[The reactogenicity and efficacy of a dried cell-free staphylococcal vaccine in treating chronic bronchitis].

The data obtained in the controlled trial of the immunotherapy of chronic bronchitis (CB) patients with lyophilized acellular staphylococcal vaccine developed at the Mechnikov Research Institute for Vaccines and Sera (USSR Acad. Med. Sci.), Moscow, are presented. The patients were divided at random into two groups; of these, one group received the vaccine and the other, placebo (0.9% sodium chloride solution). The preparations were injected subcutaneously simultaneously with traditional treatment. The vaccine was well tolerated by the patients. In the blood of the patients receiving the vaccine, in contrast to those receiving placebo, a significant increase in the level of specific antibodies determined in the passive hemagglutination test and IgG was noted. When following the remote consequences of the disease for a year after the course of immunotherapy, a significant decrease in the occurrence of aggravations, an increase in the duration of remission and a decrease in the duration of sick leaves were registered. These data indicate that the immunotherapy of CB with lyophilized acellular staphylococcal vaccine is a promising method of preventing relapses.

[The effect of immunotherapy with cell-free vaccines made from the antigens of opportunistic microorganisms on the dynamics of DNA antibody formation].

The sera of patients subjected to immunotherapy with staphylococcal vaccine and with multicomponent vaccine (i.e. the mixture of the antigenic preparations of Staphylococcus aureus, Klebsiella pneumoniae, Proteus vulgaris, Escherichia coli) were studied by the method of the enzyme immunoassay on the basis of cattle spleen DNA. Immunotherapy with staphylococcal vaccine was given to patients with dermal diseases, chronic obstructive bronchitis and pulmonary abscess. Multicomponent vaccine was introduced to patients with the infectious allergic form of bronchial asthma, moderate or severe. Immunotherapy with both preparations under study was shown to produce no accumulation of antibodies to native and denatured DNA.

[The specific activity of a multicomponent vaccine made from the antigens of opportunistic microorganisms].

Multicomponent vaccine prepared from the antigens of 4 representatives of opportunistic microflora possesses high specific activity. The passive hemagglutination (PHA) test with the use of associated diagnosticum showed that antibody titers in the sera of immunized rabbits increased 10- to 10(4)-fold in comparison with the titers observed prior to immunization. The PHA test with the use of the antigens contained in the vaccine revealed the accumulation of antibodies to each of the 4 components of the preparation in the blood sera of immunized rabbits. When stored at 4 degrees C, the vaccine was shown to retain its specific activity for 5 years (the term of observation).

[The mutagenicity of a new multicomponent vaccine made from the antigens of Klebsiella pneumoniae, Staphylococcus aureus, Proteus vulgaris and Escherichia coli].

Inactivated bacterial vaccine, containing K. pneumoniae, S. aureus, P. vulgaris and E. coli antigenic complexes were tested for mutagenicity in the test described by Ames et al. and in vivo, in experiments on mice. In Salmonella typhimurium cells, strain TA-98 and TA-100, the preparation (5-75 mg/ml) did not increase the frequency of reversions and histidine-independence either in direct experiments or after metabolic activation with rat liver homogenate. In experiments on mice the vaccine (3.3 mg/kg and 33 mg/kg) did not induce chromosomal anomalies in spermatogonia. In all experiments the mutagens used for positive control produced a mutagenic effect.

[The reactogenicity and immunological efficacy of a Klebsiella vaccine in donors].

Klebsiella vaccine, when injected subcutaneously to donors, proved to be faintly reactogenic and safe. The injection of the vaccine had no effect on changes in the morphological composition of peripheral blood and on liver function. In persons with the initially low content of IgG an increase in this characteristic was observed after immunization. No changes in the synthesis of IgE occurred in healthy donors under the influence of immunization. The vaccine was shown to be immunogenic when introduced according to immunization schedules comprising 3 and 5 injections, the titer of Klebsiella antibodies increasing 3- to 5-fold.

