Identification of two novel mutations in Chinese patients with Dyschromatosis symmetrica hereditaria.

Dyschromatosis symmetrica hereditaria (DSH) is a rare autosomal dominant cutaneous disorder characterized by a mixture of hyperpigmented and hypopigmented macules of various sizes on the extremities. Pathogenic mutations in the DSRAD gene have recently been identified. In this study, we report and identify the mutations of the DSRAD gene in two Chinese pedigrees with DSH. Two novel mutations in the functional domains of the DSRAD gene were identified and verified in two pedigrees. The c.3244A>G (H1075R) mutation was found in all patients but not in the healthy individuals from family A and c.3335_3336delAT (Y1112fs-->1112X) mutation was found in three patients but not in the healthy family members from family B. Our data suggests that these two novel mutations in the DSRAD gene could cause DSH and add new variants to the repertoire of DSRAD mutations in DSH.

[Therapeutic neovascularization with autologous bone marrow CD34+ cells transplantation in hindlimb ischemia].

OBJECTIVE: To explore whether transplantation of autologous bone marrow stem cells might augment angiogenesis and collateral vessel formation in a canine model of hindlimb ischemia. METHODS: CD34(+) stem cells were centrifugation through Ficoll and an immune magnetic cell sorting system from bone marrow (20 ml) of canine (n = 5) and induced into endothelial cells with VEGF in vitro, and expression of von Willebrand factor. Bilateral hindlimb ischemia was surgically induced in canines and Dil fluorescence labeled autologous stem cells were transplanted into the ischemic tissues. RESULTS: Four weeks after transplantation, fluorescence microscopy revealed that transplanted cells were incorporated into the capillary network among preserved skeletal myocytes. The stem cells transplanted group had more angiographically detectable collateral vessels, a higher capillary density (12.0 +/- 2.8 vs. 5.0 +/- 1.6 per field; t = 4.17 P < 0.05) and a higher ABI (0.58 +/- 0.14 vs. 0.32 +/- 0.11; t = 2.95, P < 0.05). CONCLUSIONS: Direct local transplantation of autologous bone marrow CD34(+) stem cells seems to be a useful strategy for therapeutic neovascularization in ischemic tissues in adults, consistent with "therapeutic vasculogenesis."

[A multicenter clinic study of slow-release-gliclazide in type 2 diabetes mellitus].

OBJECTIVE: To evaluate the efficacy and safety of 30 mg slow-release-gliclazide (Diamicron MR) on type 2 diabetes patients with the level of blood glucose poorly controlled formerly. METHODS: 154 type 2 diabetes patients with the level of blood glucose poorly controlled formerly were enrolled and treated with Diamicron MR for 16 weeks. The efficacy of slow-release-gliclazide was evaluated through HbA1c, FPG and 2 h PBG after 16 weeks of treatment period. The safety was evaluated through the adverse events including hypoglycemic episodes and the change of vital signs and clinic laboratory parameters. RESULTS: After 16 weeks of treatment period, the HbA1c, FPG and 2 h PBG were decreased by 1.89%, 2.31 mmol/L and 3.94 mmol/L respectively, cutting down 21.6%, 24.5% and 27.8% relatively in comparison with pre-treatment. Glucose metabolism could be significantly improved by taking once-daily Diamicron MR. CONCLUSION: Slow-release-gliclazide offers the significant treatment safely, which is important to optimize type 2 diabetes mellitus management.

Folic acid reduces adhesion molecules VCAM-1 expession in aortic of rats with hyperhomocysteinemia.

