[Klebsiella pneumoniae secreted protein-containing antigens in the system of adaptive immunity].

The study was aimed at the evaluation of the antigenic properties of K. pneumoniae secreted protein-containing antigens with a molecular weightt of 21 and 34-35 kD, obtained from supernatant culture fluid. As confirmed by the method of flow cytofluorimetry, the protein-containing fractions belonged to the secreted components of the microbial cell. The fraction with a molecular weight of 34-35 kD possessed high antigenic activity and contributed to the formation of specific antibodies after the immunization of mice. At the same time none of the protein fractions lead to an increase in the level of autoantibodies in mouse blood sera to organ-unspecific and organ-specific antigens. As revealed by the method of solid-phase, in 6 (27.3%) from 22 patients of patients with rhizomelic spondylitis had an increased level of IgG to K. pneumoniae cell-wall antigens with a molecular weight of 34-35 kD. An increase in the level of IgG to the secreted protein-containing fraction with a molecular weight of 34-35 kD was detected only in one patient (4.5%) (p<0.05).

[Immunomodulators of microbial origin enhance cytotoxicity of human mononuclear leukocytes and reduce metastatic progression of Lewis lung carcinoma in mice].

Effect of immunomodulators for microbial origin on innate immunity and antitumor system was continued to study. Immunomodificator Immunovac VP-4, purified staphylococcal toxoid and glucosaminyl muramyl dipeptide (GMDP) equally enhanced cytotoxicity of mononuclear leukocytes of peripheral blood of healthy donors. Index of cytotoxicity was 2.78, 2.77 and 2.70 respectively. Reduced metastatic progression of Lewis lung carcinoma in mice was observed after Immunovac VP-4 and GMDP administration. Effectiveness was seen when preparations administered according to schedules including their administration before implantation of the tumor. If preparations were administered number of metastases reduced in 4.4-5.6 times and size of metastases reduced in 7-10 times. Interplay between antitumor activity of studied immunomodulators and cytotoxic activity of NK-cells, which are base effectors of antitumor immune response, are discussed.

[The antigen-specific activity of a multicomponent vaccine administered orally and subcutaneously].

The antigen-specific activity of polycomponent vaccine VP-4 consisting of Klebsiella pneumoniae, Staphylococcus aureus, Proteus vulgaris and Escherichia coli antigens was studied. The same form of the vaccine was used for both subcutaneous injection and oral administration. The oral administration of the preparation to rabbits and guinea pigs induced a rise in the level of antibodies to all components of the vaccine in their blood sera, the optimum oral doses exceed only 2- to 4-fold the doses used for subcutaneous injection. The form of vaccine VP-4, developed for subcutaneous injection, may be recommended for wider trials with its oral administration.

[The protective activity of a multicomponent vaccine made from the antigens of opportunistic microorganisms when administered orally].

Multicomponent vaccine VP-4, prepared from Streptococcus, Klebsiella, Proteus and Escherichia coli antigens, was tested for protective activity by oral administration to mice with experimental Klebsiella, Proteus and S. pneumoniae infections. Vaccine VP-4, when introduced orally (in 2-3 administrations of 2 mg), was found to produce a protective effect with respect to K. pneumoniae (the effectiveness index was equal to 5.7 and 4.3). With respect to P. vulgaris, the effectiveness index was 2.7 (immunization was made in two oral administrations of 2 mg); in pneumococcal infection preliminary oral immunization with vaccine VP-4 in two administrations in a dose of 2-4 mg produced high protective effect. Thus, immunization with vaccine VP-4, introduced orally in comparatively small doses, proved to be effective in animal models used in the experiment.

[The immune reactivity of patients with chronic bronchitis on vaccinal therapy].

The treatment of 17 patients having chronic obstructive bronchitis with a combined vaccine resulted in a rise in the number of T lymphocytes and T suppressors, as well as a simultaneous increase in their functional activity, in these patients. Moreover, an increase in the ingestive activity of neutrophils and in the concentration of immune complexes was registered. Treatment with the vaccine containing Staphylococcus aureus, Klebsiella pneumoniae, Proteus and Escherichia coli antigens facilitated an increase in the titers of antibodies to most antigens contained in the vaccine and in the resistance of the body to infection. Vaccinal therapy made it possible to achieve a significant prolongation of the period of remission.

[The efficacy and reactogenicity of a cell-free staphylococcal vaccine in the immunotherapy of patients with chronic pyodermas].

The clinical effectiveness, reactogenicity and immunological activity of cell free staphylococcal vaccine in the immunotherapy of chronic pyoderma patients under the conditions of controlled trials were studied. Immunotherapy was carried out in combination with traditional methods of treatment. A group of 39 patient received cell free staphylococcal vaccine subcutaneously; the second control group (30 patients) received the commercial preparation of staphylococcal antiphagin and the third group (31 patients) was treated by traditional methods without vaccines. The study revealed that cell free staphylococcal vaccine had low reactogenicity and was well tolerated by patients; in contrast to the methods of treatment in the control groups, the vaccine stimulated an increased levels of specific antibodies and IgA in the blood of the patients. Immunotherapy with cell free staphylococcal vaccine led to clinical convalescence and remission in 1-3 years (the term of observation) in 71.8% of cases. In the two control groups such effect did not exceed 23%.

[The fractional composition of hydroxylamine preparations and of the lipopolysaccharide of a vaccinal strain of Klebsiella pneumoniae].

The fractional composition of the vaccine preparation obtained by the hydroxylamine treatment of K. pneumoniae 204 cells was studied with the use of gel chromatography in sepharose CL-6B. The preparation was shown to form 2 peaks, only the first high molecular peak being serologically active. The method for the treatment of the culture, obtained directly from a fermenter and stored in a frozen state before treatment, was proposed. In the technology of the manufacture of Klebsiella vaccine preparation the possibility of replacing dialysis with gel chromatography was shown.

[The immunotherapy of bronchial asthma with a multicomponent vaccine administered intranasally and orally].

Multicomponent vaccine prepared from opportunistic bacteria, when administered intranasally and orally to patients with bronchial asthma and chronic bronchitis, was well tolerated by the patients. Side effects induced by the vaccine in a few patients lasted for a short time and were mainly local. Immunotherapy with the vaccine led to a decrease in the severity of the course of the disease, as well as in the frequency and severity of exacerbations. In 26 out of 35 adult patients (74.3%) good and excellent effect was noted; during the observation of 20 sick children aged up to 3 years for a period of 1 year similar effect was noted in 60% of cases. Vaccinal therapy, in addition to its main effect, also contributed to a considerable drop in the morbidity rate of acute respiratory diseases among susceptible children.
