[Cell therapy as a method to correct pathogenetic disturbances in dyslipidemia and early atherosclerosis].

The article is dedicated to the pathogenetic mechanisms of the development of immune inflammation in the vascular wall, as well as hepatic functional disregulation in dislipidemia and atherosclerosis. The authors emphasize that the decompensation of the process of cell regeneration in actively functioning organs (vascular endothelium, hepatocytes) and a concomitant immune disregulation become the main factors of the progression of dislipidemia and atherosclerosis. The authors consider the conditions for and factors of cell therapy, based on the use of allogenic fetal hepatic cells and autological bone marrow cells.

[Inflammation and apoptosis in relation with the effectiveness of bone marrow cell transplantation to patients with chronic heart failure].

The authors analyze the pathogenetic significance of inflammation, immune activation, and apoptosis in the development of heart failure and the functional regeneration of left ventricular myocardium after treatment including autologic bone marrow cell transplantation. The paper shows that autologic bone marrow cell transplantation influences the same mechanisms that cause heart failure, namely inflammation, apoptosis, and neurohumoral mechanisms. Investigations have made it possible to formulate guidelines for the prediction and monitoring of the effects of treatment including autologic bone marrow cell transplantation in patients with heart failure.

First experience of the use bone marrow mesenchymal stem cells for the treatment of a patient with deep skin burns.

Female patient with extensive skin burn (I-II-IIIAB skin burn, total area 40%, area of IIIB degree 30%) was treated using transplantation of allogenic fibroblast-like bone marrow mesenchymal stem cells onto the surface of deep thermal burn. The study of wound healing dynamics after transplantation of allogenic fibroblast-like mesenchymal stem cells confirmed high tempo of wound regeneration in the presence of active neoangiogenesis. Due to this, autodermoplasty of burn wounds could be carried out with good results as early as on day 4 after transplantation of fibroblast-like mesenchymal stem cells; this led to more rapid healing of donor zones and accelerated rehabilitation of the patient.

Use of recombinant AdSV40-BetaGal adenoviral construction for monitoring of transplanted cells.

Original recombinant adenoviral construction carrying E. coli beta-galactosidase LacZ gene designed by the authors is convenient for labeling and monitoring of bone marrow mesenchymal (stromal) progenitor cells and myocardial and skin fetal cells transplanted in damaged rat tissues (in the perinecrotic zone of the myocardium and onto burnt skin surface) for their reparation. This genetic construction after pre-inactivation of endogenous beta-galactosidase allows to detect transplanted cells in the foci of injury; positive effects of transplantation on tissue reparation processes can be attributed to the presence of transplanted cells.

Recovery of contractile function of cryodamaged rat myocardium after transplantation of fetal cardiomyocytes and predifferentiated bone marrow stromal stem cells.

The effect of cell transplantation into cryodamaged rat myocardium was studied on isolated hearts by increasing functional load to the left ventricle. Transplantation of allogeneic fetal cardiomyocytes improved the function of the left ventricle under conditions of considerably increased preload. Transplantation of autologous mesenchymal stem cells repaired left-ventricular function under conditions of increased pre- and afterload.

Differentiation of bone marrow stromal stem cells into cardiomyocyte-like cells in different mammalian species.

We describe the possibility of obtaining cardiomyocyte-like cell cultures from rat, guinea pig, and human bone marrow stromal stem cells. The content of troponin I-positive cells attains 35-45% of the total number of cells in the cultures and persists at this level for up to 4 months under differentiation conditions. Spontaneous contractions of cardiomyocyte-like cells were observed after the formation of cell monolayer under differentiation conditions.

Dyslipidogenic microangiopathy in guinea pigs at early stages of atherogenesis.

We studied the effect of dyslipidemia on lipid metabolism, state of microcirculatory system, and morphological alterations in the aorta and liver of guinea pigs at the early stages of experimental atherogenesis. The important role of microcirculatory disorders in the development of regional pathology and atherosclerosis is confirmed. The proposed alimentary model can be used in the development of novel methods for prevention and treatment of atherosclerosis.

Mesenchymal bone marrow stem cells more effectively stimulate regeneration of deep burn wounds than embryonic fibroblasts.

Regeneration of deep burn wounds after transplantation of allogenic and autogenic fibroblast-like bone marrow mesenchymal stem cells and embryonic fibroblasts on burn surface was studied in 40 Wistar rats. Transplantation of allogenic and autogenic fibroblast-like bone marrow mesenchymal stem cells and transplantation of embryonic fibroblasts decreased cell infiltration of the wound and accelerated the formation of new vessels and granulation tissue in the wound in comparison with the control (burn wounds without cell transplantation). Regeneration processes were most active after transplantation of fibroblast-like bone marrow mesenchymal stem cells, in particular, autogenic cells, which was confirmed by more rapid decrease in burn surface area. Wound healing after transplantation of fibroblast-like bone marrow mesenchymal cells and embryonic fibroblasts was associated with long functioning of transplanted cells (as was shown by staining for beta-galactosidase, the cells were transfected with an adenovirus vector carrying the marker gene). It is hypothesized that more rapid regeneration of burn wounds after transplantation of fibroblast-like bone marrow mesenchymal stem cells was due to low differentiation of these cells in comparison with embryonic fibroblasts.