[The immunobiological properties of the antigenic preparations obtained from a vaccinal strain of Klebsiella pneumoniae 204 grown in media of differing compositions].

In the technology of the cultivation of K. pneumoniae vaccine strain 204 with a view to obtaining biomass for the production of antigenic preparations the traditionally used culture medium with full nutritional value has been replaced by the alternative variant of synthetic medium. The specific physiological and morphological features of this strain grown in synthetic culture medium have been studied and described. Irrespective of the composition of the culture media used for cultivation, the antigenic preparations have been shown to have no difference in their chemical composition, immunogenic and toxic properties.

[Postvaccinal reactions to the administration of a dried staphylococcal vaccine made from water-soluble antigens].

An unequivocal regularity in local and systemic reactions to multiple (in 5 injections) administration of lyophilized staphylococcal vaccine prepared from water-soluble antigens has been established in patients and in healthy adults: the number and intensity of reaction decreased after each subsequent injection. A similar tendency has been observed in the vaccinees who were examined for their IgE levels. These data are indicative of the desensitizing action of the vaccine.

[Immunostimulant preparation made from the antigens of opportunistic microorganisms in the treatment of inflammatory lung diseases].

The results of the trial of an immunostimulating preparation, consisting of Klebsiella pneumoniae, Proteus, Escherichia coli and Staphylococcus antigenic complexes, on 20 patients with acute pulmonary abscess and bronchiectasis are presented. The preparation was introduced subcutaneously in 5 injections. The preparation was found to have low reactogenicity, and in the course of immunotherapy the manifestations of systemic and local reactions became considerably less pronounced. Immunotherapy produced a good curative effect, objectively manifested by a decrease in coughing and in the amount of sputum gradually changing its character. After the fourth and fifth injections the patients no longer ejected purulent sputum. Fluoroscopic examination revealed a considerable decrease in the size of the pathological focus. The preparation stimulated immunological reactions, and immunization resulted in a considerable increase in the titer of antibodies to all components of the combined preparation, as well as in an increase in the number of functionally active T-lymphocytes, in the blood of the patients.

[Experimental study of the immunological activity of a preparation made from the antigenic complexes of opportunistic microorganisms administered by different routes].

The data obtained in the experimental study of the humoral factors of local and systemic immunity, as well as the morphofunctional changes of internal organs, after multiple subcutaneous and aerosol immunization with the combined preparation of Proteus, Staphylococcus, Klebsiella pneumoniae and Escherichia coli antigens (preparation nC-4) are presented. The subcutaneous and aerosol administration of preparation nC-4 has been found to induce an increase in the levels of antibodies to all components of the preparation in the blood serum and in respiratory tract secretions. The introduction of the preparation through the respiratory tract resulted in an earlier intensive accumulation of specific antibodies and IgA in respiratory tract secretions. The results of the quantitative cytological study of respiratory tract secretions and the morphofunctional changes of the lymphoid tissue associated with the lungs are indicative of an important role played by cell-mediated immunity factors in the formation of local postvaccinal immunity.

[Protective activity of a cell-free Klebsiella vaccine in infection in mice caused by Streptococcus pneumoniae serotypes].

The capacity of dried Klebsiella cell-free vaccine, obtained from strain No. 204 by the disintegration of microbial mass with hydroxylamine, for protecting mice from pneumococcal infection caused by S. pneumoniae, serotypes 3, 4 and 9N, has been studied. Klebsiella vaccine has been found to possess immunostimulating potency with respect to the S. pneumoniae serotypes under study. On day 5 this potency is manifested to a greater extent than 24 hours after immunization. The combination of Klebsiella vaccine with Proteus vulgaris, Staphylococcus aureus and Escherichia coli K-100 antigens enhances the stimulation of nonspecific resistance.

[Immunostimulating activity of combined antigens of Klebsiella pneumoniae, Staphylococcus, Proteus and Escherichia coli and separate components in cross infection].