To investigate effects of supplementation of folic acid on the expression of adhesion molecules VCAM-1 in the aortas of rats with hyperhomocysteinemia. Thirty male SD rats (200 +/- 20 g) were invided into 3 groups (n = 10 for each group): control group(Control), high Met group(Met) and Met plus Folate group(Met + Folate), fed. for 45 days. Plasma Hcy levels were higher with the high-methionine diet (140.68 +/- 36.87 micromol/L vs 6.47 +/- 1.10 micromol/L in control rats) an effect which was reduced by folate. Respectively, the aortic expression of adhesion molecules VCAM-1 at protein and mRNA levels were higher in the Met groups than those in the control groups or the Met + Folate groups. A high methionine diet for 45 days was sufficient to induce hyperhomocysteinemia. Folate supplementation prevented elevation of Hcy levels in the blood, and reduced expression of the adhesion molecule VCAM-1. Hyperhomocysteinemia is now regarded as one of the important risk factors for cardiovascular and cerebralvascular disorders.[Welch GN, Loscalzo J. Homocysteine and atherothrombosis. N Engl J Med 1998; 38(15):1042-50.] Several plausible mechanisms for Hcy-induecd atherosclerosis have been proposed. These include endothelial dysfunction, enhancement of oxidative stress, reduction in NO bioavailability, and augmentation of thrombus formation.[Holven KB, Holm T, Aukrust P, et al. Effect of folic acid treatment on endothelium-dependent vasodilation and nitric oxide-derived end products in hyperhomocysteinemic subjects . Am J Med 2001;110(7):536-42; Guba SC, Fonseca V, Fink LM. Hyperhomocysteinemia and thrombosis. Semin Thromb Hemost 1999;25(3):291-309.] However, the precise molecular mechanism is still unclear. Recent reports have suggested a role for inflammatory processes in the pathogenesis of atherosclerosis.[Gerard C, Rollins BJ. Chemokines and disease. Nat Immunol 2001;2(2):108-15.] Dysfunction of endothelial cells is the key process promoting inflammatory reactions. On injury, endothlial cells are capable of producing various cytokines that participate in inflammatory reactions in the arterial wall. Although results from in vitro studies suggest that Hcy, at pathophysiological concentrations, stimulates chemokine expression in vascular cells, it is unknown whether hyperhomocysteinemia can initiate similar changes, leading to enhanced momocyte adhesion/binding to the vascular endothelium in vivo.[Zeng X, Dai J, Remick DG, Wang X. Homocysteine mediated expression and secretion of monocyte chemoattractant protein-1 and interleukin-8 in human monocytes. Circ Res 2003;93(4):311-20.] On the basis of the potential pathogenic role of chemokines in atherogenesis, the objective of the present study was to investigate that homocsteine may exert its effect in part though adhesion molecules VCAM-1 and that folic acid supplementation may downregulate these inflammatory responses. Male Sprague-Dawley rats (bred from animal centers of Tongji Medical College, Huazhong Science and Technology University) aged 8 weeks were divided into 3 groups(n=10 for each group) and maintained for 45 days on the following diets before the experiments: (1) regular diet; (2) high-metheionine diet, consisting of regular diet plus 1.7% methionine; and (3) high-methionine plus folate -rich diet, consisting of regular diet plus 1.7% methionine and 0.006% folate.[Boisvert WA, Curtiss LK, Terkeltaub RA. Interleukin-8 and its receptor CXCR2 in atherosclerosis. Immunol Res 2000;21(2-3):129-d37.] Plasma and serum samples wee colleced and stored at -80 degrees C after 45 days until analysis. The plasma homocysteine concentration of rats in three groups were determined by high-pressue liquid chromatography. To detect the endothelial expression of adhesion molecules VCAM-1, the thoracic aorta was isolated and dived into segments. These segments were immersion-fixed in 10% neutral-buffered formalin overlight and then embedded in paraffin. Sequential 5 mum paraffin-embedded cross sections were prepared. Immunohistochemical analyisis was performed to detect vascular cell adhesion molecule(VCAM)-1, The fixed cryosections were immediately blcked in 10% horse serum and phosphate baffered saline(PBS) at room temperature for 30 min. Goat polyclonal andibodies against rat VCAM-1(Santa Cruz Biotechnology) were diluted 1:100 in PBS and incubated with the cryosections for 1 h of room temperature. After three washes, the sections were incubated with biotin-conjugated rabbit anti-goat immunoglobulins(Dako) at 1:250 dilution in PBS. After three washes, the samples were mounted in 90% glycerol-PBS. Photographs were taken by use of a light microscope at a mignification of x200.

Molecular cloning and functional analysis of (R)-3-hydroxyacyl-acyl carrier protein:coenzyme A transacylase from Pseudomonas mendocina LZ.

