Methoxyflavones protect cells against endoplasmic reticulum stress and neurotoxin.

Enhanced endoplasmic reticulum (ER) stress leads to cell death in various pathophysiological situations. During a search for compounds that regulate ER stress, we identified methoxyflavones, a group of flavonoids, as strong protective agents against ER stress. Analysis in mouse insulinoma MIN6 cells revealed that methoxyflavones mildly activated the eukaryotic initiation factor 2alpha and nuclear factor erythroid 2-related factor pathways, but not the XBP1 pathway, and induced downstream genes, including glucose-regulated protein (GRP) 78, a molecular chaperone in the ER. The protective effect of methoxyflavones was enhanced by agents that increase intracellular cAMP levels such as forskolin, dibutyryl-cAMP and IBMX, but suppressed by the protein kinase A (PKA) inhibitor H-89, suggesting involvement of the PKA pathway in the regulation of ER stress by methoxyflavones. Consistent with the results in cultured cells, pretreatment of mice with tangeretin, a methoxyflavone, enhanced expression of GRP78 and HO-1 without causing ER stress in renal tubular epithelium and prevented tunicamycin-induced cell death. Furthermore, preadministration of tangeretin in mice enhanced expression of GRP78 in the substantia nigra pars compacta and protected dopaminergic neurons against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine, a neurotoxin that induces both oxidative and ER stress. These results suggest that methoxyflavones play an important role in the regulation of ER stress and could be a therapeutic target for the ER stress-related diseases.

Allergy-preventive phenolic glycosides from Populus sieboldii.

Allergy-preventive activity was demonstrated for an extract of the bark of Populus sieboldii in a continuing search for allergy-preventive substances from natural sources. By bioassay-directed fractionation of this plant bark, two new phenolic glycosides, siebolside A {2-hydroxy-5-[(benzoyloxy)methyl]phenyl (6'-O-acetyl) beta-D-glucopyranoside} (1) and siebolside B {2-hydroxy-5-[(benzoyloxy)methyl]phenyl beta-D-glucopyranoside} (2), were isolated, together with three known compounds, salicin (3), sakuranin (4), and neosakuranin (5). The structures of 1 and 2 were elucidated by spectroscopic methods. Compounds 1-5 all showed allergy-preventive effects.

Growth inhibition of stilbenoids in Welwitschiaceae and Gnetaceae through induction of apoptosis in human leukemia HL60 cells.

Fifty-six stilbenoids isolated from the families of Welwitschiaceae and Gnetaceae were screened for growth inhibitory activity against HL60 cells, and two compounds (gnemonol G and gnetin I) among them exhibited a strong activity with IC(50) of 10.0 microM and 12.2 microM at 48 h incubation, respectively. The growth suppression by gnemonol G and gnetin I was found to be in part due to apoptosis which was assessed by morphological findings such as nuclear condensation and fragmentation, and DNA ladder formation in human leukemia HL60 cells.

Inhibitory activity of plant stilbene oligomers against DNA topoisomerase II.

The inhibitory activity of 40 stilbene oligomers isolated from six plant species against topoisomerase II was evaluated, of which nine compounds showed a potent inhibitory effect, stronger than daunorubicin, a topoisomerase II inhibitor, used as an anti-cancer drug. The specificity of active stilbene oligomers on topoisomerase II was assessed by their effect on DNA restriction enzyme. In particular, specific inhibitory activity was observed in alpha-viniferin 13-O-beta-glucopryranoside (2) and hemsleyanol C (13).

Melanogenesis stimulation in murine B16 melanoma cells by Kava (Piper methysticum) rhizome extract and kavalactones.

Melanogenesis stimulation activity of aqueous ethanolic extracts obtained from several different parts of five Piper species, namely Piper longum, P. kadsura, P. methysticum, P. betle, and P. cubeba, were examined by using cultured murine B16 melanoma cells. Among them, the extract of P. methysticum rhizome (Kava) showed potent stimulatory effect on melanogenesis as well as P. nigrum leaf extract. Activity-guided fractionation of Kava extract led to the isolation of two active kavalactones, yangonin (2) and 7,8-epoxyyangonin (5), along with three inactive kavalactones, 5,6-dehydrokawain (1), (+)-kawain (3) and (+)-methysticin (4), and a glucosylsterol, daucosterin (6). 7,8-Epoxyyangonin (5) showed a significant stimulatory effect on melanogenesis in B16 melanoma cells. Yangonin (2) exhibited a weak melanogenesis stimulation activity.

Nepalensinols D-G, new resveratrol oligomers from Kobresia nepalensis (Cyperaceae) as potent inhibitors of DNA topoisomerase II.

