New air-stable planar chiral ferrocenyl monophosphine ligands: Suzuki cross-coupling of aryl chlorides and bromides.

[reaction: see text] A novel class of planar chiral electron-rich monophosphine ligands has been developed. The modular design allows a short and efficient synthesis of an array of aryl-ferrocenyl derivatives carrying the donating bis(dicyclohexyl)phosphino moiety. These new ligands have successfully been applied in the palladium-catalyzed Suzuki cross-coupling of activated as well as nonactivated aryl chlorides at room temperature. The asymmetric coupling of an aryl bromide and an aryl boronic acid was also tested, giving ees up to 54%.

Modular approach to novel chiral aryl-ferrocenyl phosphines by Suzuki cross-coupling.

Two novel planar chiral and atropisomeric P,N and P,O aryl-ferrocenyl ligand systems have been developed. The strategy is short and involves a new synthetic approach to aryl-ferrocenyl compounds via a Suzuki cross-coupling procedure. The modular design can easily give access to variety of chiral mono- and bidentate ligands. Two simple derivatives of a novel chiral bidenate P,N ligand belonging to the MOPF family have been synthesized and tested in the enantioselective copper-catalyzed addition of diethyl zinc to an enone and a "difficult" diester. Moderate to excellent yields and enantioselectivities up to 58% were obtained using 1 mol % Cu(OTf)(2) and 1.5 mol % chiral ligand.

Pentamethylated and pentaphenylated azaferro- and azaruthenocenes: simple and general methodology for the preparation of enantiopure derivatives.

A methodology for the enantioselective synthesis of planar chiral 2-substituted 1',2',3',4',5'-pentamethylazaferro- and azaruthenocenes is reported. The key step is the introduction of an enantiopure chiral sulfoxide auxiliary via a selective ortho-lithiation with subsequent chromatographic separation of the resulting diastereomers. Cleaving off the sulfoxide auxiliary by treatment with t-BuLi generates an optically pure anion which may be quenched with a variety of electrophiles to give the optically pure azametallocene derivatives. In addition, it was attempted to extend the methodology to encompass 1',2',3',4',5'-pentaphenyl derivatives. It was, however, not possible to attach the chiral sulfoxide auxiliary onto the pentaphenylated azaferrocene, and for the azaruthenocene case, only one diastereomer could be isolated. Most importantly, the sulfoxide group could be cleaved off and the resulting chiral azaruthenocenyl anion was quenched with paraformaldehyde and iodine, resulting in products with ee values of 85% and 99%, respectively.

Synthesis and application of arylmonophosphinoferrocene ligands: ultrafast asymmetric hydrosilylation of styrene.

A short and efficient synthetic route to a novel class of atropisomeric and planar chiral 2-aryl-1-diphenylphosphanylferrocene ligands is presented. The modular design of the ligands allows a synthetic approach in which both the aromatic moiety and the phosphino substituent can be varied easily. This permits fine-tuning of steric and electronic properties. The ligands have been tested in the asymmetric hydrosilylation of styrene where enantioselectivities up to 90% are obtained. Optimization of the palladium-to-ligand ratio resulted in hitherto unparalleled turnover frequencies (TOF) exceeding 180 000 h(-1). The absolute stereochemistry of the ligands was determined from an X-ray structure. 2D NMR experiments in combination with ab initio calculations were used to assign the conformation of the atropisomeric biarylic scaffold.

Highly enantioselective hydrosilylation of aromatic alkenes.

Currently, the most effective and economic way to convert an alkene into an optically active alcohol is the two-step sequence: hydrosilylation/oxidation. Much work has been devoted to elucidating effective catalysts for this process, but hitherto only one effective and highly stereoselective process has been available. Herein we present a novel catalytic system for the asymmetric hydrosilylation of aromatic alkenes, giving the products in high yields and with the highest enantioselectivity (up to 99% ee) ever observed for this reaction. The reaction works efficiently for a variety of substituted aromatic alkenes, giving access after Tamao oxidation to almost optically pure benzylic alcohols in high yields.

Catalytic Approach for the Formation of Optically Active Allyl alpha-Amino Acids by Addition of Allylic Metal Compounds to alpha-Imino Esters.

A new catalytic enantioselective approach for the formation of allyl alpha-amino acid derivatives by reaction of N-tosyl alpha-imino esters with allyl stannanes and silanes catalyzed by chiral copper(I) complexes has been developed. A series of different BINAP and phosphine-oxazoline (P,N) ligands have, in combination with various Lewis acids, been tested as chiral catalysts for allylation of N-tosyl alpha-imino esters. It has been found that both type of ligands, in combination with copper(I) salts, give highly valuable unsaturated alpha-amino acid derivatives. The reaction has been investigated for different allyl stannanes and silanes, and it has been found that tri-n-butyl allyl stannane gives the best results of the simple allyl compounds tested, leading to gamma,delta-unsaturated alpha-amino acid derivatives in up to 94% yield and with up to 83% ee, which can be improved to be >95% ee by recrystallization. The reaction has also been investigated using different acyclic and cyclic allyl stannanes leading to various types of unsaturated alpha-amino acid derivatives in very high yield (up to 95%) and with up to 98% ee. The stereochemistry and absolute configurations of the allyl alpha-amino acid derivatives have been determined by X-ray analysis, and it is suggested that the reaction takes place as an ene-like reaction.

Total Synthesis of (R)-Dihydroactinidiolide and (R)-Actinidiolide Using Asymmetric Catalytic Hetero-Diels-Alder Methodology.

The total synthesis of the naturally occurring bicyclic lactones (R)-dihydroactinidiolide and (R)-actinidiolide is presented. The key step in the syntheses is the copper(II)-bisoxazoline-catalyzed hetero-Diels-Alder reaction of a cyclic diene with ethyl glyoxylate giving the hetero-Diels-Alder product in high yield and with very high regio-, diastereo-, and enantioselectivity. The total syntheses proceed via an intermediate, which also has the potential for a series of other natural products. The structure of the key intermediate is confirmed by X-ray analysis.