An inactive (R)-3-hydroxyacyl-acyl carrier protein:coenzyme A transacylase (PhaG(Pm)) was cloned from a newly isolated Proteobacteria Pseudomonas mendocina LZ. It is the first characterized native inactive PhaG protein. Sequence analysis indicated that there were only two sites where the amino acid sequence differed between this inactive protein and the functional PhaG(Pp) from P. putida. The differences were located at position 78 and in the region 109-113 in the amino acid sequence. Mutagenesis was carried out to investigate these two sites. A recombinant strain harboring a S78C PhaG(Pp) mutant accumulated polyhydroxyalkanoates (PHA) at 11.9% of the cellular dry weight, as compared to the 21.6% PHA produced by the recombinant harboring the wild-type PhaG(Pp). On the other hand, the changes in the amino acid region 109-113 of PhaG(Pp) to its corresponding region of PhaG(Pm) resulted in negligible PHA accumulation. This demonstrated that region 109-113 in PhaG is relatively important for transacylase activity, while position 78 just plays a supporting role for the enzyme. Furthermore, 3-D structural models of PhaG(Pp) and PhaG(Pm) developed by computational prediction revealed that the variation in amino acids at 109-113 leads to the destruction of the PhaG catalytic center, resulting in the loss of enzyme activity.

Resveratrol inhibits the electrical activity of subfornical organ neurons in rat.

The effects of resveratrol on the discharges of neurons in rat subfornical organ (SFO) slices were examined by using extracellular recording technique. The results are as follows: (1) In response to the application of resveratrol (1, 5, 10 mumol/L, n=65) into the superfusate for 2 min, the spontaneous discharge rate of 60/65 (92.3%) neurons was significantly decreased in a dose-dependent manner;(2) Application of L-glutamate (0.3 mmol/L) into the superfusate led to a marked increase in discharge rate of all 12 (100%) neurons in an epileptiform pattern. The increased discharges of 10/12 (83.3%) neurons were suppressed by application of resveratrol (5 mumol/L);(3) In 8 neurons, the selective L-type calcium channel agonist, Bay K8644 (0.1 mumol/L), induced a significant increase in discharge rate of all 8 (100%) neurons. The increased discharges of all 8 (100%) neurons were suppressed by resveratrol (5 mumol/L);(4) In 14 neurons, nitric oxide synthase (NOS) inhibitor N(G)-nitro-L-arginine methyl ester (L-NAME) 50 mumol/L significantly increased the discharge rate of 11/14 (78.6%) neurons. Resveratrol (5 ?mol/L) applied into the superfusate reduced the increased discharges of 9/11 (81.8%) neurons;(5) In 12 neurons, the large-conductance Ca(2+)-activated K(+) channel blocker tetraethylammonium chloride (TEA) 1 mmol/L significantly increased the discharge rate of 10/12 (83.3%) neurons. Resveratrol (5 mumol/L) inhibited the increased discharges of 9/10 (90%) neurons. These results suggest that resveratrol inhibits the electrical activity of SFO neurons. This effect may be related to its properties of blockade of L-type voltage-gated calcium channel and nitric oxide (NO) promoting, and probably has no association with large-conductance Ca(2+)-activated K(+) channel.

Changing patterns of colorectal cancer in China over a period of 20 years.

AIM: To determine whether any changes have occurred on the patterns of colorectal cancer in China. METHODS: Data from 21 Chinese articles published from 1980 to 1999, were used to analyze the time trend of colorectal cancer according to the patients' age at diagnosis, sex, the site of the tumor, stage, and the pathology. RESULTS: From 1980s to 1990s, the mean age of the colorectal cancer patients has increased. The percentage of the female patients rose. The distribution of colorectal carcinoma shows a predominance of rectal cancer. However, the proportion of proximal colon cancer (including transverse and ascending colon) increased significantly accompanied by a decline in the percentage of rectal cancer. Similarity in the percentage of distal colon cancer between two decades was revealed. In the 1990s, statistically more Stage B patients were found than those in 1980s. In addition, databases show a significant decrease in the Stage D cases. The proportion of adenocarcinoma increased, but the mucinous adenocarcinoma decreased during two decades. CONCLUSION: These findings indicate that the pattern of colorectal cancer in China has been changing. Especially, a proximal shift due to the increasing proportion of ascending and transverse colon cancer has occurred in China.