Four new resveratrol oligomers, nepalensinols D-G, were isolated from the stem of Kobresia nepalensis (Cyperaceae). The structures were determined by detailed NMR spectral analysis. The compounds were assessed for their inhibitory activity against human topoisomerase II, a potential target of anti-tumor agents. These stilbenoids showed potent inhibitory activity against human topoisomerase II with IC50 values of 5-15 microM.

Resveratrol oligomers and their O-glucosides from Cotylelobium lanceolatum.

Three new resveratrol oligomers, cotylelophenol C (1) (resveratrol tetramer) and cotylelosides A (2) and B (3) (O-glucosides of resveratrol trimer), together with four known glucosides of resveratrol oligomers (vaticasides A, B, C, D) and piceid, were isolated from an acetone soluble part of stem of Cotylelobium lanceolatum (Dipterocarpaceae). The structures of new compounds were determined by spectral data analysis. The characteristic properties observed in the NMR spectra of 1 were also discussed.

Stilbenoids of Kobresia nepalensis (Cyperaceae) exhibiting DNA topoisomerase II inhibition.

Resveratrol oligomers, nepalensinol A, B and C, were isolated from the stem of Kobresia nepalensis (Cyperaceae). The structures were established on the basis of chemical properties and spectroscopic evidence including 2D NMR spectroscopic analysis. Nepalensinol A, B and C showed a potent inhibitory effect on topoisomerase II -- stronger than etoposide (VP-16), a topoisomerase II inhibitor used as an anti-cancer drug. Nepalensinol B, in particular, exhibited the most potent activity with an IC(50) of 0.02 microg/ml.

Xanthones induce cell-cycle arrest and apoptosis in human colon cancer DLD-1 cells.

We investigated the antiproliferative effects of four structurally similar prenylated xanthones, alpha-mangostin, beta-mangostin, gamma-mangostin, and methoxy-beta-mangostin, in human colon cancer DLD-1 cells. These xanthones differ in the number of hydroxyl and methoxy groups. Except for methoxy-beta-mangostin, the other three xanthones strongly inhibited cell growth at 20 microM and their antitumor efficacy was correlated with the number of hydroxyl groups. Hoechst 33342 nuclear staining and nucleosomal DNA-gel electrophoresis revealed that the antiproliferative effects of alpha- and gamma-mangostin, but not that of beta-mangostin, were associated with apoptosis. It was also shown that their antiproliferative effects were associated with cell-cycle arrest by affecting the expression of cyclins, cdc2, and p27; G1 arrest was by alpha-mangostin and beta-mangostin, and S arrest by gamma-mangostin. These findings provide a relevant basis for the development of xanthones as an agent for cancer prevention and combination therapy with anti-cancer drugs.

A potent apoptosis-inducing activity of a sesquiterpene lactone, arucanolide, in HL60 cells: a crucial role of apoptosis-inducing factor.

Six main sesquiterpene lactones (germacranolides) from Calea urticifolia were evaluated for in vitro cytotoxicity against human tumor cell lines HL60 and SW480 cells. Among them, arucanolide and parthenolide displayed marked cytotoxicity against both cell lines. Arucanolide exhibited a low IC(50) in HL60 cells. The cytotoxic activity of arucanolide was observed at lower concentrations compared to that of parthenolide, which has been reported to be a typical and simple germacranolide. The activity was found to be mainly due to apoptosis that was assessed by morphological findings, DNA ladder formation (24 - 36 h), and flow cytometric analysis in HL60 cells. Western blotting and an apoptosis inhibition assay using caspase inhibitors did not demonstrate the activation of any caspases tested. However, the mitochondrial membrane potential of HL60 cells was lost after 24-h treatment with arucanolide, and concurrently apoptosis-inducing factor (AIF) released from mitochondria was detected by Western blot analysis. The inactivation of nuclear factor-kappaB, which has been commonly shown in parthenolide-induced apoptosis, did not occur in arucanolide-induced apoptosis. Taken together, the findings presented here indicate that arucanolide induced marked apoptosis in HL60 cells mainly by dissipating mitochondrial membrane potential, which would trigger AIF-induced apoptosis.

Four new trimeric stilbene glucosides from Welwitschia mirabilis.

Four new trimeric stilbene glucosides, mirabilosides C-F (1-4) were isolated from MeOH extract of stem and root of Welwitschia mirabilis (Welwitschiaceae) along with three known stilbenoids, resveratrol (5), gnemonoside B (6), and gnetin G (7). The structures of these compounds were elucidated by spectroscopic methods.