[The use of pre-differentiated mesenchymal bone marrow stem cells for treatment of deep burn wounds].

The opportunity of faster closing deep burn wounds by using fibroblast-like mesenchymal bone marrow stem cells (FMSC) and embryonic fibroblasts (EF) was investigated in rats. It was shown that FMSC as well as EF transplanted onto burn surfaces reduced the expression of cell infiltration but accelerated the formation of vessels de novo and granulation tissue in the wounds. These changes form the conditions for faster closing the burn wounds as compared to the control wounds (without cell transplantation). High rate of wound closing induced by FMSC and EF is thought to be caused by a long period (up to 30 days) of vital activity of the cells grafted on the burn surface. It was also found out that the rate of wound regeneration induced by FMSC was higher that that induced by EF.

An isocratic, reversed-phase HPLC method for the determination of postischemic efflux of purines and pyrimidines during reperfusion of isolated liver.

An isocratic, reversed-phase HPLC method was developed for the determination of the rates of purine and pyrimidine efflux during early reperfusion of isolated organs after non-perfusion cold conservation. The method enables determination of uric acid, cytidine, xanthine, hypoxanthine, uridine, AMP, inosine, and adenosine in liver perfusate using a standard C-18 column (25 cm length). Peaks are resolved by elution with buffer containing 1% acetonitrile, 20 mM potassium citrate (pH 6.25), and 25-55 mM tetramethylammonium. The effects of pH and solvents on peak retention times are described. As an example of the application of the method, the effects of allopurinol on the rates of postischemic efflux of purines and pyrimidines during reperfusion of liver stored in the cold for 24 h in Euro-Collins solution was studied.

[Functional activity of placental, splenic and hepatic tissues in pyo-septic diseases under conditions of extracorporeal detoxication].

Immunocorrective and detoxication treatment by plasma perfusion through fragments of alloplacental tissue (group 1), fragments of xenosplenic tissue (group 2) and isolated xenohepatocytes (group 3) was carried out as an adjuvant therapy in 61 patients with pyoseptic diseases. Control group consisted of 21 patients. Comparative analysis of effectiveness of the treatment showed, that in patients of group 2 after 5 days substantial increase of lymphocytes count was detected due to increased pool of T- and B-lymphocytes. In patients of group 1 increase in lymphocytes content occurred even in greater degree, but it occurred mainly owing to the augmentation of B-lymphocytes pool. In patients of groups 1 and 2 already to the 3rd day an increase of FAL as well IgA and IgM occurred. There was no immunocorrective effect in patients of group 3, as well as control group. However, detoxication activity, having been evaluated by bilirubin and cytolytic enzymes dynamics, by urea and kreatinine level increased in all patients of groups 1, 2, 3 in comparison with the control. It is suggested, that allogenic placental tissue could be recommended for the treatment of pyoseptic conditions, especially in patients with transplanted organs, because this tissue electively activates only humoral immunity.

[The enhancement of treatment efficacy in chronic liver failure by using hemoperfusion across donor hepatocytes and spleen fragments].

Hemoperfusion via donor hepatocytes and splenic fragments is an effective adjuvant method of treatment of hepatic insufficiency, especially in patients with autoimmune disease. Hemoperfusion effect occurs due to normalization of mesenchymoinflammatory and immunological indices as a result of positive action of hepatotropic and immunotropic factors of the spleen. The highest effectiveness of the above hemoperfusion is reached in its early use, that is before the disease becoming chronic.

[The use of perfluorocarbon emulsions for suppression of rejection reaction in kidney allotransplantation].

Successful solution of the problems of kidney transplantation is associated with further study of the pharmacological correction of the function of the kidneys. The article discusses the prospects of the use of perfluoran, a plasma substitute with a function of oxygen transport based on a perfluorocarbon emulsion, in the posttransplantation period for increasing the positive results of kidney transplantation. From the results of 24 dog experiments with allotransplantation of kidneys, a conclusion is drawn that the decrease of creatinine concentration in the blood with the use of perfluoran may be a consequence of the antiischemic properties of the emulsion. It is pointed out that thorough study of the condition of the parenchymatous organs in the body and the determination of the sequelae of the use of these blood substitutes are necessary.

[Experience in using preserved xenogeneic hepatocytes in the combined treatment of jaundice patients].

The authors give brief characteristics of the apparatus "subsidiary liver" and describe the mechanism of its work. The method of taking the liver of piglets and isolation of hepatocytes are described. The characteristics of patients are given in who the apparatus "subsidiary liver" was used. Certain parameters of blood were shown to be reduced after using the apparatus "subsidiary liver".
