Protein expression of the alpha, gamma, delta and epsilon subspecies of protein kinase C changes as C6 glioma cells become contact inhibited and quiescent in the presence of serum.

Total protein kinase C (PKC) activity and protein expression of the alpha and delta subspecies of PKC increases markedly as C6 glioma cells grow from low cell density to the contact-inhibited quiescent state (also known as G(o)) in the presence of serum. At the same time protein expression of PKC subspecies gamma and epsilon decreases while the beta I, beta II, iota and zeta subspecies did not change. Serum deprivation of growing C6 glioma cells does not induce the same changes in PKC subspecies protein expression. The findings support the growing view that there are significant differences between the G(o) states brought about by contact inhibition or serum deprivation.

Increased neuronal excitability conferred by a mutation in the Drosophila bemused gene.

We have identified a new gene in Drosophila on the basis of a mutation that affects both behavior and neuronal excitability. This mutation, called bemused (bem) is the result of a P element insertion at cytological position 85D. Flies defective at bem display greatly reduced coordination, flight ability and fertility. The bem mutation also affects synaptic transmission at the larval neuromuscular junction. In particular, bem larvae display a more rapid onset of augmentation than bem+ larvae. This effect is potentiated by the potassium channel blocking drug quinidine. The increased transmitter release that occurs in bem mutants following repetitive nerve stimulation is accompanied by the appearance of extra action potentials in the motor neuron. We conclude that the bem mutation increases neuronal excitability, possibly as a result of a defect in an ion channel structural or regulatory gene.

Congenital duodenal obstruction: an approach to diagnosis.

53 neonates with congenital duodenal obstruction presented to Our Lady's Hospital for Sick Children over a consecutive 4-year period. Of these 53, 27 presented with intrinsic obstruction, and 26 with extrinsic obstruction. Diagnostic methods and features resulting in surgically proven diagnoses are discussed.

Delayed primary anastomosis following spontaneous growth of esophageal segments in esophageal atresia.

Between 1977 and 1979, five consecutive cases of esophageal atresia had delayed primary esophageal anastomosis at ages ranging from 5 1/2 to 18 wk. Serial growth studies of the two esophageal segments were performed under fluoroscopic screening. It was observed that spontaneous growth of esophageal segments occurs in the absence of any stretching or bougienage of the esophageal ends.

Relationship between functional and histologic appearances of developing ganglion cells in the guinea pig rectum.

Time dated fetal guinea-pigs between 47 and 67 days gestation were studied for segmental propulsive activity in the rectosigmoid under fluoroscopy. Large bowel from the sacrificed fetus was studied histologically to determine distribution and morphology of ganglion cells. Contractions were first observed on day 54 and these became more frequent and vigorous after day 63. Between 47-53 days gestation circular muscle is thin and neurons consists of cluster of neuroblastic cells. Near the end of gestation the neurons arrange themselves into discrete groups and have an appearance like mature ganglion cells.

Duodenal ulcer in childhood: a continuing disease in adult life.

Thirty-one of the 37 children with duodenal ulcers were available for follow-up. Of the 28 patients treated medically, 22 had persistence of symptoms over periods up to 18 yr after the time of the initial diagnosis. Five patients required surgery elsewhere for complications, thirteen showed ulcer crater or duodenal deformity on a follow-up barium meal studies. Earlier surgery is recommended in chronic ulcer disease.

Accuracy of sonography in transient synovitis.

The presence of acute transient synovitis (TS, irritable hip) often warrants hospital admission not only to exclude underlying sepsis but also to aspirate when the intraarticular pressure might induce ischemia in the femoral head. Diagnostic doubt may affect this admission. We evaluated 68 patients in a prospective study comparing sonography with diagnostic aspiration and clinical examination. Sonography proved to be relatively accurate (81%) but was inferior to careful clinical evaluation in diagnosing hip effusions. Sonography may be helpful if diagnostic doubt exists. Aspiration under sonographic control will improve this accuracy.