Experiments on mice have revealed that the combined preparation of K. pneumoniae, Staphylococcus, Proteus and E. coli antigens possesses pronounced immunostimulating activity against the microorganisms whose antigens make up this combined preparation, as well as against Haemophilus influenzae, type b. Cross protection between the antigens making up the preparation was observed, as well as of each of its components with respect to H. influenzae, type b. The activity of the combined antigenic preparation has been shown to exceed that of its monocomponents, as manifested under the conditions of cross infection. These experimental data may serve as a prerequisite for the development of an immunostimulant for the treatment of patients with chronic pyoinflammatory diseases.

[Experimental study of the immunogenicity of a monopreparation of Klebsiella pneumoniae antigen complex and its association with Staphylococcus, Proteus and Escherichia coli antigens].

The study carried out in mice with experimental Klebsiella sepsis has revealed that Staphylococcus, Proteus and E. coli antigenic complexes used as monovaccines ensure the protection of a definite percentage of the animals from K. pneumoniae infection. The mixture of these 3 preparations possesses a higher protective potency. The immunogenic potency of K. pneumoniae antigenic complex used as a component of combined vaccines with 2 or 4 components has proved to be sufficiently high and not inferior to the potency of K. pneumoniae monovaccine.

[Protective activity of a cell-free Klebsiella vaccine in relation to different Klebsiella pneumoniae serovars].

The capacity of dried Klebsiella cell-free vaccine, obtained from strain No. 204 by the disintegration of microbial cells with hydroxylamine, for protecting mice from Klebsiella septic infection caused by the homologous serovar and 9 heterologous serovars of K. pneumoniae was studied. The newly developed preparation was found capable of stimulating immunity not only to the homologous K. pneumoniae serovar, but also to other K. pneumoniae heterologous serovars: K1, K9, K11, K16, K20, K61. The protective capacity of the preparation with respect to these serovars was not inferior to that of the vaccines prepared by the same method from the corresponding homologous strains. The capacity of the vaccine to protect mice from Klebsiella sepsis was manifested irrespective of the virulence of the strains used for challenge.

[Nonharsh methods of isolation of antigenic complexes of Klebsiella pneumoniae, and analysis of their protective activity].

Protective antigenic complexes capable of protecting mice from septic Klebsiella infection can be obtained from Klebsiella pneumoniae. The preparations obtained by the method of aqueous extraction and by the action of hydroxyl amine and autolysis have been found to possess approximately the same protective activity and to protect mice from infection with Klebsiella pneumoniae strains belonging to both homologous and heterologous serotypes.

[Combined Staphylococcus-Proteus-Pseudomonas vaccine. II. The toxicity of the vaccine in "chronic" experiments and its ability to protect animals against Proteus and Staphylococcus infections].

The combined preparation consisting of the antigenic complexes of staphylococci (1 part), Proteus (1 part) and P. aeruginosa parts) was capable of protecting mice from infection with staphylococci, Proteus and P. aeruginosa and prolonging the survival time of rabbits under the conditions of the development of staphylococcal sepsis. The staphylococcal component of the combined preparation possessed adjuvant activity, increasing the immunogenicity of Proteus antigen. The combined vaccine enhanced a short-time increase in the nonspecific resistance of the animals. The moderate toxicity of the preparation permits making multiple injections of the preparation to mice without inhibiting the weight gain of the animals.

[Immunostimulating activity of antigenic complexes isolated from various Staphylococcus strains].

The study of the immunostimulating potency of the staphylococcal antigenic complex obtained from different strains by aqueous extraction indicated that the preparations obtained from highly virulent strains showed the lowest potency after challenge with both homologous and heterologous strains. The preparations obtained from strains with low virulence stimulated resistance to highly virulent strains to a greater extent than the preparations from the same virulent strain which was used for challenge. The necessity of revising the requirements to strains used for the production of antibacterial staphylococcal preparations is substantiated.