A hybrid method for relation extraction from biomedical literature.

PURPOSE: Over recent years, there has been a growing interest in extracting entities and relations from biomedical literature. There are a vast number of systems and approaches being proposed to extract biological relations, but none of them achieves satisfactory results. These methodologies are either parsing-based or pattern-based, which are not competent to handle the grammatical complexities of biomedical texts, or too complicated to be adapted. It is well known that appositive, coordinative propositions and such grammatical structures are extremely common in biomedical texts, particularly in full texts. However, these problems are still untouched for most of researchers. METHODS: In this paper, we have proposed a new approach, which is hybrid with both shallow parsing and pattern matching, to extract relations between proteins from scientific papers of biomedical themes. In the method, appositive and coordinative structures are interpreted based on the shallow parsing analysis, with both syntactic and semantic constraints. Then long sentences are splitted into sub-ones, from which relations are extracted by a greedy pattern matching algorithm, along with automatically generated patterns. RESULTS: Our approach is experimented to extract protein-protein interactions from full biomedical texts, and has achieved an average F-score of 80% on individual verbs, and 66% on all verbs. With the help of shallow parsing analysis, pattern matching is improved remarkably. Compared with the traditional pattern matching algorithm, our approach achieves about 7% improvement of both precision and F-score. In contrast to other systems, our approach achieves performance comparable to the best. A demo system has been available at http://spies.cs.tsinghua.edu.cn.

Studies of C-S bond cleavage reactions of Re(V) dithiolates: synthesis, reactivity, and mechanism.

A series of rhenium(V) complexes, [(X)(ReO)(dt)(PPh(3))] and [(o-SC(6)H(4)PPh(2))(ReO)(mtp)], were prepared to explore electronic effects on the C-S cleavage reaction that occurs upon reaction with PAr(3) at ambient temperature [where X = S(C(6)H(4)-p-Z) (Z = OMe, Me, H, F, Cl), OPh, Cl, and SC(2)H(5), and dt is the chelating dithiolate ligand derived from 2-(mercaptomethyl)thiophenol, 1,2-ethanedithiol, 1,3-propanedithiol, 1,3-butanedithiol, and 2,4-pentanedithiol]. The scope and selectivity of the C-S activation were examined. The C-S bond cleavage to form metallacyclic Re(V) complexes with a ReS core occurs only for the complexes with mtp and pdt frameworks and X = SAr and SC(2)H(5). The difference in reactivity is due to the different donating abilities of ancillary and dithiolate ligands, especially their pi-donating ability, which plays a critical role in C-S activation. The kinetics of the C-S activation process was determined; nucleophilic attack of PPh(3) on the oxo group of the Re(V)O core appears to be the rate-controlling step. The reaction is accelerated by electron-poor ArS ligands, but is unaffected by the substituents on phosphines. A detailed mechanistic study is presented. The results represent a rare example of migration of alkanethiolate leading to the formation of alkylthiolato complexes.

Resveratrol inhibits neuronal discharges in rat hippocampal CA1 area.

