurotoxicity on human neuroblastoma cells. METHODS: The proliferation of neuroblastoma SH-SY5Y cells was determined by MTT spectrometry. (45)Ca uptake was determined by adding (45)CaCl(2) and tri-o-cresyl phosphate (TOCP) or methamidophos into the cultured medium for the SH-SY5Y cells. The cells were incubated and then lysed and finally counted in a Beckman LS 6000 liquid scintillation spectrometer. RESULTS: Methamidophos stimulated the cell proliferation of SH-SY5Y at its lower concentrations (7 x 10(-7) mol/L to 7 x 10(-6) mol/L), with an increase by 28% at 7 x 10(-7) mol/L; however, it inhibited the proliferation at higher ones (7 x 10(-4) mol/L to 7 x 10(-3) mol/L) with 62% inhibition at 7 x 10(-3) mol/L. TOCP only inhibited the cell proliferation at high concentration (with 34% inhibition at 7 x 10(-3) mol/L) and markedly inhibited calcium uptake of the cells up to 55% at higher concentrations (1 x 10(-6) mol/L to 1 x 10(-4) mol/L); while the uptake was stimulated by OPs up to 241% of increase at lower concentrations (1 x 10(-9) mol/L to 1 x 10(-7) mol/L). CONCLUSION: The interference of growth in nerve cells and disturbance of calcium homeostasis may be involved in the mechanisms of neurotoxicity of OPs.

[Expression of NGF, BDNF and NT-3 in satellite cells of dorsal root ganglion].

OBJECTIVE: To investigate the distribution of NGF, BDNF, NT-3 and their mRNA in satellite cells of dorsal root ganglion (DRG) in adult cat. METHODS: Five male adult cats were used and their L6 DRGs unilateral side were taken and sectioned into frozen sections 20 microm in thickness respectively. The sections were stained by immunohistochemistry ABC method (NGF antibody 1:1000, Sigma; BDNF antibody 1:500, Santa Cruz; NT-3 antibody 1:1500, Santa Cruz), and in situ hybridization study(cRNA probe 1:100). RESULTS: NGF, BDNF, NT-3 and their mRNA were observed in some satellite cells. The positive signal of mRNA for NGF, BDNF and NT-3 was observed in cytoplasm. However, there was difference in the subcellular localization of protein for the three factors. BDNF immunoreactivity (BDNF-IR) was located in cytoplasm, while NGF-IR and NT-3-IR were located in nucleus. CONCLUSION: The results demonstrated that some satellite cells contain NGF, BDNF and NT-3 as well as their mRNA. The difference of subcellular localization for NGF, BDNF and NT-3 indicated that the functional mechanism of each factor in satellite cells in adult cats may be different.
