Feulgen hydrolysis of normal cells and mouse ascites tumor cells.

The effect of HCl hydrolysis on the dye content (Feulgen reaction) of normal cells and mouse ascites tumor cells was examined by means of cytophotometric measurements. After 11 min of hydrolysis, 16-day-old tumor cells showed a hypotetraploid DNA line with doubling peaks. The DNA values were in the ratios of 1:2:4:8 during all the tested hydrolysis times (3 to 21 min). The size of the nucleus and the DNA concentration did not influence the hydrolysis and the dye content. However, the time of the hydrolysis considerably influenced the dye content of normal and tumor cells. The course of the curves obtained by plotting dye absorption against hydrolysis time showed an inflection of the curve at 9 min' hydrolysis time in tumor cells, whereas the inflection occurred at 8 min in mitotic cells. These inflections were statistically significant. The DNA stem-line(1) for tumor cells shifted during different hydrolysis times when compared to normal cells. The possibility is discussed of two types of DNA which differed in their acid sensitivity and which yielded atypical hydrolysis curves.

[Giant lymph nodes in combined autoimmune neutro- and thrombocytopenia].

A boy aged 15 years is described in whom a combined autoimmune neutro- and thrombocytopenia developed since the age of 11. Cell-membrane bound IgG antibodies were detected on neutrophils and platelets. A therapy with prednisone and/or immunoglobulins showed only a transient normalization of the peripheral blood values. In the course of this disease a lymph node adenopathy occurred showing histologically follicle hyperplasia with multiple plasma cells and relative atrophy of the paracortical T-cell region.

[Embryopathy and diabetic fetopathy in a premature stillborn infant. Case report and review of the disease picture].

The diabetic embryopathy syndrome comprises a number of developmental anomalies among fetuses of diabetic mothers. Fetopathia diabetica, on the other hand, is characterized by typical, hormonal, and metabolic dysfunctions and their morphological sequelae in fetuses and offsprings of diabetic mothers. We observed the combination of both these conditions in an immature stillborn fetus. The 34 year-old diabetic mother, who had been treated by insulin since age 16, was first seen at 27 weeks of gestation. Sonography revealed severe congenital malformations of the fetus, and a late abortion was induced. The stillborn female revealed the typical congenital malformations of the diabetic embryopathy syndrome, such as abnormalities of face and skull, skeletal malformations of the thorax, spine, and lower extremities, and malformations of the heart, great vessels and the genitourinary system. We, too, found the characteristic features of fetopathia diabetica, i.e. obesity, macrosomia, increased weight and size of the internal organs, polynesia and macronesia of the pancreas, increased extramedullary hematopoiesis and cellular depletion of lymphoid tissues. A review of the literature revealed various hypotheses about the etiology and pathogenesis of both conditions.

[Lung resection in the principle disease of mucoviscidosis and status following poliomyelitis--2 case reports].

Two cases of destroyed lung after repeated pneumonia are reported; one in a patient suffering from cystic fibrosis in the age of 8 years, the other in a 15 year old girl with a phrenic nerve paralysis. In both children physiotherapy as well as retreatments with appropriate antibiotics did not achieve any long-term curative effect. Therefore a pneumonectomy was indicated. In the patient with cystic fibrosis a small air leak was observed on the first postoperative day at the bronchial suture line, which was treated with an immediate rethoracotomy and correction. In the other child a pericardial effusion 4 weeks after surgery occurred. Regression was seen within two weeks under antibiotic therapy. In diseases such as cystic fibrosis and paralysis of the diaphragm a resection of the lung is justified in cases even if they are complicated by recurrent infections due to atelectasis or bronchiectasia. The preoperative investigation includes the proof of a total lung failure by scintiscan and/or measurement of a mucociliary clearance. A non altered function of the remaining lung is a prerequisite.

[DNA-ploidy and proliferation in metastatic neuroblastoma].

Nuclear DNA contents of tumor cells were measures by fluorescence cytophotometry in bone marrow and touch preparations of 26 children with neuroblastoma, registered in the NBL79 study of the German Pediatric Oncology society. In all cases tumor cells were found to have a higher DNA-content as compared to the normal diploid (2c) DNA-value of granulocytes in the same preparation. Average ploidy values of the histograms of bone marrow metastases were in the tetra- to hypertetraploid (4c-5c) range and showed unimodal distribution in the majority of cases. In the primary tumors there was also a wide distribution of DNA-values in the aneuploid range. A bimodal distribution with stemlines at 2,5c and 5c was found in a stage I, grade I, differentiated primary tumor. Various ploidy patterns found at different sites in the large tumor mass of a newborn might be interpreted as an expression of different degrees of differentiation within the same tumor. In 2 of 4 patients where primary tumor and metastatic lesions could be examined there was a higher ploidy in the 5c to 6c, or even in the 8c range in the metastatic cell population. It appears, thus, that clonal evolution of tumor cells occurs during growth and spread of neuroblastoma.

[Hypoplastic left heart syndrome--clinical and pathologico-anatomic findings].

In twenty-two cases with hypoplastic left heart syndrome the anamnestic criterias and the clinical features were analysed. The pathologic specimens were examined in thirteen cases. The relations of the left heart to the right heart were measured. The same information is to be measured by two-dimensional echocardiography intra vitam. It should be discussed whether it is possible to perform an operative procedure in some rare cases of hypoplastic left heart syndrome.

Carcinoembryonic antigen (CEA) expression in early embryogenesis: a study of the first trimester of gestation.

The immunohistological CEA positivity was studied in 711 tissue samples derived from the ecto-, meso- and entoderm of 63 embryos obtained during the first 12 weeks of gestation. It was found that CEA was expressed by derivates of all three germ layers, although the strongest reactions and the highest incidence of positivity was found in organs derived from the entoderm. In the 7th week of gestation the antigen was first seen in mesodermal and entodermal tissues; only after the 8th week the first appearance of CEA was noted, though only in 2/9 ectodermal organs examined. It therefore seems that the initially described high preponderance of CEA positivity of entodermal organs, previously observed for later stages of gestation, can be confirmed. However, not only are exceptions seen in organs of this germ layer, but CEA can also be expressed in tissues of the two other germ layers early in ontogenesis.

Vascular tumor in the heart.

A rare case of a vascular tumor originating from the uterus growing up in the inferior vena cava into the right heart with clinical signs of tricuspid insufficiency is reported. Although this vascular tumor primarily showed no signs of malignancy, consequent re-evaluation seems to be necessary in order to detect possible tumor recurrence in time.