The effects of resveratrol on the discharges of neurons in CA1 area of rat hippocampal slices were examined by using extracellular recording technique. The results are as follows: (1) In response to the application of resveratrol (0.05, 0.5, 5.0 micromol/L, n=52) into the superfusate for 2 min, the spontaneous discharge rate of 46/52 (88.5%) neurons was significantly decreased in a dose-dependent manner; (2) Application of L-glutamate (0.2 mmol/L) into the superfusate led to a marked increase in discharge rate of all 8 (100%) slices in an epileptiform pattern. The increased discharges were suppressed by application of resveratrol (5.0 micromol/L); (3) In 7 slices, perfusion of the selective L-type calcium channel agonist, Bay K8644 (0.1 micromol/L), induced a significant increase in the discharge rate of 6/7 (85.7%) slices. The increased discharges were suppressed by application of resveratrol (5.0 micromol/L); (4) In 9 slices, perfusion of nitric oxide synthase (NOS) inhibitor N(G)-nitro-L-arginine methyl ester (L-NAME, 50 micromol/L) into the superfusate significantly augmented the discharge rate of 7/9 (77.8%) slices. Resveratrol (5.0 micromol/L) applied into the superfusate reduced the increased discharges of all 7/7 (100%) neurons; (5) In 10 units, the large-conductance Ca(2+)-activated K(+) channel blocker (tetraethylammonium chloride, TEA, 1 mmol/L) significantly increased the discharge rate of 9/10 (90%) slices. Resveratrol (5.0 micromol/L) applied into the superfusate inhibited the discharges of 8/9 (88.9%) slices. These results suggest that resveratrol inhibits the electrical activity of CA1 neurons. This effect may be related to the blockade of L-type calcium channel and a subsequent reduction of calcium influx, and probably has no association with large-conductance Ca(2+)-activated K(+) channel.

Effects of chronic typical and atypical antipsychotic drug treatment on maternal behavior in rats.

Understanding the effects of antipsychotics on maternal behavior is important for understanding the poor quality of mother-infant interaction in schizophrenia. Previous preclinical work has demonstrated that acute treatment with typical and atypical antipsychotics disrupts maternal behavior. However, the effects of chronic antipsychotic treatment on maternal behavior are unknown. This issue is of importance since clinical use of antipsychotic medication requires continuous exposure to these drugs. In this study, we treated postpartum rats with haloperidol (0.25 mg/kg/day) or olanzapine (7.5 mg/kg/day), via osmotic minipumps or daily injections for 3 weeks. Maternal behavior was assessed every third day. On each observation day, maternal behavior was observed twice, once just prior to the daily injection ("trough" as this was 24 h after last injection) and again 2 h after the injection ("peak"). Daily injections of haloperidol and olanzapine significantly disrupted pup retrieval, pup licking, nest building at peak, but this effect was gone by trough. Drug administration via minipumps also disrupted these behaviors, but the effects were less severe. Pup nursing was enhanced by either method of drug administration. No evidence of sensitization or tolerance associated with chronic drug treatment was found. It is concluded that chronic antipsychotic treatment disrupts active maternal behaviors and this disruption, most likely to lead to side effects in humans, should be avoided in future drug development.

In vitro modeling of human pancreatic duct epithelial cell transformation defines gene expression changes induced by K-ras oncogenic activation in pancreatic carcinogenesis.

Genetic analysis of pancreatic ductal adenocarcinomas and their putative precursor lesions, pancreatic intraepithelial neoplasias (PanIN), has shown a multistep molecular paradigm for duct cell carcinogenesis. Mutational activation or inactivation of the K-ras, p16(INK4A), Smad4, and p53 genes occur at progressive and high frequencies in these lesions. Oncogenic activation of the K-ras gene occurs in >90% of pancreatic ductal carcinoma and is found early in the PanIN-carcinoma sequence, but its functional roles remain poorly understood. We show here that the expression of K-ras(G12V) oncogene in a near diploid HPV16-E6E7 gene immortalized human pancreatic duct epithelial cell line originally derived from normal pancreas induced the formation of carcinoma in 50% of severe combined immunodeficient mice implanted with these cells. A tumor cell line established from one of these tumors formed ductal cancer when implanted orthotopically. These cells also showed increased activation of the mitogen-activated protein kinase, AKT, and nuclear factor-kappaB pathways. Microarray expression profiling studies identified 584 genes whose expression seemed specifically up-regulated by the K-ras oncogene expression. Forty-two of these genes have been reported previously as differentially overexpressed in pancreatic cancer cell lines or primary tumors. Real-time PCR confirmed the overexpression of a large number of these genes. Immunohistochemistry done on tissue microarrays constructed from PanIN and pancreatic cancer samples showed laminin beta3 overexpression starting in high-grade PanINs and occurring in >90% of pancreatic ductal carcinoma. The in vitro modeling of human pancreatic duct epithelial cell transformation may provide mechanistic insights on gene expression changes that occur during multistage pancreatic duct cell carcinogenesis.