Two novel trimeric resveratrol derivatives from Cotylelobium lanceolatum.

Two new resveratrol (= 5-[(E)-2-(4-hydroxyphenyl)ethenyl]benzene-1,3-diol) trimers, cotylelophenols A (1) and B (2), were isolated from the stem of Cotylelobium lanceolatum (Dipterocarpaceae), together with ten known resveratrol oligomers (3-12). The structures of the isolates were established on the basis of spectroscopic analyses, including a detailed NMR spectroscopic investigation of 1 under different conditions. Compound 1 is the first resveratrol trimer with a rearranged 4-hydroxyphenyl group. Four possible biogenetic pathways towards resveratrol oligomers are proposed (Scheme).

New resveratrol tetramers from the stem bark of Upuna borneensis.

Five new resveratrol (=5-[(E)-2-(4-hydroxyphenyl)ethenyl]benzene-1,3-diol) tetramers, upunaphenols H-J (1-3) and trans-(4) and cis-upunaphenol K (5), were isolated from the stem of Upuna borneensis (Dipterocarpaceae). Their structures were elucidated on the basis of 1D- and 2D-NMR as well as FAB-MS data. Compounds 1-3 bear a rare biphenyl bond in their frameworks. Compounds 1 and 2 have an unprecedented nonacyclic fused ring system, and compounds 2 and 3 have symmetrical structures.

Inhibitory effects of xanthones from guttiferae plants on PAF-induced hypotension in mice.

The inhibitory effects of 22 xanthones from three Guttiferae plants (Hypericum patulum, Calophyllum inophyllum and C. austroindium) on exogenous platelet activating factor (PAF)-induced hypotension were examined using a blood pressure monitoring in vivo assay method. Guanandin (2), caloxanthone E (3), 1,3,5,6-tetrahydroxy-2-isoprenylxanthone (8), 6-deoxyjacareubin (11) and patulone (18) showed strong inhibition of PAF-induced hypotension, with inhibitory effects of more than 60 %. Their ID50 values were greater than that of ginkgolide B (BN-52 021), a natural PAF-antagonist from the Ginkgo biloba.

Antianaphylactic and antipruritic effects of the flowers of Impatiens textori MIQ.

The anti-anaphylactic and anti-pruritic activities of a 35% EtOH extract (IT) of the flowers of Impatiens textori MIQ. were investigated by in vivo assay. IT and apigenin (1), apigenin 7-glucoside (2) and luteolin (3), principal compounds from IT, inhibited compound 48/80 (COM)-induced by blood pressure (BP) decrease, which was an immunoglobulin (Ig)E-independent anaphylaxis-like response. Compounds 1-3 all inhibited BP decrease induced by IgE-dependent anaphylaxis. Furthermore, IT also inhibited the blood flow (BF) decrease induced by antigen-induced anaphylaxis in actively sensitized mice. IT showed a significant inhibitory effect on scratching behavior induced by COM without a central depressant. IT also significantly inhibited platelet activating factor (PAF)- and serotonin (5-HT)-induced scratching behavior and mitigated protease (PA)-induced scratching behavior. These findings showed that the flowers of I. textori can be utilized as an anti-anaphylactic and anti-pruritic agent in addition to the traditional applications of this plant.

Resveratrol derivatives from Upuna borneensis.

Four new resveratrol derivatives, upunaphenols B (1), C (4), D (5) (resveratrol tetramer) and E (6, resveratrol dimer with a C6-C1 unit), together with nine known resveratrol oligomers and resveratrol were isolated from an acetone soluble part of stem of Upuna borneensis (Dipterocarpaceae). The structures of new compounds were determined by spectral analysis including 1D and 2D NMR experiments.

A new and known cytotoxic aryltetralin-type lignans from stems of Bursera graveolens.

A new 4alpha-aryltetralin-type lignan called burseranin (1) and a known analogous lignan picropolygamain (2) were isolated along with known triterpenes, lupeol and epi-lupeol from the methanol extract of stems of Bursera graveolens, which showed a remarkable inhibitory activity against human HT1080 fibrosarcoma cells. The whole structure of 1 was established based on combined spectral studies and the absolute structure for 2 was first confirmed by CD spectral evidence. In addition, cytotoxic activities of the stem (methanol) extract and its components are evaluated in this paper.

Flavonol glycosides in leaves of two Diospyros species.

Fourteen flavonol glycosides including two new compounds were isolated from the leaves of two Diospyros plants (D. cathayensis and D. rhombifolia). The structures of isolated compounds were determined by spectroscopic analysis. The scavenging activity of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical of the isolated compounds was also investigated.
