Active conformation of 1,4-dihydropyridine calcium entry blockers. Effect of size of 2-aryl substituent on rotameric equilibria and receptor binding.

The conformational requisites at the receptor for unsymmetrically substituted phenyl-1,4-dihydropyridine calcium entry blockers are examined by screening a series of (2'-halophenyl)-1,4-dihydropyridines 1-4, with increasing bulk at the 2'-position of the phenyl ring, for their ability to relax potassium-contracted rabbit aortic smooth muscle and to competitively displace [3H]nitrendipine from its specific binding sites on guinea pig skeletal muscle. The fraction of synperiplanar rotamer in solution for these compounds, as determined by the nuclear Overhauser enhancement method, shows a positive correlation with vasorelaxant activity and receptor binding affinity. These findings are consistent with the synperiplanar rotamer of nonrigid unsymmetrically substituted phenyl 1,4-dihydropyridine calcium channel blockers being the receptor-bound conformation.

Studies directed toward ascertaining the active conformation of 1,4-dihydropyridine calcium entry blockers.

A series of unsymmetrically substituted 4-phenyl-1,4-dihydropyridine calcium entry blockers were investigated for their ability to relax potassium-contracted rabbit aortic smooth muscle and to competitively displace [3H]nitrendipine from its specific binding sites on guinea pig myocardial membranes in order to delineate the pharmacologically active conformer with respect to the position of the aromatic substituent (synperiplanar or antiperiplanar). The data show that the 1,4-dihydropyridine receptor distinguishes between 2',3'-disubstituted phenyldihydropyridines and 2',5'-disubstituted analogues as measured by changes of vasodilation and receptor affinity in vitro. The IC50 values for vasorelaxation by the analogues presented here correlate best with the Kd values for binding to the predominant receptor of two coexisting dihydropyridine binding sites in the guinea pig myocardium. We report the first observation of an antiperiplanar orientation of an o-phenyl substituent in the X-ray structure of 2-chlorophenyl analogue 3. Using nuclear Overhauser enhancement, we have developed a method that also demonstrates that an ortho (chloro or nitro) substituent on the phenyl ring does not preclude the presence of either synperiplanar or antiperiplanar phenyl rotamer in solution. These experimental findings contrast with the accepted belief that o-phenyl substituents essentially force these 1,4-dihydropyridines into the synperiplanar conformation exclusively.

RNA and homology mapping of two DNA fragments with repressible acid phosphatase genes from Saccharomyces cerevisiae.

Two EcoRI restriction fragments carrying Saccharomyces cerevisiae repressible acid phosphatase genes were analyzed. Transcripts were mapped by restriction endonuclease cleavage of glyoxal-stabilized R-loops and by gel blot hybridizations to cDNA. Homology between the two fragments was examined by gel blots and heteroduplex analysis. Each fragment carried a region of about 1.5 kilobases that coded for a repressible acid phosphatase, and these regions showed homology to one another. In addition, one fragment carried a second region of somewhat lower homology that probably codes for the so-called constitutive acid phosphatase.

Health attitudes and knowledge, and coronary risk factors in high-school children, Sydney and Inverell.

Attitudes to, and knowledge of, health and nutrition, together with presence of major coronary risk factors, were assessed in New south Wales high-school students at the start of an experimental healthy lifestyle programme. Data were obtained for 2596 children (mean age, 12.5 to 15.5 years), of whom 1971 lived in Sydney and 625 in rural Inverell. Girls had higher scores than boys on questions related to nutrition and fitness attitudes. Girls' scores on questions related to health attitudes and knowledge and nutrition attitudes and knowledge increased with age. The earliest precursors of coronary heart disease and chronic lung diseases were present in this group of children, of whom 20% to 30% were smoking cigarettes, 30% were overweight or obese, 8% to 15% of boys and 2% to 7% of girls had high blood pressure, and 9% had hypercholesterolaemia.

Isolation of yeast genes with mRNA levels controlled by phosphate concentration.

A library of DNA from the yeast, Saccharomyces cerevisiae, was constructed in phage lambda Charon 4 vector and then screened by differential plaque filter hybridization for genes induced by phosphate starvation. Two EcoRI fragments of 7.9 and 5.0 kilobase pairs that contained such genes were isolated. These cloned fragments may each carry one of the several copies of the genes for the repressible acid phosphatase of yeast. The fragments were use to examine mRNA levels of these genes in regulatory mutants of acid phosphatase.