Role of high-voltage-activated calcium channels in glucose-regulated beta-cell calcium homeostasis and insulin release.

High-voltage-activated (HVA) calcium channels are known to be the primary source of calcium for glucose-stimulated insulin secretion. However, few studies have investigated how these channels can be regulated by chronically elevated levels of glucose. In the present study, we determined the level of expression of the four major HVA calcium channels (N-type, P/Q-type, L(C)-type, and L(D)-type) in rat pancreatic beta-cells. Using quantitative real-time PCR (QRT-PCR), we found the expression of all four HVA genes in rat insulinoma cells (INS-1) and in primary isolated rat islet cells. We then determined the role of each channel in insulin secretion by using channel-selective antagonists. Insulin secretion analysis revealed that N- and L-type channels are both involved in immediate glucose-induced insulin secretion. However, L-type was preferentially coupled to secretion at later time points. P/Q-type channels were not found to play a role in insulin secretion at any stage. It was also found that long-term exposure to elevated glucose increases basal calcium in these cells. Interestingly, chronically elevated glucose decreased the mRNA expression of the channels involved with insulin secretion and diminished the level of stimulated calcium influx in these cells. Using whole cell patch clamp, we found that N- and L-type channel currents increase gradually subsequent to lower intracellular calcium perfusion, suggesting that these channels may be regulated by glucose-induced changes in calcium.

Increase of the LC-MS/MS sensitivity and detection limits using on-line sample preparation with large volume plasma injection.

Large volume injection (LVI) has systematically been studied to improve LC-MS/MS sensitivity (signal-to-noise ratio, or S/N) and detection limits. The method of LVI was combined with on-line solid phase extraction (on-line SPE) and LC-MS/MS detection for analysis of compounds directly in plasma. It was demonstrated that LVI of plasma with on-line SPE-LC-MS/MS allows for improvement of sensitivity and detection limits without compromising chromatographic peak shape and resolution and inducing significant matrix and signal suppression effects. Furthermore, sensitivity and detection limits improve linearly with the injection volume up to 100 microL. Quantification of the model compounds in plasma demonstrated comparable calibration curve statistics, precision and accuracy for 5, 50 and 100 microL plasma injections.

[Association between A49T polymorphism of SRD5A2 gene and risk of prostate cancer].

OBJECTIVE: To investigate the association between A49T polymorphism of SRD5A2 gene and risk of prostate cancer. METHODS: PCR was used to examine the A49T polymorphisms of SRD5A2 gene in the tissues of prostate cancer resected from 112 patients (CaP group) and the specimens of benign prostate hyperplasia (BPH group) resected from 89 patients. The association of A49T polymorphism with age of onset, FPSA, TPSA, F/T, T stage, and Gleason score were analyzed. RESULTS: There was no significant difference in A49T polymorphism between the CaP and BPH groups (P > 0.05). The average age of CaP patients was significantly higher than that of the BPH patients (P < 0.05). In the CaP patients, the Gleason score was significantly higher, and the age of onset was significantly lower in the AT + TT genotype than in the AA genotype (both P < 0.05) 2. The age of onset of the AA + AT group was significantly lower than that of the AA group (P < 0.05). CONCLUSION: AA + AT genotype may be of worse prognosis, however, without significant difference. Rank scoring may reflect the relation between Gleason score and A49T genotype and estimate the prognosis better than two-level discrete evaluation.

Rhenium(V) complexes with thiolato and dithiolato ligands: synthesis, structures, and monomerization reactions.

