Characterization of an anthracycline-resistant human promyelocyte leukemia (HL-60) cell line with an elevated MDR-1 gene expression.

Multidrug resistance to a variety of cytotoxic drugs is due to decreased drug accumulation at the intracellular site of drug action. When due to increased energy-dependent drug efflux, this transport change is often associated with increased expression of an efflux pump for various lipophilic compounds, for example the P-glycoprotein which is the product of the MDR-1 gene. However, previously described HL-60 human promyelocytic leukemia cell lines resistant to the cytotoxic effect of anthracyclines have been reported not to express P-glycoprotein. We have isolated, by drug selection, an anthracycline-resistant HL-60 cell line that, in comparison to parental drug sensitive cells, exhibits a multidrug resistant phenotype including diminished intracellular drug retention, cross-resistance to multiple cytotoxic drugs, increased expression of a monoclonal antibody C219-reactive 180 kDa P-glycoprotein detected by Western blot analysis as well as increased expression of MDR-1 mRNA as determined by Northern blot and solution hybridization/RNAse protection analyses. Evidence is presented that the anthracycline-resistant HL-60 cells have amplified the MDR-1 gene.

Developing an acute pain service in a multicultural setting.

Development of an Acute Pain Service in a multicultural environment can be a difficult and challenging endeavor. In addition to concerns about communicating with patients who speak a foreign language, the problem of integrating staff from many different backgrounds must be faced. Education for both patients and staff must be innovative, multifaceted, and individualized. At King Faisal Specialist Hospital and Research Centre in Riyadh, Saudi Arabia, the past 2 years have been a time of challenge and growth as an interdisciplinary team developed a pain service that meets the hospital's unique needs while fulfilling the goal of improved pain management for the patients.

Effects of cryopreservation on subsets of fetal liver cells.

Human fetal livers from 6 to 13 weeks postconception were analysed before and after cryopreservation. The percentages of cell subsets, detected by MoAbs, did not change significantly after cryopreservation. Compared with BM, fetal liver contained significantly smaller subsets of cells identified by MoAbs, with two exceptions. Fetal liver contained a mean of 47% M5 positive cells versus 31% in BM, and there was no difference in the numbers of CD34+ cells. The colony-forming capacity was studied: 53 colonies grew from 10(5) cells from fresh fetal liver compared with 51 colonies from cryopreserved cells. For fresh BM the corresponding value was 88 per 10(5) cells. Incubation time for fetal stem cells was 17-18 days while the corresponding time for BM cells was 8-10 days.

Stimulation of human myelopoiesis by leukotriene B4.

Cultivation of human mononuclear bone marrow cells for 10 days in the presence of leukotriene B4 (8 X 10(-8) - 3 X 10(-6)M) led to an increase in the formation of granulocyte-macrophage colonies. The increase varied between 19 and 122% when compared to control cells. 5S, 12S-Dihydroxy-6, 8, 10, 14-eicosatetraenoic acid (5S, 12S-DHETE), an isomer of leukotriene B4, did not stimulate colony formation. Preincubation of the cells with 5S, 12S-DHETE inhibited the stimulatory action of leukotriene B4 on the proliferation of bone marrow cells. The present study indicates that leukotriene B4 amplifies the stimulation caused by the colony stimulating factor(s) and may play a role in modulating granulocyte and macrophage poiesis by a positive feedback mechanism.

Interactive computer program for self-distributed medical questionnaire in a population health screening.

The questionnaire is an important element of a medical screening investigation, just as important as the biometrical screening tests. There are several difficulties in realizing interactive computer programs for such tasks. In the present report, we describe the design, operation and results of an interactive computer program for self-distributed medical questionnaire in which the screening subjects are informed and instructed on the terminal routines, and answer, check, confirm and record the questions direct in the computer terminals by means of simple keyboard functions. This program was possible to construct as an integrated component of the interactive computer system monitoring our multiphasic health screening in a standard minicomputer environment.

Experiences with an interactive computer program for recording biometrical health screening data.

The nurses' measurements of biometrical health screening data comprise the central elements in our multiphasic preventive medical investigation in Malmö. It is also vitally important for the information processing, evaluation and actions of the screening that all test values and other data can be supplied to a uniform computer bank without delay in terms of secondary transferral routines. In our ongoing population investigation projectin Malmö, this has been accomplished by on-line computer programs for the different components of the screening. Here we describe the practical experiences with the subroutine use by the nurses to feed the biometrical test results to the computer bank.

Interactive computer system for monitoring multiphasic health screening.

In an on-going population screening investigation at the Department of Preventive Medicine in Malmö, an interactive computer system has been developed for monitoring: (i) the electron and invitation of the health screening population; (ii) on-line registration and representation of demographic informations, test results and questionnaires; (iii) generation of the patient files and other listings; (iv) systems for diagnostic and statistical processing and representation. Ten thousand health screening investigations have now been monitored by the system, which is described in this report.