The new compound {(PhS)(2)ReO(mu-SPh)}(2), 1, was synthesized from Re(2)O(7) and PhSH and then used as the synthon for a number of hitherto unknown oxorhenium(V) compounds. Reactions between dithiols and 1 (2:1 ratio) afford {PhSReO(dt)}(2), where the dithiols, dtH(2), are 1,2-ethanedithiol (edtH(2)), 1,3-propanedithiol (pdtH(2)), 1,3-butanedithiol (pdtMeH(2)), 1,2-benzenedithiol (bdtH(2)), 2-(mercaptomethyl)thiophenol (mtpH(2)), and 2-mercaptoethyl sulfide (mesH(2)). Similar reactions carried out with a 3:1 ratio of dtH(2) to 1 afford [(ReO)(2)(dt)(3)], dt = edt, pdt. When NEt(3) was introduced prior to the 3:1 reaction between edtH(2) and 1, a compound containing an anionic complex was isolated, [PPh(4)][ReO(edt)(2)]. The new compounds were characterized analytically, spectroscopically, and crystallographically. The Re-O groups in two of the compounds, 1 and {ReO(mu-SPh)(bdt)}(2), exist in rare anti orientations; the others adopt the more familiar syn geometry, as discussed. Selected monomerization reactions of {PhSReO(dt)}(2) were also carried out: {PhSReO(dt)}(2) + 2L = 2[PhSReO(dt)L]. The rate for L = 4-phenylpyridine is given by v = {k(a)[L] + k(b)[L](2)} x [{PhSReO(dt)}(2)], as it is for the reactions of {MeReO(dt)}(2); for all of these compounds, the reaction proceeds nearly entirely by the third-order pathway. Values of k(b)/L(2) mol(-2) s(-1) at 25.0 degrees C are 5.8 x 10(2) (mtp), 2.97 x 10(3) (pdt), 4.62 x 10(5) (edt), and 3.87 x 10(5) (bdt). The rate law for the reactions of {PhSReO(dt)}(2) with L = PAr(3) is v = k(a)[L]/{1 + kappa[L]} x [{PhSReO(dt)}(2)]. For PPh(3), values at 25.0 degrees C of k(a)/L mol(-1) s(-1) (kappa/L mol(-1)) for {PhSReO(dt)}(2) are 9.64 x 10(-2) (1.87) for mtp, 3.43 x 10(-2) (0.492) for pdt, 1.91 (1.42) for edt, 1.84 x 10(-2) (0.82) for bdt, and 1.14 x 10(3) (10.6) for 1. Mechanisms are proposed that are consistent with the data obtained and with earlier work.

Discovering patterns to extract protein-protein interactions from the literature: Part II.

MOTIVATION: An enormous number of protein-protein interaction relationships are buried in millions of research articles published over the years, and the number is growing. Rediscovering them automatically is a challenging bioinformatics task. Solutions to this problem also reach far beyond bioinformatics. RESULTS: We study a new approach that involves automatically discovering English expression patterns, optimizing them and using them to extract protein-protein interactions. In a sister paper, we described how to generate English expression patterns related to protein-protein interactions, and this approach alone has already achieved precision and recall rates significantly higher than those of other automatic systems. This paper continues to present our theory, focusing on how to improve the patterns. A minimum description length (MDL)-based pattern-optimization algorithm is designed to reduce and merge patterns. This has significantly increased generalization power, and hence the recall and precision rates, as confirmed by our experiments. AVAILABILITY: http://spies.cs.tsinghua.edu.cn.

Kinetic studies of spreading DMPC vesicles at the air-solution interface using film pressure measurements.

DMPC vesicles were injected into a 50 mM NaF solution in water, and the kinetics of a monolayer formation at the air-solution interface was investigated by measuring changes of the film pressure, pi, as a function of time. The studies were carried out in temperature range from 5 to 35 degrees C. The solutions were stirred in order to eliminate the mass transport limitations. Under these conditions, the monolayer formation was controlled by the surface processes only. At temperatures above the critical temperature Tc, compression isotherms were measured and used to convert the pi-t curves into the gamma-t plots, with gamma being the surface concentration of DMPC. These kinetic data indicate that the monolayer formation involves fast rupture of vesicles and formation of a constant number of disk-shaped monolayer islands at the air-solution interface that grow with a constant radial rate. At higher coverages, the growth is restricted by the availability of the monolayer-free surface area. At temperatures below Tc, the pi-t curves cannot be converted into the gamma-t plots. Here the kinetics can be discussed only qualitatively. The data indicate that the kinetics of the monolayer formation involves two steps. Initially, an expanded film is formed. At higher film pressures, the expanded film is slowly transformed into a liquid condensed state.
